A recent study reported that a diet rich in isoflavones is beneficial for bone formation in growing rats. It therefore seemed desirable to find out whether the beneficial effect of isoflavones in ovariectomized rats could also be reproduced with same amount of isoflavones which used for growing rats. To study the effect of isoflavones, an equal amount of isoflavones which used for growing rats, on bone mineral density and bone mineral content in ovariectmized rats were performed. Forty female Sprague-Dawley rats (body weight $210{\pm}5g$) were divided into two groups, ovariectomy and sham groups, which were each randomly divided into two subgroups that were fed casein and casein supplemented with isoflavones diets for 9 weeks after operation. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone mineral density (BMD) and bone mineral content (BMC) were measured using PIXImus (GE Lunar Co, Wisconsin) in spine and femur. Serum alkaline phosphatase activity (ALP) and osteocalcin and urinary DPD crosslinks value were measured as markers of bone formation and resorption. The results of this study indicate that body weight gain and food intake were higher in ovariectomy groups than in sham groups regardless of diets. Serum Ca concentration was lower in ovariectomy groups than in SHAM groups. Serum ALP, osteocalcin, and crosslink value were increased in ovariectomy groups. Spine BMD/weight, femur BMD/weight, and femur BMC/weight of ovariectomy groups were significantly lower than SHAM groups after 9 weeks. However, isoflavones supplemented group in ovariectomy groups, serum ALP and osteocalcin concentrations, spine BMD/weight and spine BMC, femur BMD/weight and femur BMC/weight were significantly increased after 9 weeks. In conclusion, the beneficial effect of isoflavones on bone in ovareiectomized rats was shown on 9 weeks after feeding with an equal amount of isoflavones supplementation which used for growing rats.
This study conducted the following experiment to examine effects of bone metabolism on aquatic exercise, Drynariae Rhizoma and aquatic exercise with Drynariae Rhizoma. This experiment was conducted to compare bone strength, bone mineral density, weight, change of femur, osteocalcin, ALP, Ca and P effects by aquatic exercise for 6 weeks, Drynariae Rhizoma and Drynariae Rhizoma for aquatic exercise with 40 SD rats of postoophorectomy osteoporosis and it divided 10 subjects. experiment group (I) is applying postoophorectomy osteoporosis group, (II) is applying aquatic exercise group, (III) is applying Drynariae Rhizoma group and (IV) is applying aquatic exercise with Drynariae Rhizoma group. These result lead us to the conclusion that osteocalcin were showed a statically increase and blood Ca level were showed a statically decrease on other groups compare to group(I). Consequently, aquatic exercise and Drynariae Rhizoma would be lead to increment of bone metabolism on postoophorectomy osteoporosis.
We studied the effects of soy isoflavone supplements on bone metabolism marker (serum osteocalcin, urinary deoxypyridinoline) and urinary mineral excretion (urinary Ca, Mg, Zn) in 47 postmenopausal women. There were 24 participants in the treatment group and 23 in the control group. The treatment group consumed isoflavone extract capsules daily (which contained 90 mg of soy isoflavones) for 12 weeks. The study compared before and after isoflavone intake in the following areas: Physical examination, diet survey, bone metabolism marker and urinary mineral excretion. The average age of the treatment group was 64.6 years and that of the control group was 66.5 years. There were no significant differences between the two groups in terms of height, weight and body mass index. Both groups maintained a regular diet pattern in terms of their average daily nutrient intake. There were no significant differences between the treatment group (23.9 mg) and the control group (25.4 mg) in terms of daily isoflavone intake based on diet. The analysis of bone metabolism marker changes in the treatment group after 12 weeks of taking the isoflavone supplements demonstrated significant differences in the following: Serum osteocalcin (13.7 ng/mL in befor versus 6.8 ng/mL in after) and urinary deoxypyridinoline (5.9 nmol/mmol Cr in befor versus 4.5 nmol/mmol Cr in after). The subjects in the treatment group showed no significant difference in urinary Ca excretion. But the subjects showed a significant difference in urinary Mg (131.9 mg/day in befor versus 115.6 mg/day in after) and Zn (400.5 $\mu\textrm{g}$/day in befor versus 310.2 $\mu\textrm{g}$/day in after) excretion in the isoflavone treatment group at the levels of p<0.001, p<0.01, respectively. No changes were made in the intake of minerals. The composition of serum osteocalcin and urinary deoxypyridinoline, and indicators of bone metabolism, including the excretion Mg and Zn, significantly decreased. This study was done to evaluate the effectiveness of dietary calcium level(a diet which met 100% or twice the calcium level in AIN-76 diet) on preventing bone loss in ovariectomized rats. Forty female Sprauge-Dawley rats(body weight 200$\pm$5g) were divided into two groups. One group were ovariectomized(Ovx) while the others received sham operation(Sham). Thereafter, each rat group was further divided into normal calcium diet(NCD, 0.52%) and high calcium diet(HCD, 1.04%) sub-groups. All rats were fed on experimental diet and deionized water ad libitum for 8 weeks. Urinary pyridinoline & creatinine and serum estradiol, luteinizing hormone, calcium, phosphate, total protein, albumin, alkaline phosphatase and osteocalcin were determined. There were no significant differences in serum calcium. total protein and albumin in the two groups(Ovx vs Sham) of rats. Ovariectomized rats had significantly lower estradiol than sham operated rats. There was a highly significant correlation between total bone mineral density(TBMD) and overall level of esteradiol(r=0.59, p<0.05). Total bone mineral density did not correlate significantly with ALP or osteocalcin, although a negative trend was evident. However, the rats fed high calcium diet had a lower crosslinks value and osteocalcine than the rats fed normal calcium diet. An increased rate of bone turnover is usually associated with a decrease in bone mass bexause bone formation at each remodeling site is never as great as resorption. Ovariectomized rats fed high calcium diet had a lower crosslink value and osteocalcin; it means high cacium diet decreased bone turnover rate. The findings from the present study demonstrated that bone loss due to ovarian hormonal deficiency can be partially prevented by a high calcium diet. Futhermore, these findings support the strategy of the use of a high calcium diet in the prevention of estrogen depletion bone loss (postmenopausal osteoporosis).
The purpose of this study was to evaluate the daily vegetable and fruit intake status of Korean adults and to examine the relationship of vegetable and fruit intake with bone metabolism. The vegetable and fruit intake of 542 healthy male and female adults was analyzed. Then, by selecting 51 targets from the subjects, the relation of vegetable and fruit intake with serum calcium, osteocalcin and deoxypyridinoline (DPD) excretion in urine was examined. The total vegetable intake per day was 397.7 g and 333.5 g by men and women respectively for the age group of 20-29, 366.9 g and 309.2 g respectively for the age group of 30-49, 378.4 g and 325.9 g respectively for the age group of 50-64. Of vegetable varieties, leafy and stem vegetables displayed the highest intake. The order of major intake items of vegetables and fruits was found to be Chinese cabbage kimchi, onion, radish, cucumber, and welsh onion for the age group of 20-29, watermelon, Chinese cabbage kimchi, peach, potato, and onion for the age group of 30-49 and watermelon, Chinese cabbage kimchi, tomato, potato, and peach for the age group of 50-64. Of 51 targets, ${\beta}$-carotene intake displayed a significantly negative correlation with serum osteocalcin. While caloric intake as well as protein, carbohydrate, calcium, phosphorous, zinc and total food intake displayed a significantly negative correlation with DPD excretion in urine, tuber vegetable intake displayed a significantly positive correlation with DPD excretion in urine. In the future, a study will be necessary to accurately explain the relevance of vegetable and fruit intake with bone mineral density and bone metabolism. Also, efforts will be required to increase vegetable and fruit intake.

This study was carried out to investigate the effects of Cervi Pontotrichum cornu and Carthami semen on the serum osteocalcin level, bone mineral density, bone trabecular area and cortical thickness index in ovariectomized rats. Femal Sprague-Dawley rats were divided into 5 groups. Non-ovariectomized groups were divided into commertial diet fed(normal group) and calcium free basal diet fed(sham operation group). Ovariectomized groups were divided into 3groups, calcium free basal diet fed group(control group or ovariectomized group), 10% Cervi Pontotrichum cornu supplemented diet fed group and 10% Carthami semen supplemented diet fed group. After 4 weeks diet fed, serum osteocalcin level, bone mineral density, bone trabecular areas and cortical thickness index were analyzed. The bone mineral density and bone trabeculae area were significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The osteocalcin level was significantly decreased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The trabecular area(%) in epiphysis of tibia was significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. The cortical thickness index in distal diaphysis of tibia was significantly increased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. the alkaline phosphatase level was significantly decreased by Cervi Pontotrichum cornu and Carthami semen supplemented diet fed groups. These results suggest that Cervi Pontotrichum cornu and Carthami semen might have inhibitory effects on osteoporosis by increasing bone mineral density and accerelating bone formation in ovariectomized rats.

Objectives: The present study aimed at investigating the effects of Schisandrae Fructus (SF) on menopause symptoms in ovariectomized mice. Methods: 6 weeks old female mice were ovariectomized (OVX), and the OVX mice were fed with a three doses of SF (25, 100, and 400 mg/kg, respectively) or $17{\beta}$-estradiol (E2, 0.5 mg/kg) 3 times a week for 6 weeks. Body weight was measured at every week and uterus weight was also measured at the end of experiment. Lipids and osteocalcin levels in serum were analyzed. Results: The uterine weight was not changed in SF treated group, while E2 intake increased the reduced uterine weight by OVX. Although Body weight was increased since two weeks after ovariectomy compared with sham operated group, there was no change in SF or E2 treated group compared with OVX. But LDL was significantly reduced in SF (400 mg/kg) or E2 fed mice and SF or E2 treatment decreased enhanced osteocalcin level by OVX. Conclusion: These results indicated that SF would be effective in the improvement of LDL level and osteoporosis on menopause.

Osteocalcin (OC), a marker of bone turnover, displays patterns in relation to physiological and genetic factors. Here, we present an association study in a population of Chinese Holstein cattle (n = 24) with OC serum concentration as a phenotypic trait. We hypothesised that OC status is associated with phylogeny, vitamin D serum level and single nucleotide polymorphisms (SNPs). Mitochondrial DNA (mtDNA) was used as an unlinked marker to examine phylogeny and linkage to measured phenotypic traits of vitamin D and OC status. Following an association study with OC serum variability as the trait, genotyping of SNPs (n = 27) in OC-related genes was performed. Candidate SNPs were chosen in genes with an emphasis on the vitamin D and vitamin K pathways. Multivariant factor analysis revealed a correlation between vitamin D serum concentration and a SNP in the gene GC (rs43338565), which encodes a vitamin D-binding protein, as well as between a SNP in NFATc1 (rs42038422) and OC concentration. However, univariate analysis revealed that population structure, vitamin D serum levels and SNPs were not significant determinants of OC status in the studied group.
An important related question is whether arginine has influence bone metabolism. The effect of arginine supplements on bone markers and related hormones were studied in young female Sprague-Dawley rats fed either an arginine supplemented diet or control diet. Twenty four rats (body weight 83${\pm}$5 g) were randomly assigned to one of two groups, consuming casein or casein with supplemented arginine diet. All rats were fed on experimental diet and deionized water ad libitum for 9 weeks. Bone formation was measured by serum osteocalcin and alkaline phosphatase (ALP) concentrations. And bone resorption rate was measured by deoxypyridinoline (DPD) crosslinks immunoassay and corrected for creatinine. Serum osteocalcin, growth hormone, estrogen, insulin-like growth factor-1 (IGF-1), parathyroid hormone (PTH) and calcitonin were analyzed using radioimmunoassay kits. The weight gain and mean food intake were not affected regardless of diets. The rats fed arginine-supplemented diet had not significantly different in ALP, osteocalcin, crosslinks value, PTH, estradiol, and IGF-1 compared to those fed casein diet group. The arginine-supplemented group had significantly higher growth hormone and calcitonin than casein group. This study suggests that arginine is beneficial for bone formation in growing female rats. Therefore exposure to diet which rich in arginine early in life may have benefits for bone formation and osteoporosis prevention.
This study was performed to investigate the effects of electrical stimulation on femoral fracture healing in dogs. Eight healthy dogs from 4 to 5 $\beta^3$ were used in this experiment. In the treatment group, anode and cathode were connected to proximal and distal site apart from the fracture line by 2 cm and electrical stimulation was applied to the fracture site for l5minutes by 2 V, 25 Hz and for a month. The control group was connected to electrostimulator as the treatment group, but no electrical stimulation was applied. Various parameters were evaluated including radiograph and serum levels of total-ALP(TALP), bone-ALP(BALP) and osteocalcin. The radiography revealed more rapid callus formation in the treatment group than in the control by about a week. The total-ALP levels of the treatment group were higher than those of the control group from the 2nd to the 24th day(p< 0.05). The bone-ALP levels of the treatment group were significantly higher than those of the control from the 2nd to the 24th day(p< 0.05). The bone-ALP/ total-ALP ratios of the treatment group were higher than those of control throughout this experiment but there were no significance. There were no significance in the osteocalcin levels between two groups. In conclusion, the electrical stimulation on femoral fracture site was effective for bone healing in dogs.
