Introgression of Oryza minuta into Rice, Oryza sativa (벼 Oryza sativa x O. minuta 여교배 계통에서 이입 염색체단편 검정)

  • An introgression line, WH79006 was produced from a single plant from $BC_5F_3$ families from a cross between Hwaseongbyeo used as a recurrent parent and O. minuta (BBCC, Ace. No. 101154) as a donor parent, which was subsequently self-pollinated for three generations. WH79006 resembled the O. sativa parent, Hwaseongbyeo. However it differed from Hwaseongbyeo in several traits including days to heading, culm length, grain size, spikelets per panicle and fertility. These differences in the traits between WH79006 and Hwaseongbyeo can be attributed to the O. minuta introgressions. To detect the introgressions, 294 SSR markers of known chromosomal position have been used. At least, 28 introgressed chromosomal segments have been identified using SSR markers and they map to all chromosomes except chromosome 2. The size of the introgressed segments ranged from 4 to 35cM. A QTL related to culm length was detected using 75 $F_2$ plants from the Hwaseongbyeo/WH79006 cross. This QTL, cl6 located on chromosome 6 explained $9.6\%$ of the total phenotypic variation in the population. This QTL has not been detected in the previous QTL studies between Oryza sativa cultivars, indicating potentially novel alleles from O. minutan.

Telomere association of Oryza sativa telomere repeat-binding factor like 1 and its roles in telomere maintenance and development in rice, Oryza sativa L.

  • Telomeres are specialized nucleoprotein complexes that function to protect eukaryotic chromosomes from recombination and erosion. Several telomere binding proteins (TBPs) have been characterized in higher plants, but their detailed in vivo functions at the plant level are largely unknown. In this study, we identified and characterized OsTRFL1 (Oryza sativa Telomere Repeat-binding Factor Like 1) in rice, a monocot model crop. Although OsTRFL1 did not directly bind to telomere repeats $(TTTAGGG){_4}$ in vitro, it was associated with telomeric sequences in planta. OsTRFL1 interacted with rice TBPs, such as OsTRBF1 and RTBP1, in yeast and plant cells as well as in vitro. Thus, it seems likely that the association of OsTRFL1 with other TBPs enables OsTRFL1 to bind to telomeres indirectly. T-DNA inserted OsTRFL1 knock-out mutant rice plants displayed significantly longer telomeres (6-25 kb) than those (5-12 kb) in wild-type plants, indicating that OsTRFL1 is a negative factor for telomere lengthening. The reduced levels of OsTRFL1 caused serious developmental defects in both vegetative and reproductive organs of rice plants. These results suggest that OsTRFL1 is an essential factor for the proper maintenance of telomeres and normal development of rice.

Effects of the Fractions of Oryza sativa cv. Heugjinmi on Plasma Glucose and Lipid Levels in Streptozotocin-Induced Diabetic Rats (흑진미 과피 용매분획물 투여시 당뇨 유발 흰쥐의 혈당 및 혈중 지질함량에 미치는 영향)

  • We have investigated the effects of the solvent fractions from the aleurone layer of Oryza sativa cv. Heugjinmi on plasma glucose and lipid levels in streptozotocin-induced diabetic rats. Sprague-Dawley rats were divided into 6 groups, normal, diabetic control, and four experimental groups. Diabetes mellitus was induced by injection of streptozotocin(45 mg/kg body weight) into tail vein, and fraction of Oryza sativa cv. Heugjinmi was administered orally in experimental rats for 14 days. The body weight, glucose, cholesterol, HDL-cholesterol, triglyceride, free fatty acid level, and aminotransferase activities were analyzed. It was identified that acetone and anthocyanin fractions significantly reduced the levels of glucose, triglyceride, and free fatty acid in plasma of diabetic rats. And HDL-cholesterol levels of acetone and anthocyanin groups were higher than those of diabetic control group. From these data, it can be postulated that acetone and anthocyanin fractions might possess rather potent hypoglycemic and hypotriglyceridemic properties in streptozotocin-induced diabetic rats.

Identification and expression of leuD Gene in Rice (Oryza sativa L.) (벼(Oryza sativa L.)의 leuD 유전자)

  • A rice OsLeuD gene for small subunit of 3-isopropylmalate isomerase (IPMI) (EC 4.2.1.33) has been isolated. OsLeuD gene is located on 109.3 cM of chromosome 2. OsLeuD gene was expressed abundantly in metabolically active organs including leaves and developing seeds, indicating that OsLeuD gene expression is developmentally regulated. The cDNA of OsLeuD gene was coded for 257 amino acids which showed 58% and 48% homology to small subunits of IPMI in OsLeuD genes of cyanobacteria and green sulfur bacteria, respectively. The molecular character of OsLeuD is closely related to those of photosynthetic bacteria rather than those of eukaryotes including fungi and yeast. This suggests that OsLeuD gene in chromosomal genome of plants may possibly be originated from chloroplast genome.

Preliminary Study on the Osteoporosis Improvement Effect of Compounds Isolated from Oryza sativa L. root Extract in the Ovariectomized Mouse Model (난소 적출 마우스 모델에서 Oryza sativa L.의 뿌리 추출물에서 분리된 화합물의 골다공증 개선 효과에 관한 예비연구)

  • Bisphosphonates, estrogen, and calcium supplements are commonly used medications for postmenopausal osteoporosis, but they are associated with various side effects such as vaginal bleeding, deep vein thromembolism, and breast cancer. In this study, we aimed to investigate the potential of a compound isolated from the roots of Oryza sativa L. to improve osteoporosis using an ovariectomized mouse model. We isolated and identified oryzativol A, a lignan compound, through chemical analysis of an ethanol extract using a bioassay-guided fractionation protocol. We also examined the metabolism, clearance, and CYP enzyme activity of oryzativol A, and found that it showed plasma stability of over 80% at all analysis times, and indicating a low likelihood of inactivation or excretion by the CYP3A4 enzyme. Our results showed that the high-dose group of oryzativol A exhibited a significant increase in bone mineral density compared to the control group. Although the ALP concentration did not differ significantly compared to the control group, it showed a tendency to increase in the high-dose group of oryzativol A. Furthermore, the abnormal ratio of serum Ca/P, caused by osteoporosis, was improved to a level closer to that of the normal group as the dosage of oryzativol A increased. Taken together, these findings suggest that oryzativol A is stable in vivo and has potential as a therapeutic agent for osteoporosis, particularly when administered in high doses.

Screening of Gravity Inducible cDNAs in Rice(Oryza sativa L.) Cultured Cell (벼 (Oryza sativa L.)배양세포의 고중력유도성 cDNA의 탐색)

  • Two different gravity specific cDNA, namely, GSC 13 and GSC 124 with length of 1.34 and 0.67 kilobase pairs, and transcripts of 2.0 and 1.9 kilobase pairs, respectively. were isolated by differential screening and northern hybridization of the total RNA isolated from treated and untreated cultured cells showed that maximum levels of trannscripts were achieved after 4 h of gravity stress at 450, 000 x g for both, GSC 13 and GSC 124, suggesting that these mRNA could be expressed and translated into polyeptites related to the cell to extream gravity stress.

The Effect of Ginseng Saponin Fraction on the Germination and Early Growth of Rice Seed (Oryza sativa L.) (인삼사포닌이 벼의 발아 및 초기 생장에 미치는 영향)

  • The effects of ginseng saponin on the germination and early growth of rice seeds (Oryza sativa L.) were investigated. The early growth (length) of test rice seeds which were rinsed for 60 hrs in $10^{-4}$% saponin solution prior to transplantation to water agar bed was about 20% faster than that of control seeds, It was also found that the amylase activity of the seeds was most active when the seeds were rinsed in 10-4% saponin solution for 48 hrs. In vitro investigation showed that the amylase activities were stimulated about 30% by the treatment of $10^{-5}$% saponin solution compared with control group. From the above results, it seems that the ginseng saponin might activate amylase of rice seed during germination, resulting in rapid growth of rice.

  • Two new compounds 1'-(stigmast-11,20(21),25-trien 3$\alpha$,9$\alpha$-diolyl)-3'-(pimara-11,15-dien-3$\alpha$-olyl) glycerol (1) and stigmast-5-en-3$\alpha$,26-diol (2) along with known fatty acids n-hexacosanoic acid (3) and hexadecanoic acid (4), have been isolated from the methanol extract of rice hulls of Oryza sativa. The structures of the compounds were elucidated using 1D and 2D NMR spectral methods, viz; $^1H, ^{13}C, ^{13}C$-DEPT, $^1H-^1H$ COSY, $^1H-^{13}C$ HETCOR, HSQC and HMBC aided by IR, EIMS, FABMS and HRFABMS. Compound (1) showed inhibition to radish germination, growth of shoot and root length.

Black Rice (Oryza sativa L. var. japonica) Hydrolyzed Peptides Induce Expression of Hyaluronan Synthase 2 Gene in HaCaT Keratinocytes

  • Black rice (Oryza sativa L. var. japonica) has been used in folk medicine in Asia. To understand the effects of black rice hydrolyzed peptides (BRP) from germinated black rice, we assessed the expression levels of about 20,000 transcripts in BRP-treated HaCaT keratinocytes using human 1A oligo microarray analysis. As a result, the BRP treatment showed a differential expression ratio of more than 2-fold: 745 were activated and 1,011 were repressed. One of the most interesting findings was a 2-fold increase in hyaluronan synthase 2 (HAS2) gene expression by BRP. Semiquantitative RT-PCR showed that BRP increased HAS2 mRNA in dose-dependent manners. ELISA showed that BRP effectively increased hyaluronan (HA) production in HaCaT keratinocytes.
