Effect of Lythrum salicaria Extract on Body Fat Reduction: A Protocol for a Randomized, Double-Blinded, Placebo-Controlled Clinical Trial (체지방 감소에 대한 털부처꽃 추출물의 효과: 무작위배정, 이중눈가림, 대조군 비교 인체적용시험 프로토콜)

  • Objectives: Obesity is a globally prevalent public health issue. Hence, there is a need for the development of safer and more effective anti-obesity drugs. Lythrum salicaria, a traditional medicinal herb used for centuries, has been reported to improve lipid metabolism and fat accumulation. It also has a low toxicity profile. Therefore, its potential as a functional ingredient in health functional foods needs to be evaluated. Methods: In this randomized, double-blind, placebo-controlled clinical trial, 90 participants will be randomly assigned to either the experimental or control group. Each subject will orally receive L. salicaria extract (1,350 mg/day) (500 mg L. salicaria+850 mg lactose as vehicle) or lactose (1,350 mg/day) as a hard capsule formula for 84 days (12 weeks). The primary outcome will be body fat mass (kg), which will be assessed using dual-energy x-ray absorptiometry (DXA) (performed only at visits 2 and 4). Secondary outcomes include body mass index, body weight, waist-to-hip ratio, body fat percentage (%) measured using DXA, lean body mass (kg) measured using DXA (assessed only at visits 2 and 4), lipids (total cholesterol, triglyceride, high-density lipoprotein cholesterol, and calculated low-density lipoprotein cholesterol), free fatty acid, high sensitivity C-reactive protein, adiponectin, and leptin. Conclusions: This protocol will be implemented after approval of Institutional Review Board of Pusan National University Korean Medicine Hospital (approval number: PNUKHIRB-2022-08-002) and registration with the Korean National Clinical Research Information Service (CRIS) (CRIS-KCT0008060). The results of this trial will provide potential of L. salicaria as a new anti-obesity functional food with fat-reducing effects and low toxicity.

Associations between Mother and Child Temperament and Parenting Attitude (어머니와 아동의 기질과 양육태도 관계 연구)

  • Objectives: The purpose of this study was to investigate the relations between mother's temperament and child's and parenting attitude. Methods: One hundred twenty healthy mothers (above 20 ys) who has a child (7-11 ys) participated after informed consent. The temperament of the children was analyzed by the Junior Temperament and Character Inventory 7-11 (JTCI 7-11) by their mothers. The temperament of the mothers was analyzed by the Temperament and Character Inventory-Revised Short (TCI-RS) and the parenting attitude of the mothers was analyzed by the Parenting Attitude Test (PAT). The relations between mother and child temperament and parenting attitude were assessed using the Pearson correlation coefficient. Results: There were some significant correlations between maternal temperaments and child's temperaments and maternal parenting attitude. Maternal temperament and parenting attitude: Novelty seeking (NS) of mother had low positive correlations with punishment, high expectation and inconsistency. Harm avoidance (HA) had low positive correlations with high involvement and high expectation, and had weak negative correlations with supportive expression and rational explanation. Reward dependence (RD) had low positive correlation with rational explanation. Persistence (P) had low positive correlations with supportive expression, rational explanation and superintendence, and weak negative correlation with high expectation. Child's temperament and parenting attitude: NS of child had low positive correlations with achievement press, high involvement, punishment, high expectation and inconsistency. Low positive correlations were found in many sets such as HA and high expectation, RD and supportive expression and rational explanation, P and supportive expression and rational explanation. Conclusions: These results suggest that there are significant relations between mother and child temperament and parenting attitude the temperament of mother and child.

  • This study analyzed the contents of the research papers concerning the vascular dementia(VD) recorded in Chinese medical journal published over the period between 1998 and 2000. As a result, the following conclusion was drawn. The Chinese medical category of VD includes Sun-Mang(善忘), Chi-Mae, Jung-Pung-Chi-Mae and so on, and the Major mechanism of the VD is Bon-Heo- Pyo-Sil(state of deficient vital essence and excessive pestilential vapor;本虛標實), Bon(本) includes Sin-Jeong-Heo-Son(腎精虧損), Ki-Hyeol-Hyu-Heo(氣血虧虛) and Pyo(標) includes Dam-Chae-Eo-Jo(痰滯瘀阻), Ki-Chae-Hyeol-Eo(氣滯血瘀), Gan-Yang-Sang-Hang(肝陽上亢). It is mainly used DSM and evidence of brain-vascular disease in brain CT or MRI as the criterion for diagnosis in Western medicine and MMSE, HDS, ADL is used as assistant diagnostic indicator. "Principle of clinical study guidance for treatment of dementia by Chinese medicine, Western medicine"("中藥新藥治療痴呆 病的臨床硏究指導原則") is mainly used as the criterion for diagnosis in Chinese medicine. It is mainly used "Principle of clinical study guidance for treatment of senile disease by Chinese medicine, Western medicine"("中藥新藥治療老年病臨床硏究指導原則"), "Principle of clinical study guidance for treatment of dementia by Chinese medicine, Western medicine" as the criterion for treatment effect evaluation, and score change of MMSE and HDS is used as assistant evaluation indicator. The research papers reported that the use of prescriptions such as Hwan-Chong-Dan(還聰丹), Noi-Hyeol-So-Tong-Gu-Bok-Yaek(腦血疏通口服液), Yik-Gi-Gyo-Nang(益智膠囊), Cheon-Ma-Chok-Gi-Chung-Je(天麻促智沖劑), Sam-Gap-San-Ga-Mi-Bang(三甲散加味方), Seon-Ryong-Gyo-Nang(仙龍膠囊), Seong-Noi-Tang II(醒腦湯II號), Bo-Sin-Geon-Noi-Tang(補腎健腦湯), Ga-Mi-Tong-Gyu-Hyal-Hyeol-Tang(加味通竅活血湯), So-Jo-Yik-Ji-Bang(疏調益智方), Tong-Gyu-Hwal-Hyeol-Tang-Ga-Gam(通竅活血湯加減), Yik-Sin-Seong-Noi-Tang(益腎醒腦湯) led to the high efficacy.

A Clinical Study on Correlation between Cervical, Lumbar Lordosis and Low Back Pain (요통과 경추, 요추전만의 관계에 대한 임상적 연구)

  • Objectives: This study was designed to investigate the correlation between cervical, lumbar lordosis and low back pain(LBP), sex, age and duration of LBP. Methods : Cervical, lumbar lordosis(by Cobb's Method) and Ferguson's angle were measured and evaluated in LBP group and control. Radiograph was taken in lateral direction, erect position. Cobb's angle between C1-C7, C2-C7, L1-L5, L1-S1 and Ferguson's angle were measured and investigated with statistical program. Results: 1. Cervical lordosis have no relation to LBP, sex and age. 2. Lumbar lordosis and Ferguson's angle have no relation to LBP and sex. 3. Cobb's angle L1-L5 have no relation to age. Lumbar lordosis from L1 to S1(Cobb's angle L1-S1) increased in old group(Age>40) compared to young group(Age${\leq}$40). 4. In LBP group, Cobb's angle L1-S1 have no relation to duration of LBP. Lumbar lordosis from L1 to L5(Cobb's angle L1-L5) decreased in acute LBP group compared to Chronic group. Conclusions : Cervical, lumbar lordosis and Ferguson's angle have no relation to LBP and sex. As far as age is concerned, old group have larger lumbosacral lordosis than young group. Acute LBP group have smaller lumbar lordosis(Cobb's angle L1-L5) than chronic group.

  • Objectives: This study reports on the efficacy of using Korean medical treatment for suspected gastric ulcer. Methods: We treated a patient with suspected gastric ulcer using Korean medical treatments (herbal medicines, acupuncture, and herbal acupuncture therapy). We evaluated the results using a numerical rating scale (NRS) to measure abdominal pain. The degree of gastrointestinal symptoms was also evaluated using the Korean Gastrointestinal Symptom Rating Scale (KGSRS). The generic health status was evaluated using the European Quality of Life-5 Dimensions (EQ-5D) scale. Results: The NRS and EQ-5D scale scores were improved after the treatments. The change in the KGSRS scores showed that the patient's abdominal pain was relieved. Conclusions: The results of this case study suggest that Korean medicine therapies may be effective in the treatment of suspected gastric ulcer.

Relationships between Mother-Child Temperamental Interactions and Parenting Stress in Korean School-Aged Children (학령기 아동에서 어머니-아동 기질 관계와 양육스트레스 연구)

  • Objectives : The purpose of this study was to investigate the relationships between mother's and child's temper and parenting stress. Methods : One hundred twenty healthy mothers (above 20 yrs old) who has a school-aged child (7-11 yrs old) participated after informed consent. The temperament of the children was analyzed by the Junior Temperament and Character Inventory 7-11 (JTCI 7-11) by their mothers. The temperament of the mothers was analyzed by the Temperament and Character Inventory-Revised Short (TCI-RS) and the mother's parenting stress was analyzed by the Korean Parenting Stress Index (K-PSI). The relationships between mothers' and child's temperament and parenting stress were assessed using the Pearson correlation coefficient and multiple regression test. Results : There were significant correlations between maternal temperaments and child's temperaments and maternal parenting stress. There was a positive correlation between mother's harm avoidance characteristic, child's novelty seeking degree and parenting stress. On the other hand, there was a negative correlation between child's persistence and parenting stress. In addition, interactions between child and mother temperament dimensions predicted higher levels of parenting stress. Parenting stress was higher in the group of high mother's harm avoidance with child's high novelty seeking and high mother's harm avoidance with low child's persistence. Conclusions : These results are showing that there are significant relationships between mother and child temperament and parenting stress in mothers with school-aged children.

Effects of Treadmill Exercise on the Recovery of Dopaminergic Neuron Loss and Muscle Atrophy in the 6-OHDA Lesioned Parkinson's Disease Rat Model

  • This study was to determine the effect of exercise on the recovery of dopaminergic neuron loss and muscle atrophy in 6-OHDA-induced hemi Parkinson's disease model. Exercise was loaded twice per day for 30 minutes each time, at 5 days after 6-OHDA lesioning and continued for 16 days using a treadmill. Exercise significantly increased the number of tyrosine hydroxylase positive neuron in the lesioned substantia nigra and the expression level of tyrosine hydroxylase in the striatum compared with the control group. To examine which signaling pathways may be involved in the exercise, the phosphorylation of $GSK3{\beta}$ and ERK were observed in the striatum. In the control group, basal level of $GSK3{\beta}$ phosphorylation was less than in both striatum, but exercise increased it. ERK phosphorylation decreased in the lesioned striatum, but exercise recovered it. These findings suggest that exercise inactivates $GSK3{\beta}$ by phosphorylation which may be involved in the neuroprotective effect of exercise on the 6-OHDA-induced cell death. In the exercise group, weight, and Type I and II fiber cross-sectional area of the contralateral soleus significantly recovered and expression of myosin heavy chain and Akt and ERK phosphorylation significantly increased by exercise. These results suggest that exercise recovers Parkinson's disease induced dopaminergic neuron loss and contralateral soleus muscle atrophy.

Effect of DHEA on Recovery of Muscle Atrophy Induced by Parkinson' s Disease

  • Purpose: The purpose of this study was to determine the effect of dehydroepiandrosterone (DHEA) on recovery of muscle atrophy induced by Parkinson's disease. Methods: The rat model was established by direct injection of 6-hydroxydopamine (6-OHDA, 20 ${\mu}g$) into the left striatum using stereotaxic surgery. Rats were divided into two groups; the Parkinson's disease group with vehicle treatment (Vehicle; n=12) or DHEA treatment group (DHEA; n=22). DHEA or vehicle was administrated intraperitoneally daily at a dose of 0.34 mmol/kg for 21 days. At 22-days after DHEA treatment, soleus, plantaris, and striatum were dissected. Results: The DHEA group showed significant increase (p<.01) in the number of tyrosine hydroxylase (TH) positive neurons in the lesioned side substantia nigra compared to the vehicle group. Weights and Type I fiber cross-sectional areas of the contralateral soleus of the DHEA group were significantly greater than those of the vehicle group (p=.02, p=.00). Moreover, extracellular signal-regulated kinase (ERK) phosphorylation significantly decreased in the lesioned striatum, but was recovered with DHEA and also in the contralateral soleus muscle, Akt and ERK phosphorylation recovered significantly and the expression level of myosin heavy chain also recovered by DHEA treatment. Conclusion: Our results suggest that DHEA treatment recovers Parkinson's disease induced contralateral soleus muscle atrophy through Akt and ERK phosphorylation.

The Effect of Herbal Medicine on Rheumatoid Arthritis Animal Model: A systematic Review of Animal Experiment Studies Published in Korea and Overseas (한약의 류마티스 관절염 동물 모델에 대한 치료효과: 국내 및 국외 논문에 대한 체계적 문헌 고찰)

  • Objectives This study was designed to review the effect of herbal medicine on animal model with rheumatoid arthritis. Methods We reviewed studies published both in Korea and overseas, all published after 2008. We investigated 8 databases, 6 Korean databases and 2 of abroad. Searching was conducted through flowchart and 24 studies met our guideline. Datas were extrated and organized as animal model characteristics, experimental period, study design, herbal medicine used as intervention and rheumatoid arthritis indicator. Results 336 articles were primarily selected. After screening, 24 articles were left in the end. All 24 studies showed that the herbal medicine used in the animal experiment was effective on treating rheumatoid arthris, proven by macroscopic, histological, hematological and osteological indicators. Conclusions The result showed that herbal medicine is effective in treating rheumatoid arthritis through various pathways. However, study design and intervention were all different from each other and we could not specify the certain compound, material or substance of efficacy. Further study should be conducted to figure out the effectiveness of herbal medicine on rheumatoid arthritis.

Protective effect of furosin isolated from Euphorbia helioscopia against glutamate-induced HT22 cell death (등대(燈臺)풀 유래 Furosin의 glutamate에 의한 HT22 세포 사멸 억제 효과)

  • Objectives: In the brain, glutamate is the most important excitable neurotransmitter in physiological and pathological conditions. However, the high level of glutamate induces neuronal cell death due to exitotoxicity and oxidative stress. The present study investigated to evaluate a possible neuroprotective effect of furosin isolated from Euphorbia helioscopia against glutamate-induced HT22 cell death. Methods: Furosin was isolated from methanol extract of Euphorbia helioscopia and examined whether it protects glutamate-induced neuronal cell death. The cell viability was determined using Ez-Cytox assay. Anti-oxidative effect of furosin was determined by DPPH scavenging activities, and the levels of intracellular reactive oxygen species (ROS) were determined by the fluorescent intensity after staining the cells with $H_2DCFDA$. To evaluate apoptotic cell death, we performed nuclear staining and image-based cytometeric analysis. Results: The cell viability was significantly increased by treatement with furosin compared with the treatment with glutamate. Furosin showed a strong DPPH radical scavenging activity ($EC50=1.83{\mu}M$) and prevented the accumulation of intra cellular ROS. Finally, the presence of 50 and $100{\mu}M$ furosin significantly the percentage of apoptotic cells compared with glutamate treatment. Conclusion: The present study found that furosin is a potent neuroprotectant against glutamate-induced oxidative stress through inhibition of apoptotic cell death induced by glutamate. Therefore, the present study suggests that furosin as a bioactive compound of E. helioscopia can be a useful source to develop a drug for the treatment of neurodegenerative diseases and acute brain injuries.
