Effects of Rhodiola Rosea on Brain Edema and Matrix Metalloproteinase Expressions Following Intracerebral Hemorrhage in the Rat (홍경천(紅景天)이 뇌조직내출혈(腦組織內出血) 흰쥐의 뇌부종(腦浮腫)과 Matrix Metalloproteinase 발현에 미치는 영향)

  • Objectives : This study aimed at evaluation of the effects of Rhodiola rosea on brain edema and expressions of matrix metalloproteinases (MMPs) related to blood-brain barrier (BBB) disruption. Methods : Brain edema following intracerebral hemorrhage (ICH) was induced by the stereotaxic intrastriatal injection of bacterial collagenase type VII in rats (Sprague-Dawley). Then ethanol extract of Rhodiola rosea was treated once a day for 3 days. Brain edema % and water contents, and BBB leakage were examined. Immunohistochemistry was processed for MMP-9, MMP-12, and iNOS expressions in the brain sections and each immuno-labeled cells were analyzed with image analysis software. Results : 1. Ethanol extract of Rhodiola rosea reduced brain edema following ICH in rats significantly. 2. Ethanol extract of Rhodiola rosea reduced excessive brain tissue water contents following ICH in rats significantly. 3. Ethanol extract of Rhodiola rosea reduced BBB leakage in the cerebral cortex following ICH in rats. 4. Ethanol extract of Rhodiola rosea reduced cellular edema of neurons in peri-hematoma and the cerebral cortex following ICH in rats significantly. 5. Ethanol extract of Rhodiola rosea reduced MMP-9 positive cells in the cerebral cortex following ICH in rats significantly. 6. Ethanol extract of Rhodiola rosea reduced MMP-12 positive vessels in the cerebral cortex following ICH in rats significantly. 7. Ethanol extract of Rhodiola rosea reduced iNOS positive cells in the cerebral cortex and external capsule following ICH in rats significantly. Conclusions : These results suggest that Rhodiola rosea reveals protective effect against brain edema and cytotoxic edema of neurons by means of down-regulation of MMPs and iNOS expressions, and inhibition of BBB leakage.

  • We examined the radioprotection effects of acupoint (acupuncture point) stimulation during organogenesis stages of ICR mice. Pregnant mice received 1.5 Gy whole body X-irradiation on day 8 of gestation, which is the early stage of organogenesis. The embryonic death rate and teratogenesis rate by radiation were examined. Electroacupuncture to the leg acupoints and/ or transcutaneous stimulation to the back acupoints on the pregnant mice showed no protective effect against irradiation on embryonic or fetal death rate. On the contrary, the strong stimulation resulted in increase in the mortality after irradiation rather than protection. However acupoint stimulation to the pregnant mice never showed harmful effects by itself on embryos. It tended to reduce the skeletal malformations induced by X-ray irradiation. We suspect that acupoint stimulation removed the cells injured by irradiation during embryonic development, resulting in an increase in embryonic death rate and reduction in skeletal anomalies.

  • In this dissertation, I will focus on the channel entry, the effect, and the treatment throughout books of oriental medicine from ancient to modern in order to classify the medicines of the liver as main or supplementary organ. The results are as follows: 1. The kinds of the medicines of working of the liver(本臟) were 29, which were Amydae carapax(鱉甲), Chrysanthemi Flos(菊花), Cassiae Torae Semen(草決明), Plantaginis Semen(車前子), Scirpi Seu Sparganii(三稜), Ulmi Pasta Semen(蕪荑), Cinnamomi Cortex(肉桂), Fraxini Cortex(秦皮), Chaenomelis Fructus(木果), Corii Asini Colla(阿膠), Chuanxiong Rhizoma(川芎), Adenophorae Radix(沙蔘), Coicis Semen(薏苡仁), Acanthopanacis Cortex(五加皮), Zizyphi Spinosae Semen(酸棗仁), Picrorrhizae Rhizoma(胡黃連), Gentianae Radix(草龍膽), Citri Reticulatae Virdie Pericarpium(靑皮), Paeoniae Alba Radix(白芍藥), Paeoniae Rubra Radix(赤芍藥), Bupleuri Radix(柴胡), Peucedani Radix(前胡), Naturalis Indigo(靑黛), Citrus unshiu(橘葉), Rhinocerotis Cornu(犀角), Aucklandiae Radix(木香), Polygonati Odorati Rhizoma, Farfarae Flos(款冬花), Evodiae Fructus(吳茱萸), Citri Reticulatae Pericarpium(陳皮) . 2. The kinds of the medicines of working of other viscera(他臟) were 7, which were Astragaliadix(黃耆), Ginseng Radix(人蔘), pinelliae Rhizoma(半夏), Myristicae Semen, Euryales Semen, Arecae Semen, Piperis Longi Fructus. 3. Medicines, effected on the heart functioned through any other viscera are as follows: Arecae Semen works to treat Spleen Gi Entering the Liver(脾氣入肝), Piperis Longi Fructus, pinelliae Rhizoma(半夏), Euryales Semen and Myristicae Semen operate to treat Spleen Cold Entering the Liver(脾冷入肝), Astragali Radix(黃耆) and Ginseng Radix(人蔘) work to treat Spleen Vacuity Entering the Liver(脾虛入肝). In the study of concerning the medicines effected on the liver, It is considered that it dedicated to development of the medicines related to the disease of the liver and making efficient use of the medicines.

Geospatial Analysis for Oriental Medical Services of Jinju (진주시 한의원 의료서비스 공간특성분석)

  • In keeping with the increasing demand for medical services in accordance with the economic development, the use of oriental medicine is on the rise year by year. In particular, it is a tendency that the elderly over age 65 prefer the oriental medicine to the western medicine in Korea. Under the circumstance, changing into an aging society, the importance of oriental medical services has been growing. In this regard, this study evaluated the quality of medical services by conducting the density analysis and the accessibility analysis of oriental medical clinic targeting Jinju, which is one of local small and medium-sized cities. The study results reveal that oriental medical hospitals accounted for 27% of the entire medical facilities in Jinju, and there was an imbalance in the quality of the oriental medical services according to districts(called dong) because oriental medical hospitals were mainly concentrated in the central commercial areas. In particular, in terms of the analysis on the accessibility of an aging population over age 65 to the oriental medical hospitals, over 50% of the whole districts (dongs) in Jinju were blind spots in medical services, showing a very vulnerable condition. Accordingly, it is judged that systematic measures for improvement in support of the oriental medical services in local small and medium-sized cities should be provided in Korea when we consider the characteristics of Korea where an aging population is rapidly increasing.

  • Objectives : This study was intended to establish for medical treatment range of Ojeoksan and to help extending application of medical insurance through prescriptive research. Methods : We analyzed "Dongeuibogam", "Taepyeonghaeminhwajegukbang" and other books Result : 1. composition of Ojeoksan in "Dongeuibogam" is same as in "Gogeumeuigam" 2. Ojeoksan is composed of five kinds of basic prescripitions; Er Chen Tang, Ping Wei San, Ma Huang Tang, Si Wu Tang etc. and it used for five pathological factors; cold, energy, eating, phlegm, blood. 3. Property of Ojeoksan is warm so it can be used for any cold diseases. 4. Ojeoksan is used for 16 kinds of diseases including chest pain, abdominal pain etc. Conclusion : Ojeoksan can be used for cardiovascular disorders and Ojeoksan is possible to apply or to extend medical insurance coverage.

  • Objectives : Substantial evidence suggests that reinforcing effects of nicotine can be mediated by the mesolimbic dopaminergic system. It has been shown that repeated injections of nicotine produce an increase in locomotor activity and expression of the immediate-early gene, c-fos, in the dopaminergic target areas. Herbal medicine as a therapeutic intervention has been widely used for the treatment of mental dysfunction. Many studies have shown that Radix Scutellariae (RS) can affect the biochemical balance in the central nervous system. Tn order to investigate whether RS has an influence on nicotine-induced behavioral sensitization, we examined the effect of RS on nicotine-induced locomotor activity and c-fos expression in the striatum and nucleus accumbens utilizing the fos-tike immunohistochemistry (FLI). Methods : Male SD rats received RS (200mg/kg, i.p.) 30min before repeated daily injections of nicotine (0.4mg/kg, s.c.) for 7 days. This was followed by withdrawal for 3 days and one challenge for 1 day. Results : System challenge with nicotine produced a much larger increase in locomotor activity and accumbal FLI. Pretreatment with RS significantly inhibited nicotine-induced locomotor activity and FLI in the striatum and nucleus accumbens. Conclusions : These results demonstrate that reduction in locomotor activity by RS may be reflected by reduction of dopamine release and postsynaptic neuronal activity in the striatum and nucleus accumbens. Our results suggest that RS may have therapeutic effect on nicotine addiction.

The Effect of Herb-acupunctures of Bojoongiggi-tang (Buzhongyiqi-tang), Ginseng Radix, and Astragali Radix on Immune Responses in Rats (보중익기탕(補中益氣湯), 인삼(人蔘) 및 황(黃)기 약침(藥鍼)이 MTX로 약침(誘發)된 백서(白鼠)의 면역기능저하(免疫機能低下)에 미치는 영향(影響))

  • Objectives : To investigate the effects of herb-acupunctures of Qi tonification herbs or formula on the rat immune depression induced by an anticancer drug, methotrexate (MTX) Methods: Animals were divided into 5 groups; Normal control group was not given any treatment. Immune depression was induced by oral administration of 1mg/kg MTX b.i.d for 4 days in Control, Sample I, II, and III groups. We treated on CV 4 (Guanyuan) with saline, Bojoongiggi-tang (Buzhongyiqi-tang), Ginseng Radix and Astragali Radix herb-acupunctures in Control, Sample I, II, and Ill groups, respectively. In both blood and spleen, the assessment of CD4+ T-cell count, CD8+ T-cell count, CD4/CD8 T-cell ratio was performed. Results: Here we show that 3 herb- acupuncture groups have an influence, to some extent, on CD4+ and CD8+ T-cell counts and CD4/CD8 T-cell ratio in both blood and spleen. Astragali Radix herb-acupuncture, in particular, was found to have significantly increased CD4+ T-cell count in blood and CD4/CD8 T-cell ratio in blood and spleen. Conclusions: The results of this study suggest that herb-acupunctures of Bojoongiggi-tang (Buzhongyiqi-tang), Ginseng Radix and Astragali Radix may have an influence over rat immune depression induced by MTX.

Evidence of hydrolyzed traditional Korean red ginseng by malted barley on activation of receptor interacting proteins 2 and IkappaB kinase-beta in mouse peritoneal macrophages

  • Red ginseng, which has a variety of biological and pharmacological activities including antioxidant, anti-inflammatory, antimutagenic and anticarcinogenic effects, has been used for thousands of years as a general tonic in traditional oriental medicine. Here, we tested the immune regulatory activities of hydrolyzed red ginseng by malted barley (HRG) on the expressions of receptor interacting proteins (Rip) 2 and $I{\kappa}B$ kinase-beta (IKK-${\beta}$) in mouse peritoneal macrophages. We show that HRG increased the activations of Rip 2 and IKK-${\beta}$ for the first time. When HRG was used in combination with recombinant interferon-${\gamma}$ (rIFN-${\gamma}$), there was a marked cooperative induction of nitric oxide (NO) production. The increased expression of inducible NO synthase from rIFN-${\gamma}$ plus HRG-stimulated cells was almost completely inhibited by pre-treatment with pyrrolidine dithiocarbamate (PDTC), an inhibitor of nuclear factor-${\kappa}B$ (NF-${\kappa}B$). In addition, the treatment of peritoneal macrophages with rIFN-${\gamma}$ plus HRG caused significant increases in tumor necrosis factor (TNF)-${\alpha}$ mRNA expression and production. Because NO and TNF-${\alpha}$ play an important role in the immune function and host defense, HRG treatment can modulate several aspects of the host defense mechanisms as a result of the stimulations of the inducible nitric oxide synthase and NF-${\kappa}B$. In conclusion, our findings demonstrate that HRG increases the productions of NO and TNF-${\alpha}$ from rIFN-${\gamma}$-primed macrophages and suggest that Rip2/IKK-${\beta}$ plays a critical role in mediating these immune regulatory effects of HRG.

  • According to the literatural study om ryuk-jeol disease(歷節病), we obtained the result as follows : First, ryuk-jelo disease(歷節病) was stated a kind of arthralgia syndrome(痺病), Because ryuk-jeol disease(歷節病) is only joint disease while arthrlgia syndrome(痺病) affects skin, muscle, joint, bone, organs, etc. Second, ryuk-jeol disease(歷節病) can bne thought to be a category of rheumatoid arthritis in western medical science. Third, etiology factors of ryuk-jeol disease(歷節病) was classified endogenous and exogenous pathogenic factors. The formers was the deficiency in both Qi and Blood(氣血不足), the deficiency in both the liver and kidney(肝腎虧損), dam(痰), blood stasis(瘀血). The latter was Windg風), Cool(寒), Dampness(濕), Heat(熱). Forth, the therapy of ryuk-jeol disease(歷節病) was based on 'diagnosis and trentment based on over all analysis of symptoms and sings(辨證施治)'
