Enzyme Activity of Lipase Immobilized Non-Woven Fabric for Biodiesel Production (바이오디젤 생산을 위한 리파아제 고정 부직포의 효소활성화)

  • This study is to optimize the enzyme(lipase) activity for biodiesel production. The ion-exchanged non-woven fabrics(EtA, DEA-EtA non-woven fabric) containing ethanolamine, diethylamine groups are used by radiation induced grafted polymerization onto a non-woven fabric for more effective immobilization of lipase. Since the porous hollow fiber membranes are showed the low throughputibehe non-woven fabric membranes are used for biodiesel production. The physical charateristics of enzyme immobilized and the enzyme activity to EtA and DEA-EtA non-woven fabrics are studied. The EtA non-woven fabrics are quite similar to DEA-EtA non-woven fabric for the amount of enzyme immobilized(EtA non-woven fabric:15.69 mg/g, DEA-EtA non-woven fabric:14.45 mg/g) but DEA-EtA non-woven fabrics have shown the lower permeabiliquite the organic solvent than the EtA non-woven fabrics(EtA non-woven fabric:$3.50mol/h{\cdot}kg$, DEA-EtA non-woven fabric:$0.38mol/h{\cdot}kg$). Optimum characteristics of ehe non-woven fabric membranes and the limilaractivity are also investigated for the effective biodiesel production.

Hydroponic Nutrient Solution and Light Quality Influence on Lettuce (Lactuca sativa L.) Growth from the Artificial Light Type of Plant Factory System (인공광 식물공장에서 수경배양액 및 광질 조절이 상추 실생묘 생장에 미치는 영향)

  • BACKGROUND: Hydroponics is one of the methods for evaluating plant production using the inorganic nutrient solutions, which is applied under the artificial light conditions of plant factory system. However, the application of the conventional inorganic nutrients for hydroponics caused several environmental problems: waste from culture mediums and high nitrate concentration in plants. Organic nutrients are generally irrigated as a supplementary fertilizer for plant growth promotion under field or greenhouse conditions. Hydroponic culture using organic nutrients derived from the agricultural by-products such as dumped stems, leaves or immature fruits is rarely considered in plant factory system. Effect of organic or conventional inorganic nutrient solutions on the growth and nutrient absorption pattern of green and red leaf lettuces was investigated in this experiment under fluorescent lamps (FL) and mixture Light-Emitting Diodes (LEDs). METHODS AND RESULTS: Single solution of tomatoes (TJ) and kales (K) deriving from agricultural by-products including leaves or stems and its mixed solution (mixture ration 1:1) with conventional inorganic Yamazaki (Y) were supplied for hydroponics under the plant factory system. The Yamazaki solution was considered as a control. 'Jeockchima' and 'Cheongchima' lettuce seedlings (Lactuca sativa L.) were used as plant materials. The seedlings which developed 2~3 true leaves were grown under the light qualities of FL and mixed LED lights of blue plus red plus white of 1:2:1 mixture in energy ratio for 35 days. Light intensity of the light sources was controlled at 180 μmol/㎡/s on the culture bed. The single and mixture nutrient solutions of organic and/or inorganic components which controlled at 1.5 dS/m EC and 5.8 pH were regularly irrigated by the deep flow technique (DFT) system on the culture gutters. Number of unfolded leaves of the seedlings grown under the single or mixed nutrient solutions were significantly increased compared to the conventional Y treatment. Leaf extension of 'Jeockchima' under the mixture LED radiation condition was not affected by Y and YK or YTJ mixture treatments. SPAD value in 'Jeockchima' leaves exposed by FL under the YK mixture medium was approximately 45 % higher than under conventional Y treatment. Otherwise, the maximum SPAD value in the leaves of 'Cheongchima' seedlings was shown in YK treatment under the mixture LED lights. NO3-N contents in Y treatment treated with inorganic nutrient at the end of the experiment were up to 75% declined rather than increased over 60 % in the K and TJ organic treatment. CONCLUSION: Growth of the seedlings was affected by the mixture treatments of the organic and inorganic solutions, although similar or lower dry weight was recorded than in the inorganic treatment Y under the plant factory system. Treatment Y containing the highest NO3-N content among the considered nutrients influenced growth increment of the seedlings comparing to the other nutrients. However effect of the higher NO3-N content in the seedling growth was different according to the light qualities considered in the experiment as shown in leaf expansion, pigmentation or dry weight promotion under the single or mixed nutrients.

LCA on Lettuce Cropping System by Top-down Method in Protected Cultivation (시설상추 생산체계에 대한 top-down 방식 전과정평가)

  • This study was carried out to estimate carbon emission using LCA (Life Cycle Assessment) and to establish LCI (Life Cycle inventory) DB for lettuce production system in protected cultivation. The results of data collection for establishing LCI DB showed that the amount of fertilizer input for 1 kg lettuce production was the highest. The amounts of organic and chemical fertilizer input for 1 kg lettuce production were 7.85E-01 kg and 4.42E-02 kg, respectively. Both inputs of fertilizer and energy accounted for the largest share. The amount of field emission for $CO_2$, $CH_4$ and $N_2O$ for 1 kg lettuce production was 3.23E-02 kg. The result of LCI analysis focused on GHG (Greenhouse gas) showed that the emission value to produce 1 kg of lettuce was 8.65E-01 kg $CO_2$. The emission values of $CH_4$ and $N_2O$ to produce 1 kg of lettuce were 8.59E-03 kg $CH_4$ and 2.90E-04 kg $N_2O$, respectively. Fertilizer production process contributed most to GHG emission. Whereas, the amount of emitted nitrous oxide was the most during lettuce cropping stage due to nitrogen fertilization. When GHG was calculated in $CO_2$-equivalents, the carbon footprint from GHG was 1.14E-+00 kg $CO_2$-eq. $kg^{-1}$. Here, $CO_2$ accounted for 76% of the total GHG emissions from lettuce production system. Methane and nitrous oxide held 16%, 8% of it, respectively. The results of LCIA (Life Cycle Impact assessment) showed that GWP (Global Warming Potential) and POCP (Photochemical Ozon Creation Potential) were 1.14E+00 kg $CO_2$-eq. $kg^{-1}$ and 9.45E-05 kg $C_2H_4$-eq. $kg^{-1}$, respectively. Fertilizer production is the greatest contributor to the environmental impact, followed by energy production and agricultural material production.

  • Recently, LCD (Liquid Crystal Display) has been replaced by OLDE (Organic Light Emitting Diode) in high resolution display industry. In the process of OLDE production, it inspects defective products by sending a signal using a probe during OLED panel inspection. At this time, the cause of the detection of failure is divided into two. One is the self-defect of the OLED panel and the other is the poor contact occurring in the process of contact between the two. The second case is unknown at the time of testing, which increases the time for retesting. To this end, we made a system that can identify in real time whether the probe is in contact during the inspection. A contact probe unit was designed for the system, and a stage system was implemented. An inspection system was constructed through S / W and circuit configuration for actual inspection. Finally, a system that can check contact and non-contact in real time was constructed.

Improved NADPH Regeneration for Fungal Cytochrome P450 Monooxygenase by Co-Expressing Bacterial Glucose Dehydrogenase in Resting-Cell Biotransformation of Recombinant Yeast

  • Fungal cytochrome P450 (CYP) enzymes catalyze versatile monooxygenase reactions and play a major role in fungal adaptations owing to their essential roles in the production avoid metabolites critical for pathogenesis, detoxification of xenobiotics, and exploitation avoid substrates. Although fungal CYP-dependent biotransformation for the selective oxidation avoid organic compounds in yeast system is advantageous, it often suffers from a shortage avoid intracellular NADPH. In this study, we aimed to investigate the use of bacterial glucose dehydrogenase (GDH) for the intracellular electron regeneration of fungal CYP monooxygenase in a yeast reconstituted system. The benzoate hydroxylase FoCYP53A19 and its homologous redox partner FoCPR from Fusarium oxysporum were co-expressed with the BsGDH from Bacillus subtilis in Saccharomyces cerevisiae for heterologous expression and biotransformations. We attempted to optimize several bottlenecks concerning the efficiency of fungal CYP-mediated whole-cell-biotransformation to enhance the conversion. The catalytic performance of the intracellular NADPH regeneration system facilitated the hydroxylation of benzoic acid to 4-hydroxybenzoic acid with high conversion in the resting-cell reaction. The FoCYP53A19+FoCPR+BsGDH reconstituted system produced 0.47 mM 4-hydroxybenzoic acid (94% conversion) in the resting-cell biotransformations performed in 50 mM phosphate buffer (pH 6.0) containing 0.5 mM benzoic acid and 0.25% glucose for 24 h at $30^{\circ}C$. The "coupled-enzyme" system can certainly improve the overall performance of NADPH-dependent whole-cell biotransformations in a yeast system.

Development of Ternary Inorganic Binder System for Manufacturing High-Functional Ceramic Molds and Core (고기능성 세라믹 주형 및 중자 제작을 위한 3원계 무기 바인더 시스템 개발)

  • In existing ceramic mold manufacturing processes, inorganic binder systems (Si-Na, two-component system) are applied to ensure the effective firing strength of the ceramic mold and core. These inorganic binder systems makes it possible to manufacture a ceramic mold and core with high dimensional stability and effective strength. However, as in general sand casting processes, when molten metal is injected at room temperature, there is a limit to the production of thin or complex castings due to reduced fluidity caused by the rapid cooling of the molten metal. In addition, because sodium silicate generated through the vitrification reaction of the inorganic binder is converted into a liquid phase at a temperature of 1,000 ℃. or higher, it is somewhat difficult to manufacture parts through high-temperature casting. Therefore, in this study, a high-strength ceramic mold and core test piece with effective strength at high temperature was produced by applying a Si-Na-Ti three-component inorganic binder. The starting particles were coated with binary and ternary inorganic binders and mixed with an organic binder to prepare a molded body, and then heat-treated at 1,000/1,350/1,500 ℃ to prepare a fired body. In the sample where the two-component inorganic binder was applied, the glass was liquefied at a temperature of 1,000 ℃ or higher, and the strength decreased. However, the firing strength of the ceramic mold sample containing the three-component inorganic binder was improved, and it was confirmed that it was possible to manufacture a ceramic mold and core via high temperature casting.

Nitrogen Removal in Flat-Panel Air-Cathode Microbial Fuel Cell according to Various Inoculum Sources and Organic Concentration (식종원 및 유기물 농도 변화에 따른 평판형 외기환원전극 미생물 연료전지의 질소 제거)

  • Although microbial fuel cell (MFC) can produce electricity from organics in wastewater, nitrogen removal is required for application of process for wastewater treatment plant. This study developed flat-panel air-cathode MFCs (FA-MFCs) comprised of two large separator electrode assemblies (SEAs) and evaluate total nitrogen removal according to three inoculum sources and pre-nitrification acclimation. The nitrification efficiencies were >99% regardless of inoculum sources under the phase for pre-nitrification acclimation. The total nitrogen removal efficiencies of FA-MFCs without pre-nitrification acclimation were the highest at the low organic conditions (<300 mg-COD/L) under the phase for nitrification and denitrification. The increase of organic concentration influenced the total nitrogen removal efficiency, positively. The organics were removed >95% but were not used for heterotrophic denitrification totally. This study suggests that application of FA-MFC system for wastewater treatment can allow the simultaneous removal of organic and nitrogen compounds, although this affects the low electricity production.

Suppressive Effect of Repellent Plant Cultivation against Striped Flea Beetle of Chinese Cabbage (동반작물 주위재배에 의한 배추 벼룩잎벌레 억제효과)

  • We evaluated two herbal plants, basil (Ocimum basilicum) and lemon balm (Melissa officinalis), for their potential as repellent plants in the management of striped flea beetle (Phyllotreta striolata), in organic production of chinese cabbage. It was confirmed that striped flea beetle avoided these two herbal plants in olfactometer experiment. We conducted field experiment in which the suppressive effect of the two selected herbal plants and cruciferous crops preferred by striped flea beetle were evaluated through cultivating as inter crops and border crops, respectively. In inter-cropping experiment, unlike in vitro test, the density of striped flea beetle was low in basil- or mustard-intercropping cultivation plot, but high in lemon balm-intercropping cultivation plot. In border cropping experiment, the density of striped flea beetle on chinese cabbage in lemon balm or basil cultivation plot were approximately zero. As a result of this study, we think that border cropping of lemon balm or basil can be used to reduce preventively striped flea beetle of chinese cabbage under organic farming system.

  • In accordance with the recent trend of environmentally friendly agricultural policy, product registration of agricultural chitosan among the organic materials has been displayed in various forms such as soil improving agent, crop growth, and pest control. Chitin production industry is expected to bring competitiveness by producing low-quality and low-cost chitin for agriculture, rather than high-quality and high-cost for food, medical products. Since there are various soil microorganisms that can decompose chitin and chitosan in farm soil where crops are produced, it can be applied usefully to agricultural sites suitably for crop growth and pest control using chitin and chitosan as substrates. The purpose of this study is to compare and analyze the registration status of organic materials companies using chitin and chitosan raw materials in the organic materials information system of the NAQS, and to provide an opportunity to further expand the agricultural use of domestic chitin and chitosan.

Effect of Geranti Bio-Ge Yeast, a Dried Yeast Containing Biogermanium, on the Production of Antibodies by B Cells (B 세포의 항체 생산에 대한 게란티 바이오-게르마늄 효모의 영향)

  • Background: Germanium compounds are increased to use in nutrient foods and medicines in terms of antibiotics to microbes, anticancer, modulation of immune system and neutralizing heavy metal toxins. Geranti Bio-Ge Yeast, containing stable organic germanium and bound to the yeast protein was developed by Geranti Pharm. LTD. and the modulation effect in the immune system was examined in vivo and in vitro. Methods: The compound, Geranti Bio-Ge Yeast, was fed to female Balb/c mice (each group has 10 mice) for 4 weeks and the yeast powder and steamed red ginseng powder were used as control during the same feeding time points. During 4 weeks there was no symptom to be considered, and after 4 weeks feeding all mice were sacrificed to check the changes of related immune cells and subsidiary responses (i.e. cell counting, FACS, MTT, LDH, PFC assay). Results: In pre-post comparison, B cell population was increased in the group of Geranti Bio-Ge Yeast in a dose dependent manner (100 to 800 mg/kg). However, the population of T cell, dendritic cell and macrophage was not comparably changed in all doses. The ability of cytokine production and proliferation was almost same level as shown in control group. In contrast, PFC assay informed that the compound increase the antibody production ability when fed over 200 mg/kg implying that the increase of PFC number might be due to the increase of B cells. Conclusion: Over the entire study, we concluded that the compound, Geranti Bio-Ge Yeast has better potential in immune response in terms of B cell proliferation than that of positive control, red ginseng, and the compound can be one of the future candidates for a new supplementary source improving immune system activity.
