The water extracts of Samultang (Samul) has been used for treatment of ischemic heart and brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of Samul rescues cells from oxidative damages in cisplatin-induced ototoxicity. Cisplatin is a widely used chemotherapeutic agent that is also highly ototoxic. This study was designed to investigate the protective effects of Samul on ciplatin-induced ototoxicity in HEI-OC1 auditory cells and organ of Corti explant culture. Cisplatin markedly decreased the viability of HEI-OC1 auditory cells. However, treatment of HEI-OC1 cells with Samul significantly reduced cisplatin-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Cisplatin induced cytotoxicity in isolated and cultured hair cell progenitors from postnatal rat cochleae. These progenitor cells are isolated from the lesser epithelial ridge (LER, or outer spiral sulcus cell) area of pre-plated neonatal rat cochlear segments. However, Samul completely protected the morphological changes of organ of Corti and LER. Taken together, these data suggest that the protective effects of the water extracts of Samul against cisplatin may be mediated by the reduction of intracellular peroxide generation.

Basically stem cells have characteristics of multi-potency, differentiation into multiple tissue types, and self-renew through proliferation. Recent advances in stem cell biology can make identifying the stem-cell like cells in various mammalian tissues. Stem cells in various tissues can restore damaged tissue. Stem cells from the adult nervous system proliferate to form clonal floating colonies called spheres in vitro, and recent studies have demonstrated sphere formation by cells in the tympanic membrane, vestibular system, spiral ganglion, and partly in the organ of Corti. The presence of stem cells in the ear raises the possibilities for the regeneration of the tympanic membrane & inner ear hair cells & neurons. But the gradual loss of stem cells postnatally in the organ of Corti may correlate with the loss of regenerative capacity and limited hearing restoration. Future strategies using endogenous stem cells in the ear can be the another treatment modality for the patients with intractable inner ear diseases.
와우 수용기의 모세포를 파괴한 가묘에서 "전기 와우"의 내부 장치를 중이 골포내에 이입한 후에 그 전극을 고실계내에 삽입하고, "전기 와우"의 외부 장치에 폭로한 음향 자극에 대한 반응을 Electric Kymograph(Model 404 Harvard Apparastu Co.)에 묘기하여 다음과 같은 결과를 얻었다. 1) 음향 자극에 의하여 야기되는 가묘의 이개운동의 크기는 특정한 축위 안에서 자극의 강도와 평행하였다. 2) 반복 음향 자극에 의한 이개반사운동은 최초의 자극에 대해서는 약간 증대한 후, 점차 감소하는 경향이 있었다. 3)음향 자극에 의하여 야기되는 이개운동은, 절대불응기, 비교적 불응기 및 반응기 등이 관찰되었다. 4) 음향 자극에 의하여 야기되는 실험 가묘의 이개반사는 청수용기 모세포를 파괴한 후 "전기 와우"의 삽입에 의하여 "청각감"이 야기되고 있음을 시사한다.

Deiters' cells are the supporting cells in organ of Corti and are suggested to play an important role in biochemical and mechanical modulation of outer hair cells. We successfully isolated functionally different $K^+$ currents from Deiters' cells of guinea pig using whole cell patch clamp technique. With high $K^+$ pipette solution, depolarizing step pulses activated strongly outward rectifying currents which were dose-dependently blocked by clofilium, a class III anti-arrhythmic $K^+$ channel blocker. The remaining outward current was transient in time course whereas the clofilium-sensitive outward current showed slow inactivation and delayed rectification. Addition of 5 mM tetraethylammonium (TEA) further blocked the remaining current leaving a very fast inactivating transient outward current. Therefore, at least three different types of $K^+$ current were identified in Deiters' cells, such as fast activating and fast inactivating current, fast activating slow inactivating current, and very fast inactivating transient outward current. Physiological role of them needs to be established.
Cisplatin은 임상적으로 다양한 종류의 종양 치료에 사용되는 중요한 항암제 중의 하나이다. 그러나 cisplatin은 이독성, 신장독성, 골수독성, 위장독성 및 말초신경독성 등의 심각한 부작용으로 인하여 사용이 제한적이다. Cisplatin에 의한 청각장애에서 organ of Corti 외측 유모세포(outer hair cells) 손상이 유발한다. Cisplatin에 의한 세포독성에 대한 연구가 진행 중이지만 pro-inflammatory cytokine과 관련된 청각세포사멸에 대한 연구는 미비하다. 이 연구에서 cisplatin은 청각세포주 HEI-OC1 세포와 렛트 cochlear explant에서 염증성 사이토카인인 $TNF-{\alpha}$, $IL-1{\beta}$ 및 IL-6의 유전자 발현과 분비를 현저히 증가시켰다. 이들 염증성 사이토카인은 organ of Corti 청각유모세포에 직접적인 세포독성을 나타내어 외측 및 내측 유모세포와 배열을 파괴하였다. 염증반응에서 중요한 $TNF-{\alpha}$를 cisplatin을 처리한 실험군에서 immunocytochemistry를 통하여 관찰 한 결과 organ of Corti에서의 발현이 현저히 증가됨을 관찰하였다. 염증성 사이토카인에 대한 중화항체를 처리하여 cisplatin에 의한 세포독성이 현저히 감소됨을 HEI-OC1 세포와 청각유모세포에서 확인하였다. 또한 GSH, NAC와 같은 항산화제를 처리하여 세포독성이 현저히 감소됨을 확인하였다. 이상의 결과는 cisplatin에 의한 청각유모세포의 죽음에서 $TNF-{\alpha}$, $IL-1{\beta}$ 및 IL-6와 같은 염증성 사이토카인이 병리 생리학적으로 중요한 역할을 하고 있음을 시사한다.

As the results of investigation, the noise level was between 60 dB and 80 dB in the area of explosion. The residents living within 1 Km would feel uncomfortable. However, hearing loss is not happened by this range. The maximum range that the human can hear is 20,000 Hz and the maximum range that the dog can hear is between 20 Hz and 40,000 Hz. The auditory range for humans to be uncomfortable toward noise is between 1,000 Hz and 2,000 Hz. As the result of this experiment, the auditory range of dog is more wide than of a human. The change of hair cells in the Corti's organ occurred when the dog was exposed to 1,000 Hz at 100 dB for 1 month. Therefore, the structure change of the ear could happen by hearing loss because of noise, but the structure change of hair cells is the worst symptom by hearing loss because of noise.
Hearing organs have unique characteristics and have a role in processing external sensory signals. Sensory hair cells and nerve fibers in the organ of Corti can be damaged by various causes and they do not regenerate themselves. Medication used for clinical treatment for the inner ear is limited due to the anatomical structure of the inner ear. Photobiomodulation (PBM) is a therapeutic approach that uses various sources of light and the success of PBM therapy is highly reliant on the parameters of the light sources. The positive effects of PBM have been reported in various clinical fields. This paper summarizes the previously reported research on PBM for the treatment of hearing damage in animal models.
Mammalian inner ear comprises of six sensory organs; cochlea, utricle, saccule, and three semicircular canals. The cochlea contains sensory epithelium known as the organ of Corti which senses sound through mechanosensory hair cells. Mammalian inner ear undergoes series of morphogenesis during development beginning thickening of ectoderm nearby hindbrain. These events require tight regulation of multiple signaling cascades including FGF, Wnt, Notch and Bmp signaling. In this review, we will discuss the role of newly emerging signaling, FGF signaling, for its roles required for cochlear development.

Luteolin, a sort of flavonoid, has been reported to be involved in neuroprotective function via suppression of neuroinflammation. In this study, we investigated the protective effect of luteolin against oxidative stress-induced cellular senescence and its molecular mechanism using hydrogen peroxide (H2O2)-induced cellular senescence model in House Ear Institute-Organ of Corti 1 cells (HEI-OC1). Our results showed that luteolin attenuated senescent phenotypes including alterations of morphology, cell proliferation, senescence-associated 𝛽-galactosidase expression, DNA damage, as well as related molecules expression such as p53 and p21 in the oxidant challenged model. Interestingly, we found that luteolin induces expression of sirtuin 1 in dose- and time-dependent manners and it has protective role against H2O2-induced cellular senescence by upregulation of sirtuin 1 (SIRT1). In contrast, the inhibitory effect of luteolin on cellular senescence under oxidative stress was abolished by silencing of SIRT1. This study indicates that luteolin effectively protects against oxidative stress-induced cellular senescence through p53 and SIRT1. These results suggest that luteolin possesses therapeutic potentials against age-related hearing loss that are induced by oxidative stress.
