Optimal Culture Conditions for in vitro Propagation of Orostachys japonicus and Enhancement of Polysaccharide Production

  • Optimal culture conditions for efficient in vitro propagation and polysaccharide production of Orostachys japonicus were established. The highest growth yield was achieved in 1/2 MS medium, while the lowest growth yield was obtained in 4 MS medium. The patterns of polysaccharide formation were a little similar in all cases, but on MB5 medium, the po]ysaccharide contents of plant were higher than others. Among the modified nitrate levels, effective growth level were obtained in 1/4 N and 1/2 N. High contents of polysaccharide were obtained in 4 N. Different concentration of potassium and calcium did not improve the growth and polysaccharide production. The micropropagated shoots were successfully acclimatized artificial soils.

  • Rhodotorula sp. CL-82 유래 의 epoxide hydrolase 활성을 이용하여 라세믹 styrene oxide에 대한 입체선택적 가수분해 반응을 실시하였다. Rhodotorula sp. CL-82 생촉매의 입체선택적 가수분해속도를 나타내주는 반응표면 곡선에 대한 분석을 통해 pH, 반응주도, cosolvent 첨가량에 대한 최적조건을 각각 7.6,$33.3^{\circ}C$ , 3%(v/v)으로 결정하였다. 최적반응조건에서 약 4시간 정도의 반응을 통해 ee값이 99% 이상인 광학적으로 순수한 (S)-styrene oxide를 이론 수율대비 40% 수율로 얻을 수 있었다.

Polyaluminium Chloride를 이용한 미세조류 Spirogyra의 응집 최적화 (Optimization of Spirogyra Flocculation Using Polyaluminium Chloride)

  • Flocculation is known one of the effective methods for harvesting microalgae. This study was aimed to optimize the flocculation condition for decreasing the amounts of flocculant and obtaining the highest yield of algal biomass. To achieve this goal, it was optimized the flocculant concentration, reaction pH and the concentration of cell density for harvest using response surface methodology (RSM). The flocculation of microalgae, Spirogyra varians, was carried out using inorganic flocculant polyaluminium chloride. By the RSM result, the optimal flocculation condition was calculated 5 ppm of polyaluminum chloride, pH 7.5 and 0.33 of optical cell density at $OD_{640}$. The obtained recovery yield of S. varians was 97.6% at the optimal condition.

  • A novel indentation theory is proposed by examining the data from the incremental plasticity theory based finite element analyses. First the optimal data acquisition location is selected, where the strain gradient is the least and the effect of friction is negligible. This data acquisition point increases the strain range by a factor of five. Numerical regressions of obtained data exhibit that strain hardening exponent and yield strain are the two main parameters which govern the subindenter deformation characteristics. The new indentation theory successfully provides the stress-strain curve with an average error less than 5%.

  • Pleurotus eryngii(King oyster) is one of the most promising mushrooms being produced on the domestic farms. The quality as well as quantity of Eryngii is sensitively affected by micro climate factors. To safely produce high-quality Eryngii all the year round, it is required that the environmental factors be carefully controlled by well designed structures equipped with various facilities and control systems. This study was carried out at the commercial mushroom cultivation houses to find out reasonable range of each environmental factor and yield together with economic and safe structures influencing on the optimal productivity of Eryngii. This experiment was conducted from Nov. 10, 2004 to Aug. 27, 2005 in Eryngii. cultivation houses. The environmental factors measured for this study were inside/outside temperature, relative humidity and $CO_2$ concentration in Pleurotus eryngii medium. In addition, the yield and quality of mushroom were made investigation. But the optimal productivity will be evaluated by considering the quality and quantity of mushroom production, energy requirements, facility construction and management cost, etc.

완두와 보리의 엽내세포 원형질체 분이 및 융합 (The Isolation and Fusion of Pea and Barley Mesophyll Protoplasts)

  • The optimal conditions for the protoplast isolation from the leaves of pea (Pisum sativum L. cv. Sparkle) and barley (Hordeum vulgare L. cv. Baecdong) were determined in order to achieve a somatic hybridization between two species. It was revealed that the use of 0.5M sorbitol as an osmoticum was appropriate for pea. The yield of intact protoplasts was the highest (40%) when pea leaves were incubated in the enzyme solution for 4 hours. In case of barley, the optimal concentrations of cellulase, pectinase and mannitol as the enzyme solution were 2%, 1% and 0.35M, respectively. And the yield of barley protoplasts was the highest(87%) when leaves were incubated in this enzyme solution for 3.5 hours. A fusion of protoplasts from pea and barley was induced by PEG treatment enriched with calcium salts within 60 minutes.

  • Lactic acid production parameter optimization using Lactobacillus amylovorus NRRL B-4542 was performed using the design of experiments (DOE) available in the form of an orthogonal array and a software for automatic design and analysis of the experiments, both based on Taguchi protocol. Optimal levels of physical parameters and key media components namely temperature, pH, inoculum size, moisture, yeast extract, $MgSO_4{\cdot}7H_20$, Tween 80, and corn steep liquor (CSL) were determined. Among the physical parameters, temperature contributed higher influence, and among media components, yeast extract, $MgSO_4{\cdot}7H_20$, and Tween 80 played important roles in the conversion of starch to lactic acid. The expected yield of lactic acid under these optimal conditions was 95.80% and the actual yield at optimum conditions was 93.50%.

효과적인 효소 소화율을 위한 볏짚 전처리 (Pretreatment of Rice Straw for Efficient Enzyme Digestibility)

  • Rice straw was pretreated with aqueous ammonia in order to enhance enzyme digestibility. Soaking in ammonia aqueous (SAA) was conducted with 15% ammonia, at $60^{\circ}C$. for 24 h. Optimization of both saccharification conditions and enzyme loading of SAA rice straw was carried out. Especially enzyme loading test was performed using statistical method. Moreover proton beam irradiation (PBI) was also performed to overcome the problem which inhibit the enzyme digestibility at 1-25 kGy doses with 45 MeV of beam energy. Optimal condition for enzymatic saccharification was follows; pH 4.8, $50^{\circ}C$, 60 FPU of enzyme activity, 1:4 ratio of celluase and ${\beta}$-glucosidase. Also, optimal doses of PBI on rice straw and SAA-treated rice straw for efficient sugar recovery were found to be 3 kGy, respectively. When saccharification was performed with optimal condition, glucose conversion yield was 89% of theocratical maximum in 48 h, and 3 kGy of PBI was applied to SAA-treated rice straw, approximately 90% of the theoretical glucose yield was obtained in 12 h. The results of X-ray diffractometry (XRD) support the effect of both SAA and PBI on sugar recovery, and scanning electron microscopy (SEM) images unveiled the physical change of the rice straw surface since rugged rice straw surface was observed.

Improvement of Fungal Cellulase Production by Mutation and Optimization of Solid State Fermentation

  • Spores of Aspergillus sp. SU14 were treated repeatedly and sequentially with $Co^{60}$ ${\gamma}$-rays, ultraviolet irradiation, and N-methyl-N'-nitro-N-nitrosoguanidine. One selected mutant strain, Aspergillus sp. SU14-M15, produced cellulase in a yield 2.2-fold exceeding that of the wild type. Optimal conditions for the production of cellulase by the mutant fungal strain using solid-state fermentation were examined. The medium consisted of wheat-bran supplemented with 1% (w/w) urea or $NH_4Cl$, 1% (w/w) rice starch, 2.5 mM $MgCl_2$, and 0.05% (v/w) Tween 80. Optimal moisture content and initial pH was 50% (v/w) and 3.5, respectively, and optimal aeration area was 3/100 (inoculated wheat bran/container). The medium was inoculated with 25% 48 hr seeding culture and fermented at $35^{\circ}C$ for 3 days. The resulting cellulase yield was 8.5-fold more than that of the wild type strain grown on the basal wheat bran medium.
