Production of Cell Mass and Monacolin K from Monascus sp. on Rice Solid Culture (Monascus 속 균주의 균체 생산 및 고체배양에 의한 Monacolin K 생산)

  • The optimal conditions for production of Monascus sp. KM100l cell mass on submerged culture and production of monacolin K on rice solid culture were investigated. An overproducing mutant of Monascus pigments, KM 1001 mutant, from Monascus purpureus KCCM60016 was selected by NTG treatment. The optimal medium for the production of KM100l mutant cell mass is instructed to be composed of 3% glucose, 2% yeast extract, 0.1 % KH$_2$PO$_4$, 0.05% The optimal conditions for production of Monascus sp. The optimal medium for the production of KM100l mutant cell mass is instructed to be composed of 3% glucose, 2% yeast extract, 0.1 % $(KH_2PO_4$, 0.05% $MgSO_4{\cdot}7H_2O$, 0.2% L-asparagine, pH 4.5, and the optimal inoculum size and shaking speed were $1.5{\times}10^6$ spores/50 m1 medium and 150 rpm, respectively. On optimal conditions, 4.1 g/l of the cell mass was obtained at 28$^{\circ}C$ for 3 days. The mycelium were inoculated on 500 g of steamed rice using vinyl bag ($30.6{\times}44$ cm) and incubated at $30^{\circ}C$, 85% humidity for 21 days. Lactone form monacolin K was rapidly increased for 2 days and reached highest concentration of monacolin K (2,930 mg/kg) for 15 days, and monacolin K was decreased after 15 days.

  • In this study the optimal control of fed-batch glycerol fermentation is investigated based on an impulsive dynamical system. Considering the sudden increase of the glycerol and alkali in fed-batch culture of biodissimilation of glycerol to 1,3-propanediol, this paper proposes a non-linear impulsive system of fed-batch culture. The existence, uniqueness and regularity properties of piecewise solution for the system are proved. In view of the controllability of volumes of glycerol added to the reactor instantaneously, the paper constructs an optimal control model based on the nonlinear impulsive system and the existence of the optimal control is obtained. The control variables here are the moments and the sizes of jumps in the states at the discrete instants and the objective is to maximize the productivity of 1,3-propanediol over one cycle.

  • The present study has been carried out to investigate the optimal condition on lymphocyte blastogenesis of rabbit lymphocytes, whole blood culture and microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the In vitro optimal condition of rabbit lymphocytes. Data were presented to show many variables that are involved in studying the phytohemagglutinin(PHA) and lipopolysaccharide(LPS) response of rabbit lymphocyte in a microculture system. Analysis indicated that the conditions for optimal PHA as measured by incorporation of $^3H$-TdR include: (1) use of RPMI-1640 as culture medium. (2) use of $6{\mu}g$ of PHA, per culture. (3) 48-hours culture period. Conditions for optimal stimulation with LPS mitogen were similar to those used for PHA.

  • The useful fungi which secret extracellular enzymes was selected for deinking agent of old newsprint. Five fungal strains were isolated from a paper mill soil ground. The CMCase, FPase and xylanase activities of fungi on the liquid culture were investigated at optimal growth conditions. The results of this study were as follow: The optimal pH and temperature for culture growth were 4~8 and 27~$35^{\circ}C$, respectively. For screening of extracellular enzymes at optimal culture conditions the optimal culture period were less than 6-7 days. Fusarium pallidoroseum and Aspergiilus niger which shows relatively higher CMCase, FPase and xylanase activities than the other species were selected for further enzymatic deinking research.

  • A novel cathepsin B inhibitor-producing bacterium was isolated from marine sediments and identified based on its 16S rDNA sequence as Pseudomonas sp. strain PB01 (Accession No. EU126129). The growth and enzyme inhibitor production were investigated under various culture conditions. A mixture of organic nitrogen source was required for the optimal production, whereas both glucose and maltose proved to be the effective carbon sources for cathepsin B inhibitor production. Other optimal culture conditions included temperature range between 25 and $28^{\circ}C$, initial medium pH of 6.6, and shaking speed of 200 rpm. Under these optimal conditions, the maximum inhibitory activity from culture broth was approximately 50% after 30 h of cultivation. Additionally, kinetic study revealed that inhibitor production paralleled with cell growth, which suggested that the inhibitor may be a primary metabolite of that bacterium.

  • To establish the optimal culture systems for production of transferable embryos in Korean Cattle, pregnancy rates of IVF-derived blastocysts according to different culture media, culture method and culture duration were compared. Development of IVF-derived embryos to blastocysts was most effective in YS medium group co-cultre with cumulus cells. Blastocysts cultured for 6 to 8 d in vitro showed higher hatching rate and good quality. Pregnancy rates after transfer of IVF-derived blastocysts cultured for 7 or 8 d were high. Through our experiments, it is considered that improvement of culture media and culture method is necessary for mass production of blastocysts with excellent of good quality in Korean Cattle.

  • Agrocybe aegerita is Hymenomycetes fungus belonging to the order Agaricales and family of Bolbitiaceae. Much less is known about liquid culture of Agrocybe aegerita. Thus, the present study was to investigate the liquid cultural characteristics of Agrocybe aegerita my-celium. The optimal medium for the mycelial growth and density was ME medium, optimal tem-perature and initial pH were 25$\pm$1$^{\circ}C$ and 5.5, respectively. And optimal culture time for mycelial growth was 12 days. The modified optimal medium compositions were dextrin 3% (w/v), yeast extract 2% (w/w), MgSO$_4$0.05% (w/v), and KH$_2$PO$_4$0.15% (w/v). Under optimal culture conditions, the mycelial growth of modified optimal medium was higher than that of ME medium.

  • The present study has been carried out to investigate the optimal condition on the blastogenesis of guinea pig lymphocytes. A microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the in vito optimal condition of guinea pig lymphocytes. Data were presented to show many variables that are Involved in studying the responses of guinea pig lymphocyte in a microculture system to the stimulation of Concanavalin A(Con A) and lipopolysaccharide (LPS). Analysis indicated that the conditions for optimal Con A as measured by incorporation of $^3H$-TdR include : (1) use of RPMI-1640 as culture medium, (2) use of $6{\mu}g$ of Con A, per culture, (3) use of $1{\times}10^6$ cells per culture. Conditions for optimal stimulation with LPS mitogen were similar to those used for Con A.

  • This study was carried out to select the useful bacteria which secret extracellula enzymes for enzymatic deinking agent of old newspaper. CMCase, FPase and xylanase activities of the bacteria liquid culture were measured at optimal growth conditions. Clear zone test was checked on the solid culture. The results of this study were as follow: Eight strains of 28 bacteria isolated from a paper mill soil ground were shown strong CMCase and xylanase activity with the clear zone test. The optimal pH and temperature for culture growth were 6~8 and 26~$34^{\circ}C$, respectively and optimal culture period were less than 60 hours. Based on CMCase, FPase and xylanase activity, strain No. 18, 21, 22 and 28 which were relatively higher than the other strains, were selected for further enzymatic deinking research.

  • This study was carried out to determine the effects of plant growth regulators on cell culture and organogenesis from Sicyos angulatus L. using explants of leaves, stems and cotyledons. Optimal callus induction for S. angulatus was obtained on MS medium with 0.1mg/$\ell$ BA and 2.0mg/$\ell$ 2,4 -D from cotyledons, 0.1mg/$\ell$ BA and 5.0mg/$\ell$ NAA from leaves explants, Optimal media for subculture and growth of S. angulatus callus were 1/2 MS medium with 0.1mg/$\ell$ BA and 1.0mg/$\ell$ 2,4 -D for solid culture, and 0.1mg/$\ell$ 2,4-D for suspension culture. Many adventitious roots with some shoots were formed were formed from leaf and cotyledon explants of S. angulatus during callus induction with optimal combinations of plants growth regulators.
