Stabilization of Quinonoid Intermediate E-Q by Glu32 of D-Amino Acid Transaminase

  • The stable anchorage of pyridoxal 5'-phosphate (PLP) in the active site of D-amino acid transaminase (D-AT) is crucial for the enzyme catalysis. The three-dimensional structure of D-AT revealed that Glu32 is one of the active site groups that may playa role in PLP binding. To prove the role of Glu32 in PLP stability, we firstly checked the rate of the potential rate-limiting step. The kinetic analysis showed that the rate of the ${\alpha}$-deprotonation step reduced to 26-folds in E32A mutant enzyme. Spectral analyses of the reaction of D-AT with D-serine revealed that the E32A mutant enzyme failed to stabilize the key enzyme-substrate intermediate, namely a quinonoid intermediate (E-Q). Finally, analysis of circular dichroism (CD) on the wild-type and E32A mutant enzymes showed that the optical activity of PLP in the enzyme active site was lost by the removal of the carboxylic group, proving that Glu32 is indeed involved in the cofactor anchorage. The results suggested that the electrostatic interaction network through the groups from PLP, Glu32, His47, and Arg50, which was observed from the three-dimensional structure of the enzyme, plays a crucial role in the stable anchorage of the cofactor to give necessary torsion to the plane of the cofactor-substrate complex.

Microstructure and Wear Property of $Al-5Mg-X(Si,Cu,Ti)/SiC_p$ Composites Fabricated by Pressureless Infiltration Method (무가압 침투법에 의해 제조된 $Al-5Mg-X(Si,Cu,Ti)/SiC_p$ 복합재료의 조직 및 마멸특성)

  • Metal matrix composites(MMCs) reinforced with hard particles have many potential application in aerospace structures, auto parts, semiconductor package, heat resistant panels, wear resistant materials and so on. In this work, the effect of SiC partioel sizes(50 and 100 ${\mu}m$) and additional elements such as Si, Cu and Ti on the microstructure and the wear property of $Al-5Mg-X(Si,Cu,Ti)/SiC_p$ composites produced by pressureless infiltration method have been investigated using optical microscopy, scanning eletron microcopy(SEM) with EDS(energy dispersive spectrometry), hardness test, X-ray diffractometer(XRD) and wear test. In present study, the sound $Al-5Mg-X(Si,Cu,Ti)/SiC_p$(50 and 100 ${\mu}m$) composites were fabricated by pressureless infiltration method. The $Al-5Mg-0.3Si-O.1Cu-O.1Ti/SiC_p$ composite with $50 {\mu}m$ size of SiC particle has higher hardness and better wear property than any other composite with $100{\mu}m$ size of SiC particle produced by pressureless infiltration method. The hardness and wear property of $Al-5Mg/SiC_p$(50 and 100 ${\mu}m$) composites were enhanced by the addition of Si, Cu and Ti in Al-5%Mg matrix alloy.

Advanced Forecasting Approach to Improve Uncertainty of Solar Irradiance Associated with Aerosol Direct Effects

  • Numerical Weather Prediction (NWP) models such as the Weather Research and Forecasting (WRF) model are essential for forecasting one-day-ahead solar irradiance. In order to evaluate the performance of the WRF in forecasting solar irradiance over the Korean Peninsula, we compared WRF prediction data from 2008 to 2010 corresponding to weather observation data (OBS) from the Korean Meteorological Administration (KMA). The WRF model showed poor performance at polluted regions such as Seoul and Suwon where the relative Root Mean Square Error (rRMSE) is over 30%. Predictions by the WRF model alone had a large amount of potential error because of the lack of actual aerosol radiative feedbacks. For the purpose of reducing this error induced by atmospheric particles, i.e., aerosols, the WRF model was coupled with the Community Multiscale Air Quality (CMAQ) model. The coupled system makes it possible to estimate the radiative feedbacks of aerosols on the solar irradiance. As a result, the solar irradiance estimated by the coupled system showed a strong dependence on both the aerosol spatial distributions and the associated optical properties. In the NF (No Feedback) case, which refers to the WRF-only stimulated system without aerosol feedbacks, the GHI was overestimated by $50-200W\;m^{-2}$ compared with OBS derived values at each site. In the YF (Yes Feedback) case, in contrast, which refers to the WRF-CMAQ two-way coupled system, the rRMSE was significantly improved by 3.1-3.7% at Suwon and Seoul where the Particulate Matter (PM) concentrations, specifically, those related to the $PM_{10}$ size fraction, were over $100{\mu}g\;m^{-3}$. Thus, the coupled system showed promise for acquiring more accurate solar irradiance forecasts.

  • Recently, various type of nanomaterials such as nanorod, nanowire, nanotube and their core/shell nanostructures have attracted much attention in photocatalyst due to their unique properties. Among them, Type-II core/shell heterostructures have extensively studied because it has exhibited improved electrical and optical properties against their single-component nanostructure. Such structures are expected to offer high absorption efficiency and fast charge transport due to their stepwised energetic combination and large internal surface area. Thus, it has been considered as potential candidates for high efficient photocatalytic activity. In this work, we introduce a novel chemical conversion process to synthesize Type-II ZnO/ZnSe core/shell heterostructures. A plausible conversion mechanism to ZnO/ZnSe core/shell heterostructres was proposed based on SEM, XRD, TEM and XPS analysis. The ZnO/ZnSe heterostructures exhibited excellent photocatalytic activity toward the decomposition of RhB dye compared to the ZnO nanorod arrays due to enhanced light absorption and the type-II cascade band structure.

Self-assembly of ZnO Stripes Prepared by Anodization in an Ethanolic Sulfuric Acid (에탄올/황산 혼압액에서 양극산화법을 이용한 자기정렬된 ZnO 줄무늬 구조 제조 연구)

  • There are no many research reports on the preparation of ZnO by electrochemical oxidation since the zinc oxide is very easily dissolved in an acidic or basic environment, even though zinc oxides have attracted many attentions because of their optical/electrical properties. In this paper, we describe the fabrication of self-ordered stripes of ZnO by anodization of Zn in an ethanolic sulfuric acid. The formation of stripes of ZnO originating from Zn is attributed to water-selective dissolution of ZnO during anodization. We study in detail the effects of concentration of $H_2SO_4$, applied potential, anodization time, and addition of a small amount of water on the fabrication of stripes of ZnO. Mechanisms for the fabrication of ZnO stripes are discussed in terms of the above-mentioned effects.

Prediction of Residual Layer Thickness of Large-area UV Imprinting Process (대면적 UV 임프린팅 공정에서 잔류층 두께 예측)

  • Nanoimprint lithography (NIL) is the next generation photolithography process in which the photoresist is dispensed onto the substrate in its liquid form and then imprinted and cured into a desired pattern instead of using traditional optical system. There have been considerable attentions on NIL due to its potential abilities that enable cost-effective and high-throughput nanofabrication to the display device and semiconductor industry. Although one of the current major research trends of NIL is large-area patterning, the technical difficulties to keep the uniformity of the residual layer become severer as the imprinting area increases more and more. In this paper, with the rolling type imprinting process, a mold, placed upon the $2^{nd}$ generation TFT-LCD glass sized substrate($370{\times}470mm^2$), is rolled by a rubber roller to achieve a uniform residual layer. The prediction of residual layer thickness of the photoresist by rolling of the rubber roller is crucial to design the rolling type imprinting process, determine the rubber roller operation conditions-mpressing force & feeding speed, operate smoothly the following etching process, and so forth. First, using the elasticity theory of contact problem and the empirical equation of rubber hardness, the contact length between rubber roller and mold is calculated with consideration of the shape and hardness of rubber roller and the pressing force to rubber roller. Next, using the squeeze flow theory to photoresist flow, the residual layer thickness of the photoresist is calculated with information of the viscosity and initial layer thickness of photoresist, the shape of mold pattern, feeding speed of rubber roller, and the contact length between rubber roller and mold previously calculated. Last, the effects of rubber roller operation conditions, impressing force & feeding speed, on the residual layer thickness are analyzed with consideration of the shape and hardness of rubber roller.

Gaze Interaction Using Half Blink Selection and The Wearable AR System (반 깜박임 선택을 이용한 응시 상호작용과 착용형 AR 시스템)

  • In this paper, we propose a wearable augmented reality system using gaze interaction. Gaze interaction has been considered to be the potential of easy, natural and fast way of interaction and becomes a suitable way in optical see-through HMD based wearable AR. Our system recognizes user's gaze point, half blink motion and displays the information of object seen by user to HMD. Half blink selection technique avoids the Midas touch problem and represents user's intention correctly. We've developed a AR annotation system and estimated the usability of gaze interaction. The accuracy and robustness of our technique is verified on the experimental results.

Spatial Analysis of Major Atmospheric Aerosol Species Using Earth Observing Satellite Data (지구관측 위성자료를 이용한 주요 대기 에어러솔 성분의 공간분포 분석)

  • Atmospheric aerosols, small particles in the atmosphere, are one of the important parameters in climate change and human health. Additionally, accurate estimates of aerosol species are increasingly important in environmental impact assessment studies. Recent advances in global satellite remote sensing provide powerful tool for air quality monitoring. This study explores the potential usage of satellite derived data such as atmospheric aerosols for air quality monitoring as well as climate change study. The objectives of this study is to understand the general features of the global distribution of type dependent aerosols. A detailed spatio-temporal variability of the each different satellite dataset shows the variation of the global zonal average and specific geographical regions where the strong emission sources are located. Especially, significantly large aerosol amounts are observed in Asia and Africa because of the desert dust storm, anthropogenic and biomass burning emissions.

  • One-dimensional nanostructures have been researched widely because of its unique physical properties such as optical, electrical, mechanical, and chemical properties in comparison with bulk structures. Especially nanotubular structures are able to provide larger surface area, capability to load purposeful materials, and unique mechanical modulus. We reviewed the oxide nanotube technology with focusing on the method of template-directed fabrication. We can easily control of physical dimensions of nanotubes by control of nanotemplate and fabrication condition. and template-directed fabrication is ideal tool to fabricate the amount of monodisperse nanotubes. They have potentials for application in solar cell, drug-delivery, Li-ion batteries and photocatalyst. We discussed these potential applications and research trends.

The influence of preparation conditions on the electrochemical degradation of tungsten oxide thin films prepared by electron beam deposition (제작조건이 전자비임으로 제작된 텅스텐산화물 박막의 전기화학적 퇴화에 미치는 영향)

  • The electrochromic $WO_3$thin films were prepared by using the electron-beam evaporatin technique. Flms prepared at a vacuum pressure of $10^{-4}$ mbar were found to be most stable during repeated potential cycles. The chemical stability of the film in aqueous solutions was also affected by the vacuum pressure during evaporation. The redox current and the optical properties of the degraded films were affected by the thickness of the film. The 5,000$\AA$-thick films were found to be most stable, undergoing the least degradation during the repeated coloring and bleaching cycles. The origin of the mechanism dominating the degradation during the repeated coloring and bleaching cycles was the accumulation of lithium in the film, which results in decreasing redox current. Tungsten oxide films with titanium content of about 10-15 mol% was found to be most stable, undergoing the least degradation during the repeated cycles. The origin of the mechanism dominating the least degradation during the repeated cycles was the reduction of lithium ion trapping sites in the films, which results in a increased durability.
