Demonstration of the Usefulness of Optical Coherence Tomography in Imaging a Mouse Tail Model of Lymphedema

  • To investigate the usefulness of optical coherence tomography (OCT) for imaging lymphedema, we directly compared it to other histological methods in a mouse model of lymphedema. We performed detailed imaging of the lymphedema lesion on a mouse tail. We imaged the mouse tail in vivo with OCT and created histopathological samples. We constructed a spectrometer-based OCT system using a fiber-optic Michelson interferometer. The light was directed to 50:50 couplers that split the light into reference and sample arms. Backscattered light from a reference mirror and the sample produced an interference fringe. An OCT image of the lymphedema model revealed an inflammatory reaction of the skin that was accompanied by edema, leading to an increase in the light attenuation in the dermal and subcutaneous layers. Similar to OCT image findings, histological biopsy showed an inflammatory response that involved edema, increased neutrophils in epidermis and subdermis, and lymphatic microvascular dilatation. Furthermore, the lymphedema model showed an increase in thickness of the dermis in both diagnostic studies. In the mouse tail model of lymphedema, OCT imaging showed very similar results to other histological examinations. OCT provides a quick and useful diagnostic imaging technique for lymphedema and is a valuable addition or complement to other noninvasive imaging tools.

비접촉식 광생체단층촬영 기술을 이용한 맥진 연구 -맥의 빠르기, 크기 및 맥력을 중심으로- (The study of non-contact/non-invasive pulse analyzing system using Optical Coherence Tomography (OCT) for oriental pulse diagnosis)

  • Objective: Optical Coherence Tomography (OCT) has emerged as an important optical imaging modality in non-invasive medical diagnostics. Hence, the aim of this study is to measure the similarity of the diagnosis by a traditional method using doctor's hand for feeling of pulse and by the non-contact/non-invasive pulse analyzing system using OCT on Chon(寸), Kwan(關), Chuk(尺). Method: Four korean medical doctors and the non-contact/non-invasive pulse analyzing system using OCT have measured the rapidity, the dimension, and the power of pulse waves of 25 volunteers. First, four korean medical doctors measured pulse waves of volunteers. During measuring, four doctors were separated from each other and so were volunteers. And then, the pulse waves of volunteers were measured by OCT. This was performed on the right Chon(寸), Kwan(關), Chuk(尺). Results: The study showed that the traditional method and the OCT based method had the 88% matches on the values of the slow and rapid pulse condition (遲數), 64% matches on the values of the small and big pulse condition(微細弱緩大[洪]), and 72% matches on the values of the weak and strong pulse condition(虛實). Conclusions: Based on the high similarities of the measurements of two approaches, we suggest that the OCT based pulse diagnosis method is useful for compensating the traditional method for the pulse diagnosis.

Spatial Resolution Enhancement with Fiber - based Spectral Filtering for Optical Coherence Tomography

  • We report a technique that improves the spatial resolution of optical coherence tomography (OCT) by utilizing fiber-based spectral filtering. The proposed technique improves the resolution by filtering out the erbium’s characteristic peak from the amplified spontaneous emission (ASE) source spectrum, and reshaping the spectrum to Gaussian-like. We used a long period fiber grating (LPG) and an erbium doped fiber (EDF) absorber for the spectral filtering. An in-house made ASE source as well as a commercial ASE source [ASE-FL7002] was used as the OCT sources to study the proposed technique. The resolution of the OCT based on an in-house made ASE source is enhanced from 200 to 40 ㎛ with an LPG. While, the resolution of the OCT based on a commercial ASE source is enhanced from 25 to 19 ㎛ with the aid of an EDF absorber. However, sidelobes still exist in the interferogram due to imperfect spectral filtering, which limited the resolution. Further enhancement in the spatial resolution of the OCT system using the ASE source is possible with the aid of cascaded LPGs and/or carefully designed EDF absorber.

High-Resolution 3-D Refractive Index Tomography and 2-D Synthetic Aperture Imaging of Live Phytoplankton

  • Optical measurements of the morphological and biochemical imaging of phytoplankton are presented. Employing quantitative phase imaging techniques, 3-D refractive index maps and high-resolution 2-D quantitative phase images of individual live phytoplankton are simultaneously obtained without exogenous labeling agents. In addition, biochemical information of individual phytoplankton including volume, mass, and density of individual phytoplankton are also quantitatively obtained from the measured refractive index distributions. We expect the present method to become a powerful tool for the study of phytoplankton.

Identification of Fungus-infected Tomato Seeds Based on Full-Field Optical Coherence Tomography

  • The morphological changes of anthracnose (fungus) -infected tomato seeds have been studied to identify the infection and characterize its effect. Full-field optical coherence tomography (FF-OCT) has been utilized as a nondestructive but efficient modality for visualizing the effects of fungal infection. The cross-sectional images extracted from a stack of en face FF-OCT images showed significant changes with infection in the seed structure. First of all, the seed coat disappeared with the infection. The thickness of the seed coat of a healthy seed was measured as 28.2 ㎛, with a standard deviation of 1.2 ㎛. However, for infected seeds the gap between surface and endosperm was not appreciably observed. In addition, the measurements confirmed that the dryness of seeds did not affect the internal seed structure. The reconstructed three-dimensional (3D) image revealed that the permeability of the seed coat, which plays the vital role of protecting the seed, is also affected by the infection. These results suggest that FF-OCT has good potential for the identification of fungus-infected tomato seeds, and for many other tasks in agriculture.

Realization of 3-D Topographic and Tomograpic Images with Ultrahigh-resolution Full-field Optical Coherence Tomography

  • We present an ultrahigh-resolution full-field optical coherence tomography (FF-OCT) implemented with a white-light interference microscope and a detector array as an alternative OCT technique. The use of detector array allows the capture of two-dimensional en-face images in parallel without taking any lateral scanning process. The phase shifting interferometric technique with the sinusoidal phase modulation (SPM) is utilized to get the demodulated OCT images. The configuration of the system and the resolution of the obtained image are presented. The topographic images, taken with the implemented system, of a coin, an integrated circuit chip, and the tomographic images of an onion epithelium are demonstrated also. Axial and lateral spatial resolution of ${\sim}1.0{\mu}m$ and ${\sim}2.0{\mu}m$ are achieved with the system respectively.

The Development of Double Clad Fiber and Double Clad Fiber Coupler for Fiber Based Biomedical Imaging Systems

  • We report the fabrication of double clad fiber (DCF) and DCF coupler, suitable for fiber based imaging systems requiring the dual-channel transmission. Unlike the conventional DCF which uses silica for both cladding layers, the proposed DCF uses a low-index polymer for its outer-cladding layer coated over the conventional silica inner-cladding layer. The DCF is drawn with a conventional SMF preform but a low-index polymer coating is used for both jacket and outercladding of the fiber. To achieve the cladding mode coupling, a DCF coupler is fabricated by simply twisting two pieces of the proposed DCF after removing the polymer-coating at contacting regions. A cladding mode coupling ratio of 30% was achieved with a contact length of 16 cm. The proposed DCF and DCF coupler were employed in a composite optical coherence tomography (OCT) and fluorescence spectroscopy (FS) system, and both OCT images and FS signal from a plant tissue are measured simultaneously.

광학 전산화단층촬영 스캐너 사용을 위한 중합체 겔과 BANGTM 겔 선량계의 특성 비교 (Comparison of Polymer and BANGTM Gel Dosimeters to Use a Optical Computed Tomography Scanner)

  • The purpose of this study was to compare the basic radiological characteristics of in-house polymer gel and commercially-available gel (BANGTM) gel dosimeters with a spectro-photometer to use a optical computed tomography (CT) scanner. We investigated the radiological characteristics including dose linearity, absorbance spectrum, dose rate dependency and inter-and intra-reproducibility at wavelengths of 590, 600 and 630 nm. The optimal dose linearities of two gel dosimeters showed R2 value of 0.939 and 0.948 at wavelengths of 590 nm and 600 nm, respectively. For two polymer gel dosimeters, there is no peak sensitivity within the range of all wavelengths in absorbance spectrum. For in-house gel dosimeter, the dose rate dependency were within 5% for all wavelengths except for the dose rate of 100 MU/min. For BANGTM gel dosimeter, the dose rate dependency showed an error range of ±5% for all wavelengths. The inter-and intra-reproducibility of two gel dosimeters were within the range of 2.5%. We have confirmed that the two gel dosimeters was appropriate for use with a optical CT scanner.

  • 본 연구의 목표는 확산 광학 단층 촬영에 대한 기준 영상을 사용하여 동질성과 이질성을 분리하기 위한 재구성된 영상들간의 수치적 평가를 위해 구조적 유사성 지수에 기초한 알고리즘을 개발한다. 글로벌 지오메트리 및 관심 영역 평가는 유사성을 산출하기 위해 측정되었으며, 그 결과 구조적 유사성 지수의 평균이 모델 내부에 가시적 포함 여부를 판단할 수 있는 잠재적 성능을 나타낸다는 것을 알 수 있으며, 구조적 유사성 지수는 유방 구조 정보를 평가하기 위한 이미지 평가를 지원 가능한 것으로 확인 되었다.

Nd:YAG 레이저에 의한 다층 박막의 미소 점 마킹 (Spot marking of the multilayer thin films by Nd:YAG laser)

  • 콤팩트디스크(CD-R; Compact DiskRecordable)를 성분별로 분리하여 제작하고, 다층 박막 구조를 만들어서 레이저빔의 에너지를 변화시켜 가면서 조사하여 각 성분 층에서의 최적 미소 점 마킹 조건과 홈 형성 과정에 관하여 연구하였다. 본 연구는 Q-스위치 Nd:YAG 레이저를 이용하여 준비된 각 시료의 표면에 27∼373 mJ 빔을 80 $\mu\textrm{m}$의 점적 크기로 조사하여 샘플에 형성된 흠 형태를 광학현미경(OM; Optical Microscope)과 광 결맞음 단층촬영기(OCT; Optical Coherence Tomography)로 비교-관찰하여 미소 점 마킹의 형성 과정을 분석하였다. 다층 박막에서 용융된 기판 층은 짧은 시간동안 충분한 열 흐름이 발생하여 증배의 형성을 야기하며, 반사 층과 염료 층 사이에 흡수된 에너지는 염료를 용융시키고 체적을 증가시켰으며, 증배가 식으면서 표면장력의 영향 및 레이저빔에 의한 순간적인 시편의 온도상승으로 기화와 반동압력에 의한 질량흐름 때문에 두 층의 경계면에서 홈과 외륜의 발생을 설명할 수 있었다. 따라서 다층 박막에서의 미소 점 마킹의 형성 과정은 표면장력, 용융 점성력, 층 두께, 다층 박막 성분 물질의 물리화학적ㆍ광학적 성질과 관계가 있음을 알 수 있었다.
