Speckle imaging is capable of dynamic data acquisition at high spatiotemporal resolution, and has played a vital role in the functional study of biological specimens. The presence of various optical scatterers within the tissue causes alteration of speckle contrast. Thus structures like blood vessels can be delineated and quantified. Although laser speckle imaging is frequently used, an optimization process to ensure the maximum speckle contrast has not been available. In this respect, we here report an experimental procedure to optimize speckle contrast via applying different combinations of varying polarization of the illuminating laser light and multiple analyzer angles. Specifically, samples were illuminated by the p-polarization, 45°-polarization, and s-polarization of the incident laser, and speckle images were recorded without and with the analyzer rotated from 0° to 180° (Δ = 30°). Following the baseline imaging of a solid diffuser and a fixed brain sample, laser speckle contrast imaging (LSCI) was successfully performed to visualize in vivo mouse-brain blood flow. For oblique laser illumination, the maximum contrast achieved with p-polarized and s-polarized light was perpendicular to the analyzer's axis. This study demonstrates the optimization process for maximizing the speckle contrast, which can improve blood-flow estimation in vivo.
본 연구는 초소형 광학 시스템을 구현할 수 있는 설계 및 제작의 방법론을 제시하고자 한다. 초소형 광학계에서는 조명 및 결상 광학소자의 성능과 소형화가 조광면의 균질도와 결상 된 이미지의 선명도에 중요한 영향을 미치게 된다. 본 연구에서는 얇은 두께로 실효 광도를 배가시키기 위한 초박형의 LED 빔조형 렌즈를 설계 제작하였다. 상기 렌즈는 중앙부의 비구면렌즈와 외각의 전반사 프레넬 가장자리 부로 구성되어 있다. 설계된 LED 빔조형 렌즈(직경 4.7 mm, 두께 0.6 mm)는 다이아몬드 선삭으로 중앙 비구면부의 전반사(TIR) 가장자리가 정밀하게 가공되었으며, LED 의 빔각을 150 도에서 17.5 도로 축소 시켰다. 다른 광학소자들 마이크로 프리즘, 결상광학용 프레넬 렌즈, 광가이드는 다양한 마이크로 나노 크기의 제조공정으로 일체형으로 성형되었다. 시작품으로 제작된 초소형 광학계($6.8{\times}2.2{\times}2.5mm$)는 마이크로 패턴을 결상의 가능성을 보여주었고, 지문인식용 초소형광학계로서의 성능을 검증하였다.
Understanding interfacial phenomena has been one of the main research issues not only in semiconductors but only in life sciences. I have been trying to meet the atomic scale surface and interface analysis challenges from semiconductor industries and furthermore to extend the application scope to biomedical areas. Optical imaing has been most widely and successfully used for biomedical imaging but complementary ion beam imaging techniques based on mass spectrometry and ion scattering can provide more detailed molecular specific and nanoscale information In this presentation, I will review the 27 years history of medium energy ion scattering (MEIS) development at KRISS and DGIST for nanoanalysis. A electrostatic MEIS system constructed at KRISS after the FOM, Netherland design had been successfully applied for the gate oxide analysis and quantitative surface analysis. Recenlty, we developed time-of-flight (TOF) MEIS system, for the first time in the world. With TOF-MEIS, we reported quantitative compositional profiling with single atomic layer resolution for 0.5~3 nm CdSe/ZnS conjugated QDs and ultra shallow junctions and FINFET's of As implanted Si. With this new TOF-MEIS nano analysis technique, details of nano-structured materials could be measured quantitatively. Progresses in TOF-MEIS analysis in various nano & bio technology will be discussed. For last 10 years, I have been trying to develop multimodal nanobio imaging techniques for cardiovascular and brain tissues. Firstly, in atherosclerotic plaque imaging, using, coherent anti-stokes raman scattering (CARS) and time-of-flight secondary ion mass spectrometry (TOF-SIMS) multimodal analysis showed that increased cholesterol palmitate may contribute to the formation of a necrotic core by increasing cell death. Secondly, surface plasmon resonance imaging ellipsometry (SPRIE) was developed for cell biointerface imaging of cell adhesion, migration, and infiltration dynamics for HUVEC, CASMC, and T cells. Thirdly, we developed an ambient mass spectrometric imaging system for live cells and tissues. Preliminary results on mouse brain hippocampus and hypotahlamus will be presented. In conclusions, multimodal optical and mass spectrometric imaging privides overall structural and morphological information with complementary molecular specific information, which can be a useful methodology for biomedical studies. Future challenges in optical and mass spectrometric imaging for new biomedical applications will be discussed.
The tetradecameric peptide (K47-K60) near the NH$_2$-terminal region of epithelial-cell adhesion molecule (Ep-CAM) was chosen as antigenic site and a polyclonal antibody was generated, which could recognize Ep-CAM from the mouse colon tissue or the colon cancer cell, CT-26, in Western blot analysis. Then, the fluorescein isothiocyanate (FITC), a fluorescence dye, was conjugated with the affinity purified Ep-CAM antibody using thiocyanate and the amino groups of FITC and antibody, respectively. The molar ratio of FITC to antibody was estimated approximately 1.86 to 1.00 by measuring the optical densities at 492 nm and 280 nm. Ep-CAM antibody-FITC conjugate was then used for immunohistochemistry of the CT-26 cells. Judging from the shapes formed by fluorescence, the Ep-CAM antibody could delivered FITC to the surface of cells in which Ep-CAM was expressed. This result implies that Ep-CAM antibody could be also used for the tissue-specific delivery of the photosensitizer to the target protein via antigen-antibody interaction.
This study aims at designing and evaluating light source devices that can stably generate light with various wavelengths in order to make possible PDD using a photosensitizer and diagnosis using auto-fluorescence. The light source was a Xenon lamp and filter wheel, composed of an optical output control through Iris and filters with several wavelength bands. It also makes the inducement of auto-fluorescence possible because it is designed to generate a wavelength band of 380-420nm, 430-480nm, and 480-560nm. The transmission part of the light source was developed to enhance the efficiency of light transmission. To evaluate this light source, the characteristics of light output and wavelength band were verified. To validate the capability of this device as PDD, the detection of auto-fluorescence using mouse models was performed.
This experiment was focused on the characterization of anti- Toxoplasma monoclonal antibodies (mAbs) and the effect of mAbs on the parasite invasion of mouse peritoneal macrophages. Twenty eight mAbs including M110, M556, R7A6 and M62l were characterized by Ab titer, immunoglobulin isotyping and western blot pattern. Antibody titer (optical density) of 4 mAbs. Ml 10. M556. R7A6 and M62l. were 0.53,0.67, 0.45 and 0.39 (normal mouse serum; 0.19) with the same IgGl isotypes shown by Enzyme-linked immunosorbent assay (ELISA). Western blot analysis showed that Ml 10. M556. R7A6 and M62l reacted with the 33 kDa (p30),31 kDa (p28),43 kDa and 36 kDa protein. Immuno-gold labelling of mAbs M110, M556, R7A6 and M621 reacted with the surface membrane, dense granules and parasitophorous vacuolar membrane (PVM) , rhoptries and cytoplasm of tachyzoite, respectively. For in vitro assay, preincubation of tachyzoties with four mAbs, Ml 10, M556, R7A6 and M62l resulted in the decrease of the number of infected macrophages (p < 0.05) and the suppression of parasite multiplication at 18 h post-infection. Four monoclonal antibodies including Ml 10 (SAGI) were found to have an important role in the inhibition of macrophage invasion and T. gondii multiplication in vitro, and these mAbs may be suitable for vaccine candidates, diagnostic kit and for chemotherapy.
Human hydatid disease (cystic echinococcosis, CE) is a chronic parasitic infection caused by the larval stage of the cestode Echinococcus granulosus. As the disease mainly affects the liver, approximately 70% of all identified CE cases are detected in this organ. Optical molecular imaging (OMI), a noninvasive imaging technique, has never been used in vivo with the specific molecular markers of CE. Thus, we aimed to construct an in vivo fluorescent imaging mouse model of CE to locate and quantify the presence of the parasites within the liver noninvasively. Drug-treated protoscolices were monitored after marking by JC-1 dye in in vitro and in vivo studies. This work describes for the first time the successful construction of an in vivo model of E. granulosus in a small living experimental animal to achieve dynamic monitoring and observation of multiple time points of the infection course. Using this model, we quantified and analyzed labeled protoscolices based on the intensities of their red and green fluorescence. Interestingly, the ratio of red to green fluorescence intensity not only revealed the location of protoscolices but also determined the viability of the parasites in vivo and in vivo tests. The noninvasive imaging model proposed in this work will be further studied for long-term detection and observation and may potentially be widely utilized in susceptibility testing and therapeutic effect evaluation.
본 논문에서는 실시간으로 입력되는 비디오 영상으로부터 사용자의 얼굴 방향을 효율적으로 추정하는 새로운 방법을 제안하였다. 이를 위하여 입력 영상으로부터 외부조명의 변화에 덜 민감한 Haar-like 특성을 이용하여 얼굴영역의 검출을 수행하고 검출 된 얼굴영역 내에서 양쪽 눈, 코, 입 등의 주요 특성을 검출한다. 이 후 실시간으로 매 프레임마다 광류를 이용해 검출된 특징 점을 추적하게 되며, 추적된 특징 점을 이용해 얼굴의 방향성 추정한다. 일반적으로 광류를 이용한 특징 추적에서 발생할 수 있는 특징점의 좌표가 유실되어 잘못된 특징점을 추적하게 되는 상황을 방지하기 위하여 검출된 특징점의 템플릿 매칭(template matching)을 사용해 추적중인 특징점의 유효성을 실시간 판단하고, 그 결과에 따라 얼굴 특징 점들을 다시 검출하거나, 추적을 지속하여 얼굴의 방향성을 추정을 가능하게 한다. 탬플릿 매칭은 특징검출 단계에서 추출된 좌우 눈, 코끝 그리고 입의 위치 등 4가지 정보를 저장한 후 얼굴포즈 측정에 있어 광류에의해 추적중인 해당 특징점들 간의 유사도를 비교하여 유사도가 임계치를 벗어 날 경우 새로이 특징점을 찾아내는 작업을 수행하여 정보를 갱신한다. 제안된 방법을 통해 얼굴의 특성 추출을 위한 특성 검출과정과 검출된 특징을 지속적으로 보완하는 추적과정을 자동적으로 상호 결합하여 안정적으로 실시간에 얼굴 방향성 추정 할 수 있었다. 실험을 통하여 제안된 방법이 효과적으로 얼굴의 포즈를 측정할 수 있음을 입증하였다.
본 논문에서는 상업용 아임센서터치 전자칠판(Interactive Whiteboard System, IWB)을 소개한다. 이 시스템은 손가락이나 펜을 이용하여 접촉식 상호작용이 가능한 화이트보드 스크린을 통해 개인용 컴퓨터를 운용할 수 있도록 도와주는 인터페이스(Interface)이다. 제안된 인터페이스는 윈도우즈 운영체제와 상호작용하며, 온도와 조명의 변화에 적응적으로 동작한다. 제안된 시스템은 카메라에서 입력된 수광부(Optical Receptive Field)의 영상을 참조영상과 비교하여 차이를 계산하고, 그 차이를 이용하여 터치스크린의 좌표값을 계산한다. 계산된 좌표값을 기반으로 윈도우즈 마우스 이벤트를 생성하여 윈도우즈시스템으로 전달한다. 우리는 참조영상을 갱신하기 위해 두 개의 스레드(Thread)을 이용한 임계영역을 구현하고, 차이계산의 신뢰성을 위해 적응적 임계값을 이용한 참조영상의 갱신을 구현한다. 제안된 터치스크린 인터페이스를 장착한 전자칠판 시스템은 향후 국내외 시장의 성장률이 높아 전도유망한 상품이며, 시장성이 밝을 것으로 기대한다.
효율적인 바이오칩을 개발을 위해 칩 표면에 생체 물질들의 상호반응이 효과적으로 일어날 수 있는 센싱 표면의 조성과 항원-항체 반응과 같은 생체인식 반응을 정량적 신호로 전환하는 방법에 대해 연구하였다. 전기화학식 센서의 표면을 개선하기 위해 폴리아미도아민 덴드리머를 가교 물질로 도입하였다. 생체 분자들의 인식작용을 정량적인 신호로 전환하기 위해 전형적인 면역조직화학분석에서 사용된 반응들을 바이오센서에 적용한 방법론을 사용하였다 효소에 의해 촉매되는 신호화 방법은 면역반응들에 대하여 광학식 센서와 전기화학식 센서에서 공히 수행되었으며, 매우 정량적인 신호로 측정되었다. 측정된 신호들로부터 단백질 농도에 비례하는 검량곡선을 획득할 수 있었으며 다양한 면역 샘플에 대한 적용 가능성을 제시하였다.
