  • Objectives : The aim of this study was to evaluate the efficacy of Hwangryunhaedok-tang (HHT) and Gungangbuja-tang (GBT) on lipopolysaccharide (LPS)-induced mouse model of inflammation. HHT and GBT are one of the representative prescriptions of cold drug and one of the representative prescriptions of hot drug, respectively. For experimental evaluation of their efficacy, we investigated the anti-inflammatory effects of HHT and GBT on LPS-induced inflammation and the mechanisms of their action. Methods : ICR mice were given a HHT (50, 500 mg/kg), GBT (100, 1000 mg/kg) extract orally on three consecutive days. On the third day, they were administered LPS intraperitoneally (35 mg/kg), 1 h after the last sample administration. Blood and liver samples were taken 6 h after the LPS challenge. Cytokine expression and inflammation-related protein factor analyses were performed by Western blotting. Results : Oral administration of HHT significantly reduced pro-inflammatory cytokines, including interleukin (IL)-6, and interferon (IFN)-${\gamma}$ in the serum. While GBT inhibited an increase of IL-6, IFN-${\gamma}$ was not affected. Immunoblot analysis showed that LPS-induced NF-${\kappa}b$ activation was inhibited by GBT, meanwhile HHT only inhibited NF-${\kappa}b$ expression at high does (500 mg/kg). In addition, HHT and GBT inhibited LPS-induced phosphorylation of Erk1/2, Jnk and p38 MAPKs. GBT also significantly inhibited i-Nos and Cox-2 expression, and HHT inhibited only i-Nos expression. Conclusions : Both of HHT and GBT showed anti-inflammatory effects against LPS-induced endotoxemia. However, HHT significantly decreased inflammatory cytokine levels, such as IL-6 and IFN-${\gamma}$ more than GBT, while GBT significantly inhibited inflammatory proteins, including NF-${\kappa}b$, MAP Kinases, i-Nos and Cox-2, more than HHT. These results suggest that HHT and GBT regulate the different mechanisms of action and pathways, presumably by regulating cytokine levels (IL-6, IFN-${\gamma}$), NF-${\kappa}b$ activation, and several pro-inflammatory gene expression, although both of HHT and GBT have anti-inflammatory effects.

  • Objectives : The purpose of this study is to report the effect of Korean medicine on olfactory disorders due to chronic sinusitis. Methods : Two patients with chronic sinusitis complaining of olfactory disturbances, nasal obstruction, and purulent discharge were treated with herbal medicines and acupuncture treatment. The nasal endoscopic examination and paranasal sinuses x-ray series (PNS x-ray) were performed. In addition, numeral rating scale (NRS) of olfactory disturbances and degrees of nasal obstruction and purulent discharge were evaluated. Results : In cases 1 and 2, NRS of olfactory disturbances decreased from 9 to 1. Nasal obstruction and purulent discharge decreased to no symptoms or intermittent level at moderate or severe level. The endoscopic findings and PNS x-ray findings were improved after treatments. Among various treatments of Korean medicine, Gamitonggyu-tang may play a crucial role in the reduction of sinusitis and olfactory disorders. Conclusions : This case report can be used as a basis for the use of Korean medicine, Gamitonggyu-tang, for olfactory disorder due to chronic sinusitis.

  • Sipjeondaebo-tang has been traditionally prescribed a medicine as a restorative. In this study, we investigated the acute toxicity about water-extracted Sipjeondaebo-tang. Thirty rats completed 14 days of oral Sipjeondaebo-tang at the respective doses of 0(control group), 2000 and 5000 mg/kg. We observed survival rates, general toxicity, change of body weight and autopsy. To be confirmed the data for the toxicity and safety problems of oriental medicine prescription. Compared with the control group, we could not find any toxic alteration in all treated groups (2000 and 5000 mg/kg). LD50 of Sipjeondaebo-tang was over 5000 mg/kg and it is very safe to SD rats.

  • This study was carried out to investigate the protective effects of GaMiHyangSaYukGunJaTang on gastric mucosal lesions caused by indomethacin in rats. The test group was injected with indomethacin after the oral administration of GaMiHyangSaYukGunJaTang, while the control group was injected only with indomethacin. The lipid peroxidation level increased considerably in the control group, but superoxide disnurtase(SOD), catalase and glutathione(GSH) levels remarkably decreased. The following effects induced by indomethacin were observed in the stomach of the control group; mucosa hemoIThagic infarct, mucosa cell necrosis, leukocyte appearance, mucosa hemorrhagic erosion and gastric pit disawearance. In the test group, lipid peroxides level was significantly reduced compared with the control group While SOD, catalase and GSH levels considerably increased. The following effects were also observed in the stomach of the test group; mucosa hemorrhagic infarct, mucosa cell necrosis, leukocyte appearance and mucosa hemorrhagic erosion occured far less and gastric pit disappearance was not observed at all. This results suggest that GaMiHyangSaYukGunJaTang is effective in the remedy of gastric mucosal lesions as GaMiHyangSaYukGunJaTang suppresses the production of the lipid peroxides and free radicals induced by indomethacin and activates SOD, catalase and GSH.

  • Ginseng is a perennial herb widely used in China, Japan, and Korea. It is also one of the most commonly used herbal medicines in the U.S. Although it is generally considered safe to use, adverse effects associated with ginseng use have been reported. Inappropriate ginseng use, such as high dose administration, may cause insomnia, headaches, diarrhea, as well as cardiovascular and endocrine disorders. Other factors that may contribute to adverse effects of ginseng include the variety of ginseng species, variability in commercial ginseng preparations, and potential ginseng-drug interactions. To minimize adverse effects of ginseng, consumers should be advised to use it appropriately, and the herbal industry should try to provide standardized ginseng preparations.

  • Resibufogenin is a single compound isolated from the skin venom gland of the toad (Bufo bufo gargarizans cantor). Formulations containing toad venom have been widely used as complementary and alternative medicines. However, like digitalis, resibufogenin possesses both pharmacological and toxicological activities. Our previous data indicated that resibufogenin induces electro-toxicity, including delayed afterdepolarization and triggered arrhythmias at high concentration, both in cardiac fiber in vitro and in beating heart in vivo.
