  • Streptococcal diseases are known as serious problems in fresh water and marine fish culture industry worldwide. Recently, the importance of the infection of Streptococcus parauberis has been increased among the streptococcicosis since severe outbreaks in cultured olive flounder (Paralichthys olivaceus) were recorded in Korea. The aims of the present study were to investigate immunomodulatory differences between S. parauberis and S. iniae. In this study, S. parauberis isolated from diseased olive flounder was investigated on the characteristics of morphological and immune responses. Immune response of the fish to the pathogen was characterized by bactericidal activity in the serum, phagocytic activity and reactive oxygen intermediaters (ROIs) production of olive flounder. The capsules of both bacteria, S. parauberis and S. iniae seem to be associated with the ability to resist killing activity of normal sera. Also, capsulated strains could survive in the phagocytes and induce to lower ROIs production.

  • 알로에가 in vitro에서 넙치의 비특이적 면역반응에 미치는 영향을 조사하기 위하여 신장으로부터 분리된 백혈구의 호흡폭발을 chemiluminescent(CL) 반응을 이용하여 측정하였다. 알로에 분말을 0.001, 0.01, 0.1, 0.5, 1.0, $10.0{\mu}g/ml$의 농도로 MEM에 희석하여 넙치의 식세포와 함께 배양한 후 3, 6, 12, 24, 48시간째에 CL 반응을 이용하여 호흡폭발능을 측정한 결과, 알로에 $0.5{\mu}g/ml$$1.0{\mu}g/ml$의 농도에서 24시간 동안 배양한 실험구만이 알로에를 첨가하지 않은 대조구에 비해서 유의성 있게 호흡폭발능이 증가하였다. 이러한 결과는 알로에가 양식 넙치의 비특이적 면역능을 증강시킬 수 있음을 시사하며, 앞으로 in vivo 실험을 통한 검증이 필요할 것으로 사료된다.

Propagation of lymphocystis disease virus (LCDV) in the FIN cell line originated from olive flounder Paralichthys olivaceus fin

  • The present study demonstrated lymphocystis disease virus (LCDV) propagation through cytopathic effects (CPE) formation and LCDV detection in olive flounder fin (FFN) cells by polymerase chain reaction (PCR) and fluorescent antibody technique (FAT) methods. Tissue filtrates from the cluster cells produced CPE in FFN cells, which initially cells became enlarged and gradually underwent fusion en masse. Infectivity of culture grown LCDV using the FFN cells reached $10^{2.3}$ $TCID_{50}$/ml at 4 days post infection and the highest titer was measured $10^{6.5}$ $TCID_{50}$/ml at 12 days. The viral DNA was detected in the cell culture supernatants showing CPE and the CPE cells by PCR. Antigen specific strong fluorescence reacting with monoclonal antibody against the virus revealed the presence of viral antigen in the cytoplasm of infected FFN cells. These results suggest that the FFN cell line originated from the olive flounder has a susceptibility of the LCDV.

급격한 수온변화에 따른 양식 넙치(Paralichthys olivaceus)의 생리적 반응 (Physiological Response of the Cultured Olive Flounder (Paralichthys olivaceus) to the Sharp changes of Water Temperature)

  • The effects of sparp changes of water temperature (WT) on the stree response and physiological change of the cultured olive flounder (Paralichthys olivaceus) were examined by manipulating WT (3 patterns) in a running seawater culture system. In the first group (Exp. I), the WT was decreased from 18$^{\circ}C$ to 11$^{\circ}C$ within 6 hours and increased back to the original WT quickly. WT was decreased from 2$0^{\circ}C$ to 11$^{\circ}C$ within 5 hours and main-tained at 11$^{\circ}C$ for 10 hours. and then increased to 2$0^{\circ}C$ in the second group (Exp. II). In the third group(Exp. III) WT was decreased to 11$^{\circ}C$ within 5 hours (type A) or 10 hrs. (type B). In Exp. I and III, the level of serum cortisol was increased from 2.5$\pm$0.3 ng/ml and 2.6$\pm$0.9 ng/ml to 13.6$\pm$3.0 ng/ml and 12.4$\pm$3.2ng/ml, respectively, with WT decrease. However, no consistent tendency in the change of serum glucose level was shown according to WT decrease. In Exp. III, the glucose level of fish in type A was decreased until 5 hours and increased at 7 hours, then decreased until 12 hours where as the glucose level in type B was decreased until 5 hours and stayed at the level of 15.7 mg/dl. The serum osmolality was reduced with WT decrease and the response of serum electrolytes in this experiment conflicted, and a tendency in total protein, AST and ALT was not found following WT decrease. In conclusion, olive flounder responded to the stress caused by WT decrease and acclimated to this condition when the lower temperature was maintained. But there was no stress response in the blood of olive flounder when WT was increased. On the other hand, the degree of stress response in olive flounder was various according to the range and gradient of WT change.

Field experiment on effect of butaphosphan and cyanocobalamin complex on the immunity and stress of olive flounder at low temperature

  • In this study, a fish metabolic accelerator (a combination of butaphosphan and cyanocobalamin [BPC]) was injected into the muscle of the olive flounder, Paralichthys olivaceus, to investigate its effect on immunity and stress in fish maintained at low temperatures. A single dose of BPC was injected (100 mg/kg body weight) into the olive flounder, and its immunity and stress were observed after one and two weeks. Immunity tests revealed the presence of lysozyme (LZM), nitroblue tetrazolium (NBT), myeloperoxidase (MPO), anti-protease (AP), glutathione peroxidase (GPx), and total immunoglobulin (TIg). BPC injection was found to increase immunity activity compared to the control group. In particular, there was significantly high GPx activity. There was similarly high activity for MPO and GPx in the first week following the injection, followed by significant differences between the BPC-injected and control groups in the second week. There was a reduced low water-temperature stress response in the BPC-injected fish, as evidenced by the cortisol and glucose levels of the control and BPC groups. Lower levels were also observed in the BPC group than the control group during the second week. Cortisol levels were significantly lower in the BPC group than the control group. Histological examinations were conducted in the first and second weeks after the intramuscular injection of the recommended BPC dose to confirm the safety of BPC in aquaculture. There were no abnormalities observed in any tissue samples. This study confirms that the injection of BPC is safe even when used in a culture situation. BPC helps relieve stress and improves non-specific immune responses (innate immunity) in the olive flounder.

Nutrient Availability and Growth Rate Associated with Three Different Feed Types Used by Olive Flounder, Paralichthys olivaceus, Farmers in Korea

  • We estimated the digestibility and growth rate of juvenile olive flounder (Paralichthys olivaceus) on three diets: raw fish-based moist pellets, moist pellets, and extruded pellets. The diets were created using commercially available methods, and a basic formulated powder. A reference diet was used to compare feed digestibility and the fish growth rate achieved using the experimental diets. The apparent digestibility coefficients (ADCs) of protein for raw fish-based moist pellets and moist pellets were significantly higher (P<0.05) than those for extruded pellets and the reference diet. The ADCs of nitrogen-free extracts (NFE) of extruded pellets and moist pellets were significantly higher (P<0.05) than those for raw fish-based moist pellets and the reference diet. Fish that were fed with the raw fish-based moist pellets showed the greatest weight gain (452.4%), which was not significantly different from that of fish fed with extruded pellets (414.4%; P>0.05). These results indicate that the higher protein efficiency in fish fed with extruded pellets can result in good growth performance within flounder culture systems.

Effect of Recombinant Bovine Somatotropin on Growth of Olive Flounder, Paralichthys of olivaceus

  • The recombinant bovine somatotropin (rBST) was administered to the oliver flounder, Paralichthys olivaceus, to know the effects and optimal administration frequency and dosage of the hormone. The experiment was conducted with three different treatment groups (A, B and C) designated based on the duration and administration frequency of rBST and one control (D) from April 14, 1996 to March 16, 1997. The fish of hormone treated groups grew 7.86 to 10.07% (47.45 to 60.75 g in weight) better than the control at the end of the experiment (P<0.05). No significant differences in their growth were detected among treatment groups. The distinct growth improvement was recognized four weeks after completion of the first four hormone administration. When considering water temperatures measured from the experimental tanks, the effect of rBST on the flounder was greater during the period showing relatively lower temperature. The survival rates were higher in treatment groups than in the control, revealing 98.3% (A), 98.4% (B), 97.7% (C) and 93.1% (D) during the first stage of culture; 92.7% (A), 91.3% (B), 86.7% (C) and 80.0% (D) during the second stage of culture.

한약재 추출물에 배양된 유산균 배양액이 양식어류의 사료첨가제로서 성장과 비 특이적 면역에 미치는 영향 (Effects of the Culture Broth of Lactic Acid Bacteria Cultured in Herb Extracts on Growth Promotion and Nonspecific Immune Responses of Aquacultured Fish)

  • 양식 어류의 항생제 투여의 문제점을 해결하기 위하여 한 약재 추출물을 기질로 한 유산균 배양액을 양식 넙치와 돌돔의 사료에 첨가하여 성장, 혈액학적 분석, 식세포 활성, lysozyme 활성 및 어류질병세균에 대한 공격실험을 실시하였다. 성장의 결과 넙치는 실험구와 대조구강의 유의성을 보이지 않았으며, 돌돔의 경우 대조구에 비해 실험구가 약 20 g 높은 어체중 증가를 보였다. 넙치의 사료효율은 실행구가 대조구에 비해 약 25% 높았으며, 돌돔은 사료효율은 두 그룹 간에 유의성을 보이지 않았다. 어류의 혈액학적 분석 결과, GOT와 GPT는 8주 이후에 감소현상을 보였으나 glucose와 total protein은 두 그룹 간에 유의성을 보이지 않았다. 식세포 활성 또한 유의적인 차이가 없었다. lysozyme 활성은 넙치의 경우 8주부터 실험구가 대조구보다 높은 활성을 보였으며, 돌돔은 실험 12주에 대조구보다 약 3배 높은 활성을 보였다. 어류 질병 유발세균에 대한 공격실험은 넙치의 복강에 V. anguillarum을 투여하였을 시, 실험구가 대조구에 비해 18%, S. iniae을 투여하였을 시, 실험구가 23% 높은 생존율을 보였다. 돌돔의 경우, V. anguillarum의 투여경과 대조구의 비해 17%, S. iniae의 투여경과 대조구의 비해 16% 높은 생존율을 나타냈다.

  • This study was performed to determine effects of the supplementation of Kimchi lactic acid bacterial culture in extruded pellets (EP) on the growth, body composition, blood chemistry and immune response of olive flounder Paralichthys olivaceus. Four hundred eighty individuals averaging 16.1 g were randomly distributed into 12, 180 L flow-through tanks (forty fish per tank). Four concentrations of Kimchi lactic acid bacterial culture (KL) were prepared: Control (0%), 0.1%, 0.2% and 0.5%. Three concentrations (0.1%, 0.2% and 0.5%) of Kimchi lactic acid bacterial culture were each diluted to 10% of EP weight and then fully absorbed by EP for 10 minutes. Each diet was fed to triplicate groups of fish. Fish were hand-fed to apparent satiation twice a day for 8 weeks. At the end of the 8-week feeding trial, the plasma lysozyme and bacterial activities of fish were determined. In addition, the cumulative mortality of fish was monitored for 8 days after their artificial infection with Edwardsiella tarda. The weight gain, specific growth rate, feed efficiency ratio, protein efficiency ratio, protein retention, hepatosomatic index and condition factor of fish were not affected by dietary supplementation with KL. None of the proximate composition, plasma parameters, lysozyme or bactericidal activities of fish was affected by dietary supplementation with KL. However, the cumulative mortalities of fish fed EP containing 0.1% and 0.5% Kimchi lactic acid bacterial culture were relatively low compared to that of fish fed the control diet. In conclusion, dietary supplementation with KL did not effectively improve growth, feed utilization, body composition, plasma chemistry, lysozyme, bactericidal activities or immune response of olive flounder after E. tarda infection under these experimental conditions.

Neoparamoeba sp. Infection on Gills of Olive Flounder, Paralichthys olivaceus in Korea

  • Amoebic gill disease of flounder, Paralichthys olivaceus was diagnosed at commerical culture facility in South Korea. The amoeba was identified as a species of the genus Neoparamoeba based on the morpholgical characteristics of trophozoites. Transmisson electron microscopy revealed the presence of a symbiotic organism, parasome in the cytoplasm and dense glycocalyx on the surface of the trophozoites. They lacked the boat-shaped microscales on the surface and contained numerous vacuoles and channels, mitochondria in the cytoplasm. Colonization of amoebae on gill tissue elicited extensive fusion and hyperplasia of gill lamella.
