본 연구에서는 휴대전화 이용자의 시간대별 기지국 위치정보를 기반으로 하여 이용자의 이동경로를 추적하고, 기/종점을 추출하여 OD를 구축하였다. OD 구축시 통행거리가 길어질수록 통신횟수가 많아짐을 고려하여 통행거리별 평균통신횟수를 산출/반영하였고, 가정기반 통행만을 대상으로 수도권 중심의 OD를 구축하였다. 셀 단위로 집계된 휴대전화 자료를 법정동 단위로 변환하고, 이를 행정동 단위로 변환하여 본 연구에서 구축한 OD를 KTDB OD와 비교분석 한 결과, 시/군/구 단위 OD와 동 단위 OD의 상관계수는 각각 0.98, 0.85로 나타나 상당한 연관성이 있는 것으로 분석되었다.
본 연구에서는 Bacillus 유래 esterase를 생산할 수 있는 재조합 대장균을 사용하여 유가식 배양을 이용한 고농도 균체 배양을 통해 esterase 생산성을 극대화하고자 하였다. 유가식 배양 중 순수 산소의 공급을 통해 용존산소를 30% 이상 유지한 경우와 포도당농도를 1 g/l 이상 유지한 경우 각각 $OD_{600}$ 76 (35.8 g/l DCW)과 $OD_{600}$ 90 (42.4 g/l DCW)까지 균체량을 증가시킬 수 있었다. 포도당의 공급에도 불구하고 배양 후반에 세포의 성장이 정체되는 현상을 극복하기 위해 yeast extract가 강화된 추가 배지의 공급을 시도하였으며, 그 결과 $OD_{600}$ 185 (87.3 g/l DCW)까지 고농도 균체 배양이 가능함을 확인하였다. 단백질 생산 수율의 향상을 위해 성장 시기에 따라 induction에 의한 세포 성장과 esterase 생산성을 평가하였고, 그 결과 대수 성장기 후반에 induction을 유도한 경우 세포 성장 측면에서는 최대 $OD_{600}$ 190(89 g/l DCW)까지 고농도 균체 배양이 가능함을 확인하였다. Esterase 생산성 측면에서는 대수 성장기 초반에 induction 을 유도한 경우에 비해 최대 5.8배 생산성이 증가됨을 확인할 수 있었다. 따라서 본 연구를 통해 순수산소와 질소원의 공급을 통해 확립된 대장균 고밀도 배양방법을 기초로 IPTG 유도시간을 최적화 함으로써 Bacillus 유래 esterase의 최대 생산성을 확보할 수 있는 배양방법을 확립하였다.
항균력과 보습력을 동시에 가지고 있는 화장품용 첨가제인 1, 2-octanediol (OD)의 피부 문제점을 개선하기 위하여 galactoside 유도체인 1, 2-octanediol galactoside (OD-gal)를 대장균 β-galactosidase (β-gal)을 이용하여 합성하였다. 이 때, β-gal은 4.5 U/ml, OD 농도는 150 mM, pH는 7.0, 그리고, 온도는 37℃가 OD로부터 OD-gal을 합성하는 최적 조건이었다. 이 조건에 24 시간 동안 150 mM의 OD로부터 약 55.9 mM의 OD-gal이 합성되었고, 이 때, conversion 수율(mole 기준)은 약 37.2% 였다. 또한, 9 ml의 반응액에서 67.4 mg의 순수한 OD-gal을 정제할 수 있었으며, 반응액에 들어 있는 OD에서부터 정제를 포함하는 전체 합성수율은 weight 기준으로는 약 34.1% 이고, mole 기준으로는 약 16.2% 정도였다. 이러한 연구결과는 보다 안전한 화장품용 첨가제로서 OD-gal의 산업화에 기초자료로서 도움을 줄 것으로 생각된다.
An experiment was conducted to quantify the flow of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD), and to investigate diurnal pattern in SNAN flow in OD. Five ruminally cannulated Finnish-Ayrshire dairy cows in a $5{\times}5$ Latin square design consumed a basal diet of grass silage and barley grain, and that supplemented with four protein feeds (kg/d DM basis) as follows: skimmed milk powder (2.1), wet distiller' solubles (3.0), untreated rapeseed meal (2.1) and treated rapeseed meal (2.1). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 1.0 h interval during a 12 h feeding cycle. Both RD and OD were acidified, centrifuged to remove microbes and precipitated with trichloroacetic acid followed by centrifugation. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using ninhydrin assay. Free AA, peptide and soluble protein averaged 60.0, 89.4 and 2.1 g/d, respectively, for RD, and 81.8, 121.5 and 2.5 g/d, respectively, for OD. Although free AA flow was relatively high, mean peptide flow was quantitatively the most important fraction of SNAN, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis. Diurnal pattern in flow of peptide including free AA in OD during a 12 h feeding cycle peaked 1 h post-feeding, decreased by 3 h post-feeding and was relatively constant thereafter. Protein supplementation showed higher flow of peptide including free AA immediately after feeding compared with no supplemented diet. There were no differences among protein supplements in diurnal pattern in flow of peptide including free AA in OD.
Cultivation of Spirulina platensis using ammonium salts or wastewater containing ammonium as alternative nitrogen sources is considered as a commercial way to reduce the production cost. In this research, by analyzing the relationship between biomass production and ammonium-N consumption in the fed-batch culture of Spirulina platensis using ammonium bicarbonate as a nitrogen nutrient source, an online adaptive control strategy based on optical density (OD) measurements for controlling ammonium feeding was presented. The ammonium concentration was successfully controlled between the cell growth inhibitory and limiting concentrations using this OD-based feedback feeding method. As a result, the maximum biomass concentration (2.98 g/l), productivity (0.237 g/l d), nitrogen-to-cell conversion factor (7.32 gX/gN), and contents of protein (64.1%) and chlorophyll (13.4mg/g) obtained by using the OD-based feedback feeding method were higher than those using the constant and variable feeding methods. The OD-based feedback feeding method could be recognized as an applicable way to control ammonium feeding and a benefit for Spirulina platensis cultivations.
An experiment was conducted to study the effect of soluble protein supplements on concentration of soluble non-ammonia nitrogen (SNAN) in the liquid phase of ruminal (RD) and omasal digesta (OD) of Korean native steers, and to investigate diurnal pattern in SNAN concentration in RD and OD. Three ruminally cannulated Korean native steers in a $3{\times}3$ Latin square design consumed a basal diet of rice straw and corn-based concentrate (control), and that supplemented (kg/d DM basis) with intact casein (0.24; IC) or acid hydrolyzed casein (0.46; AHC). Ruminal digesta was sampled using a vacuum pump, whereas OD was collected using an omasal sampling system at 2.0 h intervals after a morning feeding. The SNAN fractions (free amino acid (AA), peptide and soluble protein) in RD and OD were assessed using the ninhydrin assay. Concentrations of free AA and total SNAN in RD were significantly (p<0.05) lower than those in OD. Although free AA concentration was relatively high, mean peptide was quantitatively the most important fraction of total SNAN in both RD and OD, indicating that degradation of peptide to AA rather than hydrolysis of soluble protein to peptide or deamination may be the most limiting step in rumen proteolysis of Korean native steers. Diurnal variation in peptide concentration in OD for the soluble protein supplemented diets during the feeding cycle peaked 2 h post-feeding and decreased thereafter whereas that for the control was relatively constant during the entire feeding cycle. Diurnal variation in peptide concentration was rather similar between RD and OD.
한국교통연구원에서 2010년 가구통행실태조사 자료를 기초로 구축한 신규 KTDB 여객자료는 대도시권 모두에 대해 PA개념을 기반으로 통행생성과 통행유인의 통행발생량과 교통존 간의 통행량 자료를 처음으로 제공하였다. 따라서, 신규 KTDB를 활용한 장래 수요예측의 분석방법은 변화된 자료형태에 적합한 PA개념의 분석방법이 적용되어야 한다. 본 연구에서는 교통정책 분석 시 반영하게 되는 장래 개발사업에 대한 통행발생량 예측과 통행분포패턴 예측 분석에 있어 PA개념의 분석 절차를 정형화할 수 있는 방법을 명확하게 제시하고, 또한 과거의 OD기반의 분석방법이 적용될 경우 그 분석결과가 PA기반의 분석방법의 결과와 다르게 나올 수 있음을 단순 예제를 통해 증명하였다. 이와 같은 분석결과의 차이는 교통정책의 의사결정에 있어 신규 KTDB 여객자료를 활용하면서 과거의 OD기반의 분석방법이 적용될 경우 정책결정에 왜곡을 가져올 수 있음을 의미하는 것이므로, 신규 자료에 대해 적합한 분석방법이 적용되어야 함을 본 연구는 강조하였다. 또한 본 연구는 신규 KTDB 여객자료에 PA기반 분석방법이 올바로 응용 적용될 수 있도록 조속히 실무분석가들에게 분석방법 지침과 기술 보급이 필요함을 주장하였다.
This study was carried out to survey the effect of Jujube extract on the growth of 3 strains of lactic acid bacteria(LBA) starter cultures in the MRS broth by the addition of 0, 0.1, 0.5, 1 and 2% extract The pH, titratable acidity and OD of LAB were investigated in order to get fundamental knowledge for the development a new product. The effects of Jujube extract on the growth of LAB were variable depending upon the LAB species and concentrations of Jujube extract significantly (p<0.05). In the results, Jujube extract enhanced the acid production and propagation by the 3 LAB strains with increasing concentration of Jujube extracts until 2.0% added was to the MRS broth medium (p<0.05). Addition of Jujube extract markedly stimulated the acid production and propagation of Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium longum.
Chaperone 분자는 세포 내에서 새로 합성된 polypeptides의 misfolding을 보호하는 역할을 가진다. 이런 chaperone 분자와의 공발현은 활성형 재조합 단백질의 생산을 증가를 기대할 수 있다. 본 연구에서는 E. cozi에서 B. macerans 유래 cyclodextrin glucanotransferase (CGTase)의 활성형 생산에 GroEL/ES chaperone과의 공발현의 효과에 대해 조사하였다. cgt와 groEL/ES 유전자출 발현하는 pTCGT1과 pGro7은 각각 T7 promoter와 araB promoter에 의해 조절되고 이들을 E. coli cell에 co-transformation시켰다. 재조합 E. coli에서 IPTG와 L-arabinose의 최적 농도를 결정하기 위해 행한 결과 1 mM IPTG, 0.3 mg L-arabinose/$m\ell$에서 가장 높은 CGTase 활성을 나타내었다. 그리고 tube에서는 L-arabinose와 IPTG를 각각 0.4~0.5 $OD_{600}$과 0.8~l.0 $OD_{600}$에서 첨가하였을 때 활성형 CGTase의 생산이 증가되었다. GroEL/ES 공발현 조건에서는 가용성 CGTase 활성이 0.7~0.73 unit/$m\ell$로 단독 발현의 0.36~0.56 unit/$m\ell$에 비해 약 1.5 배 정도 증가함을 알 수 있었다. SDS-PAGE 분석에서는 GroEL/ES 공발현 조건에서 총 CGTase의 33.6%정도가 가용성 형태로 생산됨을 알 수 있었다.
Lactobacillus acidophilus KFCC12731과 Saccharomyces cerevisiae KFCC32017을 두유 배지에 접종하여 산생성 조건을 검토하였다. L. acidophilus를 단독배양하는 것보다 S. cerevisiae와 혼합배양하는 것이 산생성이 많았다. 이 두 균을 대두유 배지에 혼합배양하였을 경우 최적조건은 다음과 같았다. 두유에 sucrose를 1,5% 또는 skim milk를 2.0~3.0% 첨가하고 L. acidophilus와 S. cerevisiae의 접종비율은 OD 600 값으로 3 : 7~8 : 2가 되게 접종하여 34$^{\circ}C$에서 12시간 이상 배양할 때 산생성이 최대가 되었다.
