Effect of interferon-γ on the fusion of mononuclear osteoclasts into bone-resorbing osteoclasts

  • Osteoclasts are multinucleated cells that are formed by the fusion of pre-fusion osteoclasts (pOCs). The fusion of pOCs is known to be important for osteoclastic bone resorption. Here, we examined the effect of IFN-${\gamma}$ on the fusion of pOCs. IFN-${\gamma}$ greatly increased the fusion of pOCs in a dose-dependent manner. Furthermore, IFN-${\gamma}$ induced pOC fusion even in hydroxyapatite-coated plates used as a substitute for bone. The resorption area of pOCs stimulated with IFN-${\gamma}$ was significantly higher than that of the control cells. IFN-${\gamma}$ induced the expression of dendritic cell-specific transmembrane protein (DC-STAMP), which is responsible for the fusion of pOCs. IFN-${\gamma}$ enhanced DC-STAMP expression in a dose-dependent manner. The mRNA expression of c-Fos and nuclear factor of activated T cells (NFAT) c1 was enhanced in the pOCs treated with IFN-${\gamma}$. Taken together, these results provide a new insight into the novel role of IFN-${\gamma}$ on the fusion of pOCs.

  • 목적 : 최근 들어 여러 가지 디지털 의료장비와 각종 의료정보시스템, 인터넷의 활용에 따라 다양한 종류의 의료정보와 지식이 엄청나게 증가하고 있고, 최신의 진료환경구축을 위해서 이러한 지식과 정보들을 통합 관리하려는 필요성과 요구가 대두되고 있다. 그러나 이러한 요구들은 기존의 종이로 된 차트나 필름 등의 기법에서는 물론이고 여러 가지 독립적인 전산 시스템들 <처방전달시스템(OCS), 의료영상저장전송시스템(PACS) 등>을 통해서도 충족되기 어려운 것이 현실이다. 왜냐하면 기존에 설치된 의료 정보 시스템들은 대체로 독자적인 처리방식과 내부구조를 가지고 있기 때문에 서로 다른 의료 정보 시스템간에 정보를 교환한다는 것이 사실상 매우 어렵거나 심지어는 불가능한 경우가 있기 때문이다. 그러나 이러한 의료 정보들을 최대한 유기적으로 활용하기 위해서는 이상의 여러 가지 의료 정보 시스템들을 완벽히 통합하는 것이 필수적이고 이러한 통합의료정보시스템이 비로소 진료에 있어서의 의사결정 과정에 중추적으로 활용될 수 있는 것이다. 특히 의료영상을 다루는 PACS에서는 다른 종류의 의료정보시스템인 OCS(처방전달시스템)와의 연계가 매우 중요하다. 그러나 이러한 중요성에도 불구하고 병원의 전산시스템이 서로 호환되지 않음으로 인해 OCS/PACS 통합은 PACS 도입과정 중에서 가장 어려운 부분으로 인식되어 온 것이 사실이다. 이제까지의 PACS에 있어서는 대량의 영상 데이터를 고속으로 처리하기 위한 기술적인 문제에 주로 관심을 기울였으며 기존의 정보 시스템과의 연계 또는 통합문제는 충분히 고려되지 않았다. 그 결과 PACS를 도입한 병원에서는 기대와는 달리 PACS 운용을 위한 많은 인력소모가 뒤따르거나 운용에 차질을 빚는 등 여러가지 문제가 발생하게 되었다. 따라서 이제까지의 PACS 개발이 기술적인 문제 해결을 위한 방향 접근에서 이루어졌다면, 이제부터는 기술적인 해결을 바탕으로 하여 병원내의 임상운용환경에서 그 효율성을 최대화할 수 있기 위하여 기존 OCS/PACS의 통합 연계성을 주 관심으로 하는 임상적인 접근이 이루어져야 할 시점이라고 파악된다. 이와 같은 OCS/PACS의 통합은 병원 내에서 일어나는 일련의 진료 과정들에 있어서 문자정보뿐 아니라 영상정보까지를 포함하는 모든 진료정보의 관리와 전달을 컴퓨터화 함으로써 진료효율을 비약적으로 형상 시키는 효과를 가져올 수 있으므로 궁극적으로는 모든 병원의 정보시스템이 지향하는 목표이기도 하다. 따라서 OCS/PACS의 통합을 구현하기 위해서는 기존 방식과 OCS/PACS 통합 방식의 차이점이 분석 되어져야 하며 이러한 분석을 바탕으로 차후 병원에 도입되는 시스템이 안정적이고 완성도 높은 병원 정보화 시스템으로 구축 되어져야 하므로 이에 따른 통합 시스템에 대한 분석을 하고자 한다.

  • Background : There were so many patients who are waiting for admission in Emergency room in spite of more than one hundred empty beds everyday. This study was conducted to evaluate admission-discharge module system by OCS which reduce empty beds. Methods : The data of bed utilization in general beds from 2004 were reviewed. For evaluation of performance at admission-discharge module system by OCS, the change of Occupancy of bed were calculated. Results : The percentage of Average Bed Emptiness was changed from 13.8% to 9.2%. The residents in surgery(100%) and in internal medicine(75.5%) approved this system. Conclusion : The personnel in hospital recognized that it was very important to manage bed. The management of beds by OCS was helpful to reduce empty beds and was important.

  • Objectives : To evaluate the effect of heath examination program which introduce web-based input system, OCS(Order communicating system) and interface with national health insurance corporation, this study was done. Method : To Exam/Order and tests separately at anytime, the experimental health examination program which had web-based questionnaire input system, on-line measurement input system, interface program with national health insurance corporation and introduced OCS was developed. Experiment and old examination program were compared in terms of satisfaction and cost. Result : The 95.7% of respondents were satisfied with web-based input system, the 95.4% satisfied with OCS use, and 97.8% satisfied with test at anytime they want. The cost and time reduction between experiment and old program were 4,159,000won and $600.3{\pm}5.75$(seconds) per man input-time. Conclusion : The experimental health examination program is good for employee and hospital. If the health examination program for non-employee will be developed, it will be done efficiently and effectively, but Internet securities will be considered.

  • Hospital information system has been widely used and increased recently for a variety of many aspects. And Order-Communicating system(OCS), as like as hospital information system, has been used in many medical care facilities, which is simple and easily accessible, useful system. Also then the use of OCS in emergency care center in YUMC has been introduced since 1996. 10, above 70% of availability is noticed at present and increasing in the use rate, is considered that it is very simple and accurate, time-saving, widely applicable system. So authors say that, after the use of OCS in emergency care center, interhospital exchange of the patient's information and also accomplishment of EMSS can be possible.

  • With the introduction of the computer, convenience at work may have increased but flexibility decreased along with a tendency for humans to become secondary to computers with regards to work schedules. As it exists, from entering the department of radiology to completion of imaging requires a great deal of time and difficulties in understanding the imaging process adds to patient dissatisfaction. Since clinics use RFID bars to track patient locations at comprehensive medical testing centers, departments of radiology may apply RFID and Worklist at the core of the OCS system in order to increase work efficiency and improve work flow. As a result of developing an RFID and Worklist centered OCS program, there was a vast improvement in patient satisfaction and work flow. Theses results are the basis of an RFID system that in the future may be connected to PACS and other systems with an intergrated Worklist. There is a need to realize a plan the can offer ubiquity to hospital work.

A Study of Okchun-San Formulation for Freeze Drying Capsulation (동결 건조법을 이용한 옥천산 캡슐 제형 개발에 관한 연구)

  • Objective : Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We performed to change OCS formulation for freeze drying capsulation. Methods The mixtures of OCS were extracted with water. finally, the filtered solution were evaporated and lyophilized to dry granules. The various ratio of excipients were studied to determine the formation for capsulizing. The samples were inspected for any difference in color, taste and appearance. Results: The prepared form of OCS were dried and weighted 260kg. The lyophilized dry powder yielding 40kg. The suitable ratio of OCS-dry powder and excipient was 10:1. The average weights of On and placebo capsules were $440{\pm}5.28mg$, $465{\pm}7.95mg$, respectively. There was no notable change in color and appearance for both capsuled samples throughout the study period. Conclusions: Therefore in can be concluded that freeze drying capsulation is appropriate form of OCS.

Performance and milk composition of dairy goats as affected by the dietary level of stoned olive cake silages

  • Objective: The current study compared the effects of dietary levels of two phase stoned olive cake (OC) in form of silage (OCS) on milk production and quality of Saanen goats. Methods: The OCS included in total mixed ration (TMR) at dry matter proportions of 0.0 (OC0), 0.10 (OC10), and 0.20 (OC20). The TMR were fed to a total of 18 goats in a completely randomized design for a period of 5 weeks. Results: Dietary treatments had no effect on the milk yield of Saanen goats, but the daily milk fat production was greater (p<0.05) at feeding OC20. The total phenolic (TP) compounds contents increased (p<0.01) in each increment of OCS in TMR and this was also reflected in the TP contents of milk. The C8:0, C10:0, C12:0, and C14:0 saturated fatty acids (FAs) in milk fat decreased (p<0.01) with increasing dietary level of OCS, but the decrease (p<0.001) in C16:0 and the increase (p<0.01) in C18:0 in milk fat occurred similarly at each inclusion level of OCS. Only OC20 reduced (p<0.05) the total saturated FA, yet the reduction (p<0.01) in n6/n3 ratio and atherogenicity index occurred in both OC10 and OC20. Conclusion: Two phase stoned OCS increases milk quality not only through modifying the milk FA composition, but also by increasing the milk TP content. These favorable changes in milk quality are closely associated with the dietary level of OCS.

  • These days, most of control system requires automazation in the all fields. Therefore OCS which can simplify control system integration is appearing. In this study, OCS consists of HMI, OS, Controller, and Communication network. (SFC and visual C++ are used as HMI, window NT is used as OS, pentium PC is used as Controller, and industrial Ethernet is used as Communication network.) Finally, We applied OCS to Level control system and Temperature control system, and verified its efficiency.

  • A wide variety of drugs and endogenous bioactive amines are organic cations (OCs). Approximately 40% of all conventional drugs on the market are OCs. Thus, the transport of xenobiotics or endogenous OCs in the body has been a subject of considerable interest, since the discovery and cloning of a family of OC transporters, referred to as organic cation transporter (OCTs), and a new subfamily of OCTs, OCTNs, leading to the functional characterization of these transporters in various systems including oocytes and some cell lines. Organic cation transporters are critical in drug absorption, targeting, and disposition of a drug. In this review, the recent advances in the characterization of organic cation transporters and their distribution in the small intestine are discussed. The results of the in vitro transport studies of various OCs in the small intestine using techniques such as isolated brush-border membrane vesicles, Ussing chamber systems and Caco-2 cells are discussed, and in vivo knock-out animal studies are summarized. Such information is essential for predicting pharmacokinetics and pharmacodynamics and in the design and development of new cationic drugs. An understanding of the mechanisms that control the intestinal transport of OCs will clearly aid achieving desirable clinical outcomes.
