COMPARISON OF KEDI-WISC AND BGT PERFORMANCE BETWEEN THE ASPERGER' DISORDER AND PDD NOS CHILDREN (아스퍼거장애와 비전형 자폐장애 아동의 KEDI-WISC와 BGT 수행의 비교)

  • Objectives:This study was conducted to compare the cognitive characteristics and visual-motor coordination ability of children with Asperger’s disorder and with those of children with PDD NOS. Methods:27 children(13 in AS group and 14 in PDD NOS group) were individually assessed using the K-WISC and BGT, and the results of those tests were analyzed. Results:The mean FSIQ of the AS group was significantly higher than that of the PDD NOS group. There was also a large discrepancy between VIQ and PIQ in the PDD NOS, while there was not significant discrepancy in the AS. The AS was distinguished from PDD NOS group by significantly higher scores in Vocabulary and Comprehension subscales and lower score in Block design. Also, when compared with the PDD NOS, the AS showed more difficulties in visual-motor coordination. Conclusion:The AS showed relatively good verbal and learning ability, while the PDD NOS relatively superior ability in visuospatial function and visual-motor coordination. The findings indicated that the K-WISC and BGT might be useful assessment tool to differentiate the AS from PDD NOS.

Anti-inflammatory and Radical Scavenging Effects of Spirodela polyrrhiza

  • Anti-inflammatory and anti-oxidant effect of Spirodela polyrrhiza (Lemnaceae), a widely used traditional medicinal plant were investigated. In macrophages nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. 85% MeOH extracts of S. polyrrhiza (0.01, 0.1, 1 mg/mL) suppressed nitric oxide production in interferone-$\gamma$ (IFN-$\gamma$) and lipopoloysaccharide(LPS)-stimulated macrophages. It also attenuated the expression of inflammatory enzymes like inducible NOS (iNOS) and cyclooxygenase-2(COX-2) as assessed by immunoblotting with specific antibodies. Moreover, the values obtained with DPPH radical, superoxide anion and NO radical scavenging assay showed that S. polyrrhiza has potent antioxidant properties as a natural ROS scavenger. The results of the present study suggest the potential use of S. polyrrhiza in the treatment of ROS-mediated chronic inflammatory diseases such as atherosclerosis and rheumatoid arthritis.

  • Nitric oxide (NO) is a newly described transmitter involved with cell to cell communication that is generated in biologic tissues by specific types of nitric oxide synthase (NOS), which metabolize L-arginine and molecular oxygen to citrulline and nitric oxide. In the skin. NO has been reported to play an important role in such diseases as psoriasis, atopic dermatitis, and contact dermatitis, as well as act as an important modulator in UVB-induced erythema. Ultraviolet B irradiation to the skin evokes an increase in NO production in the epidermis through two pathways; induction of inducible NOS, mediated by inflammatory cytokines, and elevation of constitutive neuronal NOS activity. In a cell culture system, it has been demonstrated that NO functions as a melanogen after being produced in keratinocytes in response to UVB-irradiation. NO-stimulated melanogenesis in melanocytes is mediated by the cGMP/PKG pathway. In this study, up-regulation of tyrosinase gene expression by NO-stimulation and the involvement of NO in UVB-induced pigmentation were examined. In NO-induced melanogenesis, protein synthesis and tyrosinase activity increased along with an up-regulation of tyrosinase gene expression. In an animal model, UVB-induced pigmentation in skin was suppressed by sequential daily treatments with a specific inhibitor of NOS. Thus, NO plays an important role in UVB-induced pigmentation, where its function as a melanogen is considered to be one of the mechanisms. Together with its role in the development of erythema, NO contributes to the total protective response of skin against UVB-irradiation.

A Proposal of Inclusive Framework of the Nature of Science (NOS) Based on the 4 Themes of Scientific Literacy for K-12 School Science

  • Although many researchers and science educators agree that understanding of the nature of science is essential in order for students to be a scientific literate person, it is not easy to compromise exactly what kind of understanding of the nature of science is required to achieve this goal (Smith & Scharmann, 1999). This study discusses a theoretical background about the nature of science (NOS) including some consensus views of the NOS that several important U.S. educational reform documents and science researchers have presented over the past several decades. Finally, this study proposes an inclusive framework of the nature of science based on the four categories of scientific literacy, which are (1) science as a body of knowledge, (2) science as a way of investigating, (3) science as a way of thinking, and (4) the interaction of science, technology, and society. Each category of the framework includes several statements about the nature of science to describe each theme of the NOS. This framework is comprehensive and inclusive because it is suggested by examining several major U.S. national-level documents and in the publications of science education researchers presented about the nature of science. Significantly, many of the key ideas were added into category (4) and category (3), which indicates that the current literature stresses the relationship among science, technology, and society as well as the work of scientists.

The effects of Hyunggaeyungyo-tang of suppression of iNOS production on RAW 264.7cell (Mouse cell에서 형개연교탕(荊芥連翹湯)의 iNOS 생성 억제 효과)

  • Background and Objectives : The aim of this study was to investigate anti-inflammatory and anti-oxidant effects of Hyunggaeyungyo-tang(HYT) on RAW 264.7 cells. Material and Methods : Two types of experiments were implemented for this study: first, the experiment to study the anti-oxidant effect of HYT using riboflavin; second, in vitro experiment to investigate the suppression of NF-${\kappa}$B activation using RAW 264.7 cells (I${\kappa}$B kinase and induce nitric oxide synthase mRNA expression) Results : 1. The anti-oxidant effects of HYT was dose-dependantly increased. 2. The RAW 264.7cells were treated with LPS for 1 hours prior to the addition of indicated concentrations(0.4,-1.0mg/$m\ell$) of HYT, and the cells were further incubated for 24 hours. The LPS-induced IKK & iNOS mRNA expression were dose-dependantly decreased in HYT treated RAW 264.7cells. Conclusion : The results suggest that HYT is significantly effective in the treatment of inflammation through the suppression of NF-${\kappa}$B activation and iNOS production.

Inhibition of Nitric Oxide Production by the Extracts of Hibiscus manihot (황촉규 추출물의 Nitric Oxide 생성 저해활성)

  • Anti-inflammatory activity of the extracts of Hibiscus manihot was investigated through the evaluation of its inhibitory effect on the production of inflammatory biomarkers (i-NOS, COX-2) in RAW264.7 murine macrophage cells. Among the sequential solvent fractions (hexane, dichloromethane, ethyl acetate, n-butanol and water), the fractions of dichloromethane (1 ${\mu}g/ml$) and ethyl acetate (5 ${\mu}g/ml$) showed potential inhibitory activities on i-NOS and COX-2 activity in RAW264.7 cells. These results suggest that Hibiscus manihot might have an anti-inflammatory activity through the suppression of inflammatory markers.

  • Objectives : This study was aimed to verify the cytoprotective function, antioxidative effect and inflammation genes inhibitory effects of Lycium chinense Milie. Therefore the generation of superoxide anion radical ( $O_2\;^-$), peroxynitrite ($ONOO^-$), nitric oxide (NO) and prostaglandin $E_2$ $(PGE_2)$ was investigated in the renal epithelial cells of mouse. Effects of Lycium chinense Milie on the expression of inflammation-related proteins, $IKK-{\alpha}$. $p-IKK-\alpha\beta$, $p-I{\kappa}B-\alpha$, $NF-{\kappa}B$ (p50, p65), COX-2 and iNOS, were examined by western blotting. Methods : For this study, the fluorescent probes were used, namely dihydrorhodamine 123 (DHR 123), 4.5-diaminofluorescein (DAF-2) and 2',7'-dichlorodihydrofluorescein diacetate (DCFDA). Western blotting was performed using anti-$IKK-\alpha$, anti-phospho $IKK-\alpha\beta$, anti-phospho $I{\kappa}B-\alpha$, anti-$NF-{\kappa}B$ (p50, p65), anti-COX-2 and anti-iNOS, respectively. Results : Lyciutn chinense Milie reduced $H_{2}O_{2}$-induced cell death dose-dependently. It inhibited the generation of $O_2\;^-$, $ONOO^-$, NO and $PGE_2$ in the $H_{2}O_{2}$-treated renal epithelial cells of mouse in vitro. Lycium chinense Milie inhibited the expression of $IKK-\alpha$, $p-IKK-\alpha\beta,\;p-I{\kappa}B-\alpha$, COX-2 and iNOS genes by means of decreasing activation of $NF-{\kappa}B$. Conclusions : According to above results. Lycium chinense Milie recommended to be applied in treatment for the inflammatory process and inflammation-related diseases.

The Study on Possibility of Applying Word-Level Word Embedding Model of Literature Related to NOS -Focus on Qualitative Performance Evaluation- (과학의 본성 관련 문헌들의 단어수준 워드임베딩 모델 적용 가능성 탐색 -정성적 성능 평가를 중심으로-)

  • The purpose of this study is to look qualitatively into how efficiently and reasonably a computer can learn themes related to the Nature of Science (NOS). In this regard, a corpus has been constructed focusing on literature (920 abstracts) related to NOS, and factors of the optimized Word2Vec (CBOW, Skip-gram) were confirmed. According to the four dimensions (Inquiry, Thinking, Knowledge and STS) of NOS, the comparative evaluation on the word-level word embedding was conducted. As a result of the study, according to the previous studies and the pre-evaluation on performance, the CBOW model was determined to be 200 for the dimension, five for the number of threads, ten for the minimum frequency, 100 for the number of repetition and one for the context range. And the Skip-gram model was determined to be 200 for the number of dimension, five for the number of threads, ten for the minimum frequency, 200 for the number of repetition and three for the context range. The Skip-gram had better performance in the dimension of Inquiry in terms of types of words with high similarity by model, which was checked by applying it to the four dimensions of NOS. In the dimensions of Thinking and Knowledge, there was no difference in the embedding performance of both models, but in case of words with high similarity for each model, they are sharing the name of a reciprocal domain so it seems that it is required to apply other models additionally in order to learn properly. It was evaluated that the dimension of STS also had the embedding performance that was not sufficient to look into comprehensive STS elements, while listing words related to solution of problems excessively. It is expected that overall implications on models available for science education and utilization of artificial intelligence could be given by making a computer learn themes related to NOS through this study.

Effects of Bojungchiseup-tang on Renal Expression of Water Channels, Na, K-ATPase and Nitric Oxide Synthase in Rats (보중치습탕의 백서 신장 수분채널, Na, K-ATPase, 산화질소 합성효소 발현에 미치는 영향)

  • The present study was examined the effects of Bojungchiseup-tang water extract on the renal expression of renal function regulatory proteins including aquaporin 2 (AQP 2), aquaporin 3 (AQP 3), Na, K-ATPase α1 subunit, endothelial nitric oxide synthase (ecNOS), and inducible nitric oxide synthase (iNOS) in rats. The renal expression of AQP 3 was attenuated in rats administered with Bojungchiseup-tang water extract without altered expression of AQP 2, while ecNOS was up-regualted. Oral administration of Bojungchiseup-tang water extract (40 ㎕/100 g) also attenuated the renal expression of Na, K-ATPase α1-subunit and iNOS protein. These results suggest that the diuretic and natriuretic effects of Bojungchiseup-tang maybe causely related with a decreased expression of AQP 3 and increased expression of ecNOS.
