Microbial Community Analysis of a Methane-Oxidizing Biofilm Using Ribosomal Tag Pyrosequencing

  • Current ecological knowledge of methanotrophic biofilms is incomplete, although they have been broadly studied in biotechnological processes. Four individual DNA samples were prepared from a methanotrophic biofilm, and a multiplex 16S rDNA pyrosequencing was performed. A complete library (before being de-multiplexed) contained 33,639 sequences (average length, 415 nt). Interestingly, methanotrophs were not dominant, only making up 23% of the community. Methylosinus, Methylomonas, and Methylosarcina were the dominant methanotrophs. Type II methanotrophs were more abundant than type I (56 vs. 44%), but less richer and diverse. Dominant non-methanotrophic genera included Hydrogenophaga, Flavobacterium, and Hyphomicrobium. The library was de-multiplexed into four libraries, with different sequencing efforts (3,915 - 20,133 sequences). Sorrenson abundance similarity results showed that the four libraries were almost identical (indices > 0.97), and phylogenetic comparisons using UniFrac test and P-test revealed the same results. It was demonstrated that the pyrosequencing was highly reproducible. These survey results can provide an insight into the management and/or manipulation of methanotrophic biofilms.

The inertial coefficient for fluctuating flow through a dominant opening in a building

  • For a building with a dominant windward wall opening, the wind-induced internal pressure response can be described by a second-order non-linear differential equation. However, there are two ill-defined parameters in the governing equation: the inertial coefficient $C_I$ and the loss coefficient $C_L$. Lack of knowledge of these two parameters restricts the practical use of the governing equation. This study was primarily focused on finding an accurate reference value for $C_I$, and the paper presents a systematic investigation of the factors influencing the inertial coefficient for a wind-tunnel model building including: opening configuration and location, wind speed and direction, approaching flow turbulence, the model material, and the installation method. A numerical model was used to simulate the volume deformation under internal pressure, and to predict the bulk modulus of an experimental model. In considering the structural flexibility, an alternative approach was proposed to ensure accurate internal volume distortions, so that similarity of internal pressure responses between model-scale and full-scale building was maintained. The research showed 0.8 to be a reasonable standard value for the inertial coefficient.

  • "Brokeback Mountain" deals with the love story between two homosexuals named Ennis and Jack. They never do use the word 'homosexual' but instead 'love.' They hide their love into the closet. And they conform to socially constructed gender roles. It is because they recognize that social order has punished the homosexuals severely through history. Especially, Ennis fears the homophobic heterosexual gaze. Through his paranoia, this article examines the conventional contradictory social order causing by the tragic story that is the homosexual "closet phenomenon." Such a phenomenon has resulted from the traditional patriarchal family system that is the central unit of society. Conventionally, patriarchy consists of a dominant male and non-dominant female system, based on force. Sexuality has been constructed, experienced, and understood in culturally and historically specific ways. Homosexuality has been imaged conventionally as a female disguised as a man. As such, homosexuality would violate and break such a constructed system that keeps the sexual hierarchy through male dominant construction. As homosexual, ironically with macho gender personas, Ennis and Jack are social outsiders. Through this story, Proulx suggests the conventional fixed social order is contradictory and, therefore forces the readers to re-consider the world and ponder about the future.

Comparison of Meiobenthic Faunal Communities in Seagrass Bed and Adjacent Bare Sediment (해초지와 주변 퇴적물에 서식하는 중형저서동물 군집 특성 비교)

  • To investigate the community structure and meiofaunal density in seagrass/bare non-seagrass beds, a survey was conducted at three seagrass bed locations in Doomoojin of Baegryongdo, inner harbor of Eocheongdo in May 1999, and Yulim of Dolsando for every month from February to July 1999. Meiobenthic samples were collected from sediments within seagrass beds (SB) and non-seagrass bed (or adjacent to barren sand area, NSB). Nematodes were the most dominant group among representative 13 meiofaunal groups. The sub-dominant groups were benthic for-aminiferans, benthic harpacticoids, and annelids. The highest density of meiofauna was recorded at a seagrass bed of Yulim (7,244 ind/10 $\textrm{cm}^2$ in June), and lowest density was recorded at a non-seauass bed of Baegryoungdo (438 ind/ 10 $\textrm{cm}^2$ in May). For vertical distribution, the highest density of meiofauna was recorded at 0-2 cm depth, and the density abruptly decreased with depth in all stations. The density of meiofauna in size between 0.125 m and 0.25 mm was maximum. Sediment types for the study areas ranged from sandy to sandy mud by the Folk's classification. The density of total meiofauna, the number of taxa, and the density of the dominant groups (nematodes, benthic for-aminiferans, benthic harpacticoids, annelids) between SB and NSB were significantly different. The results clearly showed the importance of seagrass bed as suitable habitat for meiofauna.

  • Basic characteristics of non-thermal plasma process to remove C2H4 and NO have been experimentally investigated with a packed-bed type reactor and an ac power supply. The performance of the non-thermal plasma generated by ac power supply was compared with that of a wire-plate type reactor equipped with a pulsed power supply. The result shows that the non-thermal plasma can be effectively generated with an AC power supply that can be easily fabricated with conventional techniques. In order to understand the basic reaction mechanisms of the non-thermal plasma process, parametric tests for different carrier gases(air and nitrogen) and for different reaction pathways have been performed. The test results show that O3 generated by non-thermal plasma plays an dominant role to oxidize C2H4 and NO over N and O radicals when these pollutant gases are carried by dry air under room temperature condition. Experimental observations, however, indicate that N and O radicals can significantly affect on the removal process of the pollutant gases under certain conditions.

Pathogens and Their Antimicrobial Susceptibilities of Endodontic Infection in Type II Diabetes Mellitus Patients (제 2형 당뇨 환자의 근관 감염에서 나타난 병원균과 그 항생제 감수성)

  • Aim: The purpose of this study was to identify dominant pathogens and their antimicrobial susceptibilities of endodontic infection in type II diabetes mellitus (DM) patients to determine effective empirical antibiotics. Methodology: Pathogens from endodontic infection in six patients with DM and in six patients without DM were cultured, identified and their antimicrobial susceptibility was tested using Vitek2 systems ($bioM{\acute{e}}rieux$, Marcy l' Etoile, France). The results were analyzed using Chi-square test and Fisher's exact test at P< 0.05 level. Results: Pathogens of opportunistic infection were dominant in DM patients (P=0.015). However, there was no significant difference of antimicrobial susceptibility between DM and non-DM patients. Relatively high percent (27%) of pathogens showed resistance to penicillin. Conclusions: More cautions should be paid to DM patients because they are prone to opportunistic infection. Penicillin is not effective in the control of endodontic infection.

  • The main problems of computational complexity in object tracking are definition of objects, segmentations and identifications in non-structured environments with erratic movements and collisions of objects. The object's information as a region that corresponds to objects without discriminating among objects are considered. This paper describes the algorithm that, automatically and efficiently, recognizes and keeps tracks of interest-regions selected by users in video or camera image sequences. The block-based feature matching method is used for the region tracking. This matching process considers only dominant feature points such as corners and curved-edges without requiring a pre-defined model of objects. Experimental results show that the proposed method provides above 96% precision for correct region matching and real-time process even when the objects undergo scaling and 3-dimen-sional movements In successive image sequences.

  • A time-energy transient characteristics of pulsed neutron transport problem with an inelastic kernel in the fast domain is solved exactly with a continuous energy transfer operator. A discrete time eigenvalue is found which is asymptotically dominant. The complete solution consists of three parts: a time-energy separable mode which is asymptotically dominant and a non-separable mode which is made up by two parts; a pure energy slowing-down transient and a mixture of time and energy transient which is negligible asymptotically.

  • This paper presents theoretical analysis of the NRRO(non-repeatable run-out) for a ball bearing with geometric imperfection. The 3DOF dynamic analysis of a ball bearing using the Newton-Raphson method is performed to calculate the displacement of shaft center. Frequency and magnitude characteristics of radial and axial vibrations are investigated. The ball form errors of the ball, the inner race, and the outer race in a HDD spindle ball bearing are precisely measured. NRRO of a ball bearing is analyzed by using the measured waviness data. It is concluded that dominant components of radial vibrations are ${\Large}f_c\;and\;2{\Larg}f_b{\pm}{\Large}f_c$, and dominant component of axial vibrations is $2{\Large}f_b$. These are generated by the size error and the second waviness of the balls.

  • New titanium alloys with a low elastic modulus have been developed for biomedical applications to avoid the stress shielding effect of the artificial prosthesis. The newly developed alloys contained the transition elements like Zr, Hf, Nb, Ta which were non-cytotoxicity elements and $\beta$ stabilizers. In the present paper the elastic moduli of Ti-xM containing Zr, Hf, Nb, Ta were evaluated by measuring the acoustic velocity (PEG). The effectiveness of the alloying elements for lowering the elastic modulus was investigated. In addition, the dominant factors for the low modulus were discussed. Ta was the most effective in lowering the elastic modulus of the alloys. The effectiveness of Hf was not acceptable for decreasing the elastic modulus. The dominant factor was the lattice parameter for Zr, and the poisson's ratio for Nb, Ta, respectively.
