Inhibitory effects of xanthorrhizol on inducible cyclooxygenase (COX-2) and nitric oxide synthase (iNOS) activity in RAW264.7 cells

Discovery of Epinastine-NSAID Hybrids as Potential Anti-inflammatory Agents: Synthesis and In Vitro Nitric Oxide Production Inhibitory Activity Study

  • A novel pharmacophore with epinastine (1) and NSAID moieties (2-5) was designed by molecular hybridization approach. The hybrid compounds 6-9 were synthesized by EDCI/HOBt or HATU-mediated coupling of 1 with salicylic acid (2), mefenamic acid (3), indomethacin (4) and naproxen (5), respectively, and were assessed for their inhibitory effect against NO production in LPS-induced RAW-264.7 macrophages in vitro. The Hybrids were found to exhibit significant NO production inhibitory effects with half-maximal inhibitory concentration (IC50) values ranging in between 15.96 ± 1.32 and 36.68 ± 2.53 μM and were non-cytotoxic to macrophages. Comparing the inhibition concentration (IC50), cytotoxicity concentration (CC50) and in vitro efficacy index (iEI), 6 (IC50 = 17.97 ± 1.92 μM; iEI = 11.13) and 9 (IC50 = 15.96 ± 1.32 μM; iEI = 12.53) were better suited than other hybrids as well as their parent compound. Our findings signify that hybrids 6 and 9 may serve as platforms for continued investigations for the development of more efficient anti-inflammatory agents.

초임계 자소자추출물의 항균효과와 바이오필름, 염증매개인자 생성 억제 효능 (The Effect of Anti-microbial and the Inhibitory Effect of Biofilm Formation and Inflammatory Factors Production of Perillae semen Supercritical Fluid Extracts)

  • 본 연구는 자소자 열수추출물과 압력은 일정하게 유지한 상태로 온도를 조절하여 추출한 3종의 자소자 초임계추출물에 대해 항산화력, 항균활성, biofilm 형성 억제능과 염증매개물질(nitric oxide), 염증성 사이토카인(interleukin-6, interleukin-8)의 생성 억제능을 평가하였다. 자소자 열수추출물과 비교하였을 때 자소자 초임계추출물이 항산화 효과는 떨어졌으나, Staphylococcus aureus(S. aureus)에 대한 minimal inhibitory concentration(MIC)이 월등히 낮은 것으로 확인되었고, biofilm 형성 억제능 또한 더 우수하였다. 염증매개물질과 염증성 사이토카인의 생성도 자소자 초임계추출물이 자소자 열수추출물에 비해 현저히 감소시킬 수 있었다. 따라서 우리는 초임계추출물중 가장 우수한 항균작용과 biofilm 형성 억제, 염증인자 생성 억제 효능을 보인 45℃로 추출한 자소자 초임계추출물이 아토피 피부염 환자의 가려움증과 경표피수분손실을 방지하기 위한 천연 기능성 소재로 적합함을 시사한다.

  • Two phenolic flavonoids were isolated from the traditional medicine of Eastern Asia, Caesalpinia sappan L (Leguminosae). Brazilin (1) showed a significant inhibitory activity against inducible Nitric Oxide Synthase (iNOS) in lipopolysaccharides (LPS)-induced macrophage RAW 264.7 cells with an IC$\sub$50/ value of 1.68 mg/ml, which is more potent than the positive control, L-N$\^$6/-(1-iminoethyl)lysine (IC$\sub$50/ 3.49 mM). On the other hand, caesalpine J (2) was found to be inactive in the present iNOS assay system despite of their structural similarities. (omitted)

  • The Barks of Ulmus davidiana (Ulmaceae) have been used for the treatment of insecticide, anti-boil and anti-fungi in Korean traditional medicine. Four phenolic compounds were isolated from 80% Acetone extracts. The structures of these compounds were elucidated as (+)-catechin, (+)-catechin 7-O-${\beta}$-D-glucopyranoside, (+)-catechin 7-O-${\beta}$-D-xylopyranoside and procyanidin B-1. These phenolic compounds showed significant nitrogen monoxide(NO) production inhibitory activity in IFN-${\gamma}$, LPS stimulated RAW 264.7 cell and also showed significant antioxidative activity on DPPH radical.   • The aim of this study was to find out the effect of hypoxic condition on the regulation of cyplal gene expression. pcyplal-Luc construct was cloned and transfected into Hepa I cells. When Hepa-I cells containing pcyplal-Luc were treated by DFO (desferrioxamine) which is iron-chelating agent, the stimulatory effect of luciferase by TCDD was decreased. This inhibitory effect of desferrioxamine on the luciferase activity was dose dependent and abolished by concomitant treatment with N$\^$G/-nitro-ι-arginine. And when cobalt chloride which is known as a hypoxia inducing chemical was administrated, the stimulatory effect of luciferase by TCDD was also decreased. This inhibitory effect of cobalt chloride on the luciferase activity was dose dependent and abolished by concomitant treatment with N$\^$G/-nitro-ι-arginine. These data showed that hypoxic condition down regulates cyplal gene expression and this might be through nitric oxide action.

  • 플라보노이드는 광범위한 생물학적 활성을 가진 중요한 식물 2차 대사 산물로, 본 연구에서 사용된 naringenin (NN)은 자몽에 가장 풍부한 플라바논의 하나로 간 보호, 항지질 과산화 및 항암 활성에 관한 연구가 보고되었다. 우리는 이전 연구에서 biorenovation 기법을 NN에 적용하여 naringenin-7-O-Glucoside(prunin), naringenin-7-O-phosphate (N7P), 6''-O-Succinyl prunin과 같은 3가지 유도체를 합성하였으며 그 중 생물학적 및 물리화학적 특성이 보고되지 않은 N7P가 NN보다 45배 증가한 수용해도를 나타냄을 확인하였다. 따라서 본 연구에서는 N7P의 추가적인 생리활성 조사하고자 RAW 264.7 세포에서 항염증 활성을 평가하였으며 N7P는 세포 독성이 나타나지 않는 농도에서 유효한 inducible NO synthase (iNOS), cyclooxygenase-2(COX-2) 억제 활성을 보였으며 이들의 발현 경로를 유의하게 억제함으로써 nitric oxide (NO) 및 prostaglandin E2 (PGE2)를 효과적으로 억제하였다. 뿐만 아니라 pro-inflammatory cytokines인 tumor necrosis factor-α (TNF-α)와 interleukin-6 (IL-6)의 발현 또한 유의하게 억제하는 것으로 확인되었다. 이러한 결과를 근거로 N7P가 다양한 염증 인자를 표적으로 하는 항염증 소재로 적용될 수 있음을 제안한다.

토양 균주 발효 추출물 Nargenicin 및 그 유도체의 항생제 대체 효과능 평가 (Biological Evaluation of Nargenicin and Its Derivatives as Antimicrobial Anti-inflammatory Agents)

  • 본 연구진은 신규 미생물인 Nocardia sp. CS682 균주에서 항균물질인 nargenicin을 확보하고, 그 유도체 5종을 확보하여 그람양성, 음성 및 다약제 내성균에 대한 항균 효능 및 LPS로 자극된 대식세포에서의 nitric oxide 생성 억제능을 확인하였다. Nargenicin 유도체들은 nargenicin 및 vancomycin에 비교하여 우수한 항균 활성을 보여주었으며, compound 4 및 5는 광범위한 항균 효능 외에 nitric oxide 생성 억제능을 보여 항균-항염효과를 가지는 dual effector로써 감염, 면역 질환에 응용 가능성을 시사하였다. Nargenicin 유도체들은 향후 염증반응에서의 면역 조절 기작에 대한 추가 연구가 필요할 것으로 사료된다.

The Inhibitory Effect of Lycii Fructus on LPS-stimulated NF-${\kappa}B$ Activation and iNOS Expression in RAW 264.7 Macrophages

  • Objective : Anti-inflammatory effects of the extract of Lycii Fructus on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophage cells were investigated. Method : In order to assess the cytotoxic effect of Lycii Fructus on the raw 264.7 macrophages 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assay was performed. Reverse transcription-polymerase chain reaction(RT-PCR) analysis of the mRNA levels of tumor necrosis factor-$\alpha$(TNF-$\alpha$) and inducible nitric oxide synthase(iNOS) was performed in order to provide an estimate of the relative level of expression of these genes. The protein level of the inhibitor of nuclear factor-${\kappa}B(I{\kappa}B)$ and nuclear factor-${\kappa}B$(NF-${\kappa}B$) activity was investigated by Western blot assay. NO production was investigated by NO detection. Result : Lycii Fructus suppressed NO production by inhibiting the LPS-induced expressions of iNOS and TNF-$^-\alpha$ mRNA and iNOS protein in RAW 264.7 macrophage cells. Also, Lycii Fructus suppressed activation of NF-${\kappa}B$ in the nucleus. Conclusion : These results show that the extract of Lycii Fructus has anti-inflammatory effect probably by suppressing iNOS expressions through the down-regulation of NF-${\kappa}B$ binding activity.

Effects of Root Extracts from Angelica gigas and Angelica acutiloba on Inflammatory Mediators in Mouse Macrophages

  • Root extracts of Angelica gigas and A. acutiloba have been used traditionally for the treatment of gynecological diseases, as well as anemia, blood stasis, and inflammatory pain, as blood tonics in Oriental medicine. In the present study, we investigated the effects of A. gigas and A. acutiloba on inflammatory mediators in mouse macrophages and compared their activities. Many studies suggest that prostaglandin $E_2$ ($PGE_2$) biosynthesis and nitric oxide (NO) production play important roles in the processes of both inflammation and carcinogenesis. Ethanolic extracts from the roots of both species exhibited significant inhibitory effects on $PGE_2$ generation in lipopolysaccharide-stimulated RAW 264.7 cells. In particular, the extract from A. gigas was more effective than that from A. acutiloba. Although neither inhibited NO generation, the extract from A. acutiloba stimulated NO generation. Our results suggest that the roots of A. gigas might possess more anti-inflammatory and/or cancer chemopreventative activity than that of A. acutiloba due to the suppression of cyclooxygenase-2 (COX2)-mediated $PGE_2$ production. In addition, A. acutiloba might exert anti-tumor activity through an increase in macrophage-produced NO.
