Effective Reconstitution of Porcine Hematopoietic Cells in Newborn NOD/SCID Mice Xenograft (돼지 골수 조혈 세포의 이종 마우스 동물 모델 생체 증식 및 분화 특성)

  • The SCID-repopulation cells(SRCs) assay has been widely used to determine the self-renewal capacity of hematopoietic stem cells (HSCs). In this study, we tested the repopulating efficiency of porcine bone marrow derived hematopoietic stem cells using nonobese diabetic/severe combined immunodieficient (NOD/SCID) mice which was inherited immunodeficiency mire with defect of T cells, B cells, and low activity of NK cells. We transplanted porcine bone marrow hematopoietic stem/progenitor cells with intraperitoneal injection into neonate NOD/SCID mice. We confirmed efficient reconstitution activity of inoculated porcine hematopoietis cells in variety of organs of NOD/SCID mice. Interestingly, pig $CD3^+$ T lymphocytes detected with high level in liver($15.6{\pm}3.7%$), spleen($5.6{\pm}3.0%$), thymus($1.5{\pm}1.3%$), and BM($2.3{\pm}0.9%$), respectively. These data imply that microenvironment of neonate NOD/SCID mice is very efficient for proliferation and differentiation of porcine T cells, and can be useful for the study of T cells development and renogeneic organ transplantation.

Production and Characterization of a Transgenic Mouse Model of Human Liver Cancer (형질 전환 기법을 이용한 인체 간암의 마우스 모델 제작 및 특성 규명)

  • Transgenic mice were generated by microinjecting a plasmid DNA containing the SV40 (simian virus 40) large T antigen (Tag) gene fused with mouse albumin promoter/enhancer sequences into fertilized one-cell mouse embryos. Among eleven founder transgenic animals, four developed hepatocellular carcinoma, two showed kidney cancer and one developed skin and brain tumors. Three stable transgenic lines, #1-2, #1-6 and #1-11 were established. Members of the lines #1-6 and #1-11 reproducibly developed liver tumors by 8 to 10 weeks of age but did not exhibit any phenotypic changes in other tissues. Histological changes loading to liver tumor formation occurred with predictable kinetics and could be classified into three distinct stages; (a) newborn to 3 weeks of age, characterized by hyperplastic hepatocytes with reduced amounts of cytoplasm without any nuclear alterations, (b) between 4 to 8 weeks of age, characterized by diffuse liver cell dysplasia without observable tumor nodules, and (c) 9 weeks of age and thereafter, characterized by hepatocellular carcinomas in the background of extensive liver dysplasia. Metastasis to the lung from a liver carcinoma was observed in #1-11 founder animal. This transgenic mouse system displays similarities with human liver cancers in a number of aspects and provides a useful model for the study of molecular events involved in hepatocarcinogenesis.

Internal Fixation of Long Bone Fractures in 6 Calves (송아지에서 발생한 긴뼈골절의 내고정 치료 6례)

  • The simple and easy treatment with cast or splint has been applied to stabilize long bone fractures in farm animals. Applying cast or splint is relatively easier than surgical fixation and may temporarily stabilize fracture site. However, the internal fixation provides strong stabilization and early recovery of limb function. In this report, we describe six cases of limb fractures in calves repaired by internal fixation. Fracture sites are various and affected ages are between 1 day and several months old. In some cases, patients were treated with cast or splint before admitting to Veterinary Medical Center. In these patients, regardless of initial treatment fractures were not immobilized. Consequently we decided to treat these patients with open reduction and internal fixation. The fracture repair with implants(bone plate, screw, cerclage wire, and intramedullary pin) were effective and satisfactory in calves which had been failed by application of cast. Although the cost of orthopedic treatment in farm animal is one of the most important considerations from the practical point of view, internal fixation has many benefits in newborn and young calves. And simplicity of postoperative management is also practically significant benefit expected from infernal fixation In conclusion, we suggest that internal fixation will produce better result than external coaptation in calve.

Congenital Neosporosis in Goats from the State of Minas Gerais, Brazil

  • Congenital $Neospora$ $caninum$ infection was diagnosed in two Saanen goat kids from two distinct herds with a history of abortion and weak newborn goat kids in the Southern region of the State of Minas Gerais, Brazil. The first kid was weak at birth, had difficulty to rise and was unable to nurse. Gross lesions of porencephaly and hydrocephalus ex vacuo were seen. Multifocal necrosis, gliosis and non-supurative encephalitis were observed in the brain. Several parasitic cysts with a thick wall that reacted strongly only with polyclonal antiserum to $Neospora$ $caninum$ were seen in the cerebral cortex, brain stem and cerebellum. The second kid was born from a $Neospora$ $caninum$ seropositive mother that aborted in the last pregnancy. It was born without clinical signs. The diagnosis of neosporosis was based on antibody titer of 1:800 to $N.$ $caninum$ by indirect fluorescence antibody test obtained from blood collected before the goat kid ingested the colostrum and $Neospora$ $caninum$ DNA was detected by polymerase chain reaction and sequenced from placenta. This is the first report of neosporosis in goats in the southeast region of Brazil.

  • Toxoplasmosis is a parasitic infection caused by the obligate parasite, Toxoplasma gondii, that is distributed throughout the world. Infection is produced by contact with animal like dog, cat, pig, cow or horse. It is also possible to produce clinical disease in the form of oropharyngeal infection through consumption of raw or incompletely cooked meat. Handling of contaminated cat litter has been found to be the major route of transmission. There are two clinical manifestations of toxoplasmosis: the congenital and the acquired form. The congenital form is characterized by hydrocephalus, chorioretinitis, convulsion, and intracerebral calcifications in the newborn. The acquired form is further subdivided into the disseminated and the lymphadenopathic types, the latter being the more common form of the disease in men. This is a case report of a Korean 8 year 2 months female congenital toxoplasmosis child. Clinical feature, patient's medical history, radiographic evaluation and dental problems are discussed in this report.

  • Objectives : Recurrent pregnancy loss occurs in approximately 0.5-3% of women. There are many studies concerning immunological factor recently. Therefore, aim of this study is to examine the latest trend of researches concerning recurrent pregnancy loss, and controlled experiment on animals about antiphospholipid antibody. Method : We referred Pubmed site by using searching word of 'recurrent pregnancy loss' (Limits : 2000.1-2004.3, animal) Results and conclusions : 1. We searched 29 papers. Immunological factor : 18 ( about antiphospholipid antibody : 10 ), Chromosomal abnormality : 6, The others : 5 2. In five papers about controlled experiment on animals, (1) Materials : 8-12 weeks old mice / 11.5-day old-rat embryos / New Zealand rabbit (2) Inductions : inject intraperitoneally with human IgG containing antiphospholipid antibodies / culture in a solution of 1 ml medium which contained IgG purified from sera of women / inject intradermally with cardiolipin (3) Treatments : inject intraperitoneally with complement component before / culture in a solution of 1 ml medium which contained IgG purified from sera of women with SLE and RPL or from healthy women / inject intradermally with TFX, 0.9% NaCl (4) Measurements : weight fetuses and placentas, calculate frequency of fetal resorption / after culture, examine the embryos / examine platelet counts, APTT and numbers of live and dead newborns, resorbed fetuses, body mass, newborn viability and survival rates.

  • To focus attention on milk and dairy products, we celebrate World Milk Day on the first day of June. Milk is a primary source of essential nutrients for newborn mammals, including humans. Milk and its by-products have been integral to human life since domestication of the dairy cow. Even though a large portion of the population is lactase impersistent, consumption of dairy products, including liquid milk, has been increasing over the last decade in Korea. Per capita annual consumption of dairy products in Korea increased to 61.3 kg in 2008, up from 49.4 kg in 1998. However, consumption of liquid milk has only increased marginally from 30.3 kg in 1998 to 35 kg in 2008. In 2007, the price of farm milk in Korea was the highest in the world. High dairy prices are forcing major manufacturers to find alternative, less expensive sources of milk, which has led to an increase in imported dairy products. Recently, the International Dairy Federation (IDF) published a special report containing the figures for world milk production, milk processing, and consumption. Understanding the current situation with regard to world diary production and the outlook for the industry could be useful during the present world economic crisis. The authors of this brief review summarize the status of dairy production in countries throughout the world, including Korea, along with some of the data published in the IDF annual report. The authors greatly appreciate the support of IDF Korea and the IDF for generously providing the data used in this paper.

The Effect of Motor Skill Training on Motor Function and Cerebellar Development After Alcohol Exposure in Neonatal Rats (운동기술훈련이 태아알코올증후군 흰쥐 모델의 소뇌 발달과 운동기능에 미치는 영향)

  • The purpose of this study was to test that motor skill training enhance motor function and cerebellar development. Using an animal model of fetal alcohol syndrome-which equates peak blood alcohol concentrations across developmental period-critifical periods for the effect of alcohol on body and cerebellar weigh was examined. The effect of motor skill training on motor function and cerebellar development of rat exposed alcohol on postnatal days 4 through 10 were studied. Newborn rats were assigned to one of two groups: (1) Control group (CG), via artificial rearing to milk formula and (2) experimental groups (EG), via 4.5g/kg/day of ethanol in a milk solution. After completion of the treatments, the pups were fostered back to lactating dams, and wearing they were raised in standard caged until they were postnatal 48 days. Rats from experimental group of postnatal treatment then spent 10 days in one of two groups: Experimental group II (EGII) was had got motor skill training (training traverse a set of 6 elevated obstacles) for 4 weeks. Experimental group I (EGI) was not trained. Before sacrificing, the rat got examined two behavioral test, body weigh and cerebellar weigh, then coronal sections were processed. The section was investigated the Purkije cell in the cerebellum using light microscope. The results of this study were as follows. 1. In body weight test, the outcome of alcohol groups were significantly lower than the normal group. 2. In cerebellar weight test, the outcome of EGI were significantly lower than CG and EGII. 3. In motor behavioral test, the outcome of EGI was significantly lower than NG and EGII. 4. In Purkinje cells counting test, the outcome of EGI was significantly lower than the NG and EGII. These result suggest that improved motor function induced by motor skill training after postnatal exposure is associated with dynamically altered expression of Purkinje cells and that is related with cerebellar function. Also, these data can potentially serve as a model for therapeutic intervention.

Role of Nitric Oxide in Leukocyte-Endothelial Interaction in Cerebral Venules during Reperfusion after Global Ischemia

  • Objective : Reactive oxygen metabolites and polymorphonuclear leukocytes have been implicated in the pathophysiology of reperfusion injury. The mechanisms involved in superoxide-mediated leukocyte adherence remain unclear, however, nitric oxide[NO] may contribute to this response. The present study is undertaken to elucidate mechamisms controlling NO based mechanisms that regulated leukocyte-endothelial interactions in the cerebral vasculature after global cerebral ischemia and reperfusion. Methods : Pial venular leukocyte adherence of anesthetized newborn piglets was quantified by in situ fluorescence videomicroscopy through closed cranial windows during basal conditions and during 2hours of reperfusion after global ischemia induced by 9minutes of asphyxia. Nitric oxide synthase[NOS] was inhibited by local window superfusion of L-nitroarginine[NA]; superfusion of sodium nitroprusside[SNP] was used to donate NO. Results : The mean number of adherent leukocytes to cerebral venules in the 9minutes asphyxia and 2hours reperfusion group were $161{\pm}19$ compared with $13{\pm}4$ in the nonasphyxial group. Superfusion of L-NA through the cranial window for 2hours resulted in leukocyte adherence similar to that observed during the initial 2hours of reperfusion after asphyxia. Leukocyte adherence was not additionally increased in asphyxic animal treated with L-NA. SNP inhibited asphyxia induced leukocyte adherence back to control levels. Conclusions : Nitric oxide inhibits leukocyte adherence to cerebral venules during the initial hours of reperfusion after asphyxia, and that NO supplementation inhibit asphyxia induced leukocyte adherence back to control levels. These results indicate that NO is an important factor in ischemia-reperfusion induced leukocyte adherence.

Implementation of Visible monkey into general-purpose Monte Carlo codes: MCNP, PHITS, and Geant4

  • Recently, a new monkey computational phantom, called Visible Monkey, was developed for non-ionizing radiation studies in animal research. In this study, we extended its applications to ionizing radiation studies by implementing the voxel model of the Visible Monkey into three general-purpose Monte Carlo (MC) codes: MCNP6, PHITS, and Geant4. The implementation work for MCNP and PHITS was conducted using the LATTICE, UNIVERSE, and FILL cards. The G4VNestedParameterisation class was used for Geant4. Then, organ dose coefficients (DCs) for idealized photon beams in the antero-posterior direction were calculated using the three codes and compared, showing excellent agreement (differences <3%). Additionally, organ DCs in other directions (postero-anterior, left-lateral, and right-lateral) were calculated and compared with those of the newborn and 1-year-old reference phantoms. Significant differences were observed (e.g., the stomach DC of the monkey was 5-fold greater than that of the 1-year-old phantom at 0.03 MeV) while the differences tended to decrease with increasing energy (mostly <20% at 10 MeV). The results of this study allows conducting MC simulations using the Visible Monkey to estimate organ-level doses, which should be valuable to support/improve monkey experiments involving ionizing radiation exposures.
