Removal of Red Tide Organisms -1. flocculation of Red Tide Organisms by Using IOSP- (적조생물의 구제 -1. IOSP에 의한 적조생물의 응집제거-)

  • This study was to examine the physicochemical characteristics of coagulation reaction between ignited oyster shell powder (IOSP) and red tide organisms (RTO), and its feasibility, in developing a technology for the removal of RTO bloom in coastal sea,IOSP was made from oyster shell and its physicochemical characteristics were examined for particle size distribution, surface characteristic by scanning electron microscope, zeta potential, and alkalinity and pH variations in sea water. Two kinds of RTO that were used in this study, Cylindrotheca closterium and Skeletonema costatum, were sampled in Masan bay and were cultured in laboratory. Coagulation experiments were conducted using various c(Incentrations of IOSP, RTO, and a jar tester. The supernatant and RTO culture solution were analyzed for pH, alkalinity, RTO cell number, IOSP showed positive zeta potentials of $11.1{\~}50.1\;mV\;at\;pH\;6.2{\~}12.7$, A positive zeta potential of IOSP slowly decreased with decreasing pNa 4,0 to 2,0. When pNa reached zero, the zeta potential approached zero, When a pMg value was decreased, the positive zeta potential of IOSP increased until pMg 3.0 and decreased below pMg 3.0. IOSP showed 4.8 mV of positive zeta potential while RTO showed -9.2 mV of negative zeta potential in sea water. A positive-negative EDL (electrical double-layer) interaction occurred between $Mg(OH)_2$ adsorption layer of IOSP and RTO in sea water so that EDL attractive force always worked between them. Hence, their coagulation reaction occurred at primary minimum on which an extreme attractive force acted because of charge neutralization by $Mg(OH)_2$ adsorption layer of IOSP. As a result, the coagulation reaction was rapidly processed and was irreversible according to DLVO (Deriaguin-Landau-Verwey-Overbeek) theory. Removal rates of RTO were exponentially increased with increasing both IOSP concentration and G-value. The removal rates were steeply increased until 50 mg/l of IOSP and reached $100{\%}\;at\;400\;mg/l$ of IOSP. Removal rates of RTO were $70.5,\;70.5,\;81.7,\;85.3{\%}$ for G-values of $1,\;6,\;29,\;139\;sec^(-1)$at IOSP 100 mg/l, respectively. This indicated that mixing (i.e., collision among particles) was very important for a coagulation reaction.

Immunocytochemistry of Metallothionein Expression in Developing Rat Liver (발생중인 흰쥐 간에서의 Metallothionein 발현에 관한 면역세포화학적 연구)

  • Metallothionein (MT) is a family of ubiquitous, low molecular weight (6-7 kDa), cysteine-rich protein with a high affinity to metal ions and has no aromatic amino acids and histidine. Some of the known functions of MT include detoxification of heavy metals and alkylating agents and neutralization of free radicals. Also, this protein may affect a number of cellular processes including gene expression, apoptosis, proliferation and differentiation. But, its actual functions are still not clear. The present study was undertaken to examine immunocytochemically the localization of MT in developing rat liver. On the day 11 of gestation, the fetal rat liver has already been formed and contained numerous oval cells with high nuclear cytoplasmic ratio, which were the progenitors of hepatic parenchymal cells, but no reaction products of MT were detected at this time. And then, positive reactions against MT started to appear predominantly in the parenchymal cells of liver from the 13th day after gestation. Reaction products, immunogold particles or brown coloration, were localized at both the nucleus and the cytoplasm of the parenchymal cells, except mitochondria. The intensity of this reaction gradually increased, and exhibited the strongest at birth. The intensity of MT staining and immunogold labelling diminished with growth, and by the 15th day after birth weak positive reaction was observed in the cells. In brief, positive reactions for MT were observed in the oval cells and the parenchymal cells during fetal stage, meanwhile they were present only in the parenchymal cells after birth. The present results suggest that MT possibly involves parechymal cell proliferation and differentiation through the storage or the supply of various metal ions in the developing rat liver.

  • Metallothionein (MT) is a family of ubiquitous, low molecular weight ($6,000{\sim}7,000D$), cysteine-rich ($30{\sim}35%$) inducible protein with a high affinity to metal ions and has no aromatic amino acids and histidine. Some of the known functions of MT include detoxification of heavy metals and alkylating agents and neutralization of free radicals. Also, this protein has been reported to involve in tumor pathophysiology and therapy resistance. MT expression may affect a number of cellular processes including gene expression, apoptosis, proliferation and differentiation. Many reports on the physiological and biochemical properties of MT have been published, but ultrastructural reports on the localization of MT in human gastric cancer tissues are extremely rare. The present study was undertaken to examine the ultrastructural features and the localization of MT within the gastric adenocarcinoma. Ultrastructures of gastric cancer cells were characterized by the high nuclear cytoplasmic ratio, the interdigitation between cells, the irregular nucleus containing much heterochromatin and the wide distribution of free ribosomes in the cytoplasm. Immunohistochemical reaction for MT was prominent in the gastric adenocarcinoma. And the immunogold labellings were more prominent within the nucleus than the cytoplasm. Particularly, immunogold particles were numerously seen at nulcleolus or nucleolar associated heterochromatin. These results suggest that MT expression by gastric cancer cells is associated with cell proliferative activity and is possibly synthesized in the cytoplasm, and then the protein is transported into the nucleus to participate in any transcriptional steps.

Comparison of Chemical Characteristics in Wet and Bulk Precipitation Collected in the Iksan Area (익산지역에서 자동 및 수동채취방식에 따른 강수의 화학적 특성 비교)

  • In order to understand the precipitation acidity and chemical composition of ion species in Iksan area as well as to know the difference of chemical characteristics in precipitation samples from the viewpoint of precipitation sampling method, precipitation samples were collected by wet-only automatic precipitation sampler and bulk manual precipitation sampler in Iksan, from March 2003 to August 2003. The mean pH of precipitation was 5.0. There was a little significant difference in the mean value of pH between automatic and manual sampler. However, pH values of some precipitation samples were lower in automatic sampler than in manual sampler, especially in case of precipitation samples with small rainfall for March 2003. The mean concentrations of each ions in precipitation were generally a little higher in precipitation samples collected by the manual sampler than in those collected by the automatic sampler because of accumulation of dry deposition on the surface of glass funnel installed at the manual sampler during the sampling period or no rainfall. Dominant species determining the acidity of precipitation, were N $H_4$$^{[-10]}$ and nss-C $a^{2+}$ for cations and nss-S $O_4$$^{2-}$ and N $O_3$$^{[-10]}$ for anions. The mean concentration of N $H_4$$^{+}$ and nss-C $a^{2+}$ were 31 $\mu$eq/L and 9 $\mu$eq/L for the automatic sampler and 40 ueq/L and 16 ueq/L for the manual sampler, respectively. In addition, nss-S $O_4$$^{2-}$ and N $O_3$$^{[-10]}$ were 27 $\mu$eq/L and 13 $\mu$eq/L for the automatic sampler and 32 $\mu$eq/L and 17 $\mu$eq/L for the manual sampler, respectively. Although the concentrations of the acidifying ions of nss-S $O_4$$^{2-}$ and N $O_3$$^{[-10]}$ were about 3 times higher than those for foreign pristine sites, precipitation acidity were estimated to be natural due to the neutralization reaction of the alkaline species of N $H_4$$^{+}$ and nss-C $a^{2+}$ with its higher concentrations. Considering the ratios of nss-S $O_4$$^{2-}$/N $O_4$$^{[-10]}$ nss-S $O_4$$^{2-}$, it was found that ammonium sulphate was dominant in Iksan precipitation. The major non-sea salt ions were maximum concentrations for March, but decreased with increasing of precipitation amount.on amount.

Seroepidemiologic Evidence for the Presence of Hantavirus in South Africa (남아프라카 지역내 한타바이러스 존재에 관한 혈청 역학적 증거)

  • Sero-epidemiologic survey has been carried out to establish serologically the presence of hantavirus in areas of South Africa. The survey was oriented to search natural infection in both of humans and wild rodents and involvement of human disease. The normal human sera were collected from the residents in urban and rural areas of Western Cape, and rural area of Eastern Cape province. The rodent sera came from various species of rodents trapped in Northern Cape and Western Free provinces. The patient sera were selected from the patients of renal failure, pulmonary syndrome and pyrexia of unknown origin (PUQ) according to diagnostic chart among the patients hospitalized in major hospitals of Cape Town area. The sera were screened and titrated by IFA test using antigens of Hantaan (HTN), Seoul (SEO), Puumala (PUU), and Prospect Hill (PH) viruses primarily. Positive cases were subjected to differential IFA test using HTN, PUU and PH antigens and plaque reduction neutralization test for further confirmation. Anti-hantavirus antibodies were detected from 2 of 352 rural, 1 of 172 urban residents of E. Cape, and 5 of 118 rural, 5 of 368 urban residents of W. Cape. The antibody was also demonstrated from 5 of 221 wild rodents, and it was appeared that 2 different species, Aethomys namaquensis and Tatem leucogaster, are involved. Among 318 patients tested, 3 who were diagnosed as chronic renal failure, acute respiratory distress syndrome (ARDS) and glomerulonephritis were proved to be positive. The reaction patterns obtained from all of these positive sera were distinct from hantaviral sero-patterns ever established. This result suggests that new viruses may exist in this area and play an possible etiologic role in human disease. The feature of serologic survey on anti-hantavirus antibody demonstrable newly from African wild rodents which are different from reservoir species in other continents elicits a conjecture that the virus may be different from known hantaviruses ever found. This fact also suggests that an expanded role in etiologic involvement with other unknown human diseases by newly emerging hantaviruses may be possible in this areas.

The Effects of Small Group Learning Using Smart Devices in Science Classes (과학 수업에서 스마트 기기를 활용한 소집단 학습의 효과)

  • The purpose of this study is to investigate the influences of small group learning using smart devices in science classes on students' achievement, learning motivation, attitude toward science lessons, and perception of small group learning using smart devices. Four 11th-grade classes (N=133) at a coed high school in Seoul were randomly assigned to a control group and a treatment group. The intervention of small group learning using smart devices emphasized collaborative writing on activity sheet. The students were taught about acid, base, and neutralization reaction for six class periods. After the instructions, an achievement test, the learning motivation test, the attitude toward science lessons test, and a questionnaire on the perception of small group learning using smart devices were administered. Two-way ANCOVA results revealed that there was a statistically significant interaction effect by their previous chemistry achievement in the achievement test scores. Only low-level students in small group learning using smart devices significantly improved their achievement probably by having the opportunities to get help from high-level students. The adjusted means of the treatment group were significantly higher than those of the control group in learning motivation and attitude toward science lessons. Students' perceptions of small group learning using smart devices tended to be positive. Educational implications of this study are discussed.

A Study on pH Reduction of Recycled Aggregates Using Coffee Waste and Its Crushability (커피박을 이용한 순환골재의 pH 저감 및 파쇄성 연구)

  • In this study, recycled aggregates crushed from waste concrete were sorted into three groups, 10-13 mm, 13-20 mm, and 20-25 mm. They were treated in different ways and then their crushability was evaluated for each treatment. Coffee waste was used for reducing their pH level. The pH of recycled aggregate was almost 11, regardless of aggregate sizes. The pH of coffee waste was nearly 5 and 10, 30, or 50 g of coffee waste was mixed with 1000 ml of distilled water and recycled aggregates. The lowest pH was about 6.2 when 50 g of coffee waste was mixed. Aggregates were treated with microwave or soaked for 1 day in vinegar (pH = 2) for neutralization reaction. Microwave treated and neutralized aggregates showed 3.3% and 6.2% higher crushing values compared to non-treated one, respectively. Neutralized treatment was more effective for crushing. In crushing tests, a sample height of 120 mm was tried, which gave 6.3% higher crushing value. A four stepped loading with each 100 kN gave 7.1% higher crushing value, compared to standard 100 mm height and 400 kN continuous loading.

The Soil and Water Pollution caused by the Weathering of Pyrophyllite Deposits: Upstream Part of Hoidong Water Reservoir in Pusan (납석광산에서 발생하는 토양 및 수질오염 실태 : 부산광역시 회동수원지 상류 지역)

  • Enoronmental problems caused by certain geologic conditions Include pollution of soil by heavy metal, acidization of souls , acid mine drainage, Pound-water pollution, and natural radioactivity, as well as zoo-logical hazards such as landslide and subsidence. The acrid mine drainage contains large amount of heavy metals nO, therefore. cause serious pollution onto the nearby drainage systems and soils. In spite of this prospective environmental danger, few studies have been done on the acid mine drainage derived from non-metallic ore deposits such as pyrophyllitefNapseok) deposits. The sudo-bearing pyrophyllite ores, alteration zones, and mine talllngs of pyrophylllte deposits produce acrid mine drainage by the okidation of weathering. Compared to the fresh host rocks, the ores and altered rocks of pyrophyllite deposits produce acidic solution which contain higher amount of heavy metals because of OeP lower buffering capacity to acrid solution. The pus of urine water and nearby stream water of pyrophyllite deposits are 2.1~3.7, which are strong- ly acidic and much lower than that (6.2~7.2) of upstream water and than that (6.8~7.6) of the stream water derived from the non-mineralized area. This study reveals that this acrid mine drainage can affect the downstream area which is 8km far from the pyrophyllite deposits, even though the drain Is diluted with abundant non-contaminated river water This suggmists that not only acid mine drainage but also the sulfide-bearing sediments originated from the pyrophyllite deposits move downstream and form acidic water through continuous oxidation reaction. The heavy metals such as Pb, Zn, Cu, Cd, Nl, Mn and Fe are enriched In the mine water of low pH, and their contents decrease as the pH of mine water Increases because of the Influx of fresh stream wainer. SoUs of the Pyrophyulte deposits are characterized by high contents of heavy metals. The stream sediments containing the yellowish brown precipitates formed by neutralization of acid mine drainage occur in all parts of the stream derived from the pyrophyllite deposits, and the sediments also contain high amounts of heavy metals. In summary, the acid mine drainage of the pyrophyllite deposits is located in the upstream part of Hoidong water reservoir in Pusan contains large amounts of heavy metals and flows into the Holdong water reservoir without any purification process. To protect the water of Holdong reservoir, the acid mine drainage should be treated with a proper purification process.

Experimental Studies on Dissolution Characteristics of a Heavy Metal(As) in Mining Waste (광산매립지에서 중금속(As)의 용출 특성에 관한 실험적 연구)

  • This study investigates the contamination mechanism of soil by drainages including acid rains around mining waste sites, and suggests the quantitative methods of prevention against soil contaminations and its alternatives. For these purposes, the dissolution of arsenic in soils, which is one of toxic heavy metals, has been examined experimentally using the artificial acidic solution. Also, in order to prevent dissolution of arsenic by acid rain, the effects of limestone for the neutrality method on the soil were investigated. The arsenic in soil specimen was dissolved by strong acidic solution below pH1.0. The maximum amount of dissolved arsenic increased with decreasing pH value. Furthermore, it was found very effective to use limestones for the neutrality method. The neutralization of limestones in acidic solution was found to follow the equation of chemical reaction-controlling formulation in unreacted-core models.

Analysis on Material Characteristics of Restored Areas with Mortar and Basis of Surface Deterioration on the Stupa of State Preceptor Jigwang from Beopchensaji Temple Site in Wonju, Korea (원주 법천사지 지광국사탑 복원부 모르타르 재료학적 특징 및 표면손상 기초 해석)

  • The Stupa of State Preceptor Jigwang from Beopcheonsa Temple Site in Wonju (National Treasure) is a representative stupa of the Goryeo Dynasty, with outstanding Buddhist carvings and splendid patterns, clearly indicating its honoree and year of construction. However, it was destroyed by bombing during the Korean War (1950-1953) and repaired and restored with cement and reinforcing bars in 1957. The surface condition of the original stone shows long-term deterioration due to the m ortar used in past restorations. In order to identify the exact causes of deterioration, the m ortar and surface contaminants on the original stone were analyzed. Portlandite, calcite, ettringite, and gypsum from the mortar were identified, and its ongoing deterioration was observed through pH measurements and the neutralization reaction test. Analysis of surface contaminants identified calcite and gypsum, both poorly water-soluble substances, and their growth in volume among rock-forming minerals was observed by microscopy. Based on those results, semi-quantitative analysis of Ca and S contents significantly influencing the formation of salt crystals was conducted using P-XRF to analyze the basis of surface deterioration, and cross-validation was performed by comparing the body stone affected by the mortar and the upper stylobate stone unaffected by the mortar. Results indicate that the elements are directly involved in the surface deterioration of the body stone.
