  • 에너지 음료는 카페인을 주성분으로 타우린, 비타민 같은 다른 energy-enhancing 성분을 함유하고 있다. 미국과 유럽에서는 글루쿠로노락톤이 에너지 음료에 첨가될수 있으나, 국내에서 의약품으로는 허가되어 있다. 따라서 식품 첨가물로는 그 사용이 금지 되어 있어, 지속적으로 수입 및 유통 음료에서 시험검사를 하여 규제하고 있다. 현재 분석법으로 사용하는 LC-PDA 법은 복잡한 유도체화 과정을 거치고, 음료 중에 당류들이 위양성 결과를 나타내기도 한다. 이런 기존 방법의 단점을 개선하기 위해 HILIC-ESI-MS/MS(hydrophilic interaction liquid chromatography coupled to electrospray ionization tandem mass spectrometry)를 이용한 분석법을 개발하고, 선택성, 직선성, 검출한계, 정량한계, 정밀도, 정확성, 재현성에 대하여 분석법 유효성 검증을 수행했고, AOAC, EURACHEM 가이드라인에 부합되는 결과를 얻었다.

Assessment of Hepatic Cytochrome P450 3A Activity Using Metabolic Markers in Patients with Renal Impairment

  • Background: The renal function of individuals is one of the reasons for the variations in therapeutic response to various drugs. Patients with renal impairment are often exposed to drug toxicity, even with drugs that are usually eliminated by hepatic metabolism. Previous study has reported an increased plasma concentration of indoxyl sulfate and decreased plasma concentration of $4{\beta}$-hydroxy (OH)-cholesterol in stable kidney transplant recipients, implicating indoxyl sulfate as a cytochrome P450 (CYP) inhibiting factor. In this study, we aimed to evaluate the impact of renal impairment severity-dependent accumulation of indoxyl sulfate on hepatic CYP3A activity using metabolic markers. Methods: Sixty-six subjects were enrolled in this study; based on estimated glomerular filtration rate (eGFR), they were classified as having mild, moderate, or severe renal impairment. The plasma concentration of indoxyl sulfate was quantified using liquid chromatography-mass spectrometry (LC-MS). Urinary and plasma markers ($6{\beta}$-OH-cortisol/cortisol, $6{\beta}$-OH-cortisone/cortisone, $4{\beta}$-OH-cholesterol) for hepatic CYP3A activity were quantified using gas chromatography-mass spectrometry (GC-MS). The total plasma concentration of cholesterol was measured using the enzymatic colorimetric assay to calculate the $4{\beta}$-OH-cholesterol/cholesterol ratio. The correlation between variables was assessed using Pearson's correlation test. Results: There was a significant negative correlation between MDRD eGFR and indoxyl sulfate levels. The levels of urinary $6{\beta}$-OH-cortisol/cortisol and $6{\beta}$-OH-cortisone/cortisone as well as plasma $4{\beta}$-OH-cholesterol and $4{\beta}$-OH-cholesterol/cholesterol were not correlated with MDRD eGFR and the plasma concentration of indoxyl sulfate. Conclusion: Hepatic CYP3A activity may not be affected by renal impairment-induced accumulation of plasma indoxyl sulfate.

고우영의 만화 <놀부뎐>의 서사 변용 양상과 흥부전의 수용문화 (Adaptation of Ko woo-young's Nolbudyeon and Reception Culture in Heungbu and Nolbu)

  • 이 연구에서는 만화가 고우영(1939-2005)을 고전에 대환 자신의 이해와 해석을 동시대인들에게 확산시킨 창조적 수용자라고 보고, 일련의 고전소설을 개작한 그의 작업 중 흥부전의 개작인 <놀부뎐>(1988)을 대상으로 서사변용 양상을 살피고 이를 가능하게 한 흥부전의 수용문화를 고찰하였다. 특히 고우영의 작품은 '어떻게 하면 과거의 문학을 현재의 고전으로서 살아있게 할 수 있는가?'하는 질문에 좋은 참조가 된다. 기존의 흥부전과 다른 특징을 드러내는 데 초점을 맞추어 사건 구성과 인물형상화, 시공간적 배경에서 작품의 서사 변용 양상을 살펴보았다. 우선이 작품은 흥부와 놀부가 태어나게 된 사연으로부터 시작하여 가족 관계 속에서 둘의 성격 형성 과정을 서사화하는 '앞이야기 이어붙이기' 방식을 취하고 있다. 이는 이 작품이 성격과 성향이 다른 형제 간의 갈등을 서사의 중심 문제로 취하고 있음을 말해 준다. 한편, 결말 처리에 있어서는 모호성과 다양한 해석 가능성을 보여주는데 이는 권선징악의 주제를 구현하던 흥부전에 대한 대화적 답변이라 할 수 있다. 인물형상화 면에서는 새로운 인물의 삽입과 기존 인물에 대한 재해석이 어떻게 이루어지는지 분석하였다. 새로운 인물인 놀순이와 연생원은 각기 자기 사연과 개성을 가진 인물로 등장하여 놀부와 흥부의 성격 형성과 인생행로에 영향을 미치면서, 형제의 서로 다른 성격을 매개적으로 드러내었다. 또한 긍정적인 요소와 부정적인 요소를 함께 갖춘 흥부와 놀부의 성격형상화를 통해 <놀부뎐>은 기존의 흥부전이 보여주는 선악관, 인간관에 대해 의문을 드러내며 겉모습으로 사람의 선악을 쉽게 판단하는 세태를 비판할 수 있었다. <놀부뎐>의 시공간을 이해할 때, 작품의 시공간적 배경은 흥부와 놀부가 살고 있는 서사세계와 작자와 독자가 살고 있는 당대가 혼융되어 있다고 할 수 있다. 흥부, 놀부의 외면은 전승되는 서사세계에서 가져왔으되, 그들이 사유 방식과 감성 구조는 현대적인 것이며, 이를 표현하기 위해서 이질적인 시공간에 속한 사물과 이와 연관된 사건들이 필요했던 것이다. 이런 시공간의 혼융은 흥부전이 옛날이야기가 아니라 동시대에 진행되는 현재의 이야기로 받아들이게 하면서 <놀부뎐>이 현재의 삶을 빗댄 이야기임을 재확인하게 한다.

Assessment of the Effects of Interactions between Climatic Conditions and Genetic Characteristics on the Agronomic Traits of Soybeans Grown in Six Different Experimental Fields

  • Soybean [Glycine max (L.) Merr.] is a species of legume native to East Asia. The interactions between climatic conditions and genetic characteristics are known to affect the agricultural performance of soybean. Therefore, the present investigation was conducted to identify the main elements affecting the agricultural performances of 11 soybean varieties/lines from China [Harbin ($45^{\circ}12^{\prime}N$), Yanji ($42^{\circ}53^{\prime}N$), Dalian ($39^{\circ}30^{\prime}N$), Qingdao ($36^{\circ}26^{\prime}N$)] and the Republic of Korea [Suwon ($37^{\circ}16^{\prime}N$), and Jeonju ($35^{\circ}49^{\prime}N$)]. The days to flowering (DTF) of soybeans with the e1-nf and e1-as alleles and the E1e2e3e4 genotype, except in 'Keumgangkong', 'Tawonkong', and 'Duyoukong', were relatively short compared to those of soybeans with other alleles. Although DTF of the soybeans was highly correlated with all climatic conditions [negative: precipitation, average temperature (AVT), accumulated temperature; positive: day-length (DL)], days to maturity and 100-seed weight of the soybeans showed no significant correlation with any climatic conditions. The soybeans with a dominant Dt1 allele, except 'Tawonkong', had the longest stem length (STL). Moreover, STL of the soybeans grown in the test fields showed a positive correlation with only DL; however, the results of our chamber test that was conducted to complement the field tests showed that STL of soybean was positively affected by AVT and DL. Although soybean yield (YLD) showed positive correlations with latitude and DL (except L62-667, OT89-5, and OT89-6), the response of YLD to the climatic conditions was cultivar-specific. Our results show that DTF and STL of soybeans grown in six different latitudes are highly affected by DL, and AVT and genetic characteristic also affect DTF and STL.

Effect of ciglitazone on adipogenic transdifferentiation of bovine skeletal muscle satellite cells

  • Ciglitazone is a member of the thiazolidinedione family, and specifically binds to peroxisome proliferator-activated receptor-γ (PPARγ), thereby promoting adipocyte differentiation. We hypothesized that ciglitazone as a PPARγ ligand in the absence of an adipocyte differentiation cocktail would increase adiponectin and adipogenic gene expression in bovine satellite cells (BSC). Muscle-derived BSCs were isolated from six, 18-month-old Yanbian Yellow Cattle. The BSC were cultured for 96 h in differentiation medium containing 5 µM ciglitazone (CL), 10 µM ciglitazone (CM), or 20 µM ciglitazone (CH). Control (CON) BSC were cultured only in a differentiation medium (containing 2% horse serum). The presence of myogenin, desmin, and paired box 7 (Pax7) proteins was confirmed in the BSC by immunofluorescence staining. The CL, CM, and CH treatments produced higher concentrations of triacylglycerol and lipid droplet accumulation in myotubes than those of the CON treatment. Ciglitazone treatments significantly increased the relative expression of PPARγ, CCAAT/enhancer-binding protein alpha (C/EBPα), C/EBPβ, fatty acid synthase, stearoyl-CoA desaturase, and perilipin 2. Ciglitazone treatments increased gene expression of Pax3 and Pax7 and decreased expression of myogenic differentiation-1, myogenin, myogenic regulatory factor-5, and myogenin-4 (p < 0.01). Adiponectin concentration caused by ciglitazone treatments was significantly greater than CON (p < 0.01). RNA sequencing showed that 281 differentially expressed genes (DEGs) were found in the treatments of ciglitazone. DEGs gene ontology (GO) analysis showed that the top 10 GO enrichment significantly changed the biological processes such as protein trimerization, negative regulation of cell proliferation, adipocytes differentiation, and cellular response to external stimulus. Kyoto Encyclopedia of Genes and Genomes pathway analysis showed that DEGs were involved in the p53 signaling pathway, PPAR signaling pathway, biosynthesis of amino acids, tumor necrosis factor signaling pathway, non-alcoholic fatty liver disease, PI3K-Akt signaling pathway, and Wnt signaling pathway. These results indicate that ciglitazone acts as PPARγ agonist, effectively increases the adiponectin concentration and adipogenic gene expression, and stimulates the conversion of BSC to adipocyte-like cells in the absence of adipocyte differentiation cocktail.

가상현실 안전교육 콘텐츠의 인터랙션 디자인 연구 (Interaction Design Study of Virtual Reality Safety Education Contents)

  • 본 연구는 가상현실 안전교육 콘텐츠의 기획과 제작에 참고할 수 있도록 내용 구성 단계별 인터랙션 디자인 특성을 도출하는 것을 목적으로 하였다. 선행연구를 토대로 VR 안전교육 콘텐츠 구성의 특성과 VR 사용자 인터페이스의 분류, 인터랙션 유형, 인터랙션 방식, 인터랙션 구성 요소들의 관계를 정리하여 인터페이스의 관계 분석 틀을 제시하였으며, VR안전교육 콘텐츠 사례 분석을 통해 비상 상황 대응절차 지식의 습득, 사고 상황 체험 학습 그리고 내용의 확인 및 평가의 세 가지 구성단계에서 각각 다음과 같은 상호작용 디자인 특징을 가지고 있음을 확인하였다. 첫째, 학습자 체험단계에서 교육 핵심 체험을 강조하기 위하여 행위 충실도를 높이고 일반적이고 반복적인 행동은 간략하게 축소하여 체험의 질을 조절하는 것이 드러났다. 둘째, 학습자가 생소한 환경에서 안전 행동 절차에 대한 정보 습득을 위하여 주요 상호작용 대상물을 쉽게 인지할 수 있도록 공간UI나 화살표나 상징을 활용한 기표, 행위를 구체적으로 지시하는 게시글 및 다감각 신호를 활용하여 사용자 체험 자유도를 낮추는 방식으로 필수 행위를 강조하고 인지적 인터랙션을 위한 비사실적 방식 인터랙션 비중이 증가하는 것으로 나타났다. 마지막으로 체험의 확인과 평가 단계에서 사고 상황 후 신체손상 등의 부정적 경험을 은유적으로 경험하도록 하는 메타UI를 활용하고 3인칭 시점이나 반복재생의 방식으로 학습자의 체험을 객관적 시각으로 환기하는 것이 활용되었다. 또한 이러한 특성과 관련하여 인터랙션 차원에 따른 요소들의 관계를 도출하여 제시하였다.

사극 드라마에 나타난 고증 의상의 현대화에 대한 대학생들의 인식 -하브루타 학습법을 중심으로- (Undergraduate Students' Perspectives towards Modernization of Historical Costume in Historical Drama -Focused on Havruta Learning-)

  • 사극 드라마는 더욱 극적인 전개를 원하는 대중들의 취향에 따라 퓨전 형태로 변화하고 있으며, 고증 의상 또한 현대적인 감각으로 표현되고 있다. 본 연구는 사극 드라마에 나타난 고증 의상의 현대화의 사례 탐색 및 학습자의 인식을 조명해보고, 향후 사극 드라마에서 고증 의상의 현대화에 대한 함의점을 제시해보는데 목적이 있다. 연구 방법은 이론적 연구와 사례 연구 및 하브루타 학습법을 통한 서술형 설문지 응답법을 병행하였다. 본 연구의 결과로 첫째, 사극 드라마에서 고증의상의 현대화에 대한 본질적인 문제는 사극 드라마의 역사적 사실을 저해하는 고증의상의 과도한 현대적 변용으로 나타났다. 둘째, 사극 드라마에서 고증의상의 현대화에 대한 대학생들의 부정적인 인식으로는 한국 고유의 정체성 상실, 드라마의 몰입도 저하, 미디어가 갖는 교육적 영향력의 견해가 주를 이루었다. 긍정적인 인식으로는 전통 문화의 인식 제고를 통한 관심 증대, 시대적 흐름의 변화에 대한 절충, 대중들의 시각적 유희 증대의 견해가 주를 이루었다. 셋째, 사극 드라마에서 고증의상의 효과적인 연출 방향에 대한 함의점은 사극 드라마 관계자들의 인식개선과 고증의상에 대한 의상디자이너의 철저한 사전 조사, 전통복식에 대한 심층적인 연구와 체계적이고 명확한 교육적 접근, 정부의 노력과 시청자들의 관심이 필요하다.

Expression of Toll-like Receptors, Pro-, and Anti-inflammatory Cytokines in Relation to Gut Microbiota in Irritable Bowel Syndrome: The Evidence for Its Micro-organic Basis

  • Background/Aims A Subset of patients with irritable bowel syndrome (IBS) may have mild inflammation due to immune activation. Toll-like receptors (TLRs) and cytokines may cause intestinal inflammation. We studied their expression in relation to gut microbiota. Methods Expression of TLRs and cytokines was assessed in 47 IBS patients (Rome III) and 25 controls using quantitative real-time polymerase chain reaction. Immunohistochemistry was further performed to confirm the expression of TLR-4 and TLR-5. Results Of 47 patients with IBS, 20 had constipation (IBS-C), 20 diarrhea (IBS-D), and 7 unclassified (IBS-U). The mRNA levels of TLR-4 and TLR-5 were up-regulated in IBS patients than controls (P = 0.013 and P < 0.001, respectively). Expression of TLR-4 and TLR-5 at protein level was 4.2-folds and 6.6-folds higher in IBS-D than controls. The mRNA levels of IL-6 (P = 0.003), C-X-C motif chemokine ligand 11 (CXCL-11) (P < 0.001) and C-X-C motif chemokine receptor 3 (CXCR-3) (P < 0.001) were higher among IBS patients than controls. Expression of IL-6 (P = 0.002), CXCL-11 (P < 0.001), and CXCR-3 (P < 0.001) were up-regulated and IL-10 (P = 0.012) was down-regulated in IBS-D patients than controls. Positive correlation was seen between TLR-4 and IL-6 (P = 0.043), CXCR-3, and CXCL-11 (P = 0.047), and IL-6 and CXCR-3 (P = 0.003). Stool frequency per week showed positive correlation with mRNA levels of TLR-4 (P = 0.016) and CXCR-3 (P = 0.005), but inversely correlated with IL-10 (P = 0.002). Copy number of Lactobacillus (P = 0.045) and Bifidobacterium (P = 0.011) showed correlation with IL-10 in IBS-C, while Gram-positive (P = 0.031) and Gram-negative bacteria (P = 0.010) showed correlation with CXCL-11 in IBS-D patients. Conclusions Altered immune activation in response to dysbiotic microbiota may promote intestinal inflammation in a subset of patients with IBS.

Ventx1.1 as a Direct Repressor of Early Neural Gene zic3 in Xenopus laevis

  • From Xenopus embryo studies, the BMP4/Smad1-targeted gene circuit is a key signaling pathway for specifying the cell fate between the ectoderm and neuro-ectoderm as well as the ventral and dorsal mesoderm. In this context, several BMP4/Smad1 target transcriptional factors have been identified as repressors of the neuro-ectoderm. However, none of these direct target transcription factors in this pathway, including GATA1b, Msx1 and Ventx1.1 have yet been proven as direct repressors of early neuro-ectodermal gene expression. In order to demonstrate that Ventx1.1 is a direct repressor of neuro-ectoderm genes, a genome-wide Xenopus ChIP-Seq of Ventx1.1 was performed. In this study, we demonstrated that Ventx1.1 bound to the Ventx1.1 response cis-acting element 1 and 2 (VRE1 and VRE2) on the promoter for zic3, which is a key early neuro-ectoderm gene, and this Ventx1.1 binding led to repression of zic3 transcription. Site-directed mutagenesis of VRE1 and VRE2 within zic3 promoter completely abolished the repression caused by Ventx1.1. In addition, we found both the positive and negative regulation of zic3 promoter activity by FoxD5b and Xcad2, respectively, and that these occur through the VREs and via modulation of Ventx1.1 levels. Taken together, the results demonstrate that the BMP4/Smad1 target gene, Ventx1.1, is a direct repressor of neuro-ectodermal gene zic3 during early Xenopus embryogenesis.

대청룡탕(大靑龍湯)이 DNCB로 유발된 생쥐의 아토피피부염에 미치는 영향 (Effects of Daecheonglyong-tang on Atopic Dermatitis Induced by DNCB in Mice)

  • Objectives : This study was designed to examine the effects of Daecheonglyong-tang(DCL) on atopic dermatitis induced by DNCB in mice Methods : The Nc/Nga mice were divided into 5 groups, and four groups excluding the normal group were applied by 2,4-dinitrochlorobenzene(DNCB), to cause AD and were orally administered with distilled water(negative control), dexamethasone(positive control), and DCL 200 or 400mg/kg once a day for 4 weeks respectively. The visual changes on skin, changes in skin tissue thickness and eosinophil infiltration were observed. IgE, Histamine, Cytokines, immune cells and the amount of gene expression of filaggrin, VEGF, $TGF-{\beta}1$, EGF were measured. Results : Dermatitis score showed a gross improvement on all DCL groups, similar to or better than positive control. All DCL groups showed no significant change in the basophils, while neutrophils and eosinophils decreased. In only DCL 400 mg/kg groups, white blood cells and mononuclear cells were decreased and lymphocytes were increased. In particular, neutrophils had similar or better effects than the positive control. In all DCL groups, IgE, Histamine, $IL-1{\beta}$, IL-4, IL-5, IL-6 and $TNF-{\alpha}$ were decreased and IL-2 was increased. In only DCL 400 mg/kg groups, IL-10 decreased and $IFN-{\gamma}$ increased. In particular, $IL-1{\beta}$ and $IFN-{\gamma}$ showed a similar rate of increase and decrease comparing positive control in DCL 400 mg/kg. $TGF-{\beta}$1 was increased in all DCL groups, filaggrin and VEGF were increased in only DCL 400 mg/kg groups. EGF did not make any changes. Epidermis, dermis thickness and eosinophil infiltration were also decreased in all DCL groups. Conclusions : By increasing Th1 cytokine and decreasing Th2 cytokine, DCL extracts appear to be effective in controlling immune response imbalances, anti-inflammatory and skin regeneration and are likely to be available as a treatment for AD.
