애기달맞이꽃(Oenothera laciniata Hill) 추출물의 항균활성 (Antibacterial Activities of Oenothera laciniata Extracts)

  • The solvent extracts of Oenothera laciniata, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservative. The O. laciniata extract by 80% ethanol was sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and butanol. The antibacterial activities and cell growth inhibition were investigated on each strain with the different concentrations of O. laciniata extracts. Antibacterial activities were shown in ethanol, ethylacetate, and butanol fraction of O. laciniata. However n-hexane, dichloromethane and water fraction showed weak antibacterial activity against the tested microorganisms. Among the five fractions, ethylacetate fraction showed the highest antibacterial activities against microorganisms tested, such as Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella Enteritidis and Salmonella Typhimurium. The polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess high antibacterial activity. The polyphenolic compounds from ethanol, n-hexane, dichloromethane, ethylacetate, butanol, and water fraction were 63.96 mg/g, 8.49 mg/g, 28.11 mg/g, 172.64 mg/g, 114.56 mg/g, and 34.91 mg/g, respectively. There are some relationships between antibacterial activity and polyphenol content in natural plant. The ethylacetate fraction could be suitable for the development of a food preservative.

한약재 및 채소류 물추출물 첨가에 의한 깍두기 숙성 적기의 연장 효과 (Effect of Hot Water Extract of Natural Plants on the Prolongation of Optimal Fermentation Time of Kakdugi)

  • To investigate the effect of natural plants on the prolongation of optimal fermentation time of kakdugi, various kakdugies, added with hot water extract from 105 kinds of natural plants(68 medicinal plants and 37 vegetables), were fermented at 20oC until optimal fermentation time. In case of control without addition, the time required to reach the optimum acidity(0.6% lactic acid) was 60 hr. Among 105 kinds tested, 48 plant(42 medicinal plants and 6vegetables) extracts decreased the acidity after 60 hr fermentation to less than 50% of control. In addition, these extracts extended the optimal fermen tation time(>120 hr) by more than 2 folds. Moreover, among the above 48 kinds, 12 sorts of extracts raised the hardness of kakdugi, by more than 30%, compared to control. And the number of lactic acid bacteria in kakdugi added with the above 12 kinds was not smaller than that of control. In sensory test, 8 kinds of medicinal plants(including Phyllostachys edulis) were found to be acceptable. Based on these results, it is suggested that kakdugi added with 8 kinds of medicinal plants was adequate in the prolongation of optimal fermentation time of kakdugi.

제주 자생식물로부터 항산화 및 화장품 기능성 소재 탐색 (Screening of Antioxidants and Cosmeceuticals from Natural Plant Resources in Jeju Island)

  • 제주 자생식물 54종을 대상을 70% 메탄올로 추출하여, 총페놀 함량, DPPH radical 소거능, xanthine oxidase(XOD), tyrosinase 저해활성을 측정한 후, 생리활성이 높은 5가지 식물을 선정하여 고압 유기용매(100% 메탄올, 13.6 MPa, $40^{\circ}C$)로 추출하여 DPPH, tyrosinase, elastase 저해활성을 측정하였다. 70% 메탄올 추출물의 총페놀 함량은 새우나무, 이질풀, 아그배나무, 자금우, 짚신나물이 250 mg GAE/g 이상으로 높은 함량을 나타내었다. DPPH radical 소거활성은 백량금이 94.1%로 가장 높았다. DPPH radical 소거활성과 총페놀 함량 사이에는 높은 상관관계($R^2=0.87$)를 나타내었다. Tyrosinase 저해활성은 이삭여뀌, 붉나무, 사방오리나무, 소귀나무가 85% 이상을 나타내었으며, XOD 저해활성은 이삭여뀌와 소귀나무가 90% 이상을 나타내었다. 고압 유기용매 추출물은 70% 메탄올 추출물과 마찬가지로 DPPH 소거능과 tyrosinase 저해활성이 높았다. Tyrosinase 와 elastase의 저해활성에 대한 $IC_{50}$은 자금우가 각각 802와 88 ppm이었고, 소귀나무가 각각 959와 66 ppm이었다.

  • For developing plant anti-inflammatory agents, extracts from Lonicera japonica were obtained and evaluated for their anti-inflammatory and analgesic activities using acute/chronic inflammatory models and writhing tests. It was found that the extracts with a modified extraction method showed higher anti-inflammatory and analgesic activities than those of the extracts based on the ancient literatures.

Inhibitory Effects of Plant Extracts on Tyrosinase Activity and Melanin Synthesis

  • In order to develop a new skin whitening agent, approximately 100 plant extracts were evaluated for their inhibitory activities against melanin biosynthesis in cultured mouse melanocyte melan-a cells. As a result, seven extracts exhibited over 50% inhibition of melanin synthesis compared to control at a concentration of $20\;{\mu}g/ml$. In particular, Aster ageratoides Turcz. var. ageratoides (branch, root, aerial, flower; $IC_{50}$ = 17.3, 6.1, 13.6, $12.9\;{\mu}g/ml$, respectively) and Physalis alkekengi var. francheti (leaf, unripen fructus, aerial; $IC_{50}$ = 6.5, 28.3, $23.9\;{\mu}g/ml$) markedly inhibited melanin synthesis. In addition, tyrosinase activity was monitored by the measurement of dopachrome formation from the oxidation of L-3,4-dihydroxyphenylalanine. Extracts of A. ageratoides Turcz. var. ageratoides (flower) and P. alkekengi var. francheti (leaf) showed the most potent tyrosinase inhibitory activity. These plants might be the potential candidate sources in the development of novel skin-whitening products.

Screening for Antioxidant Activity of Plants and Marine Algae and Its Active Principles from Prunus davidiana

  • The antioxidant activity of methanol extracts of plants and marine algae was tested by using 1,1-diphenyl-2-picrylhydrazyl(DPPH). Five plant extracts(Prunus davidiana, Eriobotrya japonica, Artemisia iwayomogi, Stirodella tolyrrhiza and Ulmus davidiana) and two algae (Ecklonia stolonifera and Symphycoladia latiuscula) were found to be the most effective in DPPH radical scavenging activity. The methanol extract obtained from the stems of Prunus davidiana was fractionated with several solvents. The ethylacetate soluble fraction exhibiting the strongest antioxidant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Antioxidant flavonoids and flavonoid glycosides were isolated and the most active ones was identified as (+)-catechin by MS, $^1H-NMR$ and $^{13}C-NMR$. Its antioxidant activity was higher than that oil vitamin C.

국내 자생 자원식물들의 항산화 활성 검색 (Screening of Natural Herb Resources for Anti-oxidative Effects in Korea)

  • 자생식물의 항산화 활성 탐색을 위하여 140종의 식물을 80% ethanol로 추출하여 활성을 확인하였다. 그 결과 며느리배꼽이 가장 높은 항산화 활성을 나타내었고(VitaminE, ORACPE=1.0), 쉬땅나무, 찔레나무, 좁쌀풀, 갈참나무, 신갈나무 그리고 싸리의 순으로 항산화활성이 높은 것으로 조사되었다(ORACPE ${\geq}$ 1.50). 또한 80% 에탄올 추출물의 용매별(n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH, water) 분획층 ORAC 결과에서도 며느리배꼽과 쉬땅나무가 각각 $1.13{\pm}0.002$, $1.10{\pm}0.004$, $3.67{\pm}0.232$, $1.62{\pm}0.049$, $1.05{\pm}0.003$; $1.02{\pm}0.007$, $v1.00{\pm}0.016$, $2.94{\pm}0.130$, $1.80{\pm}0.019$ 그리고 $1.04{\pm}0.012$로 높은 값을 나타내었다. 그 중 항산화 활성이 가장 높은 EtOAc 분획층의 유효성분 규명 등의 추가 연구가 필요하다.

Inhibitory Effect on Replication of Enterovirus 71 of Herb Methanol Extract

  • Anti-enterovirus 71 (EV 71) activities of fifteen herb plant species extracts were examined by SRB assay, among which Origanum vulgare and Rosmarinus officinalis (Anna Rosemary) extracts exhibited the activities with $IC_{50}$ of 8.28 and $8.17\;{\mu}g/mL$, respectively. Their 50% cytotoxicity concentrations ($CC_{50}$) were 691.89 and $1104.19\;{\mu}g/mL$, and the therapeutic indices were 83.56 and 135.15, respectively. Amantadine (positive control) showed anti-EV 71 activity with 50% inhibitory concentration and $CC_{50}$ of 4.46 and $145.22\;{\mu}g/mL$, respectively. Addition of the methanol extracts of O. vulgare and R. officinalis (Anna Rosemary) in EV 71-infected Vero cells strongly inhibited the formation of visible cytopathic effects without changing the normal morphology of the cells. These results indicate that methanol extracts of O. vulgare and R. officinalis (Anna Rosemary) may contain antiviral compound inhibiting the EV 71 replication.

Inhibitory Activity of Chinese Medicinal Plants on Nitric Oxide Synthesis in Lipopolysaccharide -Activated Macrophages

  • Nitric oxide (NO) produced in large amounts by the inducible nitric oxide synthase (iNOS) is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidate for the treatment of inflammatory diseases accompanied by the overproduction of NO. We prepared alcoholic extracts of Chinese medicinal plants and screened their inhibitory activity against NO production in lipopolysaccharide (LPS)-activated macrophages. Among the 80 kinds of extracts of herbal drugs, 15 extracts showed potent inhibitory activity of NO production above 80% at the concentration o$50\mu\textrm{g}/ml$. These potent extracts showed dose dependent inhibition of NO production of LPS-activated macrophages at the concentration of 50, 30,$10\mu\textrm{g}/ml$. Especially, Rhus chinensis, Senecio scandens and Wikstroemia indica showed most potent inhibition above 50% at the concentration of $10\mu\textrm{g}/ml$. These plants are promising candidates for the study of the activity-guided purification of active compounds and would be useful for the treatment of inflammatory diseases and endotoxemia accompanying the overproduction of NO.

Arginase II Inhibitory Activity from Crude Drugs

  • Arginase competitively inhibits nitric oxide synthase (NOS) via use of the common substrate L-arginine. Arginase II has recently reported as a novel therapeutic target for the treatment of cardiovascular diseases such as atherosclerosis. In our experiment, the EtOH extracts of four-hundreds extracts drugs were investigated for the arginase inhibitory activity. Among them, four extracts exhibited over 50% inhibition of arginase II activity compared to control at a concentration of 150${\mu}g/ml$. In particular, the seed of Arctium lappa, gum-resin of Boswellia carterii, aerial part of Artemisia apiacea and rhizome of Cyperus rotundus inhibited arginase II activity, with $IC_{50}$ values of 118.4, 135.4, 123.9 and 86.7${\mu}g/ml$, respectively. In addition, four plant extracts showed less than 20% inhibition of arginase I activity at 150${\mu}g/ml$. These plants might be the potential candidate materials in the development of the novel atherosclerosis drug.
