Background: Studies on the existence and activation of antioxidants in the wild Erigeron canadensis are still incomplete. Purposes: The activity of antioxidant substances was studied by extracting E. canadensis with ethyl alcohol. Methods: After washing the wild turfgrass, extraction with ethyl alcohol was used to investigate the activity of antioxidant substances using various analytical instruments. Results: The highest yield ratio of the extract was 49.3%, and it varied according to temperature and ethyl alcohol ratio. The 50:50 of water to ethyl alcohol at 180℃ was the highest yield. 100% survival rate was in the untreated group and 98-100% in the experimental group at 50 ppm or more of the extract. There was no cytotoxicity at almost all concentrations. The extract of 25 ppm was suppressed by 42% in the test group. The extract of 50 ppm reduced the free fatty acid content by 15%. Cell viability was 20% at the extract concentration of 50 ppm and 24% at 100 ppm. At an extract of 500 ppm, the free oxygen scavenging ability using DPPH and ABTs was 90-98%. When the changes in the free radical scavenging activity of DPPH and ABTs were observed in three dimensions, the antioxidant activity tended to increase at 85℃ as the temperature increased. However, at 85~130℃, it showed a rather decreasing tendency as the temperature increased. Conclusions: There were antioxidants in the ethyl alcohol extract of the wild grass, there was almost no cytotoxicity and suppressed NO production, and the scavenging function of free oxygen was also high. These results provide primary data for the various natural healing uses of the extracts of the turfgrass.

Peroxiredoxin 6 (Prx 6) is a member of the thiol-specific antioxidant protein family, which may play a role in protection against oxidative stress and in regulating phospholipid turnover. The aim of this study was to determine whether a human Prx 6/Luc vector was stably expressed and responded to antioxidants in a lung cell line (NCI-H460). To achieve this, the luciferase signal, hPrx 6 mRNA expression, and superoxide dismutase (SOD) activity were measured in transfectants with a hPrx 6/Luc plasmid after treatment with four antioxidant extracts, including Korea white ginseng (KWG), Korea red ginseng (KRG), Liriope platyphylla (LP), and red Liriope platyphylla (RLP). First, the hPrx 6/Luc plasmid was successfully constructed with DNA fragments of human Prx 6 promoter, amplified by PCR using genomic DNA isolated from NCI-H460 cells, and cloned into the pTransLucent reporter vector. The orientation and sequencing of the hPrx 6/Luc plasmid were identified with restriction enzyme and automatic sequencing. A luciferase assay revealed significant enhancement of luciferase activity in the four treatment groups compared with a vehicle-treated group, although the ratio of the increase was different within each group. The KRG- and LP-treated groups showed higher activity than the KWG- and RLP-treated groups. Furthermore, the luciferase activity against RLP occurred roughly in a dose-dependent manner. However, the level of endogenous hPrx 6 mRNA did not change in any group treated with the four extracts. The SOD activity was in agreement with the luciferase activity. Therefore, these results indicate that the hPrx 6/Luc vector system may successfully express and respond to antioxidant compounds in NCI-H460 cells. The data also suggest that the Prx 6/Luc vector system may be effectively applied in screening the response of hPrx 6 to antioxidant compounds in transgenic mice.
Optimal extraction method for antioxidant compounds from coat of grape dreg was established. Extracts were prepared with ethanol solutions containing phosphoric, formic, acetic, HCl, TFA, and citric acids. Antioxidant compounds of grape coat were determined by HPLC analysis and evaluated for antioxidizing effects using in vitro model system. Peonidin-3-glucoside content was the highest in 0.1% HCl-added ethanol extract. The extract prepared from pure ethanol without organic acids showed the highest content of cyanidin-3-glucoside among samples tested. Resveratrol and quercetin contents, the most important antioxidants, were highest in 0,1% HCl-added extract. Electron-donating ability was high in 0.1% acetic acid-added extract. SOD-like activities were 95.08% and 94.39% in 0.1% formic and 0.1% citric acid extracts, respectively. Inhibitory effects on peroxidation of egg yolk lecithin were observed in phosphoric (60.51%), citric (59,27%), and formic acid (56,77%) extracts. Hydrogen radical-scavenging activity was 59.47% in 0,1% HCl extract. Results suggest addition of 0.1% HCl in ethanol solution affords the highest yield in antioxidant compounds and antioxidant activities.

Phytochemicals in Codonopsis lanceolata leaves were saponins, triterpenes, tannins, and flavonoids, not alkaloids. The levels of total polyphenols and flavonoids in Codonopsis lanceolata leaves were measured to evaluate the antioxidant activity. C. lanceolata leaves showed strong 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and potent reducing power. In addition, C. lanceolata leaf extracts inhibited production of nitric oxide (NO) in lipopolysaccharide (LPS)-activated RAW 264.7 cells. To examine active phytochemical for antioxidant activity, aglycone fraction of C. lanceolata leaves was analyzed by high performance liquid chromatography (HPLC). Luteolin was identified as a main component of aglycone fraction and showed potent antioxidant activity as determined by a DPPH radical scavenging assay and reducing power test. These results suggest that C. lanceolata leaves are a good source of antioxidants.
This study aims to investigate the effects of hydrogen peroxide and grapefruit seed extract used as a chemical and natural disinfectants on human conjunctival cells in vitro. The main component of grapefruit seed extract is a narigin. It is one of the flavonoid types in citrus fruits and f1avonoids are widely recognized as naturally occurring(삭제) antioxidants. Cytotoxicity was determined by mitochondrial activity(MTT assay) and DNA damage was analyzed by measuring Comet assay. In LDH assay, 5% of grapefruits seed extract has been observed as a material is giving recovery effect of damaged cultured conjuctival cells by hydrogen peroxide. And also, each of concentrations has been treated simultaneously with same amounts and cytotoxicity of hydrogen peroxide and grapefruit seed extract have been estimated by LDH leakage assay after 24 hours. In conclusion, H2O2-induced cytotoxicity, apoptosis were Significantly prevented by grapefruit seed extract. It is a main component of bioflavonoids that we can simply take it as food. The present results suggest that grapefruit seed extract is a useful disinfectanct having antioxidant and antiapoptopic activity as a natural product.

This study investigated that the antidiabetic effects of banaba extracts with variety solvents selectivity in vitro and in vivo. Banaba extracts were prepared with water, 70% ethanol, 90% ethanol, 100% ethanol and water-ethanol that of extract twice times sequentially water and ethanol. Cell toxicity and insulin secretion of banaba extracts was tested by MTT (3-[4,5-dimetylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay on hamster insulinoma cell line, HIT-T15. Also we tested that insulin, body weight, blood glucose, total cholesterol, HDL-cholesterol, triglyceride and free fatty acid in streptozotocin diabetic rats. Water-ethanol extract has remarkable antidiabetic effect compare with the other banaba extracts. For water-ethanol extract has both of hydrophilic and hydrophobic antidiabetic materials from banaba. Expecially, corosolic acid, as known as unique polyphenol, has antidiabetic effect studied by many researchers till nowadays. But corosolic acid does not solve in water. Otherwise, we suggest that banaba extract of hydrophilic and hydrophobic materials (polyphenol and antioxidants) mixture more increased antidiabetic effects.
This study was carried out to investigate the antioxidative activities of Rosa davurica Pall for the purpose of development of novel antioxidant from natural products. Antioxidant activities of four different parts of Rosa davurica Pall such as fruit, leaf, stem and root were examined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The methanol extract from the root of Rosa davurica Pall showed the highest antioxidative activity among 16 samples tested. And, we also tested radical scavenging effects of 5 different extract compartments(Hexane, $CHCl_3$, EtOAc, BuOH and $H_2O$ fraction). EtOAc and BuOH fractions from the root of Rosa davurica Pall exhibited antioxidative activities higher to those of natural, ${\alpha}-tocopherol$ or synthetic antioxidants, BHT. The antioxidative substance of EtOAc fraction from the root of Rosa davurica Pall was successively purified with silica gel adsorption column chromatography and Sephadex LH-20 column chromatography. The purified active substance was isolated as crystal and identified as (+)-catechin by $^{l}H-NMR$ and $^{13}C-NMR$. This compound exhibited DPPH radical scavenging activity with the $IC_50$ value of $1.7\;{\mu}g/ml$. In the analysis of catechin content, the leaf extracts contained the highest catechin, and fruit extracts contained the lowest catechin. Considering antioxidative activity on DPPH assay, the extracts of Rosa davurica Pall showed a possibility to be used as a new material for natural antioxidant and functional food.

Abalone, a marine organism inhabiting the west coast of Korea, has antioxidant and anti-inflammatory effects, and is a resource with potential to be used in various industries such as antibiotic development and cosmetic raw materials. In this study, we chose abalone among various marine lives on the west coast. Antibacterial peptide (AMP) was separated from abalone and its derivative Ab4-7 was identified and its physiological activity was studied. The treatment of Ab4-7 in inflammatory RAW 264.7 to check the anti-inflammatory efficacy nhibited inflammatory cytokines, TLR4, TNF-α, IL-1β, COX-2, and iNOS and increased mRNA manifestation of HO-1, genes related to antioxidants. Based on the strong antioxidant and anti-inflammatory effects of Ab4-7, the effects of Ab4-7 in the inflammatory RAW 264.7 cells were identified through RT-PCR, which regulates the gene Matrix Metalloproteinase (MMPs) that induces a variety of inflammatory skin diseases by engaging in the decomposition of the extrocellular matrix metalloproteinase (ECM). Taken together, it is concluded that Ab4-7 has a powerful anti-inflammatory and antioxidant effect and can be used as a functional material for various inflammatory skin disease treatments by controlling the genes associated with matrix metalloproteinase (MMPs).

This work was carried out to investigate the antioxidative effects of Acanthopnax sessiliflorus from Jeongseon County for the purpose of development of a novel antioxidant from natural products. The antioxidant activity was determined by using electron paramagnetic resonance (EPR), not measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) which have bun used for antioxidant activity of natural sources. Although DPPH radical scavenging activity assay have been generally used for antioxidant activity, this assay is nut appropriated for determinating which radical is scavenged by extracts from natural products. Using EPR, we determinated whether A. sessiliflorus extracts from Jeongseon County scavenge specific radicals or not. On experiment of scavenging superoxide anion radical, hydroxyl radical, nitrogen dioxide and peroxinitrite. Extracts from A. sessiliflorus showed high antioxidant activities to reactive oxygen and nitrogen species. These result suggest that extracts from A. sessiliflorus act as an antioxidant by scavenging reactive oxygen and nitrogen species and used as new cosmetic ingredients for anti-oxidative stress in skin.
Contemporary society, the era of the 4th industrial revolution, has been enjoying a boom in the cosmetics industry due to the launch of cosmetics through new technology development due to the interest in the beauty industry. However, the cosmetics industry is booming, but dry eye syndrome, skin trouble, Hormonal disturbances, and many other side effects. In order to prove the universal use value of pine needles as a natural cosmetic material that can complement the side effects of existing chemical ingredients cosmetics harmful to human body, and to propose a roll model of desirable natural cosmetics, DPPH radical scavenging activity, ABTS assay, and flavonoid content were studied. The results showed that both the DPPH radical and ABTS radical scavenging activity increased in both the $70^{\circ}C$ and $100^{\circ}C$ pine water hot water extracts. The extracts from the pine tree extracts at $70^{\circ}C$ and $100^{\circ}C$ showed $32.0{\pm}2.5mg\;QE/ml$ and 116.8 A total flavonoid of ${\pm}5.5mg\;QE/ml$ was detected. Therefore, it is considered that the pine needle hydrothermal extract having a large amount of antioxidant which is advantageous to human body is highly applicable as a natural cosmetic material harmless to the human body.
