Inhibitory Effects of Leachianone A from Sophora fIavescens Ait. against Cadmium Induced Cytotoxicity (고삼으로부터 분리된 Leachianone A의 카드뮴에 대한 독성억제 효과)

  • In this study, cytotoxicity of cadmium on NIH 3T fibroblasts was utilized in order to discover antitoxic compound in methanol extract of Sophora fIavascens Ait. There were treatment groups; control (medium only), $MTT_{50}$ group and five experimental groups. MTT {3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H- tetrazoliumbromide} assay was performed to evaluate the cytotoxicity of cell organelles and $IC_{50}$ was also measured. Accordingly we have examined the detoxification effects of methanol extract of S. flavescens Ait. and leachianone A (LA) on cadmium-treated NIH 3T3 fibroblasts ($IC_{50}=\;12.5{\mu}M$) to observe morphological changes by the light microscopy. Both S. flavescens Ait. methanol extract and LA showed inhibitory effects on cadmium-induced cytotoxicity. Furthermore, LA showed dose-dependency in detoxication. From these results, it is conceivable to suggest that LA from S. flavescens Ait. methanol extract is a potential antitoxic agent.

  • In this study, hot water and ethanol, acetone, and hexane extracts of the steamed Deodeok were investigated for use as raw materials for natural cosmetics. Hot water and ethanol, acetone and hexane extracts of steamed Deodeok did not indicate cell toxicity up to 300 ㎍/mL concentration in the MTT assay. The tyrosinase inhibition effects of the ethanol extract were higher than the ascorbic acid. The efficacy of the steamed Deodeok's ethanol extract on the elastase inhibition was similar to that of quercetin at higher concentrations. In conclusion, water, ethanol, acetone and hexane extracts of the steamed Deodeok indicated strong possibility as a whitening and anti-wrinkle functional cosmetic ingredient. Among them, the steamed Deodeok's ethanol extract, which showed excellent activity, is highly available because it has high safety compared to other organic solvent extraction methods used in cosmetics, and has minimal irritation to the skin.

Dyeing Characteristics of Herb Dyed Fabrics Treated with Mordants - Focused on Sage and Rosemary Extract - (매염제 처리에 의한 허브 염색직물의 염착 특성 - 세이지와 로즈마리 추출물 중심으로 -)

  • Unlike synthetic dyes, natural dyes do not affect the human body or create atmosphere pollution that are also mothproof, antibiotic and sterilized. This study examined the changes in dye adsorption properties according to mordant types and mordanting method to qualify trends using sage and rosemary extracts. Dyeability of sage extract is often similar to rosemary extract regardless of dyeing conditions. Dyeing properties of herb colorants on cotton, silk and wool fiber and effect of dyeing conditions on dye uptake were also compared. Dyed fabric showed a yellowish color and the dyeing affinity was somewhat good with rosemary extract. The surface color of the dyed fabrics was darkened by mordant treatment. The K/S value of herb colorants of protein fiber was higher than cellulose fiber. The concentration of mordant having the highest dyeing affinity was different from the mordant type. Regardless of the type of fiber, the K/S value of premordanting was the highest in mordanting method, and the K/S value of simultaneous mordanting indicated no noticeable mordant effect. The fastness of washing of silk fabric was reduced the most by mordant treatment. The lightfastness was reduced in Al mordant treatment regardless of the type of fiber. The lightfastness of wool increased somewhat by mordant treatment.

  • The purpose of this investigation was to extract the bioactive agents from Alpinia officinarum Hance. The methanol with ethylacetate extracts alone and combined were examined for their activities against VRE (vancomycin-resistant enterococci) and pathogenic yeast in vitro. The incidence of infections caused by VRE and other pathogenic microorganisms and the importance of using novel synergistic drug combinations has become important. Previously, we reported the antimicrobial effects of the butanol extract from Lonicera japonica and have evaluated combinations of solvent extracts, with a focus on the MeOH and EtOAc extracts from A. officinarum. In the present study, enhanced inhibitory effects were achieved by employing a combination of the two solvent extracts. The MeOH and EtOAc combination was especially effective against four VRE strains: E. faecalis (K-10-22), E. faecaium (K-11-212), E. faecalis (K-10-57) and E. faecalis (K-10-361) with MIC values of 12.5, 12.5, 6.25 and 25 ${\mu}g/ml$, respectively. Thus, the combination was more effective than other antibiotics such as kanamycin, gentamicin or tetracycline against bacteria including E. coli, Staphylococcus aureus, and Micrococcus luteus. In addition, the combination was effective against yeasts such as Candida albicans, Candida tropicalis and Cryptococcus neoformans.

  • Opuntia Ficus has been an epiphyte plant since it was introduced in Cheju island as an ornamental plant two hundred years ago. And now it is grown as many as it is designated to local souvenir No.35. The fruit of this plant contains red-purple sap, which is due to be used in coloring clothes. This study aims at confirmation of use possibility as eco-friendly dyeing stuffs on extract from opuntia ficus fruits and contributing to the practical use of natural dyeing. Thus this study was investigated the dyeability of extract from opuntia ficus fruits on the silk fabric. Antioxidants were used ascorbic acid, L-cysteine, prophyl gallete. After dyeing, K/S and dye fastness were measured. The results were as follows; Maximum absorption band of extract from opuntia ficus fruits was 534.5nm. The appropriate dyeing condition was bath ratio 50:1, dyeing temperature $30^{\circ}C$, dyeing concentration 50%, dyeing time 50min, pH 3,concentration of all antioxidants 3%. Addition of antioxidants brought increase of K/S value. And K/S value and dye fastness had the greatest value when ascorbic acid was added

Component Analysis and Antioxidant Activity of Wasabi japonica Matsum Leaves (고추냉이 잎의 함유성분 분석과 항산화 활성)

  • The purpose of this study was to determine the possibility of using Wasabi japonica Matsum leaves as natural health food source. To accomplish this purpose, the contents of general and antioxidative nutrients of Wasabi japonica Matsum leaves were measured. The contents of carbohydrate, crude protein, crude lipid and ash are 53.41%, 25.00%, 7.95% and 13.64%. And the calories of Wasabi leaves was 385.23 Kcal. Total dietary fiber was 52.27%. The K was the largest mineral followed by Ca, P, Mg which means Wasabi leaves is alkali material. The contents of sinigrin and allyisothiocyanate in the wasabi leaves were 69.2 mg/g and 241.0 mg/g, respectively. Total phenol contents of the hot water extract and the 70% ethanolic extract were $19.44{\pm}0.23$ and $19.33{\pm}1.17mg$ GAE/g, respectively. The total flavonoids content of the hot water extract and the 70% ethanol extract were $7.69{\pm}0.71$ and $19.25{\pm}1.41mg$ QE/g, espectively. The general nutrients and other antioxidant bioactive materials in Wasabi japonica Matsum leaves were also potential materials for good health food.

Assessment of Antioxidant Activity of Garlic (Allium sativum L.) Peels by Various Extraction Solvents

  • This study was carried out to investigate the antioxidant activities, total phenol, and flavonoid contents of 70% methanol, 70% ethanol, and chloroform-methanol (CM, 2:1, v/v) extracts from garlic (Allium sativum L.) peels. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ${\beta}$-carotene bleaching assay, and ferric reducing antioxidant power. Total phenol and flavonoid were in a range of 32.59-63.05 mg CAE/g, 5.26-9.22 mg QE/g, respectively. Total phenol and flavonoid were predominant with the values of $63.05{\pm}0.20mgCAE/g$ and $9.22{\pm}0.17mgQE/g$ in 70% ethanol extract. Additionally, 70% ethanol extract also displayed the strongest antioxidant activity followed by 70% methanol extract and CM extract in all assays. These results showed that garlic peel can be used as natural antioxidant in nutraceutical preparations to prevent human diseases.

Antigastritic and Antiulcerative Action of the Extract of Zingiberis Rhizoma (생강추출물의 항위염 . 항궤양 작용)

  • The rhizoma of Zingiber officinale has been used as antiemetic, expectorants, stomachache relieving drugs and digestive accelerators. From the observation of antigastritic action of the methanol extract of the rhizoma, it was fractionated with hexane, chloroform, ethyl acetate and butanol, followed by bioassay on antigastritic and antiulcerative activity. The hexane and the chloroform fraction reduced significantly HCl ethanol induced gastric lesion at the dose of 370 and 210 mg/kg, p.o., respectively. On the gastric ulceration and gastric secretion in pylorus-ligated rats, the hexane fraction decreased the volume of gastric secretion and acid output, and also increased pH at the dose of 370 mg/kg, i.d.. It showed considerable curative ratio of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Zingiberis rhizoma exhibited antigastric and antiulcerative activity which might be attributable to inhibition of gastric secretion. It is revealed that the active component may be present in the hexane fraction.

Antioxidative Effects of Mushroom Flammulina velutipes Extract on Polyunsaturated Oils in Oil-in-water Emulsion

  • The antioxidative activities of the water solution of crude extract from edible mushroom enokitake Flammulina velutipes were compared with those of ascorbic acid and ascorbic acid 6-palmitate in oil-in-water (O/W) emulsions of cod liver oil. Oxidation of the emulsions was carried out at 40 and $50^{\circ}C$ in the dark. The antioxidant activities were measured by in vitro assay against oxygen uptake, 2-thiobarbituric acid value, hydroperoxide formation of the oils. Also, residual docosahexaenoic acid content was measured as indices of lipid oxidation. The cod liver oil in O/W emulsions with added enokitake crude extract (ECE) was significantly more stable against lipid oxidation than the control emulsions without the extract in terms of any oxidation indices used. Moreover, ECE provided remarkable antioxidative properties to eicosapentaenoic acid ethyl ester in emulsion system. These observations demonstrate that F. velutipes can be used as a natural antioxidant, which effectively prevents oxidation of polyunsaturated oils in emulsion system.

Lysosomal Enzyme Inhibiting Activity of Alkaloidal Fraction from Tylophora indica Leaves in Arthritic rats

  • Ethanolic extract (50% v/v) and alkaloid fraction of Tylophora indica leaves were examined for lysosomal enzyme inhibitory activity in adjuvant-induced arthritic rats. The alkaloid fraction showed statistically significant inhibition of arthritic lesions (p < 0.05) from day 18, (p < 0.025) from day 20 and (p < 0.001) from day 21 onwards in the adjuvant-induced arthritis, which was comparable to the response of standard drug Indomethacin. The ethanolic extract was less significant than the alkaloidal fraction in inhibition of arthritis. Alkaloid fraction showed significant (p < 0.001) inhibitory effect on the lysosomal enzyme activities in adjuvantinduced arthritic rats. It also significantly prevented decrease in collagen levels and synovial damage observed during arthritis and also inhibited increase in urinary excretion levels of collagen degradation products like hydroxyproline, hexosamine, hexuronic acid, etc. Both ethanolic extract as well as the alkaloid fraction, however, did not show any significant activity in normal nonarthritic rats. The ethanolic extract and the alkaloid fraction may thus be able to inhibit the progress of inflammation and inhibit the destructive activity of lysosomal enzymes on structural macromolecules like collagen etc. in the synovial capsule in joints during arthritic states. They may thus prevent synovial damage observed during arthritis.
