Effect of KW21 and water-extracted horseradish leaf combination on Nannochloropsis sp. density in laboratory scale

  • This study aims to know the effect of water-extracted horseradish Moringa oleifera leaf and KW21 fertilizer combination application on the density of Nannochloropsis sp. It was conducted in the Natural Food Laboratory of State Polytechnique, Tual. The experiment used complete randomized design with 5 treatments and 3 replications: A (25% horseradish leaf extract + 75% KW21), B (50% horseradish leaf extract + 50% KW21), C (75% horseradish leaf extract + 25% KW21), D (positive control of 100% KW21), and E (negative control of 100% horseradish leaf extract). Results showed that Treatment C yielded the best result, both the highest density of Nannochloropsis sp. and suitable harvest time.

Effect of Curcuminoids and Natural Plants Extract Mixture on the Cardiovascular System in Rats (흰쥐에서 Curcuminoid 및 이를 함유한 천연식물 혼합물이 심혈관계에 미치는 영향)

  • Antiplatelet aggregation, anticoagulant and lipid-lowering drugs are clinically widely used for secondary preventive purpose in the cardiovascular patients, but there is no primary preventive agents to prevent these diseases. With the aim of developing effective primary agents for cardiovascular diseases, we tried to formulate an optimized mixture of natural plants extract containing Theae sinensis, Camelliae sinensis, Vitis vinifera, Gingko folium and curcuminoids from Curcuma longa and to evaluate its anti-thrombotic and anti-hypercholesterolemic effects in vivo. The inhibitory effect of curcuminoids on vascular smooth muscle cell proliferation and migration were also investigated in vitro. in the animal experiments treated with hyperlipidemic diet, oral treatment of curcuminoids and natural plants extracts mixture (100 mg/kg) into male Sprague Dawley rats for 7 week simultaneously inhibited platelet aggregation as well as improved lipid profile in the blood. Compared to control group, both of curcuminoids-treated and mixture-treated groups revealed significantly decrease of total cholesterol (24.4%, 28.6%), free cholesterol (25.1%, 24.0%), cholesterol ester (14.6%, 29.0%), LDL-cholesterol (27.0%, 32.0%) and triglyceride (15.0%, 31.0%), respectively. However, both groups showed increase of HDL-cholesterol (46.6% and 51.5%) . In particular, atherogenic index of curcuminoids and mixture treatment group was significantly decreased to 47.0% and 56.0%, respectively. Furthermore, oral treatment of curcuminoids and mixture significantly inhibited collagen-induced platelet aggregation (21.1% and 29.1%, respectively), compared to control group. The anti-thrombotic values of mixture was almost similar to that of aspirin treatment (100 mg/kg) group. These results suggest that the oral treatment of curcuminoids-based natural plant extract mixture improved cardiovascular conditions in hyperlipidemic rats.

Evaluation of Antioxidant and Anticancer Activity of Steam Extract from The Bamboo Species

  • Natural plant extract has been the subject of intense research aiming in elucidating the underlying mechanisms of their chemopreventive effects upon various forms of human cancers. The objective of our study was to evaluate the natural antioxidants and anticancer agent potential of Phyllostachys. The chemical composition of steam extract from Phyllostachys was carried out using GC-MS. The steam extract of Phyllostachys was dominated by monoterpenes (62.96% - 71.36%) and sesquiterpenes (23.58% - 33.13%) as the main compounds. The antioxidant activities of the steam extract was determined using a DPPH scavenging and hydrogen peroxide scavenging activity test systems. Furthermore, the amounts of total phenolics in steam extract were determined spectrometrically The steam extract of P. pubescens and P. bambusoides were presented the high activity (69.4% and 64.0%, respectively.). The steam extract from Pyllostachys species showed a hydrogen peroxide scavenging activity of approximately 50.4% - 54.6% when compared to that of the standard gallic acid. The anticancer activities of steam extract were determined using a MTT assay. Assessment of the cytotoxic effect of the steam extract on PC-3 cells showed that the P. bambusoides (20.85%) and P. pubescens (20.41%) were superior in induced cytotoxicity compared with the steam extract of P. nigra var. henonis (1.15%). Findings from this study indicated that steam extract of P. bambusoides and P. pubescens possessed potential as medicinal drug especially in prostate cancer treatment.

Antioxidant Activities of Hot Water and Ethanol Extracts from the stem of Sorbus commixta Heal (마가목 줄기 열수 및 에탄올 추출물의 항산화 활성)

  • Objectives : In order to investigate the possibility of Sorbus commixta Heal. stem (SC) as a natural material, antioxidant activities of the hot water and ethanol extracts were examined. Methods : The samples of SC were pulverized, and fractions were extracted repeatedly three times from hot water and 70% ethanol at room temperature for 2 hours. The antioxidant activities were analyzed from total polyphenol, flavonoid contents, DPPH, ABTS, hydroxyl radical, $Fe2^+$ chelating, and nitrite scavenging activity. Results : Total polyphenol contents were significantly higher in ethanol extract group ($504.39{\mu}g/m{\ell}$) than in hot water extract group ($364.64{\mu}g/m{\ell}$). Total flavonoid contents were also significantly higher in ethanol extract group ($160.09{\mu}g/m{\ell}$) than in hot water extract group ($124.59{\mu}g/m{\ell}$). DPPH, ABTS, $Fe2^+$ chelating were slightly higher in ethanol extract gorups than in hot water extract groups, and increased in a dose-dependent manner. The hydroxyl radical scavenging activity (18.42~23.61%) of ethanol extract groups were shown to be approximately twice higher than that (7.63~10.37%) of hot water extract groups at $12.5{\sim}50{\mu}g/m{\ell}$ concentration. Nitrite scavenging activities of both ethanol and hot-water extract groups were shown to be higher in a dose dependent manner at the concentration of $12.5{\sim}50{\mu}g/m{\ell}$ at pH 3.0 than at pH 1.2, and ethanol extract groups (86.55~96.64%) had higher activity than the hot water extract groups (42.59~92.63%), which was higher than that of the control group antioxidant BHT (72.96~80.11%). Conclusions : The extracts of SC displayed antioxidant activities which suggested a natural material can be developed to functional material.

Antimicrobial Activity of Extracted by Supercritical Fluid from Origanum vulgare, Cinnamomum cassia, Chamaecyparis obtusa and Scutellariae baicalensis (오레가노, 육계, 편백 및 황금의 초임계 유체 추출물의 항균 활성)

  • The variety of functional plants has an attention for new natural food preservation and natural antiseptic development. The extracts from functional plants with various methods (ethanol extraction, hot water extraction and supercritical fluid extraction) tested antimicrobial activity against 10 strains including the pathogenic and food poisoning bacteria, the yeast and fungi. The antimicrobial activities of supercritical fluid extracts were shown higher than ethanol extract and hot water extract when tested with disc-diffusion method and minimum inhibitory concentration (MIC). Antimicrobial activity of supercritical fluid extract was two times higher than ethanol extract in Cinnaonomum cassia. In addition, the supercritical fluid extractions of Chamaecyparis obtuas and the C. cassia showed the higher yield than Origanum vulgare and Scutellariae baicalensis. The supercritical fluid extract of C. cassia showed an antimicrobial activity against all strains tested. The supercritical fluid extract of S. baicalensis showed strong antimicrobial activity on Listeria monocytogenes. Supercritical fluid extraction of O. vulgare and C. obtuas showed strong antimicrobial activity on Salmonella typhimuriium. In MIC test, C. obtuas was shown the best natural material for the preparation of natural antimicrobial agent by supercritical fluid extraction. In conclusion, these results suggest that supercritical fluid extraction technique was effective to obtain functional ingredient with higher antimicrobial activity in the development of new antimicrobial reagent from natural materials.

Anti-Termite Activity of Azadirachta excelsa Seed Kernel and Its Isolated Compound against Coptotermes curvignathus

  • Azadirachta excelsa, is a plant belonging to the same genus as Indian neem (Azadirachta indica), and its use as a pesticide is reported by few studies. Despite being a different species, it is expected to have the same biopesticide potential as A. indica. Therefore, this study aims to investigate the anti-termite activity of n-hexane and methanol extracts of A. excelsa seed kernel at various concentrations against Coptotermes curvignathus. The methanol extract demonstrated greater termicidal activity than n-hexane at doses test of 2%, 4%, and 8%. It also showed 100% termite mortality on the third day of administering the 8% dose. According to the gas chromatography with mass spectrometry data, the putative main components of the n-hexane extract were hexadecanoic acid, ethyl ester (18.99%), 9,12-octadecadienoic acid (Z,Z)- (16.31%), and 9-octadecenal (16.23%). In contrast, the principal constituents of methanol extract were patchouli alcohol (28.1%), delta-guaiene (15.15%), and alpha-guaiene (11.93%). Furthermore, limonoids profiling of A. excelsa methanol extract was determined using Ultrahigh-performance liquid chromatography coupled with quadrupole-Orbitrap high-resolution mass spectrometry. The number of limonoids identified tentatively was fifteen, such as 6-deacetylnimbin, nimbolidin C, nimbolide, 6-acetylnimbandiol, 6-deacetyl-nimbinene, salannol, 28-deoxonimbolide, gedunin, nimbandiol, epoxyazadiradione, azadirone, 2',3'-dihydrosalannin, marrangin, nimbocinol, and azadirachtin. They were the same as those reported in the seed and leaves of A. indica, but its largest component in A. excelsa was 6-deacetylnimbin. As a result, the presence of these compounds may be responsible for the anti-termite activity of A. excelsa seed kernel extract. Additionally, column chromatography of methanol extract yielded 6-deacetylnimbin, which was found to be antifeedant and termiticide against C. curvignathus.

Whitening and anti-aging effects of Cistanche deserticola extract (육종용 추출물의 미백과 항노화 효과)

  • This study assessed the whitening and anti-aging effects of the Cistanche deserticola extract, to develop a cosmetic substance. The cell viability of the Cistanche deserticola extract was evaluated in B16F10 melanoma cells by the MTT (3-(4,5-dimethylthaiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay. The cell viability of the extract was determined to be 90% at 4mg/ml concentration. Furthermore, the tyrosinase, collagenase, and elastase mRNA expression level were measured by RT-PCR, using the Cistanche deserticola extract treated B16F10 melanoma cells. At 4 mg/ml concentration, mRNA expression level of tyrosinase, collagenase, and elastase was dramatically decreased to 80.9%, 37.6%, and 70.9%, respectively. The antioxidant activity of the Cistanche deserticola extract was determined by DPPH free radical scavenging. The DPPH free radical scavenging capacities ranged from 70.6% to 82.6%, when evaluated from 2 mg/ml to 10mg/ml concentrations. The effects of whitening and anti-aging of the Cistanche deserticola extracts were examined at 2, 4, 6, 8, and 10 mg/ml concentration. Tyrosinase activities were inhibited from 66.8% to 78.5%, elastase activities were inhibited from 67.6% to 79.3%, collagenase activities were inhibited from 72.3% to 83.6%, and hyaluronidase activities were inhibited from 65.8% to 69.2%, respectively. These data suggest that the Cistanche deserticola extract is effective in whitening and anti-aging; therefore, it is considered to be a functional cosmetic material in cosmetic products.

Effect of the MeOH Extract of Adventious Root Culture of Panax ginseng on Hyperlipidemic Rat Induced by High Fat-rich Diet (조직배양산삼 부정근 메탄올 추출물이 식이성 고지혈증에 미치는 영향)

  • We have investigated the effect of MeOH extract of the adventious root culture of Panax ginseng on hyperlipidemic rats induced by fat-rich diet. We also analyzed and compared the content ginsenoisdes of the cultured ginsengs by HPLC. After oral administration of the MeOH extract to hyperlipidemic rats for four weeks, the variables including body weight, cholesterol, HDL, LDL, and triglyceride levels in serum were measured. One of the cultured ginseng roots (CBN3) decreased cholesterol and LDL-cholesterol and increased HDL-cholesterol levels in serum. HPLC analysis demonstrated that CBN3 contains more $Rb_1$ and Rd ginsenosides than any other cultured ginsengs and cultivated ginseng. These results suggest that CBN3 with high contents of $Rb_1$ and Rd may be useful in lowering blood cholesterol.

A study on the Pharmaceutical & Chemical Characteristics and Analysis of Natural Omija Extract (천연 오미자 추출물의 약리, 화학적 특성 및 분석)

  • Natural Omija belongs to magnoliaceae was known to possess natural odor, taste, color, and various pharmaceutical & chemical characteristics. Omija extraction was extracted using ethanol as a solvent. Omija extract showed a light red-violet color of viscous liquid state. Some conclusions from the result of characteristic experiment were obtained as follows. From the result of antimicrobial experiment, occurrence of staphylococuss aureus and aspergillus niger as microbes was decreased according to the passage of time. This phenomenon could know that Omija component is affected to antimicrobial effect. From the result of dye experiment, fiber dyeing showed with some ivory color after dyed to cotton and silk. This phenomenon could know that Omija component is affected to dyeing effect from observation of scanning electron microscope(SEM). From the result of instrument analysis, inorganic components of K(107.30ppm), Na(2.110ppm), Ca(0.935ppm), Mg(0.891ppm), Li(0.270ppm) etcs from Omija were detected with ICP/OES, and aromatic components of benzene(10.808), a-pinene(13.996), phenol(14.183), ${\beta}$-terpene(15.840), a-terpinolene(17.616) etcs from Omija were also detected with GC/MSD.

Effects of Houttuynia cordata Thunb Extract Inhibits on the Migration and Proliferation of Vascular Smooth Muscle Cell (어성초 추출물의 혈관 평활근 세포 이주 및 증식 억제 활성에 관한 연구)

  • Houttuynia cordata Thunb.[H.cordata]belonging to Saururaceae, is a wild medicinal herb of perennial plants, and grows well in a place with a lot of shade and moisture. The medical action of H.cordata is reported to have an antitumer effect, toxicity-suppressive effect, antifungal effect, diuretic effect, and antioxidative action, but its effect hasn't been reported on cardiovascular diseases, such as ateriosclerosis and hypertension yet. This study intended to confirm the effect of the water extract of H.cordata on the migration and proliferation of rat aortic smooth muscle cells. Such results show that the water extract of H.cordata suppresses the migration and proliferation of rat aortic smooth muscle cells. It is believed that a useful clue will be offered later to the prevention of cardiovascular diseases such as ateriosclerosis and hypertension, and the development of their medicines on the basis of the fact.
