In order to provide basic data for the development of a controlled environment cultivation system and standardization of the structures, structural status and improvement methods were investigated for the fruit tree greenhouses of grape, pear, and peach. The greenhouses for citrus and grape cultivation are increasing while pear and persimmon greenhouses are gradually decreasing due to the advance of storage facilities. In the future, greenhouse cultivation will expand for the fruit trees which are more effective in cultivation under rain shelter and are low in storage capability. Fruit tree greenhouses were mostly complying with standards of farm supply type models except for a pear greenhouse and a large single-span peach greenhouse. It showed that there was no greenhouse specialized in each species of fruit tree. Frame members of the fruit tree greenhouses were mostly complying with standards of the farm supply type model or the disaster tolerance type model published by MIFAFF and RDA. In most cases, the concrete foundations were used. The pear greenhouse built with the column of larger cross section than the disaster tolerance type. The pear greenhouse had also a special type of foundation with the steel plate welded at the bottom of columns and buried in the ground. As the results of the structural safety analysis of the fruit tree greenhouses, the grape greenhouses in Gimcheon and Cheonan and the peach greenhouses in Namwon and Cheonan appeared to be vulnerable for snow load whereas the peach greenhouse in Namwon was not safe enough to withstand wind load. The peach greenhouse converted from a vegetable growing facility turned out to be unsafe for both snow and wind loads. Considering the shape, height and planting space of fruit tree, the appropriate size of greenhouses was suggested that the grape greenhouse be 7.0~8.0 m wide and 2.5~2.8 m high for eaves, while 6.0~7.0 m wide and 3.0~3.3 m of eaves height for the pear and peach greenhouses.
This study was conducted to examine whether efficiency of oocyte production from superovulated prepubertal goats. Fifteen prepubertal and twenty adult goats, maintained in a pen under natural day length and fed hay ad libitum, were pretreated with progestagen implanted CIDR for 10 days. Superovulation treatment of the goats received twice daily intramuscular injection of a total of 70 mg FSH for 3 days from Day 8 of CIDR. All the gonadotrophin treated goats were injected with 10 mg $PGF_2{\alpha}$ on Day 8 and 400~600 IU hCG in the afternoon on Day 10. Oocytes were recovered by follicle aspiration or oviduct flushing at 35 to 40 h after hCG injection through mid-ventral incision. The in vivo matured oocytes was activated by ionomycin (5 min) and 6-DMAP (3.5~4 h). The activated oocytes were cultured in mSOF medium containing 0.8% BSA at 38.5$^{\circ}C$ in an atmosphere of 5% CO$_2$, 5% O$_2$, 90% N$_2$ for 7~8 days. There was no significant difference in the mean number of CL and in vivo matured and follicular oocytes recovered. But, quality of I + II grade follicular oocytes was lower (p<0.05) in the prepubertal goat (25.0%) than the adults (52.4%). The same results were also observed in the cleavage and blastocyst rate of activated oocytcs. The cleavage and blastocyst rate from prepubertal derived oocytes were lower (p<0.05) in the prepubertal goat (54.5%, 23.3%) than the adult goat (86.8%, 46.6%). Considering overall these results, we suggest that maturation of donor goats is a major factor affecting recovered oocytes quality and in vitro development of activated goat oocytes.
In order to save foreign currency and to domesticize the dairy products, various fresh cheeses and whey drinks were developed and some physicochemical, microbiological and sensory evaluation were performed. The yield of fresh cheese was 22.3%, while that of whey 77.7%. The pH-values of fresh cheeses were $5.90{\sim}6.49$, while those of whey drinks $6.07{\sim}6.49$, and fermented whey drinks $3.97{\sim}4.91$. The acidities of fresh cheeses were $0.09{\sim}0.26%$, while those of whey drinks $0.09{\sim}0.36%$. The contents of solid substances, protein and lactose in fresh cheeses were $25.67{\sim}34.18%$, $7.45{\sim}9.11%$ and $3.61{\sim}4.14%$, while those of whey drinks $7.39{\sim}7.70%$, $0.88{\sim}0.94%$ and $4.93(\sim}6.17%$, respectively. The lactic acid contents of whey drinks varied from $0.01{\sim}0.38%$, where the content in the fermented sample was the highest. The general colony counts of fresh cheeses were $0{\sim}30/g$, while those of whey drinks $0{\sim}80/ml$. The psychrotrophs counts of fresh cheeses were $0{\sim}20/g$, while those of whey drinks $0{\sim}60/ml$. Lactic acid bacterial counts in both products were not detected except for $97{\sim}401{\times}10^8/ml$ in fermented whey drinks. E. coli and fungi were not detected in both products. In sensory evaluation of both products, the strawberry added fresh cheese was the best of fresh cheeses, while the garlic added fresh cheese was the worst. Pure whey drink was the best of whey drinks, while the ginseng added whey drink was the worst.
In general, most of the firms do not settle down in one place for their pursuit of profit. There are many reasons for the relocation of the firms; procurement of raw material, market area, transportation cost and housing cost. The aspect of national policy, firm relocation has been systemically promoted for the purpose of logistics system efficiency. Nowadays balanced regional development has been issue. Another aspect, many countries have struggled for the preoccupancy of new place because of its production cost saving and curtailment of expenditure. The aim of this article is qualitative and quantitative analysis of relocation influence factors of domestic goods firms in Korea. This article dynamically analyzed the relocation influence factors for domestic goods firms in Korea. For the analysis this article made use of complementary survey (2006) out of the 3rd national logistics survey (2005). The complementary survey conducted pre-business district, business period, relocation reason, etc. This article dynamically analyzed from the three aspects; observation of average residence time in one business district, relocation factors, influence of market area. Analysis shows that relocation of firm is very high rate and the reasons of relocation play compositeness role. The results of determinants of location, firms approach the established market area closely and the firm's relocation is influenced by market area.
Effects of major phenolic acids identified from barley residues (straw, root) on the germination of rice and 3 paddy weeds such as Echinochloa crusgalli, Cyperus serotinus, and Potamogeton distintus were evaluated to obtain the basic informations on the development of naturally occurring herbicides. The predominant phenolic compounds extracted from barley residues in both straw and root were identified as p-coumaric, p-hydroxybenzoic, ferulic, vanillic, and salicylic acids by means of paper chromatography. Total phenol content of barley straw and root at the harvesting stage was 0.169% and 0.127% per dry weight, respectively. During the decomposing process, total phenol content slightly increased and then decreased. The germination of test plants was inhibited by treatments of 4 major authentic phenolic acids identified, most significantly on rice, and less on E. crusgalli, and C. serotinus. P. distintus, however, was markedly stimulated by them as the concentration increased, and then sprouted buds of pondweeds were changed to dark brownish color, resulting in the death as the treatment prolonged. The greater inhibitory effect appeared on shoot growth rather than germination. The aqueous extracts of barley residues showed the similar inhibitory effect on the germination and shoot growth of rice and three paddy weeds as the treatments of 4 authentic phenolic acids.
The proportion of imported wood has been now over 80% of raw materials needed in the wood industry, and therefore, many of studies on the substitution of domestic wood for imported wood and on the increasing the utilization rate of domestic species have been attempted and considered as very important projects to solve. But these trials were practically restricted by several disadvantages of domestic species. such as a small diameter, variety of species, and irregularity of wood physical or chemical characteristics, etc. From a this point of view, this study was carried out to investigate solid wood bending properties of sample trees and then to develop their end uses. The species of sample trees were Pinus densiflora S. et Z., Pinus rigida Mill., and Larix leptolepis Gord. which have large growing stocks but Pinus rigida and Larix leptolepis have not been used well because of their poor qualities. The results obtained were as follows: 1. The optimum conditions for solid wood bending processing of Pinus densiflora, Pinus rigida, and Larix leptolepis are showed in Table 6. 2. The minimum solid-bending radii of Pinus densifjora, Pinus rigida, and Larix ieptolepls were 260mm, 240mm, and 300mm, respectively in steaming treatment.

As the causative agent of Anthrax, Bacillus anthracis causes an acute fatal disease in herbivores such as cattle, sheep, and horses as well as humans. The therapeutics and prevention of anthrax currently available are based on antibiotics and the live attenuated vaccine strains, which may be problematic due to the emergency of antibiotic resistant strains or residual virulence in those vaccine strains. Therefore, it has been required to develop novel therapeutics and vaccines which are safer and applicable to humans. Recently, the development of the multivalent vaccine targeting both spores and vegetative cells of B. anthracis along with anthrax toxin has been reported. In our attempts to screen potential candidates for those multivalent vaccines, the whole genomic expression library of B. anthracis was constructed in this study. To the end, the partial digests of the genomic DNA from B. anthracis (ATCC 14578) with Sau3AI were ligated with the inducible pET30abc expression vectors, resulting in approximately $1{\times}10^5$ clones in E. coli BL21(DE3). The redundancy test by DNA nucleotide sequencing was performed for the randomly selected 111 clones and found 56 (50.5%) B. anthracis genes, 17 (15.3%) vector sequences, and 38 (34.2%) unknown genes with no sequence homology by BLAST. An inducible expression of the recombinant proteins was confirmed by Western blot. Interestingly, some clones could react with the antiserum against B. anthracis. These results imply that the whole genomic library constructed in this study can be applied for analyzing the immunomes of B. anthracis.
The present study was conducted to examine some factors affecting in vitro development and fecundity of embryos recloned with somatic cell nuclear transfer (SCNT). Fibroblast cells retrieved from the ear of a 3-week-old, cloned Korean goat (Jinsoonny) were used as karyoplast donors and serum-starvation was conducted in tissue culture medium (TCM)-199 supplemented with 0.5% FBS. Recipient oocytes were surgically collected by flushing the oviducts 35 h after hCG injection following FSH priming. The zonae pellucidae of the oocytes were partially perforated with a laser drill and a donor cell was transferred into an enucleated oocyte. The couplets were electrically fused and activated by ionomycin (5 min) and 6-DMAP (4 h). The reconstructed embryos were cultured in mSOF medium containing 0.8% BSA at $39^{\circ}C$ in an atmosphere of 5% $CO_2$, 5% $%O_2$, 90% $N_2$ for 12 to 15 h. Re-cloned embryos (2- to 4-cell stages) were surgically transferred into the oviducts of the recipients and pregnancy was subsequently diagnosed by progesterone assay and ultrasound on Days 21 and 63 of pregnancy. The fusion rate following 1st fusion pulse was higher (p<0.05) in 2nd cloning (65.9%) compared to 1st cloning (51.0%), but it was not different in the other groups. The rate of cleavage after fusion was significantly higher (p<0.05) in 1st (77.7%) than in 2nd cloning (56.0%). A total of 175 re-cloned embryos were transferred into 28 recipients. On day 21 and 60 after transfer, 11 (39.3%) and 4 recipients (17.4%) were pregnancy, respectively. In comparison of pregnancy rate by estrous synchronization, a total of 66 and 109 re-cloned embryos were transferred into 11 recipients in natural estrus and 17 recipients in induced estrus, respectively. Five (45.4%) and 2 recipients (18.2%) in natural estrus were pregnant on days 21 and 63 while 6 (35.3%) and 2 (11.8%) recipients in induced estrus were pregnant, respectively. These results show that recloning of goat can be achieved by SCNT and estrous synchronization between donor and recipient animals may be one of the major factors affecting success rate.
Blenniid fish, Istiblennius stellifer(jordan et Snyder) is distributed in the coastal waters of south-eastern Korea and Japan. Matured adults of blenniid fish were collected from the rocky shore of Namchun-dong, Nam-gu, Pusan, Korea on May 15, 1988. The fertilized eggs were incubated and the larvae were reared in laboratory. The eggs of this species were demersal and adhesive, and their diameters varied from 0.84 to 0.88 mm(mean 0.86 mm, n=30). They have a number of small oil globules. The water temperature throughout incubation ranged from 18.5 to $23.3^{\circ}C$ and salinity was maintained at $28.2-29.5\;^{\circ}/_{\circ\circ}$. The hatching took place in 130 hours after fertilization. The newly hatched larvae were 2.70 mm in total length with 11 (abdominal)+22~25 (caudal)=33-36 myomeres. The larvae absorbed the yolk material and oil globule completely in 10 days after hatching and became postlarvae. Total lengths of the larvae reached 4.65 and 5.75 mm in 10 and 13 days after the hatching, respectively.

To develop the descriptive evaluation questionnaire with high validity and reliability on the Clothing and Textiles section of the middle school Technology & Hone Economics textbook, apply it to students and analyze its results. We made out a draft for descriptive evaluation questionnaire that was based upon the concrete establishment of the goal and the range of evaluation. We also made a rubric for scoring as well as sample answer-sheets. Finally, we completed a total of twenty three descriptive evaluation questions and we applied it to sixty five 2nd-grade students in two classes in a middle school. Descriptive evaluation questionnaire exhibited the relative high validity on each question. Moreover, three graders gave the same score on each question of descriptive evaluation, suggesting that descriptive evaluation questionnaire has the high inter-grader reliability and the strong correlation. But, low academic achievement was generally observed in the subjects. They had difficulty in describing their knowledge via their own language and drawing up accurate and detailed answers. They recognized the positive aspects of descriptive evaluation questionnaire, but they felt it uncomfortable due to study-burden and description itself. To overcome these limitations, it is required that students should experience various materials related to subject contents in classes as well as textbooks, concentrate themselves on finding solutions for problems, expand their scope, and practice describe them in advance. Therefore, the additional training for description evaluation questionnaire will be necessary for the more efficient and discriminative questionnaire. Also the questionnaire with high validity and reliability should be developed and the aggressive and voluntary participation of teachers will be needed.
