Identification of Transmembrane Domain of a Membrane Associated Protein NS5 of Dendrolimus punctatus Cytoplasmic Polyhedrosis Virus

  • We examined the intracellular localization of NS5 protein of Dendrolimus punctatus cytoplasmic polyhedrosis virus (DpCPV) by expressing NS5-GFP fusion protein and proteins from deletion mutants of NS5 in baculovirus recombinant infected insect Spodoptera frugiperda (Sf-9) cells. It was found that the NS5 protein was present at the plasma membrane of the cells, and that the N-terminal portion of the protein played a key role in the localization. A transmembrane region was identified to be present in the N-terminal portion of the protein, and the detailed transmembrane domain (SQIHMVWVKSGLVFF, 57-71aa) of N-terminal portion of NS5 was further determined, which was accorded with the predicted results, these findings suggested that NS5 might have an important function in viral life cycle.

Implications of PSR J0737-3039B for the Galactic NS-NS Binary Merger Rate

  • The Double Pulsar (PSR J0737-3039) is the only neutron star-neutron star (NS-NS) binary in which both NSs have been detectable as radio pulsars. The Double Pulsar has been assumed to dominate the Galactic NS-NS binary merger rate $R_g$ among all known systems, solely based on the properties of the first-born, recycled pulsar (PSR J0737-3039A, or A) with an assumption for the beaming correction factor of 6. In this work, we carefully correct observational biases for the second-born, non-recycled pulsar (PSR J0737-0737B, or B) and estimate the contribution from the Double Pulsar on $R_g$ using constraints available from both A and B. Observational constraints from the B pulsar favour a small beaming correction factor for A (~2), which is consistent with a bipolar model. Considering known NS-NS binaries with the best observational constraints, including both A and B, we obtain $R_g=21_{-14}{^+28}$ per Myr at 95 per cent confidence from our reference model. We expect the detection rate of gravitational waves from NS-NS inspirals for the advanced ground-based gravitational-wave detectors is to be $8_{-5}{^+10}$ per yr at 95 per cent confidence. We discuss prospects of gravitational-wave detection based on our results. Implications of PSR J1906+0746, which is likely to be another tight NS-NS binary in the Galactic disc supported by recent observation, are also remarked.

Kerr cell을 이용한 옥소 레이저의 펄스폭 단축과 압축된 펄스의 증폭 (Pulse Shortening by a Kerr Cell in an Iodine Laser and the Amplification of a Shortened Pulse)

  • Kerr cell 광스위치를 옥소 레이저 증폭단에서 짧은 펄스를 발생시키는 광절단기로서 그리고 이후의 증폭기들을 광학적으로 분리하는 격리기로 사용하였다. Kerr cell 양단에 인가된 전위차에 따른 편광의 회전에 의해 입사된 펄스를 자를 수 있음을 보였으며 회로의 전기적 특성을 이용하여 케이블의 길이차에 의해 5ns와 1ns 펄스를 발생시켰다. 공진기에서 펄스폭이 긴 펄스가 발생한 경우 한개 이상의 단축된 출력 펄스가 발생함을 확인하였다. 또한 Kerr cell의 동작전압을 변화시켜 얻은 투과도를 Kerr 이론과 비교하였다. 단축된 펄스를 옥소레이저 전 증폭기를 통해 증폭하여 4ns 펄스에서 2J의 출력을 얻어 0.5GW의 최종출력을 얻었다.

SF6 가스 중 직류 고전압 하에서 부분방전 특성 (Characteristics of Partial Discharge Under HVDC in SF6 Gas)

  • This paper dealt with the measurement and analysis of partial discharge (PD) under high voltage direct current (HVDC) in SF6 gas. Electrode systems such as a protrusion on conductor (POC), a protrusion on enclosure (POE), a crack on epoxy plate and a free particle (FP) were fabricated to simulate the insulation defects. The analysis system was designed with a Time-Frequency (T-F) map algorithm programed based on LabVIEW. This can arrange the acquired PD pulses into frequency and time domain. A HVDC power source is composed of a transformer (220 V/50 kV), a diode (100 kV) and a capacitor (50 kV, 0.5 ${\mu}F$). The gap between the electrodes is 3 mm, and the $SF_6$ gas was set at 5 bar. PD pulses were detected by a 50 ${\Omega}$ non-inductive resistor. In the analysis, PD pulses were distributed below 0.5 MHz and 20 ns ~ 35 ns for the POC, 0.7 MHz ~ 1.7 MHz, below 0.6 MHz and 10 ns ~ 40 ns and 60 ns ~125 ns for the POE, below 0.1 MHz and 135 ns ~ 215 ns for the crack, and below 1.6 MHz and 250 ns for the FP.

Nonstructural NS5A Protein Regulates LIM and SH3 Domain Protein 1 to Promote Hepatitis C Virus Propagation

  • Hepatitis C virus (HCV) propagation is highly dependent on cellular proteins. To identify the host factors involved in HCV propagation, we previously performed protein microarray assays and identified the LIM and SH3 domain protein 1 (LASP-1) as an HCV NS5A-interacting partner. LASP-1 plays an important role in the regulation of cell proliferation, migration, and protein-protein interactions. Alteration of LASP-1 expression has been implicated in hepatocellular carcinoma. However, the functional involvement of LASP-1 in HCV propagation and HCV-induced pathogenesis has not been elucidated. Here, we first verified the protein interaction of NS5A and LASP-1 by both in vitro pulldown and coimmunoprecipitation assays. We further showed that NS5A and LASP-1 were colocalized in the cytoplasm of HCV infected cells. NS5A interacted with LASP-1 through the proline motif in domain I of NS5A and the tryptophan residue in the SH3 domain of LASP-1. Knockdown of LASP1 increased HCV replication in both HCV-infected cells and HCV subgenomic replicon cells. LASP-1 negatively regulated viral propagation and thereby overexpression of LASP-1 decreased HCV replication. Moreover, HCV propagation was decreased by wild-type LASP-1 but not by an NS5A binding-defective mutant of LASP-1. We further demonstrated that LASP-1 was involved in the replication stage of the HCV life cycle. Importantly, LASP-1 expression levels were increased in persistently infected cells with HCV. These data suggest that HCV modulates LASP-1 via NS5A in order to regulate virion levels and maintain a persistent infection.

Surimi 및 전분 첨가에 따른 고등어S(comber japonicus) 소시지의 품질 특성 (Effects of Surimi and Starch on the Quality Properties of Mackerel Scomber japonicus Sausage)

  • This study was conducted to evaluate the qualities of mackerel Scomber japonicus sausage prepared with Theragra chalcogramma surimi (TS, A grade), Nemipterus virgatus surimi (NS, A grade) (0, 5, 7, 10%), and 7% NS with starch (0, 1, 2, 3%). The whiteness of the mackerel sausage was significantly increased by adding TS and NS, but that of the NS with starch groups was decreased compared with the control. The hardness increased significantly with the addition of TS, whereas the NS and NS with starch groups showed no considerable differences compared with the control. The gel strength was not significantly different among the mackerel sausage additive groups. In the sensory evaluation, the mackerel sausages containing 5% TS, 7% NS, and 7% NS with 2% starch had the highest overall preference. In conclusion, these results suggest that 5% TS, 7% NS, and 7% NS with 2% starch improve the quality, texture and sensory properties of mackerel sausage.

DGS구조를 이용한 초광대역 대역통과 여파기 (A Ultra-Wideband Bandpass Filter Using DGS structure)

  • 본 논문에서는 대역통과 여파기와 저역통과 여파기를 하나의 구조로 합성하여 크기 증가 없이 초광대역 특성을 가지는 여파기를 설계 및 구현하였다. 제안한 구조는 그 구조가 간단하다. 그리고 저역 통과 특성을 얻기 위하여 이미 잘 알려진 DGS(Defected Ground Structure) 구조를 이용하였다. 기존의 대역통과 여파기와 저역통과 여파기를 직접 연결하였을 때 보다 크기를 줄일 수 있었다. 측정결과 통과대역은 $2.1GHz{\sim}10.56GHz$이고 삽입손실은 최대 0.5dB, 반사손실은 최소 20dB, 군 지연은 $0.13ns{\sim}0.35ns$ 까지 최대 변화폭 0.23ns의 변화를 보인다.

  • 수 KHz의 반복율로 발진하는 레이저광을 증폭하는 증폭기를 제작하고, 특성을 조사하였다. 증폭기로는 연속 방전하는 섬광등으로 펌핑되는 Nd:YAG 헤드를 사용하였고, 레이저 매질에서 발생하는 열 복굴절을 보상하여 이중통과 증폭기를 구성한 경우, 복굴절에 의한 레이저광 손실을 5%로 감소시켰다. 4 중통과 증폭기를 구성을 하였을 때, 반복율 5-10 KHz에서 이득은 약 3.2이었고, 펄스폭은 40 ns에서 48 ns로. 20%증가하였다.

Inhibition of the Replication of Hepatitis C Virus Replicon with Nuclease-Resistant RNA Aptamers

  • Hepatitis C virus (HCV)-encoded nonstructural protein 5B (NS5B) possesses RNA-dependent RNA polymerase activity, which is considered essential for viral proliferation. Thus, HCV NS5B is a good therapeutic target protein for the development of anti-HCV agents. In this study, we isolated two different kinds of nuclease-resistant RNA aptamers with 2'-fluoro pyrimidines against the HCV NS5B from a combinatorial RNA library with 40 nucleotide random sequences, using SELEX technology. The isolated RNA aptamers were observed to specifically and avidly bind the HCV NS5B with an apparent $K_d$ of 5 nM and 18 nM, respectively, in contrast with the original RNA library that hardly bound the target protein. Moreover, these aptamers could partially inhibit RNA synthesis of the HCV subgenomic replicon when transfected into Huh-7 hepatoma cell lines. These results suggest that the RNA aptamers selected in vitro could be useful not only as therapeutic agents of HCV infection but also as a powerful tool for the study of the HCV RNA-dependent RNA polymerase mechanism.

EM 및 아미노산액비 시용이 '설향' 딸기 모주의 생육에 미치는 영향 (Effect of EM and Amino acid Fertilizer Application on the Growth of 'Seolhyang' Strawberry Mother Plants)

  • The dry weight of mother plants' leaves had the highest increase rate in both NS (single-use) and NS+EM (mixed-use) mixed with NS 0.8 (customary use). In seafood amino acid fertilizer (SAF) application, the increase rate was highest in SAF solution at a 300-fold dilution. Mother plants' crown diameter, plant height, leaf length, leaf width, petiole length and leaf number showed the greatest growth amount when NS 0.8 (customary use) was mixed to NS (single-use) or NS+EM (mixed-use) solution. The growth was highest in SAF solution diluted 300 folds, but lowest in SAF solution diluted 100 folds. Of all inorganic nutrients, excluding sulfur, total amount of nitrogen, available phosphorus, potassium, calcium and magnesium had the highest increase rate in both NS (single-use) and NS+EM (mixed-use) with the treatment of NS 0.8 (customary use). Total nitrogen, in particular, was increased by 3.1% in NS 0.4, 6.0% in NS 0.8, and 4.5% in NS 0.8 with the application of NS+EM at a 500-fold dilution compared to NS alone. Total nitrogen amount showed the highest increase rate in SAF solution diluted 300 folds. Total nitrogen, available phosphorus, calcium, magnesium and EC in soils applied with culture solutions (NS, NS+EM) had increasing tendencies after fertilizer application. The results were comparable to those of SAF treatment. The increase rate of each inorganic nutrient composition declined in soils applied with NS+EM solution diluted 500 folds compared to NS alone.
