Solution Conformations of the Substrates and Inhibitor of Hepatitis C Virus NS3 Protease

  • Hepatitis C virus (HCV) has been known to be an enveloped virus with a positive strand RNA genome and the major agent of the vast majority of transfusion associated cases of hepatitis. For viral replication, HCV structural proteins are first processed by host cell signal peptidases and NS2/NS3 site of the nonstructural protein is cleaved by a zinc-dependent protease NS2 with N-terminal NS3. The four remaining junctions are cleaved by a separate NS3 protease. The solution conformations of NS4B/5A, NS5A/5B substrates and NS5A/5B inhibitor have been determined by two-dimensional nuclear magnetic resonance (NMR) spectroscopy. NMR data suggested that the both NS5A/5B substrate and inhibitor appeared to have a folded tum-like conformation not only between P1 and P6 position but also C-terminal region, whereas the NS4B/5A substrate exhibited mostly extended conformation. In addition, we have found that the conformation of the NS5A/5B inhibitor slightly differs from that of NS5A/5B substrate peptide, suggesting different binding mode for NS3 protease. These findings will be of importance for designing efficient inhibitor to suppress HCV processing.

실리콘 나노시트 피드백 전계효과 트랜지스터의 준비휘발성 메모리 특성 연구 (Quasi-nonvolatile Memory Characteristics of Silicon Nanosheet Feedback Field-effect Transistors)

  • 본 연구에서는 기존 상보성 금속 산화막 반도체 공정을 활용하여 제작된 실리콘 나노시트(SiNS) 피드백 전계효과 트랜지스터(FBFET)의 준비휘발성 메모리 특성을 분석하였다. 과노광공정을 이용하여 형성된 SiNS 채널층의 폭은 180 nm이고 높이는 70 nm이었다. 양성 피드백 루프를 기반으로 동작하는 SiNS FBFET의 낮은 문턱전압이하 기울기는 1.1 mV/dec, ON/OFF 전류비는 2.4×107이었다. 또한 SiNS FBFET는 50 초 동안 상태를 유지하는 메모리 특성을 보여 준휘발성메모리 소자로 활용 가능성을 제시하였다.

Zika Virus-Encoded NS2A and NS4A Strongly Downregulate NF-κB Promoter Activity

  • Since Zika virus (ZIKV) was first detected in Uganda in 1947, serious outbreaks have occurred globally in Yap Island, French Polynesia and Brazil. Even though the number of infections and spread of ZIKV have risen sharply, the pathogenesis and replication mechanisms of ZIKV have not been well studied. ZIKV, a recently highlighted Flavivirus, is a mosquito-borne emerging virus causing microcephaly and the Guillain-Barre syndrome in fetuses and adults, respectively. ZIKV polyprotein consists of three structural proteins named C, prM and E and seven nonstructural proteins named NS1, NS2A, NS2B, NS3, NS4A, NS4B, and NS5 in an 11-kb single-stranded positive sense RNA genome. The function of individual ZIKV genes on the host innate immune response has barely been studied. In this study, we investigated the modulations of the NF-κB promoter activity induced by the MDA5/RIG-I signaling pathway. According to our results, two nonstructural proteins, NS2A and NS4A, dramatically suppressed the NF-κB promoter activity by inhibiting signaling factors involved in the MDA5/RIG-I signaling pathway. Interestingly, NS2A suppressed all components of MDA5/RIG-I signaling pathway, but NS4A inhibited most signaling molecules, except IKKε and IRF3-5D. In addition, both NS2A and NS4A downregulated MDA5-induced NF-κB promoter activity in a dosedependent manner. Taken together, our results suggest that NS2A and NS4A signifcantly antagonize MDA5/RIG-I-mediated NF-κB production, and these proteins seem to be controlled by different mechanisms. This study could help understand the mechanisms of how ZIKV controls innate immune responses and may also assist in the development of ZIKV-specific therapeutics.

소아의 기능성 위장관 질환과 기질 및 성격과의 관계 (The Relationship Between Children's Temperament and Character to Functional Gastrointestinal Disorders)

  • Objectives The purpose of this study is to establish relationship between functional gastrointestinal disorders in children and Temperament and Character using objective criteria and questionnaires. Methods This study surveyed 3-6 years old 130 children who were attending the kindergarten or daycare center in the vicinity of Gangdong from May 15th, 2014 to November 30th, 2014. The Korean-translated Questionnaire on Pediatric Gastrointestinal Symptoms-Rome III (QPGS-Rome III) and The Junior Temperament and Character Inventory 3-6 (JTCI 3-6) questionnaires were used. Results 1. Between the groups, functional gastrointestinal disorder group scored significantly higher on NS, NS1, NS4, HA4, ST and ST1. 2. As the number of functional gastrointestinal disorders increase, the group scored higher on NS, NS1, NS4, HA4, ST and ST1. Conclusions This study showed functional gastrointestinal disorder group exhibited higher NS, NS1, NS4, HA4, ST and ST1 scores.

아미노실란 기능화된 MCM-41과 SBA-15 세공 내 메탈로센 담지 및 에틸렌 중합 (Immobilization of Metallocene inside the Aminosilane-Functionalized Nanopore of SBA-15 and MCM-41 and Its Ethylene Polymerization)

  • 메조포러스 물질, SBA-15와 MCM-41의 세공 표면을 유기실란인 3-aminopropyltrimethoxysilane (1NS)와 N-[(3-trimethoxysilyl) propyl]ethylenediamine (2NS)를 이용하여 그라프팅법으로 표면 기능화하였다. 표면 기능화된 메조포러스 물질에 $(n-BuCp)_2ZrCl_2$와 조촉매 methylaluminoxane (MAO)를 담지하여 에틸렌 중합을 실시하였다. SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ 촉매는 그라프팅되는 2NS의 양이 증가할수록 담지된 Zr와 Al 함량은 감소하였으며, MCM-41/2NS/(n-BuCp)2ZrCl2 촉매는 2NS의 양이 증가할수록 Zr 함량은 증가하지만 Al 함량은 감소하였다. SBA-15/2NS/$(n-BuCp)_2ZrCl_2$ 촉매의 중합 활성은 2NS의 투입량을 증가할수록 감소하였다. 이는 많은 양의 2NS가 표면에 그라프팅되는 경우 기공부피와 기공크기가 작아지게 되고 따라서 MAO와 메탈로센 촉매 담지량이 감소하게 되기 때문이다. 그러나 SBA-15보다 작은 기공을 갖는 MCM-41을 2NS로 표면 기능화되면 큰 분자 크기를 갖는 조촉매 MAO는 담지 되기 어려워 낮은 MAO 담지량을 갖게 된다. 따라서 메탈로센 담지량은 약간 증가하게 되고 이에 따라 중합 활성이 증가하였다.

Implications of PSR J0737-3039B for the Galactic NS-NS Binary Merger Rate

  • The Double Pulsar (PSR J0737-3039) is the only neutron star-neutron star (NS-NS) binary in which both NSs have been detectable as radio pulsars. The Double Pulsar has been assumed to dominate the Galactic NS-NS binary merger rate $R_g$ among all known systems, solely based on the properties of the first-born, recycled pulsar (PSR J0737-3039A, or A) with an assumption for the beaming correction factor of 6. In this work, we carefully correct observational biases for the second-born, non-recycled pulsar (PSR J0737-0737B, or B) and estimate the contribution from the Double Pulsar on $R_g$ using constraints available from both A and B. Observational constraints from the B pulsar favour a small beaming correction factor for A (~2), which is consistent with a bipolar model. Considering known NS-NS binaries with the best observational constraints, including both A and B, we obtain $R_g=21_{-14}{^+28}$ per Myr at 95 per cent confidence from our reference model. We expect the detection rate of gravitational waves from NS-NS inspirals for the advanced ground-based gravitational-wave detectors is to be $8_{-5}{^+10}$ per yr at 95 per cent confidence. We discuss prospects of gravitational-wave detection based on our results. Implications of PSR J1906+0746, which is likely to be another tight NS-NS binary in the Galactic disc supported by recent observation, are also remarked.

참갈겨니, Zacco koreanus(잉어과)의 NE형과 NS형의 서식처 분리 (Habitat Segregation between NE and NS Type of Zacco korean us (Cyprinidae))

  • 참갈겨니 NE와 NS형의 서식범위 내에서 두 형이 공서하는 지점을 조사한 결과 낙동강과 섬진강의 각 지류의 상류지점, 남해로 흐르는 일부 하천에서 발견되었다. 두 형이 공서하는 하천에서의 분포양상은 NE형은 상류 지역에 NS형은 중류 지역에 치우치는 경향을 보였다. 중간 지역에서는 같은 지점에서 동시에 출현하였으나, NE형은 여울에 NS형은 웅덩이 또는 흐름이 느린 곳에서 주로 서식하는 서식처 분리현상을 나타내었다. 유속과 출현개체수의 관계는 공서지에서는 유속의 증가에 따라 NE형은 개체수가 증가하는 반면 NS형은 감소하는 경향을 보였으나, 각각 단독으로 존재하는 곳에서는 그러한 경향이 나타나지 않았다.

裁培地域에 따른 加工用 감자의 品質에 關한 硏究 (Studies on the Quality of Processing Potatoes grown at Different Locations)

  • 국내의 감자 재배지역(栽培地域)에 알맞은 가공용(加工用) 감자품종(品種) 선납(選拉) 및 재배지역(栽培地域)간 품질성(品質性)에 관한 기초자료를 얻고자 재배지역(栽培地域)별로 유망(有望) 가공용(加工用) 품종(品種) 및 계통(系統)들의 규격서(規格薯) 수량(收量) 및 품질(品質)에 미치는 영향에 대하여 시험을 실시한 결과를 요약하면 다음과 같다. 지역에 따른 평 균 총서(總薯) 수량(收量)을 보면 대관령(大關嶺) (3,051 Kg/10a), 강릉(江陵(2,863 Kg/10a), 보성(寶城) (2,844Kg/10a)의 순이었으며 평균(平均) 규격서(規格薯) 수량(收量)은 대관령9大關嶺) (2,351 Kg/10a), 강릉(江陵) (2, 278 Kg/10a), 보성(寶性) (2,246Kg/10a)의 순으로 높았다. 품종별(品種別) 수량(收量)은 각지역 모두 Gemehip이 가장 많았으며 이는 식용(食用)으로 사용하기에 유망하였다. 비중(比重)은 대관령(大關嶺)지역이 비교적 높았으며 품종간(品種間)에는 대서(大西)와 NS1이 높았고 Gemchlp은 낮았으며, 클루코스 함량(含量)은 NS1, 대서(大西), NS2, 수미(秀美) 및 Gemchip순으로 낮게 나타났고 chip color 역시 NS1, 대서(大西), NS2, Gemchip 수미(秀美)순으로 밝아 글루코스 함량과 밀접한 관계를 보여주었다. 제품수율(製品收率)은 각 지역 모두 대서(大닌西), NS1, 수미(秀美), NS2, Gemchip 순(順)으로 나타나 비중(比重)의 순서와 일치하였다. 이상과 같은 결과로 볼 때 대서(大西)와 NS1 품종(品種)이 칩 가공(加工)용 유망품종(有望品種)으로 판단되었다.

Pulsar binaries and GW detection

  • Pulsar binaries in tight orbits are considered to emit strong gravitational waves (GWs) during the last stage of their coalescences. They form a subset of compact binary mergers, which consists of white dwarfs (WDs), neutron stars (NSs), or black holes (BHs). One of the most famous example of 'merging' pulsar binaries is the Hulse-Taylor pulsar (PSR B1913+16) discovered in 1974 by Russell Hulse and Joseph Taylor. About ten NS-NS and several tens of NS-WD binaries are known in our Galaxy. Merging binaries are rare and only a few NS-NS and NS-WD have been discovered to date. A pulsar with a black hole companion is also theoretically expected, but there is yet no detection. Within several years, direct detections of GWs from compact binary mergers will be made by laser interferometers. This will pave a way to study physics of compact binaries that cannot be reached by electromagnetic waves (EM). Pulsar binaries are of particular interest as we can use both EM and GW to probe these systems. In this talk, we present a brief overview on the Galactic pulsar populations and discuss their implications for GW detection.

Compact Binaries Ejected from Globular Clusters as GW Sources

  • Based on N-body simulations, we find out that significant fraction of dynamically formed BH-BH (10 $M_{\odot}$ and NS-NS (1.4 $M_{\odot$ ecah) binaries are ejected from globular clusters. About 30 percent of compact stars are ejected in the form of binary. The merging time of ejected binary depends on the velocity dispersion of globular cluster. Some of ejected binaries have merging time-scales shorter than Hubble time and are expected to produce gravitational waves that can be detectable by the advanced ground-based interferometers. The merger rates of ejected BH-BH and NS-NS binaries per globular cluster are estimated to be 3.5 and 17 per Gyr, respectively. Assuming the spatial density of globular clusters as 8.4 $h^3$ clusters $Mpc^{-3}$ and extrapolating to the horizon distance of the advanced LIGO-Virgo network, we expect the detection rates solely attributed to BH-BH and NS-NS with cluster origin are to be 42 and 1.7 $yr^{-1}$, respectively. Besides, we find out that BH-NS binary ejection hardly occurs in globular clusters and dynamically formed compact binaries may possibly be the source of short GRBs whose locations are far from host galaxies.
