Serotonin Synthesis and Metabolism in Dissociated Cultures of Fetal Rat Brainstem (흰쥐 태아 뇌간의 일차 세포배양에서 Serotonin의 합성 및 대사에 대한 연구)

  • We established an in vitro system of central serotonergic neurons by culturing dissociated rat embryonic (El4) brainstem cells to 14 days in vitro and monitored the serotonergic neuronal growth by measuring 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) contents in the cells with hish performance liquid chromatography with electrochemical detection (HPLC-EC). We studied also tile effects of various drugs on the contents of 5-HT and 5-HIAA, confirming in vivo reports. The 5-HT content (13 ng/mg protein) and 5-HT turnover rate (17 pmol/mg protein/h) at 14 days in vitro were in good agreement with those reported in the adult rat brain. The 5-HT content was more easily depleted with p-chlorophenylalanine, a tryptophan hydroxylase inhibitor than with NSD 1015 (3-hydroxybenzylhydrazine), an aromatic L-amino acid decarboxylase (AADC) inhibitor. Incubation of the cultures with tryptophan or 5-hydroxytryptophan increased the rate of serotonin formation implying that neither tryptophan hydroxylase nor AADC is saturated with its amino acid substrate in this in vitro system . The 5-HT content was depleted by reserpine. The 5-HT and 5-HIAA contents were increased and decreased, respectively, by monoamine oxidase inhibitors. All the above results indicate that the biochemical properties of the central serotonergic neurons in this culture system reflect reliably those of central serotonergic neurons in vivo. We suggest that measuring 5-HT and 5-HIAA contents in the primary cultured dissociated brainstem-cells with HPLC-EC is useful in the study of pharmacology as well as toxicolgy of the central serotonergic neurons.

A STUDY ON THE EFFECT OF CHINCAP IN JUVENILE SKELETAL GLASS III MALOCCLUSION (유년기 III급 부정교합자에서 이모장치의 치료 효과에 관한 연구)

  • The purpose of this study was to investigate the changes in the craniofacial skeleton subsequent to chincap therapy in the juvenile skeletal Class III malocclusion with more appropriate control samples. The experimental group consisted of 29 Korean children(14 males, 15 females) who had skeletal Class III malocclusion with prognathic mandible and were undergone chincap thorny from the beginning of treatment. The control group was composed of 21 Korean children(10 males, 11 females) who had no orthodontic treatment, but with similar skeletal discrepancies to experimental group. Lateral cephalometric radiographs at the age of 7, and 2 years later were analyized and compared with student's t-test(p<0.05). The results of this study were as follows; 1. The control group without chincap therapy had not shown any improvement of the skeletal discrepancies, but had grown to be much severe. This means that the untreated Class III patient with prognathic mandible would not be corrected by growth. For the experimental group with chincap therapy, the anterior-posterior skeletal discrepancies and mandibular prognathism were both improved. 2. Neither significant restraint nor acceleration of growth was found in the cranial base and maxilla by chincap treatment. 3. The inhibition of mandibular growth could not be accepted, but the changes of the direction of growth and morphological changes were found. 4. Vertical growth tendency was increased with chincap therapy. 5. When Putting together the results of the analyses , it seems to be the rotation and displacement of the mandible that the major treatment effects of chincap we. The changes of the direction of growth and the morphological changes also seems to contibute to the treatment effect partly. In summary, the chincap doesn't restrain the mandibular growth. But, it is considered as a useful treatment modality for correction of skeletal discrepancies and reduction of mandibular prognathism in growing Class III patients with madibular prognathism.

Effect of N-Acetylcysteine on the Supetoxide Release, Chemotaxis from the Neutrophils and Glutathione Level of Plasma and Neutrophils (N-Acetylcysteine이 호중구의 Superoxide, Chemotaxis 및 혈장과 호중구의 Glutathione에 미치는 영향)

  • Background: N-acetylcysteine(ACE) is used both orally and intravenously in a variety of experimental pathologies resembling human disease states which exhibit endothelial toxicity as a result of oxidative stress, including acute pulmonary oxygen toxicity, septicemia and endotoxin shock. Despite these observations in vivo, it is not certain how this thiol drug produces its protective effects. ACE is a cysteine derivative which is able to direct1y react with oxygen radicals and may also act as a cysteine and glutathione(GSH) precursor following deacetylation. In this paper, we tried to know whether the therapeutic doses of ACE can modify the inflammatory function of the neutrophils and can increase the glutathione level of plasma in chronic obstructive pulmonary disease(COPD) patients. In addition, the effect of ACE to the purified neutrophil in terms of superoxide release and glutathione synthesis were observed. Method: Firstly, we gave 600mg of ACE for seven days and compare the release of superoxide, luminol-enhanced chemiluminescence from the neutrophils, neutrophil chemotaxis, and plasma GSH levels before and after ACE treatment in COPD patients. Secondly, we observed the dose dependent effect of ACE to the purified neutrophil's superoxide release and GSH levels in vitro. Results: 1) Usual oral therapeutic doses(600mg per day) of ACE for seven days did affect neither on the neutrophil's superoxide release, chemiluminescence, chemotaxis, nor on the plasma GSH concentration in the COPD patients. 2) ACE decreases the purified neutrophil's superoxide release and increase the GSH production in dose dependent fashion in vitro. Conclusion: Despite the fact that oral ACE treatment did not affect on the neutrophil's inflammatory function and plasma GSH concentration in COPD patients in usual therapeutic doses, it decreases the superoxide release and increases the GSH production from the isolated neutrophils in high molar concentrations. These findings suggest that to obtain an antioxidative effects of ACE, it might be needed to increase the daily dosage of ACE or therapeutic duration or change the route of adminisration in COPD patients.

The Usefulness and Safety of Natural Stent in a Canine Model of Tracheal Stenosis (Nd-YAG laser를 이용한 기관협착 동물모델에서 Natural 스텐트의 안전성 및 유효성 -Dumon 스텐트와의 비교 실험-)

  • Background : In order to investigate the usefulness and safety of Natural Stent, we performed this study in a canine model of tracheal stenosis induced using Nd-YAG laser. Materials and Methods : After tracheal stenosis was induced in 12 Mongrel dogs using Nd-YAG laser, either Dumon (n=6) or Natural (n=6) stent was inserted into the trachea. To assess the degree of stent migration and mucostasis, bronchoscopy was performed every week for 4 weeks, after which all stents were removed. One week after stent removal, tracheal stenosis was evaluated by bronchoscopy. Results : The degree of stent migration was not different between the dogs with Dumon stent ($3.0{\pm}0.8$) and those with Natural ($2.0{\pm}1.0$), nor was the degree of mucostasis, at Dumon ($1.7{\pm}0.5$) and Natural Stent ($1.5{\pm}0.6$), respectively. One week after stent removal, the degree of tracheal stenosis was not different between the Dumon ($1.5{\pm}0.5$) and the Natural group ($1.0{\pm}0.4$). In addition, there was no death and the degree of tracheal stenosis remained always within the safe limit (less than 2.0) in all animals. Conclusion : In a canine model of tracheal stenosis induced using Nd-YAG laser, the usefulness and safety of Natural Stent were similar to those of Dumon Stent. A clinical trial is necessary to document the usefulness and safety of Natural Stent in patients with tracheal stenosis.

Physicochemical Properties of Jadeite Powder and Its Application to Cosmetic Formulations (경옥가루의 물리화학적 특성 및 화장품 제형 응용 연구)

  • In this study, physicochemical properties of the natural jadeite powder were investigated and also the wash-off pack and liquid foundation containing the jadeite powder were prepared. In addition, each of these formulations was evaluated by various functional effects, sensory evaluation, stability and skin safety. In the wash-off pack, the far-infrared ray emissivity and radiation energy values increased as a function of the jadeite powder content. At a 3% jadeite powder content, the skin temperature increased by about $0.5^{\circ}C$ when the jadeite powder-containing formulation was applied to the skin. Besides, the chroma of the liquid foundation containing the jadeite powder more clearly expressed the original color of the skin. Moisture content measurements of the wash-off pack and liquid foundation containing the jadeite powder showed the highest moisture uptake of 5.0% and 63.0%, respectively. In sensorial test, the wash-off pack formulations containing the jadeite powder demonstrated improved affinities toward a skin, adherency, and moistness and combatted itching. The liquid foundation containing jadeite powder showed also improved affinities except for the coverage when compared to control formulations. Furthermore, the stability evaluation for 8 weeks revealed neither discoloration nor separation phenomenon for the formulations containing the jadeite powder. Moreover, the pH was found to be stable up to 8 weeks and the viscosity up to 4 weeks. Skin safety assessments showed that all formulations containing the jadeite powder were non-irritating. These results suggest that the jadeite powder as an inorganic pigment may serve as a new multi-functional cosmetic ingredient with stability and safety.

EFFECT OF BISPHOSPHONATE ON OSTEOBLAST DIFFERENTIATION (Bisphosphonate가 조골세포 분화에 미치는 영향)

  • Bisphosphonates inhibit bone resorption in vivo and in vitro. Currently proposed mechanism of action of bisphosphonates involves both direct effect on osteoclasts and indirect effect through the mediation of osteoblasts. Recent understanding of molecular mechanism of osteoclastogenesis indicates that osteoclast differentiation is quite tightly regulated by signaling molecules from differentiating osteoblasts. Therefore this investigation was designed to elucidate the effect of bisphosphonate on osteoblast differentation. For this purpose, in vitro effects of etidronate and alendronate on the expression of Cbfa1 a master control gene of osteoblast differentiation, several bone marker genes, and formation of calcified nodules were evaluated. To evaluate the effect of bisphosphonate on calcified nodule formation, osteoblasts isolated from rat calvaria were cultured in a-MEM containing $10^{-4},\;10^{-5},\;10^{-6}M$ of etidronate or $10^{-6},\;10^{-7},\;10^{-8}M$ of alendronate for 15 days, and then stained by alizarin red to determine mineralization. To evaluate the effect of bisphosphonate on osteoblast differentiation, osteoblast cells were cultured in a-MEM containing $10^{-4},\;10^{-5},\;10^{-6}M$ of etidronate or $10^{-6}$ M of alendronate for 8 days. And then total RNA was extracted and northern blot analysis was done to examine the expression of Cbfa1, type I collagen, alkaline phosphatase, osteopontin and osteocalcin. The results were as follows: 1. Etidronate suppressed the calcification of bone nodule in dose dependent manner, while alendronate didn't. 2. The expression of Cbfa1 was decreased dose dependently by etidronate, but increased by alendronate. 3. Etidronate suppressed the expression of type I collagen, osteopontin and osteocalcin in dose dependent manner however alendronate promote the expression of osteoblast marker gene. 4. The expression of alkaline phosphatase was not affected either etidronate nor alendronate. These results suggest that etidronate suppressed the expression of Cbfa1 in dose dependent manner, and consequently the expression of osteoblast marker genes, such as type I collagen, osteopontin and osteocalcin were also suppressed in similar manner. And finally this decreased expression of osteoblastic marker gene prevent calcined bone nodule formation.

Peripheral Blood Eosinophil Counts and Serum ECP in Adolescents with Long-term Asthma Remission and Persistent Bronchial Hyperresponsiveness : Comparison with Adolescents with Symptomatic Asthma (기관지 과민성을 가진 장기간 천식 관해 상태의 청소년에서 혈액 내 호산구와 혈청 호산구 양이온 단백에 대한 연구)

  • Purpose : Bronchial hyperresponsiveness(BHR) in asthma is thought to be a consequence of underlying airway inflammation. But the mechanism responsible for persistent BHR in adolescents with long-term asthma remission is poorly understood. The aim of this study was to examine whether BHR in adolescents with asthma remission is associated with peripheral blood eosinophilia and/or increased serum levels of eosinophil cationic protein(ECP). Methods : We studied 35 adolescents with long-term asthma remission(neither symptoms nor medication during the previous two years) who have persistent BHR(remission group) and 35 adolescents with symptomatic asthma(symptomatic group) who were matched for methacholine provocative concentration producing a 20% fall in $FEV_1(PC_{20})$ with subjects in the remission group. The peripheral blood eosinophil counts and serum ECP concentrations were compared between these two groups. Correlations between $PC_{20}$ and peripheral blood eosinophil counts or serum ECP concentrations were assessed in these two groups. Results : Peripheral blood eosinophil counts and serum ECP concentrations were significantly lower in the remission group than in the symptomatic group($273{\pm}108$ vs. $365{\pm}178/{\mu}L$; $16.3{\pm}9.4$ vs. $26.5{\pm}15.1{\mu}g/L$, both, P<0.05). $PC_{20}$ was correlated with peripheral blood eosinophil counts and serum ECP concentrations in the symptomatic group(r=-0.385, P=0.022; r=-0.439, P=0.008), but not in the remission group(r=-0.292, P=0.089; r=-0.243, P=0.159). Conclusion : BHR in adolescents with long-term asthma remission is not associated with peripheral blood eosinophilia or an increase in serum ECP concentration, which suggests that BHR in this clinical setting may not be attributed to airway eosinophilic inflammation. Further studies including direct assessment of airway inflammation are needed to confirm this conclusion.

Phylogenetic Relationships of Jeju Dogs to Other Domestic and Foreign Dog Breeds Determined by Using mtDNA D-loop Sequences (mtDNA D-loop의 염기서열에 의한 제주견과 우리나라 재래견 및 외국견품종과의 유연관계)

  • Phylogenetic relationships of Jeju dogs to other domestic and foreign dog breeds were assessed using mtDNA D-loop sequences. Neighbor-joining trees were constructed using complete sequences (970 bp excluding the tandem repeat region) determined for five Cheju, four Jindo, four Sapsaree, five Pungsan, two of each East and West Laika dogs (Canis familiaris), two gray wolves (Canis lupus) and two coyotes (Canis latrans) and also published complete sequences for dogs. Coyote sequences were used as outgroups. In addition, a total of 214 haplotypes of 598bp D-loop sequences from 30 dog breeds were collected from GenBank and used to investigate genetic structure of population. In the analyses of full D-loop sequence variation and the phylogenetic trees constructed by neighbor-joining method, neither haplotypes nor clades specific for any domestic dog breeds were observed. The inter-species sequence variation (4.51%) between domestic dogs and wolves was much higher than the intra-species sequence variation within domestic dogs (1.63%) and wolves (3.64%). The divergence of the dog and wolf occurred approximately 1~2 million years ago based on these values. The taxa of Jeju dog breed in the phylogenetic tree are clustered separately and intermingled with other taxa of breeds, suggesting that active crossbreeding of Jeju dogs with other domestic breeds.

Changes in $A_{1}$, Adenosine Receptor-Adenylyl Cyclase System of Rat Adipocytes Fellowing Induction of Experimental Diabetes by Streptozotocin Treatment (Streptozotocin으로 당뇨병을 유발시킨 쥐의 지방세포에 나타나는 $A_{1}$, Adenosine Receptor-Adenylyl Cyclase System의 변화)

  • Adenosine receptors in rat adipose tissues have been reported to be of $A_{1}$ subclass, and their stimulation leads to inhibition of adenylyl cyclase, resulting in inhibition of lipolysis. In the present study we investigated changes in $A_{1}$ adenosine receptor-adenylyl cyclase system of adipocytes following induction of experimental diabetes in rats. One week following experimental diabetes were induced by intravenous injection of streptozotocin (50 mg/kg body wt.), adipocytes from rats $(170{\sim}230g)$ fed ad libitum were isolated using collagenase. When adipocytes were incubated for 1 h with 1 unit/ml adenosine deaminase and $1\;{\mu}M$ isoproterenol, and assayed for glycerol formation, it was found that the inhibition of lipolysis in diabetic adipocytes by $(-)-N^{6}-(R-phenylisopropyl)adenosine$ (PIA), an $A_{1}$, adenosine receptor agonist, was twice that of control adipocytes. In an effort to delineate the mechanism(s), $[^{3}H]PIA$ binding to adipocytic membranes from diabetic and control rats were determined. Neither the affinities nor numbers of $A_{1}$ adenosine receptor were significantly different from each other (Best fit parameters for the one-site model are: $K_{d}=0.51{\pm}0.09nM$ and $B_{max}=1.60{\pm}0.12\;pmoles/mg$ protein for control membranes; $K_{d}=0.54{\pm}0.21\;nM$ and $B_{max}=1.72{\pm}0.31\;pmoles/mg$ protein for diabetic membranes). However, the inhibiton by PIA of the isoproterenol-stimulated adenylyl cyclase activities was found to be 1.9 times higher in adipocytic membranes from diabetic rats than those from controls. These results suggest that the increased sensitivity of inhibition of lipolysis to PIA in adipocytic membranes from diabetic rats is due to changes in signal transduction pathways, rather than alterations of $A_{1}4 adenosine receptor molecules themselves.

Present and Future of the Journal of Distribution Science (유통과학연구의 현재와 미래)

  • The recent announcement of the National Research Foundation of Korea (NRF) to cease journal accreditation operations as of the end of the year 2014 can easily influence the future of many research journals in Korea. Although this plan has not yet been formalized or structured, its facilitation would be the major turning point for the current Korean research and scholarly journals and publications. In addition, the NRF's plan to select and fund top 20 or more research journals over the five year period beginning 2015 suggests that the competition will most likely increase among Korean journals. Each journal would need to develop its unique strategy to improve and strengthen its competitiveness to become or maintain its position as a major research journal in Korea. The association of Korean Distribution of Science (KODISA) and its research journal, Journal of Distribution Science (JDS), has been continuously improving its reputation as a reputable journal in the distribution and related fields since its establishment in 1999. Due to demand, JDS has had to undergo several changes in its publication cycle first from semiannual publication to quarterly, then finally to monthly publications in 2012, and has become one of the major social science journals in Korea. Furthermore, with the redesigning of its webpage with English language in July of 2011, KODISA has made the published journals freely accessible and available to both domestic and foreign researchers, scholars, practitioners, and learners. These changes have resulted in the rapid increase in the bounce rate and the number of journal submissions by foreign scholars, with four research articles having been submitted by foreign scholars just in March of 2012 alone. However, although the changes and outcomes have resulted in a reasonable success so far, the achievement may only become a short-term success without continuously developing, improving, and implementing both effective and efficient strategies through critical, thorough, and frequent examinations and evaluations of both KODISA and JDS. As such, the purpose of this research is to carefully examine both KODISA and JDS to identify problematic factors and to develop appropriate strategies to change or modify those problems for further strengthening and improving their reputation and status. The paper examines and analyzes the past, present, and future of KODISA and JDS and their managerial, operational, and systematic procedures and operations. The narrow scope of research and inefficiencies in promoting the association and the journal and the improvement of impact factors are identified as the notable problems that could hinder JDS from being included in SCOPUS or SSCI in the near future. This type of examination and exploration has not been previously conducted, so the major limitation of this paper can be identified as not meticulously elaborating on the problems nor proving detailed recommendations based on the existing researches. This article asserted that solving the problem of the narrow scope of research would lead to facilitation of resolving other inefficient problems. Inclusion of international academic disciplines to the distribution and their related fields would be the viable initiation of expanding the research area, and this strategy could promote the journal as well as improve its impact factors. The narrow scope of research seems to be a good research topic and merit further exploration as an individual research project, because this kind of research could yield the creation of new understandings or theories.
