Comparative Studies on the Effects of Fresh, White and Red Ginseng Their Effects on the Content of Several Components in Rat Serum and the Production of Lipid Peroxide in Rat Liver (수삼, 백삼 및 홍삼의 효과에 대한 비교 연구 흰쥐의 혈청내 몇가지 성분 함량 및 과산화지질생성에 미치는 영향)

  • Seventy percent ethanol extracts were prepared from 6-year-old fresh, white and red ginseng cultivated in Kangwha, and administered orally into rats for the comparison of their effects. And their effects on the production of lipid peroxide in the livers of ethanol-administered rats were measured. Red ginseng administered group showed the largest body weight increase. However, fresh, white, red ginseng extract administered group showed no significant change in concentration of total lipid, triglyceride and free cholesterol in serum. White and red ginseng extract administeration decreased blood-sugar levels significantly. Lipid peroxide content in livers of white and red ginseng administered groups was decreased significantly. Red ginseng administered group showed greater decrcase in lipid peroxide content than that of white ginseng administred group. The increase of lipid peroxide content in the livers of ethanol-administered group was inhibited by administration of fresh, white, and red ginseng extract. And the strongest-inhibitory action was observed in red ginseng administered group. Therefore, it is supposed that red ginseng has the most powerful antiaging effect.

Fermented Product Extract with Lentinus edodes Attenuate the Inflammatory Mediators Releases and Free Radical Production

  • Lentinus edodes contains functional metabolites such as polysaccharopeptides, lectins, and secondary metabolites. Fermented soybean paste is representative fermented materials in Korea, and is gradually increasing due to various biological activities. In the present study, ethanol extracts of fermented products with/without L. edodes were designated as SPL and SP, and prepared to develop safer and therapeutic functional foods with antioxidant and anti-inflammatory activities for treatment of inflammatory disorders. SP and SPL extracts exhibited antioxidant effects via inhibiting radical activities. Inflammatory mediators, nitric oxide (NO), prostaglandin E2 (PGE2), interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor (TNF)-α, and inducible nitric oxide synthase (iNOS) production and nuclear factor-kappa B (NF-κB) activation were down-regulated by two extracts. SPL extract more strongly enhanced the antioxidant and anti-inflammatory activities than SP extract. Its' activities shown more longer fermentation period and more strong inhibitory effects. Taken together, our results suggested that fermented product with medicinal plant has synergic effect and SPL can be a potential candidate for treatment of inflammatory bowel diseases.

Antioxidant and Melanin-Reduction Effect of Mixed Extracts of Bixa orellana, Ammi majus, and Glycyrrhiza glabra

  • The desire of modern people to maintain a healthy and beautiful appearance is increasing day by day along with the increasing interest in skin health and the demand for functional cosmetics. Accordingly, research on functional cosmetic materials with few side effects and excellent efficacy is being actively conducted. Therefore, this study tried to verify the antioxidant and whitening effects of the mixed extracts of Bixa orellana, Ammi majus, and Glycyrrhiza glabra, whose efficacy has been individually verified. Extracts (BAG-1~4) with different extraction methods such as steaming, fermentation, and ultrasonication were prepared for 3 types of natural plants, and antioxidant and whitening effects of these extracts were confirmed. For this purpose, antioxidant, tyrosinase activity, melanin production and stability experiments were conducted. Extracts (BAG-1~4) had no cytotoxicity, and antioxidant and whitening effects were confirmed. BAG-4 extracted by steaming and fermentation showed the best efficacy. It seems that enzymes such as lipase, protease, and amylase increase phenol components by various yeasts involved in the fermentation process, thereby improving antioxidant and melanin production inhibitory effects. It was confirmed that the three types of natural plant extracts could be used as safe and functional cosmetic materials.

Effect of ${\gamma}-ray$ Irradiation on the Activities of Monoamine Oxidase in Rat Brain and Liver (방사선 조사가 쥐의 뇌와 간의 Monoamine Oxidase 활성도에 미치는 영향)

  • In order to evalute the effects of radiation on mammalian neuronal system, we have examined the effect of gamma-ray radiation on the monoamine oxidase (MAO) activity in monoaminergic neurons. Following the whole body irradiation, MAO activity in the rat brain was measured as well as in the liver for the comparative studies between the neuronal and nonneuronal system. The effects of some radiation protectors and sensitizers were also examined in addition to the $O_2$ effect. The results can be summarized as follows. 1) The MAO activity of rat brain was minimally affected by the radiation dose up to 1,700 cGy Radiation dose above 2,500 cGy inhibited the brain MAO activity by no less than $l0\%.$ MAO-A form was found to be particularly sensitive to radiation. The liver MAO was somewhat inhibited (by about $5\%$) but hardly dependent on the dose of radiation. 2) The inhibitory effect on the brain was initiated immediately by the radiation dose of 2,500 cGy. On the contrary, for the liver, the inhibitory effect became apparent only 2 days after irradiation. 3) Two days after a dose of 2,500 cGy, Vmax and Km of the brain mitochondrial MAO decreased. For liver, Vmax decreased while Km increased, which indicates the kinetic patterns for the neuronal and nonneruronal systems are not affected similarly by radiation. 4) The effect of several known radiation protectors and sensitizers on MAO activity was tested ut no definite results were obtained. The level of -SH group increased in some degree upon radiation but not by the compounds. 5) MAO activity was not affected by $O_2$ concentration, while an elevated level of lipid peroxidase was found under the same condition. The results described here indicate that characteristics of MAO, one of the most important central nervous system enzymes, are liable to radiation, which is partially differentiated from the liver MAO. Also indicated are that the -SH groups are hardly related to the effect of radiation but the production of the lipid peroxide seems to be somewhat correlated to the effect of radiation.

Inhibitory Effect of Inflammatory Cytokines Secretion of Cheonggeumganghwa-tang in Mast cell (비만세포에서의 청금강화탕의 염증성 세포활성물질 분비 억제 효과)

  • Cheonggeumganghwa-tang(CGT) has been used for the purpose of prevention and treatment of bronchial asthma and allergic asthma in Korea. To investigate the biological effect of CGT, the author examined cytotoxicity and inflammatory cytokines secretion with human mast cell line, HMC-1. HMC-1 was stimulated with phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187. CGT by itself had no effect on viability of HMC-1. The effects of CGT on the secretion of tumor necrosis factor-alpha (TNF-α) and interleukin (IL)-6 from HMC-1 were evaluated with enzyme-linked immunosorbent assay (ELISA). CGT (1 ㎎/㎖) inhibited PMA plus A23187 -induced TNF-α and IL-6 secretion, by 93.86 ± 2.05%, 68.69 ± 2.86%, respectively. CGT also inhibited the NF-κB (p50) expression. Taken together, these results suggest that CGT inhibit the production of inflammatory cytokines in HMC-1 cells through blockade of NF-κB activation.

Anti-inflammatory Effect of Red Ginseng through Regulation of MAPK in Lipopolysaccharide-stimulated RAW264.7 (Lipopolysaccharide로 유도된 RAW264.7 세포에서 MAPK에 의한 홍삼추출물의 항염증 효과)

  • Inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) are important inflammatory mediators implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed to determine whether Red Ginseng (RG) could modulate $I{\kappa}B$-kinase, iNOS and COX-2 gene expression and immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). RG extract suppressed the expression of LPS-induced $I{\kappa}B$, iNOS, COX-2, and immune responses in a dose-dependent manner. It also showed an anti-inflammatory effect by inhibiting NF-${\kappa}B$ immune response induced by LPS treatment. Inhibitory effect of RG on LPS-induced inflammation was mediated by suppressed phosphorylation of ERK, JNK and p38 through the regulation of the mitogen-activated protein kinase (MAPK) pathway leading to a decreased production of NO, iNOS, COX-2 and NF-${\kappa}B$. The results implied the role of RG as an inflammation regulator and its possible application for curing inflammatory diseases.

  • Codonopsis Lanceolata (CL) has been widely used in Oriental medicine for treatment of chronic inflammatory diseases, such as bronchitis, cough, and spasm; however, the mechanism of its anti-inflammatory activity has not been clarified. In this study, therefore, we investigated the inhibitory effect of CL on LPS-induced inflammation. The effect of CL was analyzed by ELISA, RT-PCR and Western blotting in LPS-stimulated RAW264.7 cells. We found that CL suppressed not only the mRNA expression of pre-inflammatory cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase (COX)-2, but also the phosphorylation of ERK1/2, JNK1/2 and p38 MAPK. These results suggest that CL exerts an anti-inflammatory effect through the regulation of the mitogen-activated protein kinases (MAPK) pathway, thereby decreasing production of pre-inflammatory cytokines, NO, and PGE2.

The effect of Baeckji, Seasin, Baeckgangjam and Myunkanbang on skin-whitening (백지, 세신, 백강잠 및 면간방이 피부미백에 미치는 영향)

  • To develope new natural substances for whitening agent, Baeckji(Angelica dahuria;BK), Seasin(Asarum sieboldii;SS). Baecjgangjam(Bombycis corpus;BGJ) and Mynkanbang(MKB) were selected and extracted by hot water and 70% EtOH, respectively. Hydrothermal and ethanolic extracts of BK, SS, BGJ and MKB were evaluated for anti-oxidative effect, tyrosinase activity, melanogenesis of B16 melanoma cells and changes in level of tyrosinase expression by using Western blotting. All hydrothermal and ethanolic extracts showed scavenging activities of free radicals against 1,1-diphenyl-2-picrylhydrazyl(DPPH) but no inhibitory effect on tyrosinase activity. Ethanolic extracts of BK, SS, and BGJ and especially highly, those of MKB inhibited production of melanin in B16 melanoma cells and were able to reduce the level of tyrosinase expression in B16 melanoma cells. These results suggest that ethanolic extracts of BK, SS, BGJ and MKB can be an effective whitening agent from natural plant.

Inhibitory Effects of Esculetin Through the Down-Regulation of PI3K/MAPK Pathway on Collagen-Induced Platelets Aggregation (Esculetin이 PI3K/MAPK 경로 하향 조절을 통해 collagen 유도의 혈소판 응집 억제에 미치는 효과)

  • Platelet activation plays a major role in cardiovascular disorders (CVDs). Thus, disrupting platelet activation represents an attractive therapeutic target on CVDs. Esculetin, a bioactive 6,7-dihydroxy derivative of coumarin, possesses pharmacological activities against obesity, diabetes, renal failure, and CVDs. In other report, the effect of esculetin has been examined in human platelet activation and experimental mouse models, and esculetin inhibited collagen- and arachidonic acid-induced platelet aggregation in washed human platelets. However, it had no effects on other agonists such as thrombin and U46619, and its mechanism is not also clearly known. This study investigated the effect of esculetin on collagen-induced human platelet aggregation, and we clarified the mechanism. Esuletin has effects on the down regulation of PI3K/Akt and MAPK, phosphoproteins that act in the signaling process in platelet aggregation. The effects of esculetin reduced of TXA2 production and phospholipase A2 activation, and intracellular granule secretion including ATP and serotonin, leading to inhibit platelet aggregation. These results clearly clarified the effect of esculetin in inhibiting platelet activity and thrombus formation in humans.

Studies on the Production of Amino Acids by Methanol-Utilizing Bacteria (Methanol 자화세균에 의한 아미노산의 생성에 관한 연구)

  • About 600 strains of bacteria which can utilize methanol as the sole source of carbon were isolated from natrual sources, and their ability to produce amino acids on the medium containing methanol as the sole source of carbon was tested by paper chromatography, by which 3 strains were screened by virtue of their relatively shperior amino acid production. These were tentatively identified as follows ; one strain belonged to the genus methylomonas, while the remaining two were members of the genus Hyphomicrobium. Further studies on their growth characteristics were carried out with relation to amino acid production. According to these experiments, the following results were obtained. 1) No vitamin added was effective for the growth enhancement. 2) Optimum initial concnetration of methanol was proved to be one per cent (v/v), and no conspicuous effect of feeding methanol on the growth of these bacteria ould be observed, provide that the initial methanol concentration was controlled to 1 per cent. 3) Among the nitrogen sources tested, 0.1 per cent of ${(NH_4)}_2SO_4$ enabled the best growth. 4) Addition of 0.001 per cent $MgSO_4.$ $7H_2O$ largely increased the growth of all three strains. Other metal ions tested were either not effective or strongly inhibitory for the growth. 5) Addition of 0.1 per cent each of yeast extract, corn steep liquor, beef extract, and peptone also increased the growth markedly. 6) About $75{\mu}$g/ml of alanine could be obtained finally after 64 hr culture of Hypm. A on the medium lcontaining 0.1 per cent yeast extract and 1 percent methanol. Approximately $40.{\mu}$g/ml of glutamic acid by Hypm. GA and $43.{\mu}$g/ml of alanine by Mtlm. A were produced at the same culture conditions.
