The Study of Cyclophosphamide Metabolite $^{15}N-Isophosphamide$ Mustard (항암제인 Cyclophosphamide의 중간체인 $^{15}N-Isophosphoramide$ Mustard에 관한 연구)

  • The each nitrogen site of ifosfamide metabolite isophosphoramide mustard was synthesized with isotope enriched nitrogen. $Gylcine-^{15}N$ was converted to $2-chloroethylamine-^{15}N$ hydrochloride which was then reacted with phenyl dichlorophosphate to provide $N,N'-bis(2-chloroethyl)phosphordiamidic-^{15}N_2$ acid phenylester(50%, $PhO(O)^{15}N(CH_2CH_2Cl_2)$. Catalytic hydrogenation of this phenyl ester followed by the addition of cyclohexylamine (CHA) provided $IPM-^{15}N$ as the CHA salt(70%).

Overexpression of afsR and Optimization of Metal Chloride to Improve Lomofungin Production in Streptomyces lomondensis S015

  • As a global regulatory gene in Streptomyces, afsR can activate the biosynthesis of many secondary metabolites. The effect of afsR on the biosynthesis of a phenazine metabolite, lomofungin, was studied in Streptomyces lomondensis S015. There was a 2.5-fold increase of lomofungin production in the afsR-overexpressing strain of S. lomondensis S015 N1 compared with the wild-type strain. Meanwhile, the transcription levels of afsR and two important genes involved in the biosynthesis of lomofungin (i.e., phzC and phzE) were significantly upregulated in S. lomondensis S015 N1. The optimization of metal chlorides was investigated to further increase the production of lomofungin in the afsR-overexpressing strain. The addition of different metal chlorides to S. lomondensis S015 N1 cultivations showed that CaCl2, FeCl2, and MnCl2 led to an increase in lomofungin biosynthesis. The optimum concentrations of these metal chlorides were obtained using response surface methodology. CaCl2 (0.04 mM), FeCl2 (0.33 mM), and MnCl2 (0.38 mM) gave a maximum lomofungin production titer of 318.0 ± 10.7 mg/l, which was a 4.1-fold increase compared with that of S. lomondensis S015 N1 without the addition of a metal chloride. This work demonstrates that the biosynthesis of phenazine metabolites can be induced by afsR. The results also indicate that metal chlorides addition might be a simple and useful strategy for improving the production of other phenazine metabolites in Streptomyces.

  • The rate constants for the nucleophilic addition of potassium cyanide to α,N-diphenylnitrone and its derivatives (p-OCH3, p-CH3, p-Cl, and p-NO2) were determined by ultraviolet spectrophotometer at 25℃, and the rate equations which can be applied over a wide pH range were obtained. On the basis of pH-rate profile, adduct analysis, general base catalysis and substituent effect, a plausible mechanism of this addition reaction was proposed: At high pH, the cyanide ion to carbon-nitrogen double bond was rate controlling, however, in acidic media, the reaction proceeded by the addition of hydrogen cyanide molecule to carbon-nitrogen double bond after protonation at oxygen of a,N-diphenylnitrone. In the range of neutral pH, these two reactions occured competitively.

Comparison of Reduction Effect of Contaminants and Odor According to DO Change and EM Addition in Maturation Process of Piggery Slurry (양돈장 분뇨의 부숙 과정에서 DO 변화와 EM의 첨가에 따른 오염물질 및 악취 저감효과의 비교)

  • It was compared the reduction effect of contaminants and odor according to DO change and EM (effective microorganisms) addition in maturation process of piggery slurry. The maturation processes were divided into three cases as follows: R-1 was operated at $2.5mg/{\ell}$ of DO without the addition of EM, R-2 was operated at $7.5mg/{\ell}$ of DO without the addition of EM and R-3 was operated at $2.5mg/{\ell}$ of DO with the addition of EM. The addition of EM was more effective than the increase of DO for the reduction of CODcr, $NH_3-N$ and T-N in the maturation of piggery slurry. In addition, the reducing effect of odor intensity appeared high even in the short-term maturation period in case of adding EM and one could not even smell the stimulating odor of piggery slurry.

  • Duggal-Jeon-Kubrusly([2]) introduced Hilbert space operator T satisfying property ${\mid}T{\mid}^2{\leq}{\mid}T^2{\mid}$, where ${\mid}T{\mid}=(T^*T)^{1/2}$. In this paper we extend this property to general version, namely property B(n). In addition, we construct examples which distinguish the classes of operators with property B(n) for each $n{\in}\mathbb{N}$.

  • In this paper, we consider the hamiltonian properties of m$\times$n (m$\geq$2, n$\geq$3) mesh networks with two wraparound edges on the first row and last row, called M$_2$(m, n), in the presence of a faulty node or link. We prove that M$_2$(m, n) with odd n is hamiltonian-connected and 1-fault hamiltonian. In addition, we prove that M$_2$(m, n) with even n is strongly hamiltonian laceable and 1-vertex fault tolerant strongly hamiltonian laceable.

  • Let R be a ring with identity $1_R$ and let U(R) denote the group of all units of R. A ring R is called locally finite if every finite subset in it generates a finite semi group multiplicatively. In this paper, some results are obtained as follows: (1) for any semilocal (hence semiperfect) ring R, U(R) is a finite (resp. locally finite) group if and only if R is a finite (resp. locally finite) ring; U(R) is a locally finite group if and only if U$(M_n(R))$ is a locally finite group where $M_n(R)$ is the full matrix ring of $n{\times}n$ matrices over R for any positive integer n; in addition, if $2=1_R+1_R$ is a unit in R, then U(R) is an abelian group if and only if R is a commutative ring; (2) for any semiperfect ring R, if E(R), the set of all idempotents in R, is commuting, then $R/J\cong\oplus_{i=1}^mD_i$ where each $D_i$ is a division ring for some positive integer m and |E(R)|=$2^m$; in addition, if 2=$1_R+1_R$ is a unit in R, then every idempotent is central.

  • Ti1-xAlxN films were successfully deposited on high speed steel and silicon wafer by plasma-assisted chemical vapor deposition using a TiCl4/AlCl3/N2/Ar/H2 gas mixture. Plasma process enabled N2 gas to nitride AlCl3, which is not possible in sense of thermodynamics. XPS analyses revealed that the deposited layer contained Al-N bond as well as Ti-N bond. Ti1-xAlxN films were polycrystalline and had single phase, B1-NaCl structure of TiN. Interplanar distance, d200, of (200) crystal plane of Ti1-xAlxN was, however, decreased with Al content, x. Al incorporation into TiN caused the grain size to be finer and changed strong (200) preferred orientation of TiN to random oriented microstructure. Those microstructural changes with Al addition resulted in the increase of micro-hardness of Ti1-xAlxN film up to 2800Kg/$\textrm{mm}^2$ compared with 1400Kg/$\textrm{mm}^2$ of TiN.

Characterization of CH4-oxidizing and N2O-reducing Bacterial Consortia Enriched from the Rhizospheres of Maize and Tall Fescue (옥수수와 톨페스큐 근권 유래의 메탄 산화 및 아산화질소 환원 세균 컨소시움 특성)

  • CH4-oxidizing and N2O-reducing bacterial consortia were enriched from the rhizosphere soils of maize (Zea mays) and tall fescue (Festuca arundinacea). Illumina MiSeq sequencing analysis was performed to comparatively analyze the bacterial communities of the consortia with those of the rhizosphere soils. Additionally, the effect of root exudate on CH4 oxidation and N2O reduction activities of the microbes was evaluated. Although the inoculum sources varied, the CH4-oxidizing and N2O-reducing consortia derived from maize and tall fescue were similar. The predominant methanotrophs in the CH4-oxidizing consortia were Methylosarcina, Methylococcus, and Methylocystis. Among the N2O-reducing consortia, the representative N2O-reducing bacteria were Cloacibacterium, Azonexus, and Klebsiella. The N2O reduction rate of the N2O-reducing consortium from maize rhizosphere and tall fescue rhizosphere increased by 1.6 and 2.7 times with the addition of maize and tall fescue root exudates, respectively. The CH4 oxidization activity of the CH4-oxidizing consortia did not increase with the addition of root exudates. The CH4-oxidizing and N2O-reducing consortia can be used as promising bioresources to mitigate non-CO2 greenhouse gas emissions during remediation of oil-contaminated soils.
