Brevibacterium flavum ATCC 14067과 Corynebacterium glutamicum ATCC 13032의 원형질체 융합에 의한 L-Methionine의 생산 (L-Methionine Production by Protoplast Fusion of Brevibacterium flavum ATCC 14067 and Corynebacterium glutamicum ATCC 13032)

  • 본 연구는 Brevibacterium flavum ATCC 14067 및 Corynebacterium glutamicum ATCC 13032간의 protoplast fusion을 행하여 L-methionine의 생산성을 검토하고 발효조건을 개선하기 위하여 연속배양을 행하였다. N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) $500{\mu}g/ml$로 처리하였으며 B. flavum K-104($Thr\;Met\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}$)와 C. glutamicum B-70($Thr\;Hos\;Km^{r}\;Et^{r}\;Sm^{r}\;Tm^{r}$)의 변이주를 분리하였다. 이들 변이주에 $500{\mu}g/ml$의 lysozyme을 처리하였을 때 원형질체 형성을 및 재생율은 각각 99% 및 $64{\sim}66%$를 나타내었으며 융합 빈도는 3% PVP를 함유한 35% PEG 용액에서 $3.5{\times}10^{5}$을 나타내었다. Sodium alginate로 고정화시킨 융합주 BFCG 37은 72시간 회분배양에서 0.89g/l의 methionine을 생산하였고 연속배양에서는 $18.75mg/^{1}hr\;^{1}$의 L-methionine를 안정적으로 생산할 수 있었다.

  • 사파이어 기반의 GaN LED의 광추출 효율을 시뮬레이션을 이용하여 정량적으로 평가하였다. 각각의 LED는 ray-tracing 시뮬레이션을 이용하여 광추출 효율을 계산하였다. 본 연구에는 PSS(patterned sapphire substrate) LED와 flat LED의 비교 분석을 통하여, LED에서의 사파이어 기판의 패턴이 광추출에 미치는 영향을 설명하였다. 또한, 각각의 칩에서 반사막의 반사도가 광추출에 미치는 영향을 분석하고, 그 원인을 시뮬레이션을 이용하여 설명하였다. 한편, 사파이어 패턴에 의한 광추출효율의 변화 효과를 공기(air)와 실리콘(silicone) 분위기에서 시뮬레이션을 수행하였다. 이러한 시뮬레이션 기술을 통해 광추출 효율의 개선 정도를 정량적으로 평가할 수 있었으며, 이러한 연구가 향후 고효율 LED 개발에 도움을 줄 것으로 판단된다.

Development of a Multiplex PCR Method to Detect Fungal Pathogens for Quarantine on Exported Cacti

  • Major diseases in grafted cacti have been reported and Fusarium oxysporum, Bipolaris cactivora, Phytophthora spp. and Collectotrichum spp. are known as causal pathogens. These pathogens can lead to plant death after infection. Therefore, some European countries have quarantined imported cacti that are infected with specific fungal pathogens. Consequently, we developed PCR detection methods to identify four quarantined fungal pathogens and reduce export rejection rates of Korean grafted cacti. The pathogen specific primer sets F.oF-F.oR, B.CF-B.CR, P.nF-P.nR, and P.cF-P.CR were tested for F. oxysporum, B.cactivora, P. nicotinae, and P. cactorum, respectively. The F.oF-F.oR primer set was designed from the Fusarium ITS region; the B.CF-B.CR and P.nF-P.nR primers respectively from Bipolaris and Phytophthora ITS1; and the P.cF-P.CR primer set from the Ypt1protein gene region. The quarantine fungal pathogen primer pairs were amplified to the specific number of base pairs in each of the following fungal pathogens: 210-bp (F. oxysporum), 510-bp (B. cactivora), 313-bp (P. nicotinae), and 447-bp (P. cactorum). The detection limit for the mono- and multiplex PCR primer sets was 0.1 ng of template DNA under in vitro conditions. Therefore, each primer set successfully diagnosed contamination of quarantine pathogens in export grafted cacti. Consequently, our methodology is a viable tool to screen contamination of the fungal pathogen in exported grafted cacti.

스티렌과 불소함유 아크릴레이트의 공중합 및 공중합체의 특성 (Copolymerization and Characteristics of Styrene and Fluorine-Containing Acrylate)

  • 탄화수소의 수소 부분이 불소로 치환된 과불소알킬기(perfluoro group : $CF_3(CF_2)_nCH_2CH_2$-; n=5, 7, 9, 11)를 함유하고 있는 perfluoroalkylethyl acrylate(FA)와 styrene을 bulk 상태에서 $60^{\circ}C$를 유지하며 $\alpha$, $\alpha$'-azobisisobutyronitrile(AIBN)을 개시제로 하여 각각의 단량체의 반응 몰비를 변화시키며 전환율을 10% 이내로 제한하여 공중합체를 합성하였다. 또한 반응 몰비의 변화에 따른 중합된 공중합체내의 반복단위의 함량사이의 관계와 Helen-Tudos법을 이용하여 단량체 반응성비 $r_1$, $r_2$를 구하였다. FT-IR과 $^1H-NMR$을 통해 두 단량체의 공중합 여부와 구조를 정성적으로 확인하였고 공중합체의 조성은 $^1H-NMR$분석을 통해 구하였다. 단량체의 비율에 따른 열적 특성은 DSC와 TGA를 통하여 분석하였으며 공중합체와 소량의 공중합체가 첨가된 PMMA의 접촉각을 측정하여 공중합체의 표면특성을 나타내는 표면자유에너지를 계산하였다.

Nitrogen and Fluorine Co-doped Activated Carbon for Supercapacitors

  • Activated carbon has lower electrical conductivity and reliability than other carbonaceous materials because of the oxygen functional groups that form during the activation process. This problem can be overcome by doping the material with heteroatoms to reduce the number of oxygen functional groups. In the present study, N, F co-doped activated carbon (AC-NF) was successfully prepared by a microwave-assisted hydrothermal method, utilizing commercial activated carbon (AC-R) as the precursor and ammonium tetrafluoroborate as the single source for the co-doping of N and F. AC-NF showed improved electrical conductivity ($3.8\;S\;cm^{-1}$) with N and F contents of 0.6 and 0.1 at%, respectively. The introduction of N and F improved the performance of the pertinent supercapacitor: AC-NF exhibited an improved rate capability at current densities of $0.5-50mA\;cm^{-2}$. The rate capability was higher compared to that of raw activated carbon because N and F codoping increased the electrical conductivity of AC-NF. The developed method for the co-doping of N and F using a single source is cost-effective and yields AC-NF with excellent electrochemical properties; thus, it has promising applications in the commercialization of energy storage devices.

  • In [1, 2], described a Chebyshev series method for the numerical solution of integral equations with three automatic algorithms for computing tow regularization parameters, C/sub f/ and r. Here we describe a Fourier series expansion method for a class singular integral equations with Hilbert kernel and constant coefficients with using a new automatic algorithm.

Auto Regressive모델링 기반의 특징점 추출과 Support Vector Machine을 통한 조기수축 부정맥 분류 (Feature Extraction based on Auto Regressive Modeling and an Premature Contraction Arrhythmia Classification using Support Vector Machine)

  • 부정맥 분류를 위한 기존 연구들은 분류의 정확성을 높이기 위해 신경망, 퍼지, 시계열 주파수 분석, 비선형 분석법 등이 연구되어 왔다. 이러한 방법들은 분류율를 향상시키기 위해 정확한 특징점과 많은 양의 신호를 처리해야 하기 때문에 데이터의 가공 및 연산이 복잡하며, 다양한 부정맥을 분류하는데 어려움이 있다. 본 연구에서는 AR(Auto Regressive) 모델링 기반의 특징점 추출과 SVM(Support Vector Machine)을 통한 조기수축 부정맥 분류 방법을 제안한다. 이를 위해 잡음을 제거한 ECG 신호에서 R파를 검출하고 QRS와 RR 간격의 특정 파형 구간을 모델링하였다. 이후 최적 세그먼트 길이(n1, n2), 최적 차수( p1, p2)의 4가지 AR 모델링 변수를 추출하고 SVM을 통해 Normal, PVC, PAC를 분류하였다. 연구의 타당성을 입증하기 위해 MIT-BIH 부정맥 데이터베이스를 대상으로 한 R파의 평균 검출 성능은 99.77%, Normal, PVC, PAC 부정맥은 각각 99.23%, 97.28, 96.62의 평균 분류율을 나타내었다.

아세트아미노펜 액상좌제의 제초 및 생물학적 동등성 평가 (Preparation and Bioequivalence Test of Acetaminophen Liquid Suppository)

  • A novel in situ-gelling and mucoadhesive acetaminophen liquid suppository was developed to improve the patient compliance of conventional solid suppository. In this study, acetaminophen liquid suppository, Likipe $n_{R}$, [aminophen/Poloxamer 407/Poloxamer 188/so4ium alginate (5/15/19/0.6%)] with relation temperature at 30-36 "C and suitable gel strength and bioadhesive force, dissolution pattern similar to conventional solid type suppository, Suspe $n_{R}$, was developed. Furthermore, the bioequivalence of two acetaminophen products was evaluated in 16 normal male volunteers (age 22-27 yr, body weight 56-72 kg) following sidle rectal administration. Test product was Likipe $n_{R}$ suppository (Dong-Wha Pharm. Corp., Korea)and reference product was Suspe $n_{R}$204-212 suppository (Hanmi Pharm. Corp., Korea). Both products contain 125 mg of acetaminophen. Four Suppositories of the test and the reference product were administered to the volunteers, respectively, by randomized two period cross-over study (2$\times$2 Latin square method). The determination of acetaminophen was accomplished using HPLC. Average drug concentrations at each sampling time and pharmacokinetic parameters calculated were not significantly different between two products (p>0.05); the area under the curve to last sampling time (24 hr) (AU $Co_{-2}$4h/) (30.14$\pm$8.64 vs 27.98$\pm$ 6.53 $\mu$g .h/ml), maximum plasma concentration ( $C_{max}$) (3.29$\pm$0.87 vs 3.60$\pm$0.66 $\mu$g/ml) and time to maximum plasma concentration ( $T_{max}$) (2.91 $\pm$0.55 vs 2.69$\pm$0.60 h). The differences of mean AUCo $_{24h}$, C-a. and T-between the two products (7.18%, 9.58% and 7.53%, respectively) were less than 20%. The power (1-7) and treatment difference ($\Delta$) for AU $Co_{24h}$, $C_{max}$ and $T_{max}$ were more than 0.8 and less than 0.2, respectively at $\alpha$=0.1. The confidence limits for AU $Co_{24h}$, $C_{max}$ and $T_{max}$ (-0.81 ~13.55%, -1.56~ 17.60 and -3.81 ~18.87%, respectively) were less than $\pm$ 20% at $\alpha$=0.1. These results suggest that the bioavailability of Likipe $n_{R}$ suppository is not significantly different from that of Suspe $n_{R}$ suppsitory. Therefore, two products are bio-equivalent based on the current results.results.lts.sults.results.lts.

  • The advantage of application polymer instead of conventional porcelain as the housing material for arrester is that light weight, shorter section length of arrester would be possible due to the use of an insulated hanger (mold compound). also, polymer arrester has very good characteristics in design versatility as well as excellent resistance to moist ingress and pollution performance. The finite element method is widely used to calculate for electromagnetic devices. In this paper, we perform the each conditional analysis of PC with an advanced graphical user interface(GUI) using the vector fields simulator. And we try to analyze electromagnetic fields through the conventional two dimensional analysis in case of high voltage contamination on the electrode of a arrester.

  • In this paper, we study the following p&q-Laplacian problem with critical Sobolev and Hardy exponents {$-{\Delta}_pu-{\Delta}_qu={\mu}\frac{{\mid}u{\mid}^{p^*(s)-2}u}{{\mid}x{\mid}^s}+{\lambda}f(x,\;u)$, in $\Omega$, u=0, on $\Omega$, where ${\Omega}\;{\subset}\;\mathbb{R}^{\mathbb{N}}$ is a bounded domain and ${\Delta}_ru=div({\mid}{\nabla}u{\mid}^{r-2}{\nabla}u)$ is the r-Laplacian of u. By using the variational method and concentration-compactness principle, we obtain the existence of infinitely many small solutions for above problem which are the complement of previously known results.
