Trends in Mycotoxin Contamination of Cereals and Cereal Products in Korea (국내산 곡류와 곡류 가공품의 곰팡이독소 오염 동향)

  • In this review, the mycotoxin contamination of Korean cereals and their products is analyzed by crop based on scientific publications since 2000. Barley, rice, and corn were investigated heavier than the others. The common mycotoxins occurred in all cereals and their products were deoxynivalenol and zearalenone. Nivalenol was detected in all samples analyzed but more frequently or mainly in barley, rice, and oat. Fumonisin was commonly detected in corn and sorghum but also in adlay, millet, and rice. Adlay and millet were similar in the contamination pattern that fumonisin and zearalenone were the most frequently detected mycotoxins. Zearalenone was the most commonly detected mycotoxin with concentrations higher than the national standards (maximum limit), followed by deoxynivalenol, and aflatoxin. However, most occurrence levels were below the maximum limits for respective mycotoxins. This result shows that barley, rice, corn, sorghum, millet, and adlay are more vulnerable to mycotoxin contamination than other cereals and therefore continuous monitoring and safety management are necessary.

Developmental and Neurobehavioral Effects of Mycotoxin Fumonisin B1 in Rats

  • The objective of this experiment is to investigate neurobehavioral and developmental effects of fumonisin B1 (FB1) after prenatal FB1 administration in rats. FB1 (0.8 or 1.6 mg/kg) was orally exposed to pregnant rats during gestational days 13 to 20, whereas the vehicle alone was administered to control group. Maternal and offspring body weights, physical landmarks of incisor eruption, eye opening, testes descending and vaginal opening, open field activity, running wheel activity, and complex maze performance were included as endpoints for developmental and neurobehavioral measurement. Maternal body weights were not signfficantly altered after FB1 exposure. Percentage of maternal weight gain difference between control and 1.6 mg/kg FBI groups was about 4%. Pre- and post-weanling weight of offsprings after prenatal exposure to FB1 was not signfficantly changed, suggesting that FB1 at 0.8 or 1.6 kg/kg doses may not cross the placenta. Significant gender difference in running wheel activity on postnatal days 57 to 63 and complex maze performance on postnatal days 75 to 78 was observed.

  • Effects of combined treatments of radiation-heat and radiation-chemicals on the survival of mycotoxin-producing molds Aspergillus flavus and Penicillum islandicum conidia were investigated. Combined treatments of gamma-radiation and heat (10 minutes at $50^{\circ}C\;or\;55^{\circ}C$) showed a synergistic effect, causing remarkable decreases in $D_{10}$ values and induction doses in the two molds. Combined treatments of gamma-radiation and chemicals (sorbic acid) had no synergistic effect. Asp. flavus was more resistant to heat and sorbic acid than was Pen. islandicum.

Heterotrimeric G protein signaling and RGSs in Aspergillus nidulans

  • Heterotrimeric G proteins (G proteins) are conserved in all eukaryotes and are crucial components sensing and relaying external cues into the cells to elicit appropriate physiological and biochemical responses. Basic units of the heterotrimeric G protein signaling system include a G protein-coupled receptor (GPCR), a G protein composed of ${\alpha},\;{\beta},\;and\;{\gamma}$ subunits, and variety of effectors. Sequential sensitization and activation of these G protein elements translates external signals into gene expression changes, resulting in appropriate cellular behaviors. Regulators of G protein signaling (RGSs) constitute a crucial element of appropriate control of the intensity and duration of G protein signaling. For the past decade, G protein signaling and its regulation have been intensively studied in a number of model and/or pathogenic fungi and outcomes of the studies provided better understanding on the upstream regulation of vegetative growth, mating, development, virulence/pathogenicity establishment, and biosynthesis of secondary metabolites in fungi. This review focuses on the characteristics of the basic upstream G protein components and RGS proteins, and their roles controlling various aspects of biological processes in the model filamentous ascomycete fungus Aspergillus nidulans. In particular, their functions in controlling hyphal proliferation, asexual spore formation, sexual fruiting, and the mycotoxin sterigmatocystin production are discussed.

  • Doenjang (Korean fermented soybean paste) is one of important fermented foods in Korea. Doenjang has been traditionally manufactured from meju which is fermented rectangular shape of crushed cooked soybeans. The main microorganisms involved for meju fermentation are Bacillus subtilis and molds such as Rizopus sp., Mucor sp. and Aspergillus sp. We have already reported that Doenjang is free from mycotoxin, especially, aflatoxin B$_1$contamination during the manufacturing process of the Deonjang.(omitted)

Analysis of Total Aflatoxin, Ochratoxin A, Zearalenone, Deoxynivalenol and T-2 Toxin Contamination in Nuts (시중유통 견과류의 총아플라톡신, 오크라톡신 A, 제랄레논, 데옥시니발레놀, T-2 독소의 오염도 조사)

  • In the current study, 109 commercial nut samples were collected from different Korean markets and analyzed for the contamination of 5 different mycotoxins (aflatoxin, ochratoxin A, deoxynivalenol, zearalenone, and T-2 toxin) using ELISA kits. The results revealed that the most frequently detected mycotoxin was zearalenone (n=36, 33%), followed by aflatoxin (n=31, 28.4%) and ochratoxin A (n=30, 27.5%). Deoxynivalenol and T-2 toxin were also detected in 22 (20.3%) samples, respectively. Among 109 nut samples, 33 samples (30.3%) were contaminated only with one kind of mycotoxin, whereas 43 samples had at least 2 kinds of mycotoxins. Two samples were contaminated with as many as 4 different mycotoxins, and they were both walnuts. Although the monitoring results revealed the amount of aflatoxin contamination was under the safety criteria, there is no current safety guideline for other kinds of mycotoxins or multiple contaminations in Korea. Therefore, further studies should be performed to reveal the distribution of mycotoxin in different foods and propose appropriate safety guidelines for Korean markets.

사료 내 곰팡이독소의 객관적 접근 필요

  • 우리나라의 사료관리법 상에서 곰팡이독소(Mycotoxins)의 규제 현황을 보면 이전에는 아플라톡신만을 유해물질 규제 대상으로 하다가, 오크라톡신을 추가하여 현재까지 규제 대상독소로 규정하고 있다. 그러나, 이미 사료공장과 관련 업체에서는 현행의 사료관리법에서 유해물질로서 규제하는 곰팡이독소보다도 규제대상이 아닌 후모니신이나 T-2독소 및 제아레나론을 비롯한 곰팡이독소가 중국산 곡물원료에서 더 문제가 된다는 사실을 외국기관에 직접 분석 의뢰한 샘플에서 나온 사실과 각종 정보경로를 통해서 알고 현재는 항곰팡이제와 더불어서 곰파이독소흡착제거제(Mycotoxin binders)를 병용해서 사용하는 것이 일반화되고 있다.

  • The effect of patulin, a mycotoxin, on the growth of Escherichia coli cell was investigated. E. coli cell elongation usually shown in SOS-response for DNA repair was induced by 20 mg of patulin per ml. After staining the E. coli chromosome with fluorescence dye(DAPI, 4', 6-diamino-2-phenyl-indole), chromosomal DNA partitioning was not affected by patulin. The observation indicateds that patulin acts as a DNA damaging agent which is effective for E. coli cell elongation introduced by the inhibition of septum formation.

Mating Behavior, Mycotoxin Production, and Vegetative Compatibility of Gibberella fujikuroi Species Complex from Sorghum in Korea

  • Fusarium isolates of Gibberella fujikuroi species complex were obtained from sorghum grown in five provinces of Korea in 1996 and 1997. These isolates were characterized based on their mating behavior, mycotoxin production, and vegetative compatibility. Only three mating populations (A, D, and F) were recovered from a total of 155 isolates examined. The relative frequency of the mating populations was significantly different: F was predominant (80%), while D and A were observed at low frequencies of 9% and 3%, respectively. Female fertile isolates were more common within F (44 our of 124) than D (2 out of 14), while none of the five A isolates were female fertile. The inbreeding effective population sizes ($\textrm{N}_e$)for mating type and male/hermaphrodite ratios in mating populations A and D produced significant amounts of fumonisins, while F isolates produced none or only traces of fumonisin B$_1$. In contrast. F isolates produced higher amounts of moniliformin (average of 3,820 ppm) than A and D isolates (averages of 77 and 1,819 ppm, respectively). Fifty-one isolates were tested for vegetative compatibility using nitrogen non-utilization mutants of each isolate, and 44 vegetative compatibility groups (VCGs) were identified. A single VC type (VC1) was found in all of the five A isolates examined. Six of the D isolates examined consisted of three VC types: two for VC2, two for VC3, and the rest for VC4. All of the F isolates tested were incompatible in every combination and , thus, each constituted a unique VCG.

Studies on the Toxigenic Fungi and Detoxification Methods Isolated from Korean Foodstuffs (한국(韓國) 식품중(食品中)의 유독성(有毒性) 진균(眞菌)의 검색(檢索) 및 해독(解毒)에 관(關)한 연구(硏究))

  • These studies were carried out to detect the presence of mycotoxin fungi and detoxification of toxins in various kinds of grain and foodstuffs in Korea. The experiments were divided into three parts: mycologic, toxicologic and electron microscopic study. The resuIts were summarized as follows: 1. From the 210 various, samples(local grains, 150 samples; rice-cakes, 50 samples; meju, 10 samples), 1,205 colonies or fungi were isolated. In 1,127 of 1,205 colonies, it was possible to identify 21 genera. Among the identified strains, the predominant genera were Aspergillus sp.(28.5%), Penicillium sp.(27.1%) and Mucor sp.(7.8%). 2. In cytotoxicity test on HeLa cells, 25 strains showed severe toxic effects among the 240 strains of experiments. 3. In histopathologic test on mice, 21 strains showed severe toxic effect to mouse liver cells among the 240 tested strains. 4. In electron microscopic studies of HeLa cells and mouse liver cells from animal which had been treated with crude toxin, the liver cells showed the cytoplasmic change: dillatation or vacuolization of endoplasmic reticulum, swelling of mitochondria and disappearance of mitochondrial cristae, increased number of lipid and glycogen particles. Nucleus and nucelar envelope alterations, were also noted. 5. In the detoxification study with ultraviolet irradiation, 90% of the toxic substances were denatured by the ultraviolet light irradiation for 24 hours. 6. As a mass screening, the cytotoxicity test of HeLa cells and histopathologic study of mice liver cells treated with culture filtrates, employed and feasible to detect mycotoxin producing fungi.
