Artificial Cultivation and Antioxidant Activity of Korean Native Mushroom Pycnoporus coccineus (한국산 간버섯의 인공재배 및 항산화 활성 평가)

  • The mycelial growth of P. coccineus strain was good in PDA and YMA, but mycelial growth was low in MEA. Light irradiation during the incubation period affected the pigment formation and density of mycelia. Mushroom of P. coccineus strain was able to produce fruiting bodies in both bottle and bag cultivation, and oak sawdust was found to be the most suitable substrate for spawn culture and cultivation. In artificial cultivation using sawdust medium, fruiting body was grown to the extent that visual observation was possible from the 15th day, and it formed about 5 days fast in the treatment group with low relative humidity. From 40 to 45 days of mushroom development, mature fruiting bodies could be harvested, and the lower relative humidity of the growing room favored mushroom development and growth. Antioxidant activity of fruiting bodies harvested from artificial cultivation showed that ABTS radical scavenging activity of bottle-cultivated and wild fruit bodies were shown at 505㎍/㎖ and 515㎍/㎖, respectively. However, fruiting bodies harvested in bag cultivation were high at 910㎍/㎖. As a result of DPPH radical scavenging activity, all extracts were found to be inactive, exhibiting IC50 value of more than 2,000㎍/㎖ concentration. The ethyl acetate extract of mushrooms obtained from bottle cultivation showed the highest activity with 1,550㎍/㎖ IC50 value. Methanol extract of wild fruit bodies had the highest ABTS radical scavenging activity at the same concentration (10mg/㎖).

  • In this study, we investigated the microbial community of oyster mushrooms at different growth stages at the species level. Gram-positive bacteria were predominant in the presterilized medium. On the other hand, Gram-negative bacteria were predominant in the culture-completed medium, post-harvest medium, and fruiting bodies. In addition, Pseudomonas tolaasii, which is known to cause disease in mushrooms, was confirmed in the cultured medium, post-harvest medium, and fruiting bodies, and it was determined that the mycelium culture stage was contaminated, and the reason why no disease occurred was Sphingobacterium psychroaquaticum. It was confirmed that this was because the growth of Pseudomonas tolaasii was suppressed by producing a component called tolacin. As a result of confirming the diversity of microorganisms, it was confirmed that the presterilization medium contains a variety of microorganisms compared to other growth stages, and the diversity decreases in the order of culture completion medium, fruiting body, and post-harvest medium. showed a trend. As a result of microbial similarity analysis, it was confirmed that the cultured medium and the post-harvest medium showed similar microbial communities, and in the case of fruiting bodies, there were some similarities but overall differences.

Growth promoting effect on Tricholoma matsutake mycelium by bacteria from fairy Ring in Bonghwa-gun, Korea (국내 봉화 송이 자생지 내 균환 유래 토양세균의 송이균사체 생장촉진 효과)

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

Studies on the Cellulolytic Enzymes Produced by Ganoderma lucidum in Synthetic Media (합성배지(合成培地)에서 불로초(不老草)가 생산(生産)하는 섬유소(纖維素) 분해효소(分解酵素)에 관한 연구(硏究))

  • Factors affecting the productivity of cellulolytic enzymes and the mycelial growth of Ganoderma lucidum CAFM 9065 were examined in synthetic media. Among the carbon sources tested, Na-CMC was the best for the production of avicelase CMC ase, and cellobiose for ${\beta}-glucosidase$. Soluble starch and cellobiose were the best for the mycelial growth. The optimum concentration of Na-CMC for the production of the enzymes was 1.0 %, and mycelial growth increased remarkably with the higher concentration of Na-CMC. Glucose inhibited the production of the enzymes, but stimulated the mycelial growth. Among the nitrogen sources used, peptone was the most effective for the production of the enzymes, and the appropriate concentration of peptone was 0.2%. The mycelial growth was stimulated with the increase of the concentration of peptone up to 0.5%. The optimum concentration of $KH_2PO_4$ for the production of the enzymes and mycelial growth was 0.3 and 0.2%, respectively. The optimum concentration of $MgSO_4{\cdot}7H_2O$ for the production of the enzymes and mycelial growth was 0.02%. The production of the enzymes was facilitated by folic acid at a low concentration (0.03 mg/l), and mycelial growth by inositol. The optimum temperature for the production of the enzymes and mycelial growth was $30^{\circ}C$. The optimum pH for the production of avicelase and ${\beta}-glucosidase$ was 5.0 equally and CMCase 5.5. The activities of avicelase and CMCase were the highest at 8 and 10 days of culture, respectively and that of ${\beta}-glucosidase$ at 16 day culture. The growth of mycelium was the highest at 12 days of culture at pH 5.0.

Antioxidant and Anti-inflammatory Effects of Phellinus linteus HN1009K (목질진흙버섯 Phellinus linteus HN1009K의 항산화 활성과 항염증효과)

  • Novel Phellinus linteus HN1009K (HK1009) has been characterized on speedy mycelial growth and yearly artificial cultivation. In this study, antioxidant and anti-inflammatory activities of ethanol extract from mycelium and fruiting body of P. linteus HN1009K (HK1009) were compared to those of P. linteus (PL, Korea Sangwhang) and P. baumii (PB, Jangsu Sanghwang) as controls. HK1009 extract showed higher DPPH free radical scavenging activity in both mycelial and fruiting body samples than those of PL and PB. However, in xanthine oxidase assay, antioxidant effect was highly measured on mycelial samples of PL and HK1009. Mycelial extract of HK1009 induced high inhibitory effect on LPS-induced nitric oxide (NO) production in RAW264.7 cells comparing to PL and PB without toxicity on the cells. The results suggest that P. linteus HN1009K possesses superior antioxidant activity and anti-inflammatory effects.

Isolation of Antimicrobial Substance by Produced Bacillus sp. SD-10 with Antagonistic Activity Towards Mushroom Pathogens (버섯병원균에 대한 길항세균 Bacillus sp. SD-10이 생산하는 항균물질의 분리)

  • Bacillus sp. SD-10 was investigated to develope biological pesticides for control of mushroom diseases. Bacillus sp. SD-10 showed high antifungal activity when cultured at 35∼4$0^{\circ}C$ for 30∼4$0^{\circ}C$. The culture filtrate of the bacterium inhibited the growth of mycelium of T. virens which is a kind of mushroom pathogene. On the test of inhibition of spore germination of T. virens, more than 5% of the culture filtrate in the media inhibited completely the germination of the spores. An antimicrobial substance, UPX-1 was purified from the culture filtrate of the Bacillus. From the $^1H$-NMR and $^{13}C$-NMR spectrum analysis, the substance was indentifed as disaccharide composed to six carbon sugars. UPX-1 has not only strong antifungal activity against T. virens but also antibacterial activity against Pseudomonas tolaassi.

Cultural and Morphological Characteristics of a New White Button Mushroom Cultivar 'Saedo'

  • Since development of the first Korean button mushroom cultivar 'Sae-Ah' in 2010, mushroom farmers have demanded an improved cultivar. Although a new cultivar, 'Saejeong', was developed in 2011, this cultivar has thus far been difficult to cultivate in typical mushroom farms. To solve this problem, another new cultivar, 'Saedo', was developed in 2012. This new cultivar was selected by genetic analysis from crossing the heterokaryon line A175 and the homokaryon line ASI1346-15. The heterokaryon A175 was a crossed line between strains ASI1038-211 and ASI1346-20. The mycelium of 'Saedo' showed good growth on compost dextrose agar at $25^{\circ}C$. The morphological traits such as the pileus (cap) and stipe of the 'Saedo' cultivar were thicker than those of the reference control 'Saejeong', and the production yield was increased by 1.89-fold compared to that of the reference control. A field experiment of the 'Saedo' cultivar has been ongoing since 2013. The yield of the 'Saedo' cultivar was found to be substantially higher than that of the reference control during the $2^{nd}$ flush.

Effects of Dissolved Oxygen on Fungal Morphology and Process Rheology During Fed-Batch Processing of Ganoderma lucidum

  • Controlling the dissolved oxygen (DO) in the fed-batch culture of the medicinal mushroom Ganoderma lucidum led to a 2-fold increase of the maximum biomass productivity compared with uncontrolled DO conditions. By contrast, extracellular polysaccharide (EPS) production was two times higher under oxygen limitation (uncontrolled DO) than under increased oxygen availability (controlled DO). Morphologically, dispersed mycelium was predominant under controlled DO conditions, with highly branched hyphae, consistent with the enhanced culture growth noted under these conditions, whereas in the uncontrolled DO process mycelial clumps were the most common morphology throughout the culture. However, in both cultures, clamp connections were found. This is an exciting new finding, which widens the applicability of this basidiomycete in submerged fermentation. In rheological terms, broths demonstrated shear-thinning behavior with a yield stress under both DO conditions. The flow curves were best described by the Herschel-Bulkley model: flow index down to 0.6 and consistency coefficient up to 0.2 and 0.6 Pa $s^n$ in uncontrolled and controlled cultures DO, respectively. The pseudoplastic behavior was entirely due to the fungal biomass, and not to the presence of EPS (rheological analysis of the filtered broth showed Newtonian behavior). It is clear from this study that dissolved oxygen tension is a critical process parameter that distinctly influences G. lucidum morphology and rheology, affecting the overall performance of the process. This study contributes to an improved understanding of the process physiology of submerged fermentation of G. lucidum.

  • Auxotrophic mutants were obtained by UV-irradiation to mycelium of Ganoderma lucidum. Induction rate of auxotrophs was 5.78%. Interspecific fusion products of protoplasts were obtained by polyethylene glycol induced fusion of protoplasts from auxotrophic mutants of Ganoderma lucidum and Ganoderma applanatum. Fusion products were selected by means of the comparison with the mycelial growth rate and colony morphology. Fusion products were confirmed by mycelial morphology and esterase isozyme pattern. Some segregants were observed and fusion product produced fruit bodies.

Preparation and Treatment of Sulfur Dioxide Gas Generating Agent for Storage of Grape Fruits (포도 저장을 위한 아황산가스 발생제의 제조 및 처리 방법)

  • In order to establish the method to prepare and use the sulfur dioxide gas generating agent in low temperature storage of grape fruits, the $SO_2$ generation capacity from various sulfite compounds was investigated, and the method to regulate $SO_2$ gas generation and to detect the gas was developed. The conidial germination and mycelium growth of Botrytis cinerea which causes gray mold disease during grape fruit storage was completely inhibited at the $SO_2$ gas concentration of 400 ppm and 3200 ppm, respectively. Sodium hydrosulfite generated the most amount of $SO_2$ gas among the investigated 5 different sulfite or bisulfite compounds. By adjusting the number of pinholes on packaging film of the compound or by adding pH adjusting agent, e.g. citric acid or phosphates, it was possible to regulate the amount and duration of $SO_2$ gas generation from the compound. Because malachite green was quantitatively discolored by $SO_2$ gas, the solution or impregnated paper with the compound could be practically utilized as a indicator detecting $SO_2$ gas. Finally, when Muscat Bailey A grape was stored at low temperature with $SO_2$ gas generating agent, the disease incidence was reduced after storage.
