  • Lentinula edodes is one of the most widely consumed edible mushrooms in Korea. Mating in L. edodes is regulated by a tetrapolar system, and two unlinked genetic loci, A and B, are known to be major determinants of the mating types, as reported in other heterothallic basidiomycetes. The A locus of L. edodes encodes a pair of homeodomain (HD) transcription factors. The highly variable N-termini of these HD transcription factors contribute to the diversity among the A mating types. In this study, we developed a cleaved amplified polymorphic sequence (CAPS) marker to discriminate 11 different A mating type alleles predominant among both cultivated and wild strains. Amplification of the variable region of the A locus followed by digestion with HaeIII and EcoRI restriction enzymes enabled successful discrimination among the 11 A mating type alleles. We also evaluated the applicability of this method in the identification of two A mating types of a dikaryotic strain.

  • Beech mushroom (Hypsizygus marmoreus) is one of the most popular mushrooms in Japan, China, and Korea because of its delicious taste, nutritious value, and unique crunchy texture. In 2018-2019, unusual signs on the mushroom were observed in mushroom farms in Gyeongnam Province. The main signs were the inhibition of mycelial growth and primordial formation on the surface of mushroom media. When green mold caused contamination at early stage, the rate of pinhead formation by the mushroom greatly decreased to 46.5-71.5%. Conidia of the causal green mold were ellipsoidal, dark green, and measured 2.0-3.1㎛ long. The optimum temperature for mycelial growth of the green mold was 25℃. The phylogenetic tree obtained from the internal transcribed spacer (ITS) rDNA sequences showed that the isolated green mold corresponded to Penicillium brevicompactum (100.0%). This is the first report of green mold contamination caused by P. brevicompactum on H. marmoreus.

  • Master packaging system is a technology combining primary and secondary packaging to preserve the fresh produce in the supply chain. Master packaging system with tailor-designed gas transfer and $CO_2$ absorber of $Ca(OH)_2$ was applied to fresh shiitake mushroom in its supply from farm to retail store. The temperature, humidity and package atmosphere were monitored through the distribution and/or storage until the packages were opened to measure the mushroom quality. Conventional perforated individual packages without secondary master pack were prepared and travelled the same path for comparison purpose. While high temperature history was observed in some initial period of actual practice of the mushroom transportation and storage unexpectedly, the package atmosphere around the produce in the master packaging system was maintained at modified atmosphere consisting of $O_2$ concentration of 0.4 to 4.2% and $CO_2$ concentration of 0.7 to 1.7%, which is known to be beneficial for the mushroom preservation. While curing the mushrooms with precooled drying was effective for quality preservation, positive effect of master packaging system could be apparent for the uncured mushroom. Harmonized combination of curing treatment, master packaging system and temperature management was suggested for the best quality preservation of the fresh shiitake mushroom.

  • Recently, Pleurotus eryngii, King oyster mushroom is the main export item in South Korea mushroom industry. For appropriate transportation to Europe and USA markets, it is necessary to elucidate suitable conditions of storage and treatment of post-harvest during long time shipping. Whole mushrooms were packed with polypropylene anti-fog film (30 um) without trimming. The range of package weight and oxygen permeability of film were 200 g~400 g and 2000~5000 $cc/m^2$, 24 h,atm, respectively. The weight loss ratio was increased with increasing storage time. There were no big differences in weight loss ratio according to package weight and oxygen permeability of film at low temperature storage($4^{\circ}C$). In case of package weight is 400 g and oxygen permeability of film is 3000 $cc/m^2$, 24 h, atm. As a results of freshness, the optimum packing weight and oxygen permeability of film of P. eryngii were 400 g and below 2,000 $cc/m^2$, 24 h, atm, respectively.

  • Recently, the incidence of oak wilt disease has been increasing in Korea, resulting in an increasing number of dead trees. In this study, we performed sawdust bag cultivation of shiitake (Lentinula edodes) using oak wilt-diseased logs and measured the antioxidant activities of the resulting mushrooms. For sawdust media, 4 types of logs (healthy, infected, dead, and fumigated ones) were used. As inocula, three strains of L. edodes (NIFoS 764, Sanjo 701ho, and Sanmaru 2ho) were used. The productivity of L. edodes from dead logs was similar to that from healthy logs. Except for the Sanmaru 2ho strain, fumigated logs and infected logs yielded a lower productivity of L. edodes compared to healthy logs. In terms of antioxidant activity, L. edodes grown on sawdust from fumigated logs showed higher values than those from other logs.

  • The oyster mushroom is a wide cultivar among cultivated edible mushrooms in Korea. But, due to the excess of domestic production, the price has been falling. This study has been conducted to develope new variety oyster mushroom(Pleurotus ostreatus) which have a long term storage to export in foreign market as well as domestic. 'Gonji-7ho', a new variety of oyster mushroom, for the bottle culture, was bred by mating with monokaryons isolated from 'Nongmin-59ho' and 'MT07156'. In the characteristics of fruit body, pilei were round type and gray and stipes were white color and soft. The fruit body growth was vital and uniform. When fruit-body was stored at 4 degrees after packing with plastic vinyl, storage period was extended 7 days longer than 28 day of chunchu-2ho. The yield was 166 g per a bottle(¢65, 900 ml).

  • The aim of this study was to evaluate the clinical efficacy and safety of fermented medicinal herbs. A search of the China National Knowledge Infrastructure (CNKI), PubMed databases and Korean Journal of Oriental Medicine in 2000-2011 located 11 randomized controlled trials (RCTs) that investigated the clinical efficacy of fermented medicinal herbs. Domestic RCTs reported clinical efficacy on improvement of immune responses and clinical safety on usage of fermented medicinal herbs in subjects suffering from cerebral hemodynamics. Countries other than Chinareported studies on the cause of esophageal cancer and on local inflammatory reactions. In China, studies were reported on the effectiveness of fermented medicinal herbs on scapulohumeral periarthritis of the stasis type, chronic superficial gastritis, dysuria induced by benign prostatic hyperplasia of deficiency of kidney yang, diabetic nephropathy, essential hypertension, and benign prostate hyperplasia. These results indicate that fermented medicinal herbs have obvious clinical effects in some diseases and no adverse reactions. Therefore, we need to initiate more fermentation research with useful bacteria, fungi, and mushrooms to produce fermented medicinal herbs. Both governments and research authorities should focus on research involving fermentation of medicinal herbs.

  • The windrow turner, widely used for outdoor fermentation of mushrooms in Europe, has been improved by using rice straw instead of wheat straw in accordance with Korea's actual situation. It was compared with conventional excavator work, and the results were as follows. Agitation performance was $81m^3/hr$, which was 2.8 times higher than $28.6m^3/hr$ of excavator. As a result of the temperature distribution in the pile at the end of the fermentation stage, it was found that the temperature at the lower part of the pile was $5^{\circ}C$ higher than that of conventional pile. This was more favorable for aerobic fermentation. Meanwhile, ash ratio of prototype ($30.9{\pm}1.1%$) was higher than that of control $28.4{\pm}1.6%$. In the case of prototype turner agitation, the yield of mushroom cultivation was $880kg/66m^2$, that was 22.9% higher than the conventional control yield of $716kg/66m^2$.

  • Wild Lentinula edodes (shiitake) strains are an important breeding material for developing new varieties suitable for various consumer preferences and environmental conditions. The morphological characteristics of 100 wild shiitake strains collected from 14 regions were investigated in a sawdust block cultivation. The characteristics of their fruiting bodies were diverse. Among them, 38% of the caps was convex, 33% was broadly umbonate, 27% was applanate, and 2% was shallowly depressed. In terms of their stipes, 55, 23, and 22% were the column, clavate, and funnel type, respectively. The shape of the cap was the most diverse on Mt. Hwaak, whereas the shape of the stipe showed various phenotypes from various regions. Even among strains collected from the same forest, different types of mushrooms were observed. In conclusion, wild shiitake strains that have adapted to various environmental stress can be used to develop new varieties. Thus, securing various genetic resources and studying their traits are important for researching breeding technology.

  • In this study, the "Insect Smart Farm Air Conditioning System" is designed and proposed for the control of breeding environment of Protaetia brevitarsis seulensis larvae. The proposed "Insect Smart Farm Air Conditioning System" separates the breeding room from the air conditioning room. It is a system that creates an environment optimized for breeding and distributes it into a breeding room. When controlling the environment through air-conditioning and humidifiers in insect farms, temperature and humidity vary from part of the breeding room to part. The solution to the problem can be suggested as a solution to the difficulty of producing white-spotted flower mounds of uniform size and weight when selling edible insects. By using the "Insect Smart Farm Air Conditioning System," the temperature difference can be reduced by 6℃ and the humidity difference by 24.7% compared to the environmental control of existing insect farms. The temperature and humidity of different parts of the breeding room were improved. Provide the optimal environment of Protaetia brevitarsis seulensis larvae at all times and ensure uniform CO2 concentration. It can be expected to increase output through annual production and increase income for insect farmers. The proposed "Insecting Smart Farm Air Conditioning System" also controls the set temperature, humidity and CO2. Environmental control of the breeding of other edible insects and the reproduction of mushrooms that require environmental control in breeding or breeding will also be possible.
