An auxin-producing bacterial strain, designated 4-3, was isolated from waste button mushroom compost in Boryeong-si, Chungnam. The strain 4-3 was classified as a novel strain of Leucobacter tardus, based on chemotaxonomic and phylogenetic analyses. TLC and HPLC the isolated L. tardus strain 4-3 produced indole-3-acetic acid (IAA), the auxin. Maximum IAA productionof $94.3mg\;L^{-1}$ was detected for bacteria cultured in R2A medium with 0.1% l-tryptophan, incubated for 24 h at $35^{\circ}C$. Negative correlationwas observed between IAA production and pH of the culture medium, indicating that the increase inIAA caused acidification ofthe medium. The effect of supplementation with varying concentrations of l-tryptophan, a known precursor of IAA, was also assessed. production was maximal at 0.1% l, but decreased at lconcentrations above 0.2%. To investigate the plant growth-promoting effects of the bacterium, L. tardus strain 4-3 culture broth was used to inoculate water cultures and seed pots of mung bean. We found thatadventitious root induction and root growth were 2.2-times higher in thethan in the non-inoculated plants.
We investigated the effect of nutritional and environmental factors on Ophiocordyceps longissima mycelial growth. The longest colony diameter was observed on Schizophyllum (mushroom) genetics complete medium plus yeast extract, Schizophyllum (mushroom) genetics minimal medium, and Sabouraud dextrose agar (SDA); however, malt-extract yeast-extract agar, SDA plus yeast extract, yeast-extract malt-extract peptone dextrose agar, SDA, oatmeal agar, and potato dextrose agar showed higher mycelia density. A temperature of $25^{\circ}C$ was optimum and 7.0 was the optimum pH for mycelial growth. Colony diameter was similar under light and dark conditions. Maltose and yeast extract showed the highest mycelial growth among carbon and nitrogen sources respectively. The effect of mineral salts was less obvious; however, $K_3PO_4$ showed slightly better growth than that of the other mineral salts tested. Among all nutrition sources tested, complex organic nitrogen sources such as yeast extract, peptone, and tryptone were best for mycelial growth of O. longissima. Ophiocordyceps longissima composite medium, formulated by adding maltose (2% w/v), yeast extract (1% w/v), and $K_3PO_4$ (0.05% w/v) resulted in slightly longer colony diameter. In vitro mycelial O. longissima growth was sustainable and the production of fruiting bodies could be used for commercial purposes in the future.

Pholiota nameko is one of the four major profitable mushrooms along with oak mushroom, winter mushroom, and oyster mushroom. It contains abundant proteins, carbohydrates, organic acids and vitamins. Its unique taste and flavor as well as its nutritional features make it widely favoured. Mushroom complete medium was the optimal medium for mycelial growth of Pholiota nameko. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and 5.0, respectively. The best carbon sources for mycelial growth were glucose and mannose, and the best nitrogen sources were yeast extract, peptone, asparagine, etc. The 8:2 ratio mix of oak sawdust and wheat bran was the best for the bottle cultivation. The best mushroom was yielded after 30 days incubation. The best yield was produced with 850g of medium weight in a PP bag and bottle.
Metacordyceps yongmunensis is a newly reported species from Korea, which is very similar to Cordyceps species in morphological characters. It grows on large lepidopteran pupa, and numerous white stromata grow on a single host. Mycelial growth characteristics of M. yongmunensis isolates were studied in different media and at different temperatures. Also, different carbon sources, nitrogen sources, and mineral salts were tested for mycelial growth of M. yongmunensis. Schizophyllum (mushroom) genetics complete medium plus yeast extract, Schizophyllum (mushroom) genetics minimal medium, and Martin's peptone dextrose agar produced longer colony diameters and more compact mycelial density than other media. The optimum temperature for mycelial growth was $25^{\circ}C$. Carbon sources such as sucrose, soluble starch, dextrose, glucose, dextrin, maltose, and fructose showed better mycelial growth, whereas peptone, yeast extract and tryptone resulted in the best mycelial growth of all of the nitrogen sources tested. All of the mineral salts tested showed similar growth as the control, except $K_2HPO_4$ which showed longer colony diameter and more compact mycelial density. The compact colonies were white and cottony with a greenish margin. The results showed that M. yongmunensis is an easy fungus to growas it grew from 30 to more than 50 mm in 2 wk.

Phenylalanine ammonia-lyase (PAL) gene is known to be expressed in plants, and is involved in the differentiation, growth and synthesis of secondary metabolites. However, its expression in fungi remains to be explored. To understand its expression in mushroom fungi, the PAL gene of the edible mushroom Flammulina velutipes (Fvpal) was cloned and characterized. The cloned Fvpal consists of 2,175 bp, coding for a polypeptide containing 724 amino acids and having 11 introns. The translated amino acid sequence of Fvpal shares a high identity (66%) with that of ectomycorrhizal fungus Tricholoma matsutake. Distinctively, the Fvpal expression in the mycelium was higher in minimal medium supplemented with L-tyrosine than with other aromatic amino acids. During cultivation of the mushroom on sawdust medium, Fvpal expression in the fruit body correspondingly increased as the mushroom grew. In the fruiting body, Fvpal was expressed more in the stipe than in the pileus. These results suggest that F. velutipes PAL activity differs in the different organs of the mushroom. Overall, this is first report to show that the PAL gene expression is associated with mushroom growth in fungi.
The medium used for bottle cultivation of mushrooms is made according to a specific workflow; this includes mixing the raw materials, adjusting the moisture content, placing the mixture into bottles, and sterilizing and cooling the medium. Medium composition should be kept constant throughout the year. placed in the bottle, choice of materials, moisture content, weight of the medium, bulk density, nitrogen content, three-phase medium, such as suitable conditions. To achieve constant medium composition a comprehensive model has been established in accordance with the technology developed in the work process with respect to medium compositions piecemeal. By comprehensively applying the various methods to establish medium composition, our country can expect to grow at over four seasons and overcome the many difficulties in mushroom mycelial growth periods, cultivation periods, quantity, and quality.
The nutritional value and yield of mushrooms depend on the substrate on which it is grown. This study sought to biofortify Pleurotus floridanus with calcium supplements and assess its effect on the yield and calcium levels. The experiment was set up in a 2 × 5 factorial and replicated thrice in a completely randomized design. Two calcium supplements, OML and OMW, were added to two growth media. The examination of total dry weight yield showed that calcium supplements OML and OMW in the sawdust medium containing wheatbran in the ratio 1:10 had a mean value of 4.37 g, which was significantly higher (P < 0.05) than that in the control (1.29 g). However, in the sawdust-only medium, there was no significant difference (p > 0.05) in the application of treatments. No significant difference (p > 0.05) was observed between the calcium types in both growth media. The mineral analysis showed that calcium levels were increased in harvested mushrooms with the addition of calcium OML and OMW to the growth media.
This study was carried out to obtain the basic data for artificial culture of veiled lady mushroom (Dictyophora spp). The optimal conditions for the mycelial growth were $25^{\circ}C$ and pH 5.0 for all isolates except the optimal temperature of $30^{\circ}C$ for D. echinovolvata ASI 32002 and Phallus rugulosus. The optimal medium for Dictyophora spp. was PBA (potato bamboo sawdust extract agar) medium. The strain ASI 32002, D. echinovolvata, grew faster than. D. indusiata ASI 32003 and Phallus rugulosus ASI 25007 on the medium. Carbon sources such as glucose, maltose and inuline were favorable for stimulating a mycelial growth of the two strains of ASI 32002 and ASI 32003. Asparagine and glutamine appeared to be favorable to the strain ASI 32002 and ASI 32003, where as alanine, one of nitrogen source also favorable to the strain ASI 32002. The optimum C/N ratio of the two isolates of ASI 32002 and ASI 32003 was about 25 : 1 when 2% glucose as carbon source was mixed with the basal medium. While, in the case of 4% as carbon source, the optimum C/N ratio was about 30 : 1.

The fruiting body of honey mushroom, Armillaria gallica, was collected from Gastrodia elata cultivated fields. Pure cultures were isolated from fruiting body tissue of the mushrooms, and cultured on MCM (mushroom complete medium) or PDA (potato dextrose agar) medium. Then, 12 different types of mycelial growth characteristics such as growth rate, colony morphology and rhizomorph formation were obtained. The vitality of the mycelial growth and rhizomorph formation of the fruiting body culture isolates were better on MCM than PDA, suggesting that the optimal culture medium for A. gallica mycelia was MCM. To observe the feature of colony morphology, the subculture of isolates were incubated on MCM. Consequently, we could find the segregated or differentiated colony morphology from isolate type 11 that was similar morphology to isolate type 12. For phylogenetic analysis of the 12 isolates, RAPD (Random Amplified Polymorphic DNA) were performed. The isolate type 12 was not only shown different band patterns of RAPD variation in other 11 isolates, but also commercial strain known as Chunmagyun No. 1. Among the tissue culture isolates of fruiting, strains with better mycelial growth characteristics than Chunmagyun No. 1 were selected. We expect that the new strain can be substituted to commercial strain Chunmagyun No. 1.
Cauliflower mushroom ($Sparassis$$crispa$) is being cultivated as a functional mushroom since the mushroom contains larger amount of ${\beta}$-glucan than other edible or medicinal mushrooms. In this study, as a fundamental study for the cultivation process of cauliflower mushroom, we assayed the mycelial growth and the productivity of the mushroom cultivated on the sawdust-based medium made of larch ($Larix$$kaempferi$) by 4 kinds of medium densities and 6 kinds of particle sizes. Low densities of sawdust-based media (0.68~0.72 g/$cm^3$) showed high variations in mycelial growth at early stage on sawdust-based medium. The coefficient of variance for the mycelial growth decreased from higher than 40% at the 3rd week to lower than 10% at the 7th week. High density of sawdust-based medium (0.80 g/$cm^3$) showed relatively lower variation in the mycelial growth at early stage with less than 30% of coefficient of variance, the high density of sawdust-based medium was thought to be quite stable compared to the lower densities of sawdust-based medium. From the viewpoint of mushroom productivity especially for the goods (excluding bottom of fruiting body), 0.76 g/$cm^3$ was better than any other densities; the return rates of fruiting body from each medium were 12.2, 13.6, 13.1, and 12.0% for 0.80, 0.76, 0.72, and 0.68 g/$cm^3$, respectively. By the way, it took about 40 days for harvesting from primordium formation with the sawdust-based medium excluded the particles less than 1 mm, while it took about 70 days with the medium including the particles less than 1 mm. The yield from the sawdust-based medium with the particles less than 1 mm was also quite less than any other sawdust-based medium especially for the goods. Therefore, we recommend that the sawdust-based medium with larch for the cultivation of cauliflower mushroom be prepared as 0.76 g/$cm^3$ in medium density with excluding the particles less than 1 mm.
