We monitored the occurrence of fungal species on bed-logs inoculated with oak mushroom (Lentinula edodes) and the environmental conditions of temperature and humidity in the cultivation houses during monsoon. Six fungal species, viz., Cladosporium sp., C. cladosporioides, C. anthropophilum, Pleosporales sp., Trichoderma harzianum, and Acremonium sp., wereidentified from the cultivation houses located in Jangheung, Jeonnam province. This identification was confirmed by performing nucleotide sequence analysis of the internal transcribed spacer and 28S rDNA regions. Our study presents significant findings that can help in preventing fungal damage induced by inappropriate temperature and humidity in oak mushroom cultivation houses.

The occurrences of the major diseases and the densities of air-born microbes were surveyed in the cultivation facilities for oyster mushroom (Pleurotus ostreatus), king oyster mushroom (Pleurotus eryngii), and enoki mushroom (Flammulina velutipes) in different areas of Korea. Green mold disease was most often developed in oyster mushroom bed cultivation with the disease incidence rate of approximate 10% while the disease incidences from bottle and plastic envelop cultivation were less than 1~2%. In the bed cultivation, the major air-born microbes in the growth room were Aspergillus, Penicillium, Trichoderma, and Curvularia with the total fungal population density of 567~1,297 CFU/$m^3$ . However, only Trichoderma and Penicillium were detected in the growth rooms and innoculation rooms of bottle and plastic envelop cultivation with the densities of 350~700 CFU/$m^3$ and 160~260 CFU/$m^3$, respectively. The bacterial diseases become evident in the growth rooms of bottle and plastic envelop cultivation with the approximate incidence rate of 10%. The identified bacterial species were Brevibacillus levelkil, Rhizobium radiobacter, Brevundimonas vesicularis, Pseudomonas mosselii, Microbacterium testaceum. Sphingomonas panmi, Sphingomonas yabuuchiae, Paracocus dinitrificans, Curtobacterium flaccumfaciens pv. flaccumfaciens and some unidentified bacteria with the densities of 40~6,359 CFU/$m^3$ in the growth rooms and 9 CFU/$m^3$ in the inoculation room. This study indicated that the green mold disease by fungal strains was the major mushroom disease in the bed cultivation and suggested that the contamination of bacteria and fungi together in the growth media could result in severe production loss. The plastic envelope and bottle cultivation were evidenced to be less susceptible to such contaminations.
Sodium hypochlorite alkaline was tested against Pseudomonas tolaasii causing bacterial blotch on cultivated oyster mushroom (Pleurotus ostreatus). The minimum inhibitory concentration of sodium hypochlorite against P. tolaasii contained active chlorine (AC) at 1.4 mg/l on plate assay. The highest cultivation yield was obtained from the treatment of AC 5.7 mg/l. Treatment of sodium hypochlorite at the rate of higher than AC 11.4 mg/l resulted in reduced yields at the harvest. However, the population of total bacteria on the bed surface treated with AC 5.7 mg/l of sodium hypochlorite was maintained to some extent. Inhibitory concentration against total bacteria on the bed surface was over AC 22.8 mg/l. Mushroom mycelium was damaged and its growth strongly inhibited at the concentration of AC 200 mg/l. Mushroom caps showed yellowish symptom by chemical injury by treatments of AC 74.1 mg/l or higher. Sporocarps infected by P. tolaasii were irrevocable at any concentration of sodium hypochlorite. Routine watering with AC 5.7 mg/l from mushroom initiation to the end of picking resulted in reduced bacterial blotch incidence of 40% and 86% at two mushroom farms. The treatment resulted in higher quality mushroom production compared to that conventionally watered with tap water alone.
The process of cultivation and production of oak mushroom (Lentinula edodes (Berk.) Pegler) on sawdust surface beds were investigated. Sawdust surface bed cultivation is the method by which oak mushrooms are cultured and produced on sterilized sawdust surface bed without using bags. The bed was made by inoculating with 3 to 1 ratio of bed sawdust to oak mushroom mycelial inoculum. The sawdust bed medium with 65% water content was pasteurized at $65^{\circ}C$, inoculated with sawdust spawn and spread on the surface on vinyl film in cultivation shed. During 78 days of cultivation period, water content in the medium varied from 61 to 72%, its pH decreased from 5.6 to 3.9~4.6 and ergosterol concentration increased to $0.33{\sim}0.59{\mu}g/g$. $CO_2$ concentration in the medium rapidly increased to 8.06% in two weeks. In seven weeks the medium surface started browning and $CO_2$ concentration increased to about 5.63%. Until 11th week the $CO_2$ concentration was maintained at 6~7%. After removing the plastic cover on the bed for ventilation in 12 weeks, $CO_2$ within the bed reduced dramatically to 1.5%. In the cultivation shed the internal temperature was $7.1{\sim}29^{\circ}C$ and humidity was 27.3 to 100%, while bed temperature ranged $11.6{\sim}30^{\circ}C$. Oak mushroom fruiting started from late July, in 120 days after bed establishment in late March and continued for approximately 100 days until early December with eight cycles of irrigation treatment. The mushroom yield of the eight cycles were 288~352 kg during the 1st (7/29~8/4) to 3rd cycle (9/3~9/7), 800 kg at the 4th cycle (9/19~9/24), 1,296~1,853 kg during 5th (10/3~10/8) to 7th cycle (4.11~11/9) and 990 kg at 8th cycle (11/23~12/7). Total production was approximately 7.4 tons from 33.0 tons of oak sawdust medium, thus harvest efficiency of the mushroom production was approximately 22.4%.
${\gamma}$-Aminobutyric acid(GABA) is a four carbon non-protein amino acid that has several well-known physiological functions, such as a postsynaptic inhibitory neurotransmitter in the brain and induction of hypotensive and tranquilizer effects. A lactic acid bacterium was isolated from button mushroom bed, which is showing high GABA productivity by TLC or HPLC analysis. The strain was identified as Lactobacillus hilgardii by analysis of 16S rDNA gene sequence. When the maximum production of GABA by L. hilgardii was investigated with various concentration of monosodium glutamate, the yield of GABA reached to be 53.65 mM at 1% mono sodium glutamate (MSG) in flask cultivation. A Glutamate decarboxylase (GAD) enzyme, which was known to convert MSG to GABA, was purified from a cell-free extract of L. hilgardii and the molecular weights of purified GAD was estimated to 60,000 by SDS-PAGE. The optimum pH and temperature of GAD were at pH4.6 and at $37^{\circ}C$, respectively. The GAD activity was increased by the addition of sulfate ions such as ammonium sulfate, sodium sulfate and magnesium sulfate, indicating that the increase of hydrophobic interaction causes the increase of GAD activity.
Purpose: Recently, domestic and abroad consumption of mushroom has been increasing. Especially, oyster mushroom has been the most consumed product, sharing one third of the mushroom market. The air temperature differences between relative positions of the mushroom farms were needs to be minimal. However, in reality, the air temperature differences ranged from 2 to $5^{\circ}C$. Because of this, the mushrooms are non-uniform growth as well as decrease in both quality and quantity. Although air circulators have been employed by oyster mushroom farms to minimize air temperature differences, no experiments have been performed to illustrate the effect of the air circulators. Methods: This experiment is designed to analyze the effect of the air circulation by constructing a prototype air circulator and measuring the air temperature when the circulator was position at different heights (50 cm, 150 cm, 200 cm) from the floor in the center. Result: The horizontal plane air temperature of the first growing bed when the air circulator was installed 50cm above the floor in the center, once not using the air circulators and the other time using the air circulators, yielded the air temperature differences of $8.6^{\circ}C$ and $1.8^{\circ}C$ and deviations of 2.82 and 0.60, respectively. The third growing bed's air temperature differences were $10.0^{\circ}C$, $1.6^{\circ}C$ and deviations 3.28, 0.64, each respectively. These outcomes proved that the use of air circulators minimized the air temperature difference and deviation. The use of air circulators helped minimize the air temperature differences and the derivations in oyster mushroom farm. Conclusion: The use of air circulators helped balance the air temperature distribution in oyster mushroom farm.
This study was conducted to select organic materials (OM) and nitrogen sources in composting of waste mushroom bed from Agaricus bisporus. We examined physio-chemical properties of the organic materials and the mixture ratio for preparing the wasted mushroom bed (M) compost. The carbon content of sawdust was higher than those of rice straw (R) as OM source and the nitrogen content was high in the order of fowl manure (F)>> pig manure (P)> cow manure (C). The compost was prepared to maintain the criteria of above 25% organic matter and then the change of their ingredients was estimated during the process of fermentation. The temperature of waste mushroom bed+pig manure+rice straw (MRP) treatment was varied fast throughout fermentation, on the other hand the temperature of waste mushroom bed+pig manure+sawdust (MSP) treatment was steadily elevated to the middle of composting. The pH of the compost was somewhat high to pH 8.5~9.0 at the early stage, but decreased to 7.5 at the end stage of composting. The content of OM after fermentation was decreased to the level of 19~21% in rice straw, but the sawdust treatment maintained 25~27% organic matter. The waste mushroom bed+fowl manure+rice straw (MRF) treatment, which contains 26.2% organic matter and 0.68% nitrogen, was the highest among them. The volume of compost was reduced to 50% by using rice straw as organic matter, but reduced to 30% by using the sawdust. The contents of heavy metal in the compost were suitable within the legal criteria. The number of microorganisms were higher in the rice straw than those in the sawdust. It was high in the order of fowl manure> pig manure> cow manure. The major groups consisted of aerobic bacteria, gram negative bacteria and Bacillus sp. and their populations after fermentation were increased to $1{\times}10^1{\sim}1{\times}10^2\;cfu\;g^{-1}$ rather than those before fermentation. Therefore we concluded that the waste mushroom bed+fowl manure+sawdust (MSF 3:9:1 v/v/v) treatment was suitable combination for high organic matter and nitrogen source, and the periods of composting were 50~60 days.
Effects of substrate bed interior environments on mushroom qualities were investigated in oyster mushroom cultivation facilities in which either Reversible Air-Circulation Fans (RACF) blowing air in two directions (upwards and downwards) or customary Convection Fans (CF) with air blowing only upwards were operated throughout the cultivation period. Two days before harvest, the deviation ranges of the bed interior temperature and relative humidity in the facility using RACF were in the ranges of 1.0-1.3℃ and 7.8-9.0% in the first growing cycle, and within 0.7-1.1℃ and 10.0-11.4% in the second cycle. In the facility using CF, the ranges of variation in the indoor environment parameters (5.8-6.4℃ and 21.3-23.1% in the first growing cycle, and 3.4-5.7℃ and 14.6-18.3% in the second growing cycle) were much enlarged compared to those associated with RACF. These results strongly indicate that RACF significantly enhances air uniformity. Some mushroom qualities differed between growing cycles. For instance RACF in the first cycle gave somewhat better qualities than CF, but some qualities, like pileus diameter and stipe length, were slightly lower than those described for CF in the second cycle when the cultivation substrate weakened. The observation that some qualities worsened under RACF conditions, despite better air uniformity during the growing cycle, revealed the possibility that downward wind may exert a non-negligible negative effect on mushroom growth. Therefore in the future, making wind measurements on the interior and exterior of substrate beds is necessary to obtain insights into their influences on mushroom qualities. The RACF operation manual needs to be edited to convey this necessity.
An auxin-producing bacterium, Klebsiella michiganensis Jopap-1, was isolated from a button mushroom bed in Buyeo-Gun, Chungcheongnam-Do. The strain Jopap-1 was classified as a novel strain of K. michiganensis based on a chemotaxonomic and phylogenetic analysis. The isolated K. michiganensis Jopap-1 was confirmed to produce indole-3-acetic acid (IAA), which is one of auxin hormones by TLC and HPLC analyses. The maximum concentration of IAA ($96.05mg\;L^{-1}$) was detected in the culture broth incubated in R2A medium containing 0.1% L-tryptophan for 48 h at $35^{\circ}C$ by HPLC quantity analysis. A negative relationship between IAA production and pH variation was estimated to show that the increase of IAA caused acidic pH in the culture. The effect of the supplement on L-tryptophan (precursor of IAA) production was observed to be highest at 0.1% concentration, but was significantly lowered above a concentration of 0.2%. To investigate the growth-promoting effects on the crops, the culture broth of E. michiganensis Jopap-1 was infected to water cultures and seed pots of mung bean and lettuce. Consequently, the adventitious root induction and root growth of mung bean and lettuce were observed to be 2.1 and 1.8 times higher than those of the control.
Anamorph and teleomorph of Diatrype stigma on the bed-log inoculated with oak-mushroom (Lentinula edodes) in Korea were observed and identified. As the characters of anamorph of Diatrype stigma, conidia were hyaline, falcate or slightly curve, and $1{\sim}1.5{\times}12.5{\sim}17.5\;{\mu}m$, and conidia layer was confirmed under the bark. About teleomorph, perithecia $150{\sim}200{\times}450{\sim}500\;{\mu}m$, with ostiole ($30{\sim}40\;{\mu}m$, three- to four-sulcate). Size of ascus containing 8 ascospores was $5{\sim}6{\times}30{\sim}40\;{\mu}m$. Ascospores were hyaline to pale brown, $1.5{\sim}2{\times}5{\sim}7.5\;{\mu}m$ and mostly biguttulate.
