강원도에서 채취한 야생 뽕잎의 생육 시기별 resveratrol 함량 (Seasonal Resveratrol Contents of Wild-type Mulberry Leaves Collected from Gangwon Province in Korea)

  • 야생 뽕나무의 자원화 및 이용기술을 개발하기 위해 강원도 원주, 인제, 양양 3개 지역에서 생육 단계별로 채취한 야생 뽕나무의 뽕잎을 대상으로 레스베라트롤 함량을 분석함으로써 뽕 품종의 육종 효율을 높이는 동시에 기능성 및 이용 효율이 기대되는 뽕 유전자원을 선발하고자 하였다. 야생 뽕잎의 레스베라트롤 함량은 오디에 비해 평균 함량이 상당히 낮았으나 장려뽕에 비해서는 높은 경향을 나타냈다. 생육시기별 야생 뽕잎의 평균 레스베라트롤 함량은 추기의 뽕잎이 춘기보다 함량이 높았다. 즉 춘기 5개엽기 어린잎의 평균 레스베라트롤 함량은 $344{\mu}g$/100 g이었으며 뽕잎의 생육시기가 경과함에 따라 $356{\mu}g$/100 g DW, $273{\mu}g$/100 g DW, $253{\mu}g$/100 g DW 이었다. 추기 뽕잎의 레스베라트롤 함량은 시기가 경과함에 따라 $438{\mu}g$/100 g DW, $476{\mu}g$/100 g DW, $598{\mu}g$/100 g DW로 증가하였다. 따라서 레스베라트롤의 원료로서 뽕잎을 이용하고자 하는 경우 봄뽕보다는 가을뽕을 이용하는 것이 바람직하며, 수세 후 바로 실온에 음건하여 사용하는 방법이 레스베라트롤의 함량이 높을 것으로 판단되었다. 원주 지역의 No. 3 개체의 야생 뽕잎은 생육시기 모두 평균 함량보다 높았으며 전체 평균 $539{\mu}g$/100 g DW으로서 10개체의 뽕나무 중 가장 높은 함량을 나타냈다. 다음으로 인제 지역의 No. 7 개체의 뽕잎은 전체 평균 $491{\mu}g$/100 g DW으로 높았다. 따라서 두 개체의 뽕나무는 레스베라트롤 함량이 높은 육종소재 및 이용 가능성을 검토할 만한 자원으로 선발하였다.

YK-209 뽕잎이 Streptozotocin 유발 당뇨쥐 간조직의 항산화계에 미치는 영향 (Effects of YK-209 Mulberry Leaves on Antioxidative Defense System of Liver in Streptozotocin-Induced Diabetic Rats)

  • YK 209 뽕잎이 STZ 유발 당뇨쥐 간조직의 항산화계에 미치는 영향을 관찰코져 하였다. Sprague Dawley 흰쥐 수컷을 정상군(normal group)과 STZ 유발 당뇨군으로 나누고 당뇨군은 다시 식이내 YK-209뽕잎 함량에 따라 각각 YK-209뽕잎을 공급하지 않은 당뇨대조군(DM group),0.1% 뽕잎 공급군(DM-0.IY group), 0.2% 뽕잎 공급군(DM-0.2Y group) 및 0.4% 뽕잎 공급군 (DM-0.4Y group)으로 나누어 자유섭식 시켰다. 3주후 STZ으로 당뇨를 유발한 후 9일째에 희생하였다. 단위체중당 간무게는 정상군에 비해 당뇨군 모두 유의적으로 증가되었으나 뽕잎 비공급 당뇨군(DM group)에 비해 뽕잎공급군들은 감소되었다. 간조직중의 SOD 활성은 정상군에 비해 당뇨유발군에서 유의적으로 감소하였으며 뽕잎공급으로 인한 차이는 없었다. GSHpx 활성은 정상군에 비해 DM군은 7.3%저하되었으나, DM-0.1Y, DM-0.2Y군은 정상군 수준이었다. 간조직 중의 지질과산화가는 정상군에 비해 DM군은 144%증가되었으나 DM-0.1Y, DM-0.2Y군은 정상군 수준이었다. 리포푸신 함량은 정상군에 비해 DM군에서 100%증가되었으나 YK-209 뽕잎 공급군은 DM군에 비해 유의적으로 감소되었다. 간조직 중의 superoxide radical(O$_2$ ) 함량은 정상군에 비해 DM군에서는 81% 증가되었으나 DM-0.1Y군과 DM-0.4Y군은 정상군과 유의적인 차이가 없었다. 이상의 결과에서 YK-209 뽕잎은 항산화계를 강화시켜 조직의 산화적 손상을 감소시키므로서 산화적 스트레스 억제 효과가 있었다.

뽕잎분말이 고콜레스테롤 식이 투여 흰쥐의 지질대사에 미친 영향 (Effects of Mulberry Leaves Powder on Lipid Metabolism in High Cholesterol-Fed Rats)

  • 본 연구에서는 뽕잎분말이 고콜레스테롤 섭취시 생체 내에서의(in vivo) 지질대사 개선에 미치는 효과를 알아보고자 하였다. 흰쥐에 콜레스테롤 1%인 사료를 급여하여 고지혈증을 유발시키면서 뽕잎분말(0, 5, 10%)을 해당사료에 첨가하여 4주간 섭취시켜 혈중 지질성분을 분석하여 비교하였다. 식이섭취량과 시작체중은 군간에 유의차가 없었는데, 체중증가량과 사료효율은 10% 뽕잎첨가군(HC10M)에서 가장 낮았다. 고콜레스테롤 투여군(HC)의 혈청 중성지방, 총 콜레스테롤, LDL-cholesterol 농도는 정상대조군(C)을 포함한 다른 실험군들(HC5M, HC10M)에 비해 유의적으로 높았다. 뽕잎분말이 급여된 실험군들(HC5M, HC10M)의 이들 지질 함량은 정상군보다 높았지만 HC군에 비해서는 유의적으로 낮은 수준을 보였다. HDL-cholesterol 함량은 반대의 상반된 결과를 보였다. 심혈관계인자는 고콜레스테롤을 급여한 모든 군(HC, HC5M, HC10M)은 정상대조군(C)과 비교할 때 유의적으로 높아 심혈관계질환의 위험성이 높은 것으로 나타났으며, 실험군들(HC5M, HC10M)이 고콜레스테롤 급여 대조군(HC)보다 유의하게 낮았는데, 10% 첨가된 실험군 (HC10M)이 5% 실험군(HC5M)보다 더 효과적이었다. 혈청 AST 활성은 정상대조군에 비해 실험군(HC)에서 유의적으로 높았으며, 뽕잎분말이 투여된 실험군들(HC5M, HC10M)의 경우 정상 대조군(C)에 비해 높았지만 10% 투여군(HC10M)은 정상대조군 수준으로 낮게 나타났다. 혈청 ALT 활성은 정상대조군과 실험 군들간에 유의차가 없었다. 혈청 creatinine 수준은 대조군과 실험군사이에 유의적인 차이가 없었으며 모두 정상수준을 보였다. 고콜레스테롤을 투여한 실험군들(HC, HC5M, HC10M)의 간의 총 지질, 중성지방 및 총 콜레스테롤함량은 정상대조군(C)에 비해 각각 43, 27, 55%씩 증가하였다. 이러한 지방축적 현상은 뽕잎분말의 첨가에 의해 유의적으로 감소하였으며, 그 효과는 농도에 의존적이었다, 고콜레스테롤 투여군(HC)의 분변 중 총 지질과 중성지방 농도는 정상대조군(C)에 비해 다소 높았고, 총 콜레스테롤의 경우는 유의적으로 높게 나타났다. 그러나 뽕잎첨가군(HC5M과 HC10M)의 총 지질, 중성지방 및 콜레스테롤 농도는 콜레스테롤 투여군(HC)에 비해 분변으로 배설량이 증가하는 현상을 보였다. 이상의 결과를 살펴보면 뽕잎 분말은 고콜레스테롤식이를 섭취하는 경우 혈청과 간장의 지질수준은 떨어뜨리고, 분변중 지질배설량을 상승시키는 효과가 현저한 것으로 보인다.

Mathematical Constants for Non-Destructive Rapid Method of Leaf Area Determination in Mulberry (Morus spp.)

  • Mathematical constants for multiplication with leaf length (I) or breadth (b) or l ${\times}$ b have been worked out for determining leaf area in promising mulberry genotypes viz., Chinese White, S-146, Chak Majra and Sujanpur Local of sub-tropical India. When pooled, the mathematical constants worked out were 8.1132, 10.1019 and 0.5992 for multiplication with leaf length, breadth and l ${\times}$ b, respectively, for genotypes bearing un-lobbed leaves and 6.9447, 8.2761 and 0.5009 for multiplication with leaf length, breadth and l ${\times}$ b, respectively for genotypes bearing lobbed leaves. Leaf area can be worked out by using any constant by multiplying either with leaf length or breadth or both (l ${\times}$ b). Estimated leaf areas worked out were found significantly and positively correlated with actual leaf area (r=999$^{**}$). The suggested present non-destructive method by using mathematical constants is very quick and alternative to electronic leaf area meter for spot leaf area determination in mulberry which is the only food source for mulberry silkworm in sericulture industry.

Nutritional Evaluation of Fodder Tree Leaves with Goats

  • Two experiments were conducted to evaluate the nutritional value of common fodder tree leaves with goats fed at 50% of total ration. In experiment 1, leaves from six fodder tree species i.e., Ailanthus aitissima, Elaeagnus angustifolic, Morus alba (Mulberry), Populus spp, Robina pseudoacacia and Salix babylonia were harvested in spring and winter from northern areas of Pakistan. Chemical composition and apparent in situ dry matter digestibility (DMD) of fodder tree leaves were measured. Results showed that crude protein (CP) values were higher (p<0.05) in all the species during spring compared to winter (17.9% vs 12.0%). The concentration of NDF in Elaeagnus and Robinia was higher in spring, whereas no seasonal difference was found in other species. In situ DMD was higher (p<0.05) in Ailanthus and Populus at spring while it was higher (p<0.05) in Elaeagnus, Mulberry and Robinia at winter. There was no (p<0.05) seasonal effect on in situ DMD of Salix. In experiment II, four iso-nitrogenous and iso-caloric rations viz., A, B, C and D were prepared containing 50% (winter harvested) sun dried leaves of Salix, Robinia, Mulberry and Elaeagnus, respectively and 50% concentrate. Dry matter and crude protein intakes were higher (p<0.05) given ration A (Salix) whereas DM and CP digestibility was lowest (p<0.05) given ration B. Nitrogen retention was higher (p<0.05) given ration A. Goats fed on fodder tree leaves and concentrate showed moderate intake and digestibility.

잎채소 분말을 첨가한 초콜릿의 품질특성 및 항산화 활성 (Quality Properties and Antioxidant Activities of Chocolate with Variable Leaf Powder)

  • Purpose: The aim of this study was to investigate the effects of various leaf powders on chocolate by analyzing quality characteristics, antioxidant activities, and sensory characteristics. Methods: Pine needles, mulberry leaves, ramie leaves, barley leaves, and squash leaves were freeze-dried, powdered, and added to the chocolate. Results: Water contents of all leaf powder-added groups were significantly higher than that of the control (15.78%). The pH values of chocolate with leaf powder were lower than that of the control (6.33) while that of mulberry leaf chocolate (6.56) was higher than that of the control. In color measurement, L-value, a-value, and b-value were highest in pine needle chocolate (30.31, 3.52, and 4.78, respectively). The ${\Delta}E$ value (total color difference compared to white board) was the highest in squash leaf chocolate (69.51) and significantly different from that of the control (67.69). The hardness of leaf powder-added chocolate was lower than that of the control (6.84 N), except for pine needles. In the antioxidant activity analysis, chocolate with pine needles and squash leaves had higher polyphenols, flavonoids, and ABTS radical scavenging antioxidant activity than those of the control. Overall consumer acceptability was the most preferred in the control compared to the other groups. However, chocolate with mulberry and barley leaves also received high scores (5.60 and 5.40, respectively) for overall acceptability. Conclusion: It can be concluded that chocolate with barley leaves is desirable for making functional chocolate due to its high consumer preference and positive effects on improving antioxidant activities.

수종 잎식품 건조물의 급이가 고Cholesterol 혈종 흰쥐의 혈청지질에 미치는 영향 (The Effect of Dry Edible Leaves Feeding on Serum Lipids of Hypercholesterolemic Rats)

  • In other to investigate the hypolipidemic effect of dry edible leaves on serum lipids, four kinds of powdered leaves(persimmom, mulberry, perilla, and soy bean) were added to experimental diets at the levels of 5%(w/w). The diets were fed to 30 male Sprague-Dawley rats, aged 4 weeks and weighing 60$\pm$5g, for 3 weeks. Food intake was the largest in Group 1(control), with 5% of cellulose powder, while food efficiency was the highest in Group 2(persimmon). Total cholesterol level in serum was lower in Group 5(soybean) and Group 4(perilla) than the other groups. In comparison with Group 1, HDL-cholesterol level was very high in Group 4 and atherogenic index was low in Groups 4 and 5. Free cholesterol level was the highest in Group 2, and was the lowest in Group 1. Both LDL and LDL-cholesterol levels were the lowest in Group 1, while they were very high in Groups 2 and 3(mulberry). Triglyceride level was the lowest in Group 4, and the highest in Group 1. Except Group 3, all groups had lower phospholipid level than Group 1. Compared with Group 1, calcium level in serum was low in Groups 3, 4 and 5, magenesium level was low in Groups 2, 3 and 5, phosphate level was high in Group 4, and iron level was exceptionally low in Groups 4 and 5. In conclusion, Group 4(perilla) and Group 5 (soybean) showed a favorable effect in atherogenic index and serum levels of various lipids.

Characterization of DNJ production for large-scale fermentation of mulberry leaf

  • Mulberry leaves containing 1-deoxynojirimycin (DNJ) known to be a strong inhibitory effect for ${\alpha}-glucosidase$. Thus, DNJ has been recognized as a potentially important source for prevent or treat hyperglycemia. More effective method for the DNJ high-production is needed because DNJ content of natural mulberry leaf are as low as 0.1%. Many researchers have studied for the DNJ high-production in mulberry leaves such as the harvest season, fermentation using microorganisms, optimal culture conditions, and optimal extraction conditions. In order to provide for useful data that is anticipated at the level of industrial scale, we investigated ${\alpha}-glucosidase$ inhibitory activity, pH value and DNJ content in large-scale based on the optimal culture conditions for mulberry leaf fermentation of small-scale in our previous study. The ${\alpha}-glucosidase$ inhibitory activity, pH value, and DNJ content in this study were measured from the mulberry leaf fermentation broth for 7 days. During mulberry leaf fermentation, the ${\alpha}-glucosidase$ inhibitory activity and DNJ content was increased until 2 to 4 days, but after 4 day was decreased. The pH value showed a decreasing trend up to 2 day, and little changes in 2 to 4 days. However, the pH was started to increase after 4 days.

Development of powder with increased rutin content from mulberry leaves for the application of food materials

  • We studied on improvement method of rutin content using mulberry leaf powder. Mulberry leaves were collected and then hot-air dried and powdered for experiment. As a result, we have developed a pre-treatment method that extracts mulberry leaf powder with water or fermented alcohol with reflux extractor and then increases the rutin content by improving the process. Citric acid (0.1 ~ 1%) and 1000 ml fermented alcohol (50 ~ 95%) or water (10 ~ 50 times) was extracted with 100 g of mulberry leaf powder using a reflux extraction device ($80{\sim}90^{\circ}C$, 1 hour, twice). The extracts were collected, filtered and concentrated. For the recrystallization, the concentrate was dissolved by adding distilled water and allowed to stand at a low temperature. Then, the supernatant was discarded by centrifugation, and only the residue was lyophilized to prepare a final powder. As a result, regardless of the concentration of citric acid added, the content of rutin was higher in 90% fermented alcohol extract. Whereas, in the case of extracting with water, citric acid 0.5% was added to water 25 times as much as the weight of mulberry leaf powder, and 2274.4 (mg / 100g) of rutin content was highest in the case of refluxing twice at $80^{\circ}C$ for 1 hour. The powder with increased rutin content is expected to be applicable to various foods as a food additive. In addition, it can contribute to the improvement of the farm income by promoting consumption of mulberry leaf while satisfying the consumers' desire for functional food intake.
