Viability of Vibrio parahaemolyticus and V. vulnificus isolated from human in cultured olive flounder (Paralichthys olivaceus) (Vibrio parahaemolyticus와 V. vulnificus 사람 분리균의 넙치, Paralichthys olivaceus에서의 생존율)

  • Vibrio parahaemolyticus and V. vulnificus are known to be infected to human via fisheries products. Therefore, food safety of fisheries products is important for public health and fish industry. This paper was conducted to know how well these human isolates can survive in olive flounder (Paralichthys olivaceus). The growth of V. parahaemolyticus and V. vulnificus showed about 50~60% reduced rates at 25℃ than at 37℃ and did not show any differences according to NaCl concentration of media except the increasing in the growth of V. vulnificus in medium containing 3% NaCl. Artificial infection of 1×106 CFU/fish was carried out to confirm the sensitivity of olive flounder against V. parahaemolyticus and V. vulnificus. After 1 week from injection, no fish was dead. To evaluate nonspecific defense of olive flounder against V. parahaemolyticus and V. vulnificus, the antibacterial potency of serum and epidermal mucus were tested. The number of the vibrios exposed to serum obtained from olive flounder significantly decreased after 3 hours, and epidermal mucus showed decrease of the bacteria over than 90% until 12 hours from exposure. Phagocytosis of head kidney leucocytes of healthy olive flounder against V. parahaemolyticus and V. vulnificus showed in over 70% of leucocytes at the 2 hours. Therefore, cultured olive flounder only as vehicle for human pathogen in environmental water is well developed its antibacterial potency against human pathogens, so the viability of V. parahaemolyticus and V. vulnificus in cultured olive flounder was considered very low.

Sexual Behaviors, Estrus Detection and Conception of Heifers Synchronized by Progesterone Intravaginal Device(Prid) and Synchromate-B

  • Seventy four Holstein heifers were randomly assigned over three trials to PRID-7+PG-6 and Synchromate B-9 regimens to synchronize estrus cycle for embryo transfer. Sexual behaviors; moounting, standing, orientation, chin-resting, sniffing, licking, rubbing and butting, vaginal swelling and mucus discharge were observed between 06-08, 12-14 and 18-20 h on 1st day and 00-02, 06-08, 12-14 and 18-20 h on 2nd day after removal of hormones. Synchromate-B treatment (81.6%) showed higher synchronized estrus rate than PRID treatment (77.8%) during observation period. Standing estrus was observed within 74 h after PG injection in PRID and within 52 h after removal of implant in Synchromate-B. About 68% of heifers in PRID and 74% of heifers in Synchromate-B showed standing estrus between 0-14 h on 2nd day after removal of the hormones. Synchromate-B resulted in a tighter synchrony of standing estrus than PRID. Incidence of average mounting and standing per head during observation period was 22.3 and 16.6 in PRID and 28.1 and 13.6 in Synchromate-B. The PRID showed peak in active mounting at 18-20 h on 1st day, however, the Synchromate-B showed at 0-2 h on 2nd day after removal of hormone. Active standing was shown between 18 h on 1st day to 20h on 2nd day in PRID, however, between 0-14 h on 2nd day after removal of hormone is Synchromate-B. There was slight difference in pattern of active mounting and standing during estrus between PRID and Synchromate-B. Conception rate of synchronized heifers transferred with fresh and frozen embryos by non-surgical and surgical methods was higher in synchromate-B (62.5%) than in PRID (38.5%). Chin-resting showed highest incidence among 6 sexual behavioral components in the both treatments. Synchromate-B showed higher incidence of chin-resting (16.6) than PRID (10.7). Synchromate-B group showed also higher incidence of orientation, sniffing and butting than PRID group. Synchromate-B resulted in more active sexual behaviors than PRID. The pattern of incidence of chin-resting, licking and butting was almost symmetrical in PRID with their peak values at 6-8 h on 2nd day, however in Synchromate-B chin-resting and sniffing was symmetrical with their peak values at 12-14 h on 2nd day after removal of hormone. There was tendency to increase vaginal swelling according to time passage of synchronized estrus in the both treatments. Incidence of mucus discharge in Synchromate-B was slightly higher than in PRID. Twenty to 40% was false negative in conception rate by tall painting before re-estrus day as judged by rectal palpation.

Assessment of Treatment Response in Patients With Severe Asthma Using Visual and Quantitative Analysis of Chest CT

  • Objective: To evaluate the role of visual and quantitative chest CT parameters in assessing treatment response in patients with severe asthma. Materials and Methods: Korean participants enrolled in a prospective multicenter study, named the Precision Medicine Intervention in Severe Asthma study, from May 2020 to August 2021, underwent baseline and follow-up chest CT scans (inspiration/expiration) 10-12 months apart, before and after biologic treatment. Two radiologists scored bronchiectasis severity and mucus plugging extent. Quantitative parameters were obtained from each CT scan as follows: normal lung area (normal), air trapping without emphysema (AT without emph), air trapping with emphysema (AT with emph), and airway (total branch count, Pi10). Clinical parameters, including pulmonary function tests (forced expiratory volume in 1 s [FEV1] and FEV1/forced vital capacity [FVC]), sputum and blood eosinophil count, were assessed at initial and follow-up stages. Changes in CT parameters were correlated with changes in clinical parameters using Pearson or Spearman correlation. Results: Thirty-four participants (female:male, 20:14; median age, 50.5 years) diagnosed with severe asthma from three centers were included. Changes in the bronchiectasis and mucus plugging extent scores were negatively correlated with changes in FEV1 and FEV1/FVC (ρ = from -0.544 to -0.368, all P < 0.05). Changes in quantitative CT parameters were correlated with changes in FEV1 (normal, r = 0.373 [P = 0.030], AT without emph, r = -0.351 [P = 0.042]), FEV1/FVC (normal, r = 0.390 [P = 0.022], AT without emph, r = -0.370 [P = 0.031]). Changes in total branch count were positively correlated with changes in FEV1 (r = 0.349 [P = 0.043]). There was no correlation between changes in Pi10 and the clinical parameters (P > 0.05). Conclusion: Visual and quantitative CT parameters of normal, AT without emph, and total branch count may be effective for evaluating treatment response in patients with severe asthma.

Clinical Evaluation of Intrauterine Insemination with Washed Sperm in Infertile Patients (불임환자의 세척정자에 의한 자궁내 인공수정의 임상적 평가)

  • The sucess of intrauterine inseminations with washed human spermatozoa was evaluated in 92 patients. In according to indication of insemination, intrauterine inseminations of women with hostile cervical mucus yield a 35.3% and 12.8% with male factors and 27.3% with unexplained infertility. In ovulation induction group with variable agents including clomiphen, human menopausal gonadotropin (HMG) and GnRH analog, the pregnancy rate was 27.8% (22/79) and in natural cycle group, 15.4% (2/13). The fetal loss rate in insemination group was 12.5% (3/24). Multiple pregnancies were 7 cases and ovulation induction were performed in 6 cases among them. Intrauterine insemination with washed human spermatozoa therefore represents an effective and safe procedure selected infertile couples.

Nurse's Perceptions and Educational Intentions Regarding Natural Childbirth Control Methods (서울시내 일부 병원 간호사의 자연출산조절 방법에 대한 인식과 교육의도)

  • Purpose: This study was designed to identify nurses' perceptions and educational intentions regarding natural childbirth control (NCC) methods. Method: The participants were 313 nurses working at three general hospitals in Seoul. They were asked to complete a questionnaire composed of scales measuring knowledge and perceptions childbirth control methods, awareness of bodily changes in ovulation phase, perceptions of fetal life, which were developed by the author. Also, Chang's (2002) Sexual Autonomy Inventory was utilized. The data were analyzed by the SAS program. Results : Methods of condom and rhythm were considered to be more useful for childbirth control than other methods. Only 34-54% of them knew exactly about the NCC methods using menstrual cycle, body temperature, and mucus. The mean scores of sexual autonomy and awareness of bodily changes in ovulation phase were 3.8 and 3.4 out of 5, in respect. Educational intention was different statistically by the age, marital status, future intention to use NCC methods, and perception of educational need for NCC methods. Conclusion : Nurses' perceptions and educational intentions regarding NCC were low, especially in nurses who were married. It is recommended to encourage nurses to learn NCC methods for clients education.

Effect of Gamiyukgunja-tang on Secretion and Gene Expression of Airway Mucin (가미육군자탕(加味六君子湯)이 호흡기 뮤신분비 및 뮤신 유전자 발현에 미치는 영향)

  • In the present study, the author intended to investigate whether Gamiyukgunja-tang (Jiaweiliujunzi-tang, GYGT) significantly affect both mucin release from and MUC5AC gene expression in cultured hamster tracheal surface epithelial (HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of GYGT to assess the effect on 3H-mucin release. Possible cytotoxicity of the agent was assessed by measuring lactate dehydrogenase (LDH) release. Total elution profiles of control spent media and treatment sample through Sepharose CL-4B column were analysed and effect of GYGT on MUC5AC gene expression in cultured HTSE cells were investigated. GYGT did not affect mucin release from cultured HTSE cells. GYGT did not show significant cytotoxicity. GYGT also did not affect the secretion of the other releasable glycoproteins with less molecular weight than mucin. GYGT increased the expression level of MUC5AC gene. We suggest that the effect of GYGT with their components should be further investigated through ongoing research.

Effects of Socheongryong-tang and Kamichihyo-san on Mucin Secretion from Airway Goblet (소청용탕 및 가미치효산이 평흡기 배장세포로부터의 뮤신 분비에 미치는 영향)

  • In the present study, the author intended to investigate whether two oriental medical prescriptions named socheongryong-tang(SCRT) and Kamichihyo-san(KCHS) significantly affect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with ³H-glucosamine for 24 hrs and chased for 30 min in the presence of SCRT or KCHS to assess the effect of each agent on ³H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of SCRT and KCHS on contractility of isolated tracheal smooth muscle were investigated. The results were as follows: (1) SCRT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) KCHS significantly increased mucin release without cytotoxicity; (3) SCRT and KCHS did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of SCRT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the possible inhibitory effects on mucin release from the viewpoint of management of hypersecretion of airway mucus.

Effect of Dietary Supplementation with Galla rhois Extract on Lysozyme and Antibacterial Activity of the Cutured Catfish (Silurus asotus) (양식 메기의 라이소자임 활성 및 항 균력에 미치는 오배자 추출물의 투여 효과)

  • We are aim to evaluate lysozyme and antibacterial activity of cultured catfish, Silurus asotus, that fed supplementally with Galla rhois extracts for eight weeks. Lysozyme activity in the spleen and serum of administrated group was higher than not administrated group, but in mucus of the lysozyme activity was no regular than other organ. The lysozyme activity of the spleen, kidney, serum of administrated fishes were increased after 2 weeks and that was highest after 8 weeks. Ht and GLU in serum of administrated fishes were gradually increased but GOT was decreased after 8 weeks. There is no significant differences in HB (Hemoglobin) and TP (Total Protein) each groups. Furthermore, there is no pathohistological changes of kidney and liver of tested fishes. The cumulative survival rates of administrated group after intraperitoneal injection of Aeromonas veronii with $6.5{\times}10^6cfu\;mL^{-1}$ was presented 33% in 9 days. As the Results, Galla rhois extracts has any beneficial effects for immunity elevation and antibacterial activity in catfish, Silurus asotus.

Chronic osteitic rhinosinusitis as a manifestation of cystic fibrosis: A case report

  • A 28-year-old male patient with a history of cystic fibrosis (CF) was referred to the University of Connecticut School of Dental Medicine for an evaluation of a cystic lesion in the right maxilla using cone-beam computed tomography (CBCT). CF is an autosomal recessive disease characterized by an abnormal production of viscous mucus, affecting the mucociliary clearance. The CBCT scan revealed a large cystic lesion in the right maxilla extending from the right maxillary second molar to the midline in the region of the right central incisor with a significant buccal expansion. Further evaluation revealed complete opacification of the paranasal sinuses with medial bulging of the lateral maxillary sinus walls. The maxillary and sphenoid sinuses also appeared hypoplastic. The peculiar finding seen in this case was the presence of marked sclerosis and an increase in the thickness of the adjacent bony framework. This report aimed to describe the common sinonasal findings associated with CF and its underlying pathophysiology.

Surgical Treatment of a Plunging Ranula using the Intraoral and Submandibular Approach (구강 내 접근과 하악하 절개를 통한 몰입성 하마종의 수술적 치료)

  • Purpose: A plunging ranula is relatively uncommon and represents a mucus escape reaction occurring from a disruption of the sublingual salivary gland. It is a common condition found in young adults, even though the reported age range is 2 - 61 years. We report our experience of a complete excision of a plunging ranula via the intraoral and submandibular approach. Methods: A 23-year-old man had a large protruding mass in the right submandibular area. Initially, the protruding mass appeared bilaterally but the left side disappeared spontaneously. The MRI findings revealed a homogenous fluid attenuation mass in the submandibular space, suggesting a ranula. The sublingual gland was extirpated through the intraoral approach and the ranula excised totally via the submandibular approach. Results: The patient had an uneventful postoperative course without infection, paralysis and tongue sensory changes, etc. The pathology findings were characteristic of a pseudocyst without a lining epithelium or endothelium but with a vascular fibro-conective tissue wall filled with mucinous fluid. No recurrence was observed on the submandibular area during the 8 month follow-up period. Conclusion: The combined intraoral approach and submandibular approach is an effective and highly recommended method for sublingual gland extirpation and complete excision of a plunging ranula.
