Kalopanax pictus was cultured in vitro to find out optimal condition for embryogenic cells proliferation in liquid media rapidly. Embryogenic cells were induced from leaves and petiols of Kalopanax pictus. Optimum culture medium appeared to be a 1/2MS medium supplemented with 2.0mg/L 2,4-D and 0.1mg/L BA. To find out optimal conditions, embryogenic cells were cultured some condition as different concentrations of 2,4-D, medium and sucrose. There was cultured on 1/2MS liquid medium containing different concentration of 2,4-D. When embryogenic cells were cultured on 1/2MS liquid medium supplemented with 1.0mg/L 2,4-D, cell propagation rate was higher than other concentration of 2,4-D. When embryogenic cells were cultured on different media that MS, Gambols B5, N6, White, SH medium, observed the highest multiplication rate among Gambols B5 and White medium. To find out of effect of sucrose to embryogenic cells propagation, we tested cells under different concentrations. Optimal concentration of sucrose appeared to be a basal medium added 3% sucrose. Above results suggest that optimal conditions for proliferation of embryogenic cells were established Gambols B5 and White medium added 1.0mg/L 2,4-D and 3% sucrose. There is every possibility achieving embryogenic cells proliferation via bioreactor culture system in Kalopanax pictus.
This study was conducted to investigate an optimal condition for shoot proliferation and regenerate shoots from in vitro leaflet and embryogenic calli from in vitro roots in rose. The effect of BAP on shoot proliferation was somewhat different depending upon genotypes or gelling agents. Leaflets with petiole cut from donor shoots which had been cultured in MS medium supplemented with 0.1 $mg{\cdot}L^{-1}$ NAA for six weeks was effective for regeneration of adventitious buds(ABs) as well as shoot elongation of Rosa hybrida cv. Sweet Pink. Culturing seven leaflet explants per petri plate($100mm{\times}15mm$) was effective for regeneration of ABs. Embryogenesis was shown in the calli induced from roots of Rosa hybrida cv. Sweet Pink cultured in the SH medium supplemented with 11 $mg{\cdot}L^{-1}$ 2, 4-D for four weeks. Color of calli induced from roots was yellow although their color was a little different as type of basal medium.
Panax ginseng is an important medicinal plant that has been used worldwide for geriatric, tonic, stomachic, and aphrodisiac treatments. Ginsenosides contained in the ginseng root are the main substances having active functions for human body. The price of ginseng is very expensive due to a complex process of cultivation, and the yield of ginseng is limited, which cannot meet the demand of the increasing market. Researchers have applied plant biotechnology to solve the problems but there are still things to be determined towards ginsenoside production by large-scale adventitious root culture. In this experiment, 5 to 20 liter bioreactors were employed to determine optimal conditions for adventitious root culture and ginsenoside production of Panax gineng. Callus was induced from the ginseng root on MS agar medium containing 1.0 mg. $L^{-1}$ 2,4-D and 0.1 mg. $L^{-1}$ kinetin. Then the callus was cultured on MS agar medium supplemented with 2.0 mg. $L^{-1}$ IBA, 0.1 mg. $L^{-1}$ kinetin, and 30 g. $L^{-1}$ to induce adventitious roots. The maximum root growth and ginsenoside production were obtained in 1/2 MS medium. 2.0 mg. $L^{-1}$ naphthalene acetic acid resulted in greater root growth than 2.0 mg $L^{-1}$ indole-3-butyric acid. Ginsenoside content increased with 2.0 mg. $L^{-1}$ benzyl adenin or kinetin. High concentrations of benzyl adenin (above 3.0 mg. $L^{-1}$ ) decreased the adventitious root growth and ginsenoside productivity. N $H_{4}$$^{+}$ inhibited the ginsenoside accumulation, while high concentrations of $K^{+}$, $Mg_{2}$$^{+}$, and $Ca_{2}$$^{+}$ increased it. N $H_{4}$$^{+}$ at 0.5 and 1.0 times of the normal amount in 3/4 SH medium resulted in the greatest biomass increase, but the highest ginsenoside productivity was obtained when N $O_{3}$$^{-}$ was used as the sole nitrogen source in the medium. Most microelements at high concentrations in the medium inhibited the root growth, but high concentrations of MnS $O_4$enhanced the root growth. Root dry weight increased with increasing sucrose concentrations up to 50 g. $L^{-1}$ , but decreased from 70 g $L^{-1}$ Ginsenoside productivity was maximized at the range of 20 to 30 g. $L^{-1}$ sucrose. In the experiment on bioreactor types, cone and balloon types were determined to be favorable for both adventitious root growth and ginsenoside production. Jasmonic acid was effective for increasing ginsenoside contents and Rb group ginsenosides mainly increased. These results could be employed in commercial scale bioreactor cultures of Panax ginseng.x ginseng.

Micropropagation of Ulmus davidiana Planch was established via adventitious shoot formation from the segments of adventitious roots. Adventitious roots were produced directly from root segments of seedlings on a 1/2 SH medium plus various concentrations of IBA. The maximum growth of adventitious roots was observed in the presence of 2.0 mg/L IBA. After the segments of adventitious roots were cultured on various cytokinins (zeatin, 2-iP, BA, kinetin) and cytokinins plus auxin (IBA), formation of adventitious shoot was investigated. Among cytokinin treated, kinetin was the most effective on both adventitious shoot induction and number of shoots. Especially, 2.0 mg/L kinetin was the best to increase adventitious shoot induction (95.8%) and a number of shoots (8.4). Adventitious shoots were rooted on 1/2 WPM medium and the plantlets were acclimated 100% on composed soil (peatmoss : vermiculite 1 : 1).
In order to acquire 3 components data which has the good signal to noise ratio with only one shot, 3-C geophones were used, As a result, the vertical component showed the distinct first arrival of P-wave, and the horizontal component was improved the signal to noise ratio of S-wave, while was attenuated P-wave. The 2-D Poisson's ratio section was computed from P- and S-wave cell velocities included velocity tomograms of the P- and S-waves. The Poisson's ratio values were computed in the range of $0.2{\~}0.3$. With one shot, we can obtain 2-D distribution of dynamic Poisson's ratio as well as velocity tomograms of P- and S-waves.

In the course of our continuing search for biologically active components from Korean medicinal plants, we isolated the main compound, luteolin 5-glucoside from aqueous fraction of Ajuga spectabilis. The structure was elucidated by the basis of $^1H$ and $^{13}C$ NMR and TOF ESI-MS data. Quantitative analysis of luteolin 5-glucoside was carried out on a XBridge C18 column ($S-5{\mu}m$, $4.6{\times}250mm$) with gradient elution composed of acetonitrile:water. The results exhibit that the average content of main compound in A. spectabilis were 0.048%. Oxidative stress plays a major role Alzheimer's disease (AD) and other neurodogenerative disease. AD is major health problem and there is currently no clinically accepted treatment to cure or stop its progression. Pretreatment with luteolin 5-glucoside markedly attenuated $H_2O_2$-induced cell viability loss in a dose-dependent manner. Luteolin 5-glucoside also inhibited the formation of intracellular reactive oxygen species in SH-SY5Y. The results suggest that luteolin 5-glucoside from A. spectabilis has protective effects against oxidative stress-induced cytotoxicity, which might be a potential therapeutic compound for treating and/or preventing neurodegenerative disease implicated with oxidative stress.
Novel polythiol materials of urethane lens series for plastic optical lens were synthesized from polyol materials via thioisouronium of thiourea with c-HCl in refluxing aqueous solution, in which polythiol material was carried out from hydrolysis of thioisouronium by ammonia water. Their structure properties were identified by EA, EI-MS, FT-IR, $^1H\;and\;^{13}C$ NMR spectroscopies and TGA. Their ophthalmic lenses as polythiourethane material were prepared by thermal curing to an injected glass mold using the evenly solutions of diisocyanates series (TDI, XDI, HDI or IPDI) with polythiols. Polythiourethane shows that the strong stretching mode for SH group of polythiol disappeared in FT-IR spectra after thermosetting polymerization. Thermal deformation starting temperature of ophthalmic lenses was determined by TMA. Ophthalmic lenses made from characteristic polythiol and diisocyanate series have transparency, colorless and good impact strength, in which thermal resistance and impact strength of ophthalmic lenses were influenced by diisocyanate series. Physical properties of ophthalmic lens have contrast thermal resistance with impact strength. The property of thermal resistance and impact strength for respective ophthalmic lenses was examined by TMA and drop ball test.

Ginseng(Panax ginseng C.A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng callus and hairy roots grow vigorously and may Produce the same or more biologically active compounds for human health than natural ginseng roots. Therefore, ginseng callus and hairy roots can be used for commercial purposes. Polyacetylene, one of anti-cancer compounds in ginseng, was not detected in the callus cultured on the medium containing 2, 4-B, but cells derived from the callus growth was excellent, The ginseng calli cultured on the medium containing 2mg11 CPA and 0.05mg/1 BA was grown vigorously and produced panaxydol, one of ginseng polyacetylene. The biosynthesis of polyacetylene in callus was not affected by addition of NAA and sucrose in media. The SH medium was better than the MS medium for ginseng callus growth and biosynthesis of panaxydol. Another ginseng anti-cancer compounds, ginsenoside-Rg$_3$, Rh$_1$and Rh$_2$ were detected in ginseng hairy roots by heat treatment. Those of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes Rl000 $A_4$T in dark condition after one month of culture. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark(22$^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5 ι Erlenmeyer flasks, 1ι roller drums, 10ι jar-fermenters, and especially in 20ι air-lift .culture vessels. All heat treatments had remarkably different ginsenoside contents. Eleven ginsenosides were determined in heat treatment, eight in freeze dried hairy roots. Contents of ginsenoside-Rbl , Rb2, Rc, Rd. Re, Rf, and Rg$_1$tested in all heat treatments were less than those of freeze dried hairy roots. Contents of glnsenoside-Rg$_2$ in heat treatment for 1 hour at 105$^{\circ}C$ was 4.92mg/g dry wt, 3.9 times higher than 1.27 mg/g dry wt of freeze dried hairy roots. The optimum condition of heat treatment for the production of ginsenoside-Rg$_3$and Rhl was 2 hours at 105$^{\circ}C$, and ginsenoside content was 2.58mg/g dry wt and 3.62mg/g dry wt, respectively. The production of ginsenoside-Rh2 was the highest in heat treatment for 2 hours at 105$^{\circ}C$ among treatments examined, and ginsenoside-Rh$_2$content was 1.08mg/g dry wt.

This study was performed to classify habitat types depending on environmental factors and to find out distribution characteristics of functional feeding groups of aquatic insects which were collected at that habitat types. Field survey was conducted twice in a year for every spring and fall from 2007 to 2008 for 38 sites in the Geum River. During the field survey 15 environmental factors were measured at each 38 sites and analyzed by similarity analysis method to classify habitat types. The result of similarity analysis showed that the 38 sites could be grouped into 7 classes like as C1 and C3 class belong to Head water(HD), C2 and C4 and C5 class belong to Middle stream(MS), C6 and C7 class belong to Large River(LR) based on euclidean distances 4. And also, we could extract the main environmental factors affecting the classification of habitat types such as Stream Width and Elevation of physical environmental factors, Water Temperature, Conductivity and DO of chemical environmental factors, percentages of Sand, Silt and Gravel of substrate factors. Total 142 species of aquatic insects in 46 families, 9 orders were collected during the field surveys and the occurrence number of species and individuals showed high correlation with the Velocity factor and the percentage of Sand factor of each habitat types. In addition, correlation analysis between functional feeding groups and environmental factors represented that (1) Filtering-collectors(FC) affected by Velocity, Stream Width and Silt, (2) Gathering-collector(GC) affected by Velocity, (3) Predator(P) affected by Elevation, Velocity, Boulder, Conductivity and Sand, (4) Plant-piecer(PP) affected by Water Width and Silt, (5) Scraper(SC) affected by Elevation and Conductivity, (6) Shredder(SH) affected by Elevation, Boulder, DO, pH, Conductivity and Water Temperature respectively. As a result of this study, Elevation, Stream Width, Velocity, Conductivity, Water Temperature and percentage of Sand factors which were deduced by stepwise multiple regression analysis had correlations($r{\geqq}0.600$, p<0.01) with biota community inhabitation. Therefore these six environmental factors were regarded as major environmental factors that might affect highly the distribution of functional feeding groups in stream ecosystem of the Geum River.
As it has been reported that the depolarization-induced norepinephrine (NE) release is modulated by activation of presynaptic $A_1-adenosine$ heteroreceptor and various lines of evidence indicate the involvement of adenylate cyclase system in $A_1-adenosine$ post-receptor mechanism in hippocampus, it was attempted to delineate the role of adenylate cyclase system in the $A_1-receptor-mediated$ control of NE release in this study. Slices from rat hippocampus were equilibrated with $[^3H]-NE$ and the release of the labelled products was evoked by electrical stimulation.(3 Hz, $5Vcm^{-1}$, 2 ms, rectangular pulses). The influence of various agents on the evoked tritium-outflow was investigated. $N^6-Cyclopentyladenosine$ (CPA), a specific $A_1-adenosine$ receptor agonist, in concentrations Tanging from 0.1 to $10{\mu}M$ decreased the $[^3H]-NE$ release in a dose-dependent mauler without any change of basal rate of release. 8-Cyclopentyl-1,3-dipropylxanthine (DPCPX, $2{\mu}M$), a selective $A_1-receptor$ antagonist, inhibited the CPA effect. The responses to N-ethylmaleimide $(3&10{\mu}M)$, a SH-alkylating agent of G-protein, were characterized by increments of the evoked NE-release and the CPA effects were completely abolished by NEM pretreatment. Forskolin, a specific adenylate cyclase activator, in concentrations ranging from 0.1 to $30{\mu}M$ increased the evoked and basal rate of NE release in a dose-dependent manner and the CPA effects were inhibited by forskolin pretreatment. Rolipram $(1&10{\mu}M)$, a phosphodiesterase inhibitor, did not affect the evoked NE release but reduced the CPA effect. And 8-bromo-cAMP $(100&300{\mu}M)$, a membrane permeable cAMP analogue inhibited the CPA effect significantly. These results suggest that the $A_1-adenosine$ heteroreceptor plays an important role in NE-release via nucleotide-binding protein $G_i$ in the rat hippocampus and that the adenylate cyclase system might be participated in this process.
