Analysis of the Genetic Relationship among Mulberry (Morus spp.) Cultivars Using Inter-Simple Sequence Repeat (ISSR) Markers

  • Mulberry (Morus spp. family: Moraceae) has prime importance in the sericulture industry, and its foliage is the only natural feed of the silkworm Bombyx mori L. Traditional classification methods using morphological traits were largely unsuccessful in assessing the diversity and relationships among different mulberry species because of environmental influences on the traits of interest. For these reasons, it is difficult to differentiate between the varieties and cultivars of Morus spp. In the present study, inter-simple sequence repeat (ISSR) markers were used to investigate the genetic diversity of 48 mulberry samples genotyped using nine ISSR primers. The ISSR markers exhibited polymorphisms (53.2%) among mulberry genotypes. Furthermore, similarity coefficient estimated for these ISSR markers was found to vary between 0.67 and 0.99 for the combined pooled data. The phenogram drawn using the UPGMA cluster method based on combined pooled data of the ISSR markers divided the 48 mulberry genotypes into seven major groups. No genetic association was found in the collection area, and there was a mixed pattern between the mulberry lines. The hybridization between different mulberry species is highly likely to be homogenized due to natural hybridization.

Radical Scavenging Activities of Phenolic Compounds Isolated from Mulberry (Morus spp.) Cake

  • A methanol extract of mulberry cake prepared from mulberry fruits (Morus spp.) was shown to have strong scavenging activities against DPPH, superoxide and hydroxyl radicals. Eleven phenolic compounds were isolated from the mulberry cake by a combination of Diaion HP-20, silica gel (or polyamide), Sephadex LH-20 column chromatographies, preparative HPLC and TLC. Their chemical structures were characterized as procatechuic acid (PCA), caffeic acid (CA), cyanidin 3-O-$\beta$-D-glucopyranoside (CyG) and cyanidin $3-O-\beta­D-rutinoside$ (CyR), rutin (RT), isoquercitrin (IQT), astragalin (AG), quercetin (QT), morin (MR), di-hydroquercetin (DHQ), and 4-prenylmoracin (PM) by spectral analysis and the published data. Most of the phenolic constituents were effective scavengers of DPPH, superoxide and hydroxyl radicals, and especially caffeic acid and 4-prenylmoracin showed potent superoxide and hydroxyl radical scavenging activity, in which their activities were higher than that of the well-known antioxidant, BHT (p< 0.05). Dehydroquercetin and quercetin also exhibited strong superoxide and hydroxyl radical scavenging activities. These results suggest that mulberry cake containing antioxidant phenolic compounds may be useful as natural antioxidants in functional foods and cosmetics.

  • Anatomical and chemical properties of three mulberry species (Morus spp) were analyzed as an alternative row material for the paper mulberry (Broussonetia kazinoki) . The pulping and papermaing characteristics of bast fiber and whole stalks by three different pulping processes, conventional alkali, alkali-hydrogen peroxide, and sulfomethylated, for the Hanji were investigated. The fiber dimension of M.bombycis was the biggest of the species. The fiber length of upper part and the fiber width and cell wall thickness of lower part were bigger than the others. The extractives and ash contents of bast fiber were higher than those of whole stalk , and holocellulose and lignin contents of whole stalk were higher than those of bast fiber. The pulp yields of M.alba bast fiber, M.bombycis whole stalk, and sufomethylated pulping were higher than the others.

Mathematical Constants for Non-Destructive Rapid Method of Leaf Area Determination in Mulberry (Morus spp.)

  • Mathematical constants for multiplication with leaf length (I) or breadth (b) or l ${\times}$ b have been worked out for determining leaf area in promising mulberry genotypes viz., Chinese White, S-146, Chak Majra and Sujanpur Local of sub-tropical India. When pooled, the mathematical constants worked out were 8.1132, 10.1019 and 0.5992 for multiplication with leaf length, breadth and l ${\times}$ b, respectively, for genotypes bearing un-lobbed leaves and 6.9447, 8.2761 and 0.5009 for multiplication with leaf length, breadth and l ${\times}$ b, respectively for genotypes bearing lobbed leaves. Leaf area can be worked out by using any constant by multiplying either with leaf length or breadth or both (l ${\times}$ b). Estimated leaf areas worked out were found significantly and positively correlated with actual leaf area (r=999$^{**}$). The suggested present non-destructive method by using mathematical constants is very quick and alternative to electronic leaf area meter for spot leaf area determination in mulberry which is the only food source for mulberry silkworm in sericulture industry.

  • Excellence of the Korean traditional paper(Hanji) can be proved by literatures and legacies handed down from generations. However, with the problems pertaining to a majority of traditional manufactures, Hanji industry was declined because of low productivity, insufficient supply of raw material , and reduced demand. therefore, modernizxation of the Hanji technology and development of new uses are very important. This study was carried to investigated the papermaking characteristics of Morus spp.(M.alba, M.Bombycis, and M.Ihou) for new Hanji, modernize manufacturing process of Hanji by grafting on developed modern pulping and papermaking technology, develop the various uses, and establish the foundation for development of high value-added products. Hanji is made from M.Ihou, while stalk , and sulfomethylated pulp gave better sheet formation and higher brightness than those of the others. Physical properties of Hanji which made from bast fiber pulps were better than those of whole stalk pulps. Hanji which made from M.bombycis bast fiber and whole stalk pulps were not so good sheet strength as other species. Sulfomethylated whole stalk pulps were shown better sheet strength than alkali and alkali-hydrogen peroxide whole stalk pulps.

Simple Self Trap Cropping System to Control Tukra Mealy Bug (Maconellicoccus hirsutus Green) Incidence on Mulberry (Morus spp.)

  • A field study on self trap cropping system in controlling tukra mealy bug Maconellicoccus hirsutus Green) incidence on mulberry (Morus spp.; V-1 variety; $3'{\times}3'$ spacing) gardens was undertaken in selected sericulture farmers' mulberry gardens of Pydeti village (Parigi Mandal, Anantapur District, Andhra Pradesh, India). The study was conducted for three years (2004, 2005 and 2006). Four treatments (T0; control/no treatment, T1; self trap cropping rows with 10 row interval, T2; self trap cropping rows with 10 row interval and treating these rows only with 0.5% neem oil emulsion and T3; self trap cropping rows with 10 rows intervals and treating all the mulberry rows including self trap cropping rows with 0.5% neem oil emulsion) were considered. The results indicated that the incidence increased in T0 and T1 while the same suppressed in T2 and T3. The suppression (%) of tukra incidence between T2 and T3 was not significant. Hence, T2 only was recommended to farmers as its economical viable practice. The results are discussed based on the importance of tukra, its suppression and cost of treatment.

Evaluation of ISSR and RAPD Markers for the Detection of Genetic Diversity in Mulberry (Morus spp.)

  • The present study was carried out to evaluate the ISSR and RAPD markers for their efficiency as genetic marker systems to establish the relationships between 18 mulberry genotypes. A total of 36 from 56 (64%) RAPD primers and 12 from 48 (25%) ISSR primers produced reproducible amplification patterns. A high proportion of polymorphic bands ranging from 44 to 91% was observed respectively with RAPD and ISSR markers. The average Resolving Power (Rp) of ISSR primers was higher than RAPD primers. The ISSR primers, UBC 825, 868 and 873, and RAPD primers, UBC 712, 720 and 729, possessed the highest Rp values and could in each instance distinguish all the 18 genotypes. Similarity matrix values were estimated based on Jaccards coefficient, considering 109 polymorphic ISSR and 212 polymorphic RAPD bands and two dendrograms were constructed. The dendrograms obtained with ISSR and RAPD markers distinguished the eight exotic genotypes from the ten indigenous (Indian) genotypes. A significant correlation value (r=0.959; p=0.001) for the cophenetic matrix between the RAPD and ISSR matrices was observed. The results indicated that the ISSR and RAPD markers could assist in the differentiation of genotypes and permit the determination of genetic distances that might be exploited by mulberry breeders in improvement programs.

최소가공기술을 이용한 오디 과실주스의 제조 (Preparation of Minimally Processed Mulberry (Morus spp.) Juices)

  • 오디 과실을 이용한 고부가가치의 가공식품의 개발에 관한 연구 일환으로 먼저 최소가공기술을 이용한 오디 과실주스 제조 방법을 조사하였다. 오디 과실의 물추출액을 면포여과, 감압여과(Whatman No. 4 filter paper를 이용한) 및 원심분리를 각각 실시한 결과 원심분리가 가장 좋은 청징효과를 나타내었다. 그리고 여러 여과보조제 및 청징제를 첨가한 후 원심분리하여 얻은 오디 과실즙의 색의 변화는 처리한 여과보조제, 청징제 및 청징 효소의 종류에 따라 차이가 있었으며, 특히 casein, gelatin, chitin, chitosan 및 PVPP는 오디 색소를 강하게 흡착하였으며, 그리고 청징효소 중 ${\alpha}-amylase$ 처리구가 가장 좋은 청징효과를 나타내었다. 다음, 오디 과즙을 원심분리한 후 $0.01\;{\mu}m$ 막여과를 실시한 결과 오디주스의 청징효과가 매우 우수하였으나 한외여과방법은 오디 색소의 강한 흡착으로 적절하지 않았다. 한편, 4가지 갈변저해제 처리에 따른 오디 과실주스의 갈변억제 효과를 측정한 결과 200ppm sodium hydrosulfite 처리구가 가장 갈변억제 효과가 우수하였으며, 아울러 0.1% L-ascorbic acid(L-AsA) 및 citric acid(CA)도 갈변억제 효과가 있었다. 그리고 위의 3가지 갈변저해제를 2중 복합병행 처리한 결과 0.1% L-AsA+0.1% CA 처리구가 가장 좋은 갈변억제 효과를 나타내었다. 마지막으로 관능검사를 실시한 결과 오디주스의 최적 당/산비는 40-50이었으며, 또한 상업적 살균 온도($85-90^{\circ}C$)에서 처리시간은 10분이 가장 적합함을 알 수 있었다.

Antagonistic and growth promotion potential of endophytic bacteria of mulberry (Morus spp.)

  • Endophytes provide multifarious benefits such as promotion of plant growth and yield, suppression of phyto-pathogens, phosphate solubilising and fixation nitrogen. A study has been carried out to explore growth promotion and antifungal activities of endophytes of mulberry (Morus spp.). Endophytic bacteria were isolated from mulberry plants and studied their cultural, morphological characters, growth promotion as well as their antifungal activity against Rhizoctonia bataticola and Fusarium oxysporum , two mulberry root rot associated pathogens. Except two isolates, all bacteria were colourless and the colony size of eight isolates was small. The margin of five isolates was irregular and the consistency of three isolates was creamy, six isolates was slimy and one was mucoid. Texture of seven isolates was convex and others were flat. Eight isolates were gram positive and the rest Gram negative, five were cocci and others were bacilli (rod shaped). Four isolates were motile and all were catalase positive and only three isolates were oxidase positive. Spore staining was positive only for two isolates. The growth promotion study showed that there was significant difference in root length and seedling length. The antagonistic effect of the bacterial isolates was tested against R. bataticola showed significant (p <0.05) influence of the bacteria, days after inoculation and their interaction on the inhibition of fungal growth. The isolate En-7 completely inhibited the fungus followed by En-5 (66.67%). The bacterial isolates significantly (p <0.05) inhibited growth of F. oxysporum in PDA. The mean inhibition was higher (70.45%) in case of En-7 followed by En-8 (68.65%) and En-10 (66.44%). The study reveals that some endophytic bacteria associated with mulberry have growth promotion and antifungal activity and could be explored for promotion of mulberry growth and managing root rot disease.

Comparison of Radical Scavenging Activity of Extracts of Mulberry Juice and Cake Prepared from Mulberry (Morus spp.) Fruit

  • Radical scavenging activity of water and methanol extracts of mulberry juice and cake prepared from mulberry fruit (Morus spp.) was evaluated using three in vitro assay systems. Mulberry fruits were homogenized with $0.5\%$ trifluoroacetic acid (TFA) in distilled water, filtered with cheeze-cloth and centrifuged to yield mulberry juice and cake. Mulberry juice was evaporated and solubilized in $0.5\%$ TFA in distilled water or $0.5\%$ TFA in $80\%$ aqueous methanol, followed by filtration and evaporation to obtain water (WMJ) and methanol (MMJ) extracts of mulberry juice. Mulberrry cake also was extracted with the above same solvents, and thereby finally obtaining water (WMC) and methanol (MMC) extracts of mulberry cake. Among four extracts, the MMC showed the most potent radical scavenging activity against DPPH radical $(IC_{50}=167.45\;{\mu}g/mL)$, and superoxide $(IC_{50}=36.18\;{\mu}g/mL)$ and hydroxyl radicals $(IC_{50}=467.08\;{\mu}g/mL)$. The WMC also exhibited stronger radical scavenging activity than those of two other mulberry juice extract, WMJ and MMJ. Meanwhile, the MMJ exerted stronger three radical scavenging activity than the WMJ. Total phenolic content of the water and MeOH extracts from mulberry cake was higher than that of the water and MeOH extracts from mulberry juice. Thus, these results suggest that the extracts of mulberry cake with high dietary phenolics may be useful potential source of natural antioxidant as radical scavenger.
