Detection, modulation, and transmission of sweet taste in regulation for energy homeostasis

  • Perception of sweet compounds is important for animals to detect external carbohydrate source of calories and plays a crucial role in feeding behavior of animals. Recent progress in molecular genetic studies provides evidence for a candidate receptor (heterodimers with taste receptor type 1 member 2 and 3: T1R2/T1R3), and major downstream transduction molecules required for sweet taste signaling. Several studies demonstrated that the sweet taste signal can be modulated by a satiety hormone, leptin, through its receptors expressed in a subset of sweet-sensitive taste cells. Increase of internal energy storage in the adipose tissue leads to increase in the plasma leptin level which can reduce activities of sweet-sensitive cells. In human, thus, diurnal variation of plasma leptin level parallels variation of taste recognition thresholds for sweet compounds. This leptin modulation of sweet taste sensitivity may influence individuals' preference, ingestive behavior, and absorption of nutrients, thereby plays important roles in regulation of energy homeostasis.

Broadband Light Absorption Using Gap Plasmon Resonance

  • A gap surface plasmon resonator have received considerable attention because it can dramatically enhance the absorption of the electromagnetic field. However, whereas most of studies were just focused on the absorption within a narrow range of wavelength, few studies have been performed for the broadband absorption in the visible range. Therefore, in this study, we discuss methods that can induce broadband light absorption using gap plasmon resonance in visible regime. The gap plasmon resonator will offer great potential for appplications to solar cells and bioimaging.

Alkoxy-amine-aluminum 유도체에 의한 키랄 분자 인식 (Chiral Molecular Recognition by Alkoxy-amine-aluminum Derivatives)

  • The enantioselective reduction of representative prochiral alkyl-aryl ketones with a new chiral alkoxy-amine-aluminum derivatives from aluminum hydride and ${\alpha},{\alpha}$-diphenyl-${\beta}$-amino alcohols, such as (S)-(-)-2-amino-3-methyl-1,1-diphenylbutan-1-ol(AMDPB) and (S)-(-)-2-(diphenylhydroxy-methyl)pyrrolidine(DPHMP), in THF at $0^{\circ}C$ was studied. In the reduction of alkoxy-amine-aluminum derivatives, acetophenone, propiophenone, isopropiophenone, and butyrophenone are reduced to corresponding aromatic secoundary alcohols with 34~60 % enantiomeric excess of (S)-isomers. For such ketones, the optical induction was enhanced by increasing a size of alkyl groups.

고감도 압저항 외팔보 센서를 이용한 Liposome의 검침 (Rapid detection of liposome by piezoresistive cantilever sensor)

  • Liposomes are microscopic spherical vesicles that form when lipids are hydrated and have been widely used for biochemical assay, drug delivery and molecular imaging. In particular, they are well known for artificial cell membranes to study cellular functions such as cell fusions and membrane proteins. Here, we firstly report the detection of liposomes by the highly sensitive microfabricated piezoresistive cantilever sensor chip and the phosphatidylserine recognition protein C2A which is chemically immobilized on the sensor surface. The signal created from the bending motion of piezoresistive cantilever after the liposome attachment has been monitored in real time.

  • The purpose of the study is to develop an integrated knowledge structuring system for the domain of engineering, in which ontology-based literature mining, knowledge acquisition, knowledge integration, and knowledge retrieval are combined using XML-based tag information and ontology management. The system supports combining different types of databases (papers and patents, technologies and innovations) and retrieving different types of knowledge simultaneously. The main objective of the system is to facilitate knowledge acquisition and knowledge retrieval from documents through an ontology-based dynamic similarity calculation and a visualization of automatically structured knowledge. Through experimentations we conducted using 100,000 words economic documents reported in the "Go! Japan" project for analyzing Japanese industrial situation, and 100,000 words molecular biology Papers, we show the system is Practical enough for accelerating knowledge acquisition and knowledge discovery from the information sea.

Ab Initio Study of Conformers of p-tert-Butylcalix[4]crown-6-ether Complexed with Alkyl Ammonium Cations

  • The structures and energies of p-tert-butylcalix[4]crown-6-ether(1) in various conformers and their alkyl ammonium complexes have been calculated by ab initio HF/6-31G quantum mechanics method. We have tried to obtain the relative affinity of partial-cone and 1,3-alternate conformers of 1 for alkyl ammonium guests by comparison with its cone-shaped analogue. We have also calculated the relative complexation efficiency of these host-guest complexes focusing on the binding sites of $crown-\sigma-enther$ moiety or benzene-rings pocket of the host molecule 1. These calculations revealed that the crown moiey has better complexation efficiency than upper rim part of calyx[4]arene that is in similar trend to the cone-shaped complexes.

  • PDZ domains are molecular-recognition elements that mediate protein-protein interactions. The PDZ domain was discovered originally as a common motif present in three structurally related proteins: PSD-95 (postsynaptic density protein), Dlg (discs-large protein) and ZQ-1 (zonula occludens-1). The PDZ domain is globular domain, containing about 80-100 amino acids, and a conserved motif with two alpha helices and six beta strands. Most of them bind selectively to the C-termini of the interacting proteins at the complexes of signaling molecules and membrane associated receptors.(omitted)

Fabrication of Functional Nanomaterials by Peptide Self-Assembly

  • The self-assembly of peptide-based building blocks into nanostructures is an attractive route for fabricating novel materials because of their capacity for molecular recognition and functional flexibility as well as the mild conditions required in the fabrication process. Among various peptide-based building blocks forming nanostructures, the simplest building blocks are aromatic dipeptides like diphenylalanine, which can readily self-assemble into nanotubes in aqueous solutions at ambient conditions. Recently, we have developed a high-temperature solid-phase self-assembly process for diphenylalanine. Through this novel process, we succeeded in the growth of vertically well-aligned, uniform nanowires from amorphous peptide thin film. To demonstrate the versatility of our approach, we also fabricated a micropattern of peptide nanowires by combining our solid-phase growth method and simple soft lithographic techniques. We believe that our studies on peptide self-assembly will provide a new horizon for peptide-based nanofabrication.

The Preparation of D-Phenylalanine Imprinted Microbeads by a Novel Method of Modified Suspension Polymerization

  • Molecularly imprinted polymeric microbeads (MIPMs) were prepared by the suspension and modified suspension polymerization methods using D-phenylalanine as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross-linker, toluene as the porogen, polyvinyl alcohol as the stabilizer, and sodium dodecyl sulfate as the surfactant. The addition of a surfactant to the conventional suspension polymerization mixture decreased the mean particle size of the MIPMs and increased the adsorption selectivity. For the modified suspension polymerization method, the mean particle size of the MIPMs was smaller than the particle size of MIPMs prepared via conventional suspension polymerization. Moreover, the adsorption selectivity improved considerably compared to the adsorption selectivities of MIPs reported previously.

  • The beta-turn has been implicated as an important conformation for biological recognition of peptides or proteins. We adapted the concept of general Ca atom positioning from the cluster analysis and recombination of each ideal beta-turn conformation pattern by Garland and Dean (1. Computer-Aided Molecular Design, 1999, 13, 469) as one strategy of designing non-peptide beta-turn scaffolds. (omitted)
