Development of a Solubilization Product Containing Oleanolic Acid with Hydrogenated Lecithin (천연 유래 수첨 레시틴을 이용한 올레아놀릭산 가용화 제형 개발)

  • Recently, the number of customers who want natural cosmetics without petroleum originated cosmetic ingredients is increasing over the world, however, there are few natural cosmetics that claim functionality in the products. Because there are few functional cosmetic ingredients from nature and though so, it is very difficult to stabilize the final products manufactured by using those functional cosmetic ingredients in the long period. This study is on stabilization of oleanolic acid, insoluble anti-wrinkle cosmetic ingredients from nature, with hydrogenated lecithin in the solubilization product, aiming for developing a formulation of solubilization products which can be certified as organic cosmetics. In case of other synthetic solubilizers, they showed unstability at $45^{\circ}C$ within 2 ~ 3 days, however, for natural origin hydrogenated lecithin a stable product was made without particle size change during 4 weeks. The stability of oleanolic acid in the solubilization product was measured for 24 weeks at $25^{\circ}C$ and $40^{\circ}C$, both samples showed over 90 wt% value of the initial one, which can prove that oleanolic acid was stabilized in the product without any degradation. Also through clinical tests, the effect of moisturizing, anti-wrinkle, safety were confirmed.

Isolation of a New Agar Degrading Bacterium, Maribacter sp. SH-1 and Characterization of its Agarase (신규 한천분해세균 Maribacter sp. SH-1의 분리 및 효소 특성조사)

  • In this study, we isolated a new agar-degrading marine bacterium and characterized its agarase. An agardegrading marine bacterium SH-1 was isolated from seawater, collected from the seashore of Namhae in Gyeongnam province, Korea, and cultured in marine agar 2216 media. It was identified as Maribacter. sp. SH-1 by phylogenetic analyses, based on 16S rRNA gene sequence. The extracellular agarase was extracted from culture media of Maribacter sp. SH-1 and characterized. Its relative activities were 56, 62, 94, 100, and 8% at 20, 30, 40, 50, and 60℃, respectively, whereas 15, 100, 60, and 21% relative activities were observed at pH 5, 6, 7, and 8, respectively. Its extracellular agarase exhibited maximum activity (231 units/l) at pH 6.0 and 50℃, in 20 mM Tris-HCl buffer. Therefore, this agarase would be applicable as it showed the maximum activity at the temperature at which the agar is in a sol state. Furthermore, the agarase activities remained over 90% at 20, 30, and 40℃ after 0.5 h exposure at these temperatures. Thin layer chromatography analysis suggested that Maribacter sp. SH-1 produces extracellular β-agarase, as it hydrolyzes agarose to produce neoagarooligosaccharides, such as neoagarohexaose (34.8%), neoagarotetraose (52.2%), and neoagarobiose (13.0%). Maribacter sp. SH-1 and its β-agarase would be useful for the production of neoagarooligosaccharides, which shows functional properties, like skin moisturizing, skin whitening, inhibition of bacterial growth, and delay in starch degradation.

  • At present, coffee consumption amount is annually on the rise in Korea, which results in about 0.27 million tons of coffee waste annually. They are mostly classified as food waste and deserted with moisture contained, being a serious environmental issue. Existing slope greening techniques, which are vegetation based soil-media hydroseeding measures, have problems such as lack of coherence, dryness or lack of organic matters. Therefore in order to assess usability of Spent Coffee Ground (SCG), medium-to long-term growth test was conducted under the indoor and outdoor conditions. According to the result of growth test, when SCG was mixed with existing base materials, moisturizing power increased and organic matter content was reinforced, promoting germination and growth in a medium term. Among others, under the condition when supply of water was discontinued, withering rate was lower than existing base materials and diverse phenomena resulting from lack of nutrition decreased. Therefore, SCG as a waste organic matter with abundant nitrogen has the characteristic of inhibiting early growth but was found to have a quality favorable to long-term growth resulting from water containing ability and the supply of organic mater and is judged to be a material to replace or complement existing base materials.

Inhibitory effect of Mojaban (Sargassum fulvellum) extract on ultraviolet B-induced photoaging in hairless mouse skin (무모 마우스 피부에서 모자반 추출물의 자외선비(ultraviolet B)에 의해 유도된 광노화 억제 효과)

  • Ultraviolet radiation is a major cause of photoaging, characterized by wrinkle formation and skin dehydration. Mojaban (Sargassum fulvellum), belonging to the brown algae of the Sargassaceae family, exerts anti-inflammatory, antioxidant, and anti-atopic activities. Particularly, Mojaban extract alleviated ultraviolet B (UVB)-induced inflammatory responses in human keratinocytes and BALB/c mice. Here, the dorsal skin of hairless mice was exposed to UVB for photoaging. The mice were administrated 300 mg/kg Mojaban extract (S. fulvellum extract; SFE) daily. Phenotypically, SFE reduced wrinkle formation and epidermal thickness and increased skin elasticity. SFE increased hydroxyproline content by upregulating the expression of collagen-related genes and downregulating matrix metalloproteinase expression. Furthermore, SFE increased skin moisture content and inhibited transepidermal water loss. SFE upregulated the expression of involucrin, loricrin, and transglutaminase for cornified envelope formation as well as the expression of filaggrin and caspase-14 for natural moisturizing factors. Collectively, SFE may be employed as a nutraceutical ingredient for anti-photoaging.

The Skin Improvement Effect of Facial mask pack using Chickpea natto fermented with Bacillus subtilis natto (Bacillus subtilis natto 균주로 발효한 병아리콩 낫토를 이용한 mask pack의 피부 개선 효과)

  • In order to utilize health food chick bean as a raw material for cosmetics, the skin improvement effect of a mask pack made of chickpea Natto fermented with Bacillus subtilis Natto strain was studied. Powder obtained by lyophilization of chickpea Natto was applied to a facial mask pack as a paste formulation. This Natto pack was applied to subjects in their 50s and 60s to investigate their effect on skin improvement. As the number of treatments of Natto pack increased, the skin was improved and the improvement effect was remarkable after 4 treatments. After four times treatments, moisture content and sebaceous secretion increased by $8.4{\pm}3.6%p$ and $4.0{\pm}2.3%p$, respectively. Skin pores and wrinkles were also decreased by $1.8{\pm}0.3%p$ and $1.8{\pm}0.9%p$, respectively. Skin pigmentation decreased by $1.3{\pm}0.2%p$ and skin tone also increased from 55.2% to 55.9%. These results indicate that Natto product obtained by fermenting chickpea has the effect of improving skin such as moisturizing, pore, wrinkle, pigmentation, sebum secretion and skin tone, and can be used as a raw material for various functional cosmetics.

The physical properties of the cosmetic hydrogels affected by adding various celluloses (셀룰로오즈 첨가에 따른 미용용 하이드로겔의 물성 연구)

  • Hydrogels are natural polymer networks that can contain huge quantities of water and many cosmetical ingredients. Their hydrophilic functional groups creates a matrix, which allows high efficacy in delivering active ingredients into the skin. In industry, hydrating properties and strength of the hydrogels are of great interest in manufacturing hydrogel mask packs. We have used the cellulose in various forms such as powder, cotton fiber and cellulase treated cotton fiber to investigate the property changes of cellulose/hydrogel sheets. When 0.1% and 0.3% of cellulose powder were added to hydrogels, tensile strength of hydrogel sheets were decreased by 10% and 14% respectively. Vise versa, when 0.5 ~ 2 cm of cotton fibers were added, tensile strength of hydrogel sheets were significantly increased by about 20%. The hydrogels which contain cotton fibers also gave an excellent moisturizing effect. Especially cellolose/hydrogels containing cellulase-treated cotton fibers showed the best effect on retaining moisture content increasing upto 380% in comparison with the one containing untreated cotton as well as excellent dispersibility.

Cosmeceutical Properties of Extracts of Torreya nucifera and Alpinia henryi and Formulation Characteristics of Mask Pack Containing Extracts of These (비자 및 초두구 추출물의 코스메슈티칼 특성 및 이를 포함한 마스크팩 제형특성)

  • The extracts of antioxidants, torreya nucifera and alphinia henryi, were tested for properties as a fragrance material and applied to a mask pack formulation to study the fragrance properties. The DPPH antioxidant test of hot water and ethanol extract confirmed that the ethanol extract had superior antioxidant efficacy compared to the hot water extract. It was confirmed that the optimal mixing ratio as a raw material for the mask pack of torreya nucifera and alphinia henryi was 3:7 as a result of the DPPH antioxidant test. As a result of the cytotoxicity test, the cell viability was good as it showed 103.30% at 0.5 ug/mL, 104.25% at 1 ug/mL, 102.56% at 5 ug/mL, and 99.17% at 10 ug/mL compared to the untreated group. As a result of the patch test on the mask pack formulation, the skin irritation index of 0.02 was judged as a non-irritation product in the skin irritation primary stimulation human application test. In the evaluation of skin moisturizing, it showed a significant increase rate of 19.178% compared to before the sample adaptation. Evaluation of the change over time in the sheet mask pack formulation confirmed the formulation stability without viscosity and pH change for 12 weeks at low temperature(4℃), room temperature(25℃), and high temperature(45℃).

A Study on Synthesis of Organic Plant Surfactant and Its Solubilizing Action on Bergamot Oil (유기농 식물성 계면활성제의 합성과 베르가못오일에 대한 가용화력에 관한 연구)

  • The study is on the cosmetic solubilizing power of organic plant surfactants. The blended high purity polyglyceryl-10 oleate and polyglyceryl-10 stearate mixtures were synthesized using organically certified raw materials to develop surfactants having excellent solubilizing power. The mixture is called "Solubil ORG-1300". The appearance of this material is a pale yellowish paste, with a specific odor. The specific gravity was 1.12 and it was high purity that acid value was 0.072±0.1. The HLB value of this natural surfactant was averaged = 15.1 and calculated through the Griffin equation. Mechanically it is explained how organic surfactant are available with fragrance and oils. The solubilizing test was determined by eye evaluation method through the dissolving performance test for the two oils and measured the transmittance at 890 nm using a UV spectrophotometer to measure the transparency. The results showed that the concentration of surfactant needed to make Bergamot oil available requires approximately more 2 times. It was also found that the concentration of surfactant needed to make the tocoperyl acetate available was about 8 times higher. Experiments on the solubility resulting from pH changes showed stabilized usable solubilizing power even in acidic areas of pH=3.5, neutral areas of pH=7.2, and alkaline areas of pH=1.5. Experiments on the solubility according to pH variation showed good solubility stabilized in acidic areas of pH=3.5, neutral areas of pH=7.2, and alkaline areas of pH=11.5. As an application of cosmetics, the company successfully developed a prescription for moisturizing activity based on these results, it is expected that a wide range of applications will be available for skin care, baby lotion, sensitivity or atopic skin cosmetics.

Natural Oleosomes Loading Emulsion Technology -Loading Oleosomes as Delivery Systems for Improved Cosmetic Efficacies- (천연 Oleosomes를 이용한 에멀젼 기술)

  • Natural safflower oleosomes are small ($1{\sim}3{\mu}m$) spherical shaped "reservoir", inside which the seed stores triglycerides for use as a future energy source. The surface of the oleosome is covered with a high molecular weight ($20{\sim}24$ KDa) oleosin protein which has been demonstrated to have emulsification properties. Traditionally, oleosomes from oil bearing seeds such as safflower were simply crushed to liberate the oil within. Our patented DermaSphere technology allows for the isolation of oleosomes in the intact state. Once isolated, these materials can be used in skin care formulations to deliver the emolliency, occlusivity, and anti-oxidant effects typically associated with safflower oil. However, because of the presence of the emulsifying oleosin protein covering the spherical oil body, oleosomes have self-emulsification property as well as can emulsify other oil phase in typical oil-in-water (O/W) emulsion. The oleosomes can literally serve as the entire non-active portion of the oil phase of a typical skin care product. Most importantly, natural oleosomes can be loaded with other oil-soluble active materials and can therefore be used as delivery systems for improved cosmetic efficacies. Oleosomes can be loaded with various actives, such as fragrances, vitamins, inset repellents, and UV chromophores. The loaded oleosomes can be utilized to either protect the active ingredients within the formulation itself or to allow for control release of those actives over time.

A Study on Utilization of Waste Organic Matter for Slope Protection (비탈면보호를 위한 폐유기물질의 활용성에 대한 연구)

  • Coffee consumption in Korea has been currently growing every year, and as a result, approximately 0.2 million tons of Spent Coffee Grounds (SCG) are being created every year. SCG, which is waste organic material, is often classified as food waste and an annual amount of 0.27 million ton is discarded while containing moisture and provoking serious environmental issue. Physico-chemical characteristics of SCG were analyzed in this study and medium and long-term growth experiments were conducted in order to evaluate its utilization potential. According to the experiment results, mixing SCG into the previous base material resulted in accelerated germination and growth in the mid-term compared to previous base material alone, despite slower germination or growth in early stage. Especially, it showed lower withering rate and decrease in various symptoms that are caused by nutrition shortage in case of discontinued sprinkling, etc., compared to the previous base material. Hence, while SCG has a feature of hindering early development due to its feature of waste organic material that is rich in nitrogen, its benefit for long-term growth coming its moisturizing ability and supply of organic matter was confirmed in the study. On balance, SCG is believed to be a material that can replace or complement the previous base materials.
