The Role of PK/PD Modeling and Simulation in Model-based New Drug Development (모델 기반학적 신약개발에서 약동/약력학 모델링 및 시뮬레이션의 역할)

  • In the recent, pharmacokinetic (PK)/pharmacodynamic (PD) modeling has appeared as a critical path tools in new drug development to optimize drug efficacy and safety. PK/PD modeling is the mathematical approaches of the relationships between PK and PD. This approach in new drug development can be estimated inaccessible PK and PD parameters, evaluated competing hypothesis, and predicted the response under new conditions. Additionally, PK/PD modeling provides the information about systemic conditions for understanding the pharmacology and biology. These advantages of PK/PD model development are to provide the early decision-making information in new drug development process, and to improve the prediction power for the success of clinical trials. The purpose of this review article is to summarize the PK/PD modeling process, and to provide the theoretical and practical information about widely used PK/PD models. This review also provides model schemes and the differential equations for the development of PK/PD model.

Knowledge-Based Model for Forecasting Percentage Progress Costs

  • This study uses a hybrid estimation tool for effective cost data management of building projects, and develops a realistic cost estimation model. The method makes use of newly available information as the project progresses, and project cost and percentage progress are analyzed and used as inputs for the developed system. For model development, case-based reasoning (CBR) is proposed, as it enables complex nonlinear mapping. This study also investigates analytic hierarchy process (AHP) for weight generation and applies them to a real project case. Real case studies are used to demonstrate and validate the benefits of the proposed approach. By using this method, an evaluation of actual project performance can be developed that appropriately considers the natural variability of construction costs.

  • The attempt to use a 3D model each field such as design, structure, construction, facilities, and estimation in the construction project has recently increased more and more while BIM (Building Information Modeling) that manages the process of generating and managing building data has risen during life cycle of a construction project. While the 2D Drawing based work of each field is achieved in the already existing construction project, the BIM based construction project aims at accomplishing 3D model based work of each field efficiently. Accordingly, the solution that fits 3D model based work of each field and supports plans in order to efficiently accomplish the relevant work is demanded. The estimation, one of the fields of the construction project, has applied BIM to calculate quantity and cost of the building materials used to construction works after taking off building quantity information from the 3D model by a item for a Quantity Take-off grouping the materials relevant to a 3D object. A 3D based estimation program has been commonly used in abroad advanced countries using BIM. The program can only calculate quantity related to one 3D object. In other words, it doesn't support the take-off process considering quantity of a contiguous object. In case of temporary materials used in the frame construction, there are instances where quantity is different by the contiguous object. For example, the formwork of the temporary materials quantity is changed by dimensions of the contiguous object because formwork of temporary materials goes through the quantity take-off process that deduces quantity of the connected object when different objects are connected. A worker can compulsorily adjust quantity so as to recognize the different object connected to the contiguous object and deduces quantity, but it mainly causes the confusion of work because it must complexly consider quantity of other materials related to the object besides. Therefore, this study is to propose the solution that automates the formwork 3D modeling to efficiently accomplish the quantity take-off of formwork by preventing the confusion of the work which is caused by the quantity deduction process between the contiguous object and the connected object.

  • This study aims at improving work reliability. It proposes a way to overcome the limitations of current scheduling methods by providing a new framework, CPAM(Construction Process Analysis Model) based on the lean principles. It suggests methods which improve work reliability and production effectiveness with variability control methods. Also it suggests methods which reduce inventories of materials and equipment and WIP(Work In Process) using two techniques; Lookahead Schedule and Weekly Work Plan. The contribution of this research also includes that it assumes planning as a process of reducing uncertainty and maximizing throughput, counter-posing plan reliability to resource redundancy as alternative strategies for managing in conditions of uncertain work flow.

  • Asbestos is a durable and heat-resistant building material used in various building materials such as slate, ceiling tex, and spray paint. It has been banned since 2009 after found to be a first-class carcinogen that causes various cancers and asbestos lung disease. Since workers are likely to be exposed to asbestos in the process of dismantling and removal of asbestos-made building materials and facilities, laws and work standards are proposed by the Ministry of Employment and Labor to ensure the safety of asbestos dismantling work. In addition, prior studies on exposure levels and analysis methods have been conducted in this regard mainly for residents. However, the relation between the results of the risk assessment of the process conducted during the asbestos investigation and the work is still ambiguous for the safety of workers. Therefore, this study proposes a process model development methodology that considers work risk based on the results of a survey from asbestos dismantling companies.

A Decision Support Systems Design for Process Control (공정통제용 의사결정지원 시스템)

  • This paper deals with the case analysis of second order processes under sampled-data. Proportional Integral-Derivative(PID) control, and development of Decision Support Systems(DSS) for such processes. In this paper three techniques were described for identifying the dynamics of closed loop stable processes. The first, called pulse testing is a frequency-domain method, which yields the frequency response diagram of an open loop process. The second is a time-domain method which yields the gain and time constants of the process model. The third technique is based on step response and gives the parameters of PID controllers. The development of DSS design programs consisting of above three techniques will provide very powerful tools in the microcomputer based process control.

  • Sensorless electric superchargers have recently been actively developed to provide a large amount of oxygen to engines in order assist the combustion process for miniaturizing the engines and improving fuel efficiency. The model-based sensorless method for surface-mounted permanent magnet synchronous motors has a disadvantage in that the system may become unstable due to parameter variations in low-speed operation and the rapid-acceleration section. An electric supercharger requires fast response to improve the engine response delay, such as the turbocharger turbo-rack. Therefore, the responsiveness must be improved to use the model-based sensorless system. The position compensation algorithm designed in this study is controlled by converting the position error into the beta, which is the angle formed by the d-axis and the stator current during sudden speed change. In this study, we improved the response of the model-based sensorless system through the algorithm and verified the algorithm validity by applying the algorithm to an actual dual-motor supercharger.

A Design of Development Process Model of Product Lines for Developing Embedded Software (임베디드 소프트웨어 개발을 위한 제품계열 중심의 개발프로세스 모델 설계)

  • Recently, the requirements of the embedded software are getting diverse as the diversity of embedded software application fields increases. The systematic development methods are issued to deal with the dependency between hardware and software. However, the existing development methods have not considered the software's close connection to hardware and the high-level reusability for common requirements of several similar domains. In this paper, we propose a design method of development process model of product lines to support an efficient development method for embedded software. For this, we firstly suggest a domain scoping method and an IDEF0(Integration DEFinition)-based business model for extracting the efficient requirements. Next, we present a component deriving method based on the service architecture and an architecture design method after considering the hardware dependency. And we explain the artifacts of MSDFS(Multi Sensor Data Fusion System) at each design step in order to show how the proposed model can be applied to the embedded software development.

  • Use case diagram is used for defining inter-action between users and systems in object-oriented modeling. It is very important to defining users' requirements for efficient software development. In this paper, we propose a object-oriented modeling process using hierarchical use case analysis for designing class model. First, We define many use case diagrams by several hierarchical modeling level. And next, we can also design class model using the use case diagrams. Our proposed modeling process provides interaction between use case model and class model. That can make us to check the modeling process during the software development. Using the proposed object-oriented modeling we can develop software based on users' requirements. It is very useful for class modeling.
