Study of Interrelationship in Five Mimic-Animal Boxing and Viscera (방생도인(仿生導引) 오금희(五禽戱)와 장부(臟腑)와의 상관성 연구)

  • Five mimic-animal boxing(五禽戱, FAB) is physical fitness exercises conducted by imitating the motions of tiger, deer, bear, monkey, and bird, devised by the famous doctor Hwa-Ta(華陀) in the Three Kingdoms of China. FAB is one of bionic-physical and breathing exercises(傍生導引) by combined automassage as a practice for health care. Although FAB have effects of health control and medical treatment, but interrelationship of five animals(五禽) of FAB and five viscera(五臟) of five elements(五行) is complicated. So, the present was designed to investigate the progress of FAB, movement-feature of FAB, and the interrelationship of five animals and five viscera. further I will develop value by sports-treatment medicine in oriental medicine. The results were as follows ; FAB is physical exercises of dynamic exercises and new-dynamic exercises. Feature of China-FAB have each movement by imitating the motions of each animals, movements of Formosa-FAB were mixed. In philosophical viewpoint, Five animals (tiger, deer, bear, monkey, bird) were attached to metal, wood, water, earth, fire of five elements, and lung, liver, kidney, spleen, heart of five viscera. In viewpoint of breathing exercises, Five animals (tiger, deer, bear, monkey, bird) were attached to water, wood, earth, fire, metal of five elements, and kidney, liver, spleen, heart, lung of five viscera. Although FAB have effects of health control and medical treatment, FAB have indication and contraindication. Therefore, I think that mechanism of treatment should reason with Oriental medicine doctor in FAB.

  • The representation of shamanism or shaman in Korean cinema has changed according to eras. Since the 2000s, shamanism and shaman appear more frequently than before as the main subjects and characters in movies. The full-scale exploration of shamans usually takes places in documentary works, such as <'Mudang'>(2002), (2006) and (2011). Shamans are summoned in more various appearences in feature films. In this article, we will compare and analyze the representations of shamanism and shaman in recent Korean occult genre films. (2015), (2016), and (2017) are works that utilize new subjects and visual effect styles that were difficult to see in Korean horror films before. In these three films, the meaning of shaman and gut shows distinct differences from each others. Through the analysis of these films, we would like to explore the characteristics and possibilities of Korean occult films.

  • The aesthetics of Korean pantomimic mask-dance-drama is studied in this thesis. The scripts of the pantomimic mask-dance-drama were recorded generally, explanatorily and interpretively. The emotion and psychology are expressed by the look and body language. The mimic expression of the pantomimic mask-dance-drama was analyzed intensively. The speech drama is richer and deeper than the pantomimic mask-dance-drama at the aspect of the power of expression.

Hsa-miR-422a Originated from Short Interspersed Nuclear Element Increases ARID5B Expression by Collaborating with NF-E2

  • MicroRNAs (miRNAs) are a class of small non-coding RNAs that regulate the expression of target messenger RNA (mRNA) complementary to the 3' untranslated region (UTR) at the post-transcriptional level. Hsa-miR-422a, which is commonly known as miRNA derived from transposable element (MDTE), was derived from short interspersed nuclear element (SINE). Through expression analysis, hsa-miR-422a was found to be highly expressed in both the small intestine and liver of crab-eating monkey. AT-Rich Interaction Domain 5 B (ARID5B) was selected as the target gene of hsa-miR-422a, which has two binding sites in both the exon and 3'UTR of ARID5B. To identify the interaction between hsa-miR-422a and ARID5B, a dual luciferase assay was conducted in HepG2 cell line. The luciferase activity of cells treated with the hsa-miR-422a mimic was upregulated and inversely downregulated when both the hsa-miR-422a mimic and inhibitor were administered. Nuclear factor erythroid-2 (NF-E2) was selected as the core transcription factor (TF) via feed forward loop analysis. The luciferase expression was downregulated when both the hsa-miR-422a mimic and siRNA of NF-E2 were treated, compared to the treatment of the hsa-miR-422a mimic alone. The present study suggests that hsa-miR-422a derived from SINE could bind to the exon region as well as the 3'UTR of ARID5B. Additionally, hsa-miR-422a was found to share binding sites in ARID5B with several TFs, including NF-E2. The hsa-miR-422a might thus interact with TF to regulate the expression of ARID5B, as demonstrated experimentally. Altogether, hsa-miR-422a acts as a super enhancer miRNA of ARID5B by collaborating with TF and NF-E2.

Production of Selenium Peptide by Autolysis of Saccharomyces cerevisiae

  • Selenium-containing peptide (selenium peptide) was produced by autolysis of total proteins of Saccharomyces cerevisiae grown with inorganic selenium. Selenium peptide exhibited antioxidant activity as a glutathione peroxidase (GPx) mimic, and its activity was dependent on the hydrolysis methods. The GPx-like activity of the hydrolyzed selenium peptide increased 2.7-folds when digested by protease, but decreased by acid hydrolysis. During the autolysis of the yeast cell, the GPx-like activity and selenium content increased 4.3- and 2.3-folds, respectively, whereas the average molecular weight (MW) of selenium peptide decreased 70%. The GPx-like activity was dependent on the MW of selenium peptide and was the highest (220 U/mg protein) at 9,500 dalton. The maximum GPx-like activity (28,600 U/g cell) was obtained by 48 h of autolysis of the cells, which were precultured with 20 ppm of selenate. Selenium peptide showed little toxicity, compared with highly toxic inorganic selenium. These results show the potential of selenium peptide as a nontoxic antioxidant that can be produced by simple autolysis of yeast cells.

  • A ganglion cyst contains clear high viscous fluid within dense fibrous connective tissue wall communicating with joint or tendon sheath, but is not frequent around the knee joint. A hard mass at the medial side of the knee joint of the 73 years old woman, mimic to hard tumor mass was diagnosed as the ganglion using ultrasonography. Ultrasonography was a useful diagnostic tool of the cystic lesion although the consistency of the mass was hard enough to be suspected as a tumor mass.

Crystal Structure and Biochemical Characterization of Xylose Isomerase from Piromyces sp. E2

  • Biofuel production using lignocellulosic biomass is gaining attention because it can be substituted for fossil fuels without competing with edible resources. However, because Saccharomyces cerevisiae does not have a ${\text\tiny{D}}$-xylose metabolic pathway, oxidoreductase or isomerase pathways must be introduced to utilize ${\text\tiny{D}}$-xylose from lignocellulosic biomass in S. cerevisiae. To elucidate the biochemical properties of xylose isomerase (XI) from Piromyces sp. E2 (PsXI), we determine its crystal structure in complex with substrate mimic glycerol. An amino-acid sequence comparison with other reported XIs and relative activity measurements using five kinds of divalent metal ions confirmed that PsXI belongs to class II XIs. Moreover kinetic analysis of PsXI was also performed using $Mn^{2+}$, the preferred divalent metal ion for PsXI. In addition, the substrate-binding mode of PsXI could be predicted with the substrate mimic glycerol bound to the active site. These studies may provide structural information to enhance ${\text\tiny{D}}$-xylose utilization for biofuel production.
