본 논문에서는 개발된 범용 분자 시뮬레이션 시스템에 대해 기술하고자 한다. 본 연구에서 개발된 분자 시뮬레이션 시스템의 가장 큰 장점은 다른 무엇보다도 Langevin dynamics simulation이나 dissipative particle dynamics simulation 기법을 도입하여 all-atom 모델뿐만 아니라coarse-grain 모델까지도 다룰 수 있도록 설계하였고 따라서 미시영역은 물론 중간영역에서 일어나는 현상까지도 시뮬레이션 할 수 있도록 설계하였다는 점이다. 이를 통해 하나의 통합된 분자 시뮬레이션 시스템으로 생체막 내에서 마취제의 분포, 단백질 접힘 현상, 마이크로 채널 내에서 생체고분자의 구조와 유동 특성 등과 같이 미시영역에서부터 중간영역에 이르는 다양한 현상을 연구할 수 있게 되었다 개발된 시스템을 이용하면 molecular dynamics simulation에 기반한 분자 시뮬레이션 시스템으로는 불가능한 여러 중요한 바이오/나노 시스템을 시뮬레이션 할 수 있을 것으로 기대한다 마지막으로 벤치마크 결과를 통해 개발된 분자 시뮬레이션 시스템의 성능을 측정하였고 성능 최적화를 위한 병목지점을 조사하였다.

The Fluorescence-Activated Cell Sorter (FACS) is a well-established instrument used for identifying, enumerating, classifying and sorting cells by their physical and optical characteristics. For a miniaturized FACS device, a disposable plastic microchip has been developed which has a hydrodynamic focusing chamber using soft lithography. As the characteristics of the spatially confined sample stream have an effect on sample throughput, detection efficiency, and the accuracy of cell sorting, systematic fluid dynamic studies are required. Flow visualization is conducted with a laser scanning confocal microscopy (LSCM), and three-dimensional flow structure of the focused sample stream is reconstructed from 2D slices acquired at $1\mutextrm{m}$ intervals in depth. It was observed that the flow structure in the focusing chamber is skewed by unsymmetrical velocity profile arising from trapezoidal cross section of the microchannel. For a quantitative analysis of a microscopic flow structure, Confocal Micro-PIV system has been developed to evaluate the accelerated flow field in the focusing chamber. This study proposes a method which defines the depth of the measurement volume using a detection pinhole. The trajectories of red blood cells (RBCs) and their interactions with surrounding flow field in the squeezed sample stream are evaluated to find optimal shape of the focusing chamber and fluid manipulation scheme for stable cell transporting, efficient detection, and sorting
A syringe pump or a device using high electric voltage has been used for controlling flows inside a LOC (lab-on-a-chip). Compared to LOC, however, these microfluidic devices are large and heavy that they are burdensome for a portable ${\mu}-TAS$ (micro total analysis system). In this study, a new flow control technique employing pressure regulators and pressure chambers was developed. This technique utilizes compressed air to control the micro-scale flow inside a LOC, instead of a mechanical actuator or an electric power supply. The pressure regulator controls the output air pressure by adjusting the variable resistor attached. We checked the feasibility of this system by measuring the flow rate inside a capillary tube of $100{\mu}m$ diameter in the Re numbers ranged from 0.5 to 50. In addition, the performance of this flow control system was compared with that of a conventional syringe pump. The developed flow control system was found to show superior performance, compared with the syringe pump. It maintains automatically the: air pressure inside a pressure chamber whether the flow inside the capillary tube is on or off. Since the flow rate is nearly proportional to the resistance, we can control flow in multiple microchannels precisely. However, the syringe pump shows large variation of flow rate when the fluid flow is blocked in the microchannel.
Agglutination is one of the most commonly employed reactions in clinical diagnosis. In this paper, we have designed and fabricated nickel mold insert for injection molding of a microfluidic lab-on-a-chip for the purpose of the efficient detection of agglutination. In the presented microfluidic lab-on-a-chip, two inlets for sample blood and reagent, flow guiding microchannels, improved serpentine laminating micromixer(ISLM) and reaction microwells are fully integrated. The ISLM, recently developed by our group, can highly improve mixing of the sample blood and reagent in the microchannel, thereby enhancing reaction of agglutinogens and agglutinins. The reaction microwell was designed to contain large volume of about $25{\mu}l$ of the mixture of sample blood and reagent. The result of agglutination in the reaction microwell could be determined by means of the level of the light transmission. To achieve the cost-effectiveness, the microfluidic lab-on-a-chip was realized by the injection molding of COC(cyclic olefin copolymer) and thermal bonding of two injection molded COC substrates. To define microfeatures in the microfluidic lab-on-a-chip precisely, the nickel mold inserts of lab-on-a-chip for the injection molding were fabricated by combining the UV photolithography with a negative photoresist SU-8 and the nickel electroplating process. The microfluidic lab-on-a-chip developed in this study could be applied to various clinical diagnosis based on agglutination.
The organic light emitting diode (OLED) is proposed as the novel source in the microchip because it has ideal merits (various wavelengths, thin-film structure and overall emitting) for the integration. In this paper, we fabricated the finger-type pin photodiodes for fluorescence detection and the advanced microchip with OLED integrated pn the microchannel. The finger-type in the diode design extended the depletion region and reduced the internal resistance about 31.2% than rectangular-type. The photodiodes had a 100pA leakage current and a 8720 sensitivity $(I_{Light}/I_{Dark})$ at -1 V bias. The interference filter with 32 layers ($SiO_2$, $TiO_2$) was directly deposited on the photodiode. The OLED was fabricated on the ITO coated glass and was bonded with LOC. The application of thin-film OLED increased the excitation efficiency and simplified the integration process extremely. The prototype device of this application had a superior sensitivity of 100nM-LOD in the fluorescence detection.
본 논문에서는 다수의 전기장 분포가 생성되는 단일 미세유로를 이용한 폐암세포 전기천공 및 활성도 분석칩을 제안하였다. 종래의 세포 전기천공 분석칩은 다수의 전기장 분포를 형성하기 위해 다수의 전극패턴 또는 다수의 미세유로를 필요로 하여 구조가 복잡하였다. 반면, 제안된 세포 전기천공 및 활성도 분석칩은 한 쌍의 전극 사이에서 계단 형상으로 폭이 변화하는 단일 미세유로를 이용하여 다수의 전기장 분포를 형성함으로써 간단한 구조로 세포 전기천공 및 활성도를 분석할 수 있다. 제안된 세포 전기천공 및 활성도 분석칩은 0.3kV/cm에서 0.5kV/cm까지 5단계의 전기장이 발생되도록 설계하였다. A549와 H23의 두 종류의 비소세포 폐암세포주를 이용한 성능실험 결과, 활성을 유지하면서 전기천공된 세포의 비율이 0.4kV/cm의 전기장에서 각각 $26.6{\pm}0.7%$ 및 $51.4{\pm}3.0%$로 가장 높은 값을 보였다. 제안된 세포 전기천공 및 활성도 분석칩은 세포의 형질주입 연구를 위한 집적화된 세포칩으로 응용될 수 있다.
In droplet-based microfluidic systems, in-droplet preconcentration of a sample is one of the important prerequisites for biochemical or medical analysis. There have been a few studies on preconcentration in a moving droplet, but they are limited to practical applications since 1) their method are time-consuming or 2) they require specific properties such as electric and magnetic properties. In this study, we demonstrated the position control of polystyrene particles of 5 and $10{\mu}m$ in diameter inside a moving water-in-oil droplet using traveling surface acoustic waves. Since the frequencies for effective control of each diameter were found, microparticles with no labels could be utilized. In addition, the proposed method enabled on-demand preconcentration inside a polydimethylsiloxane microchannel. In-droplet preconcentration of microparticles was realized by splitting a mother droplet with manipulated particles at a downstream bifurcation zone. Given these advantages, the proposed system is a promising acoustofluidic lab-on-a-chip platform for preconcentration inside a droplet.
사각공동 및 채널이 형성된 마이크로구조 표면을 활용하여 수조비등 임계열유속에 대한 중력 및 모세관 압력의 영향에 대해 연구하였다. 마이크로 공동구조는 모세관 압력에 의한 액체유동을 억제하는 역할을 하였고, 마이크로 채널구조는 비등표면으로의 1차원적인 액체유동을 유발하는데 기여하였다. 이를 통해 임계열유속과 모세관 유동의 상관관계를 정량화할 수 있었다. 비등표면으로의 액체공급을 위한 원동력은 중력수두 및 모세관 압력에 의해 유발될 수 있다. 본 연구에서는 수조비등 실험 및 가시화 데이터의 분석을 통해 수조비등 표면에서의 핵비등을 유지할 수 있는 액체공급은 중력 수두 및 모세관 압력과 밀접한 상관관계를 가지고 있음을 확인할 수 있었다.
본 연구에서는 직경이 수백 nm로부터 수 ${\mu}m$에 이르는 균일한 크기의 액적을 생성하는 마이크로 플루이딕 플랫폼이 설계되었다. 미세한 액적을 생성하기 위하여 T-정션과 유동집속 장치가 마이크로�a푸이딕 채널로 통합되었다. 상대적으로 큰 수성 액적들이 상류의 T-정션에서 생성되어 유동집속 장치로 이송되는데, 여기에서 각각의 액적은 압력과 점성응력의 작용에 의하여 목표로 하는 크기로 잘게 쪼개진다. 이러한 구성은 내부 유체의 매우 느린 유량과 유동집속 영역에서 내부 및 외부 유체 사이의 높은 유량비를 가능하게 한다. 본 마이크로플루이딕 장치는 약 $1\;{\mu}m$ 크기의 직경을 가지는 액적들을 3%보다 작은 표준 편차로 생성할 수 있음이 제시되었다.
$500{\mu}m$의 수력직경을 가진 마이크로 채널에서 유동 비등 시 물에 대한 마찰 압력 강하를 측정하기 위한 실험적 연구를 수행하였다. 실험은 열 유속 $100-400kW/m^2$, 증기건도 0-0.2 그리고 질량 유속 $200-600kg/m^2s$의 범위에서 이루어졌다. 유동 비등 시 마찰 압력 강하는 두 가지 모델을 사용하여 예측된다. 즉, 두 상의 속도가 동일하다고 가정한 균질 모델과 두 상 사이에 서로 다른 속도를 가지는 분리류 모델로 분류된다. 실험결과 이상 마찰 승수는 질량 유속이 증가함에 따라 감소한다는 것을 알 수 있었다. 측정된 압력 강하 데이터는 매크로 스케일과 미니/마이크로 스케일에서 제안된 기존의 여러 상관식들과 비교하였다. 균질 모델은 본 연구에서 고려한 실험 조건에서 29.4 %의 평균 오차내에서 마찰 압력 강하를 예측하였다.
