Bacillus species were isolated from swine manure to develope the microbial additive suitable for the rapid com-posting. The 3 of 4 isolated strains were identified as Bacillus cereus KD-2, B. pumilus KD-3, and B. licheni-formis KD-4. Bacillus sp. KD-1 was, however, not highly identical with any Bacillus sp. The isolated strains were analyzed their growth rates, enzyme activities, and antibacterial activities. The maximum growth tem-peratures of KD-1, KD-2, KD-3 and KD-4 were $45^{\circ}C$, $50^{\circ}C$, $53^{\circ}C$, and $55^{\circ}C$, respectively. The activities of pro-tease or amylase in mixed culture of 4 strains were similar in the range of $37^{\circ}C$ to $53^{\circ}C$ and activities of lipase in the range of $37^{\circ}C$ to $42^{\circ}C$ were twice higher than those of lipase in the range of $47^{\circ}C$ to $53^{\circ}C$. The antibacterial activity of KD-l, KD-2, or KD-3 against each other was not detected. That of KD-4 against KD-1, KD-2, or KD-3 was, however, detected. The organic compound and C/N ratio of compost fermented by the mixed culture were determined as 61.9% and 22.4%, respectively. The concentration of the ammonia gas was 12.35 mg/l in the liquid compost.

Biocatalytic modification of natural resources can be used to generate novel compounds with specific properties, such as higher viscosity and reactivity. The production of hydroxy fatty acids (HFAs), originally found in low quantities in plants, is a good example of the biocatalytic modification of natural vegetable oils. HFAs show high potential for application in a wide range of industrial products, including resins, waxes, nylons, plastics, lubricants, cosmetics, and additives in coatings and paintings. In a recent study, Pseudomonas aeruginosa strain PR3 was used to produce 7,10-dihydroxy-8(E)-octadecenoic acid (DOD) from oleic acid. This present study focused primarily on the utilization of three natural nut oils obtained from the Philippines -pili nut oil (PNO), palm oil (PO), and virgin coconut oil (VCO)- to produce DOD by P. aeruginosa strain PR3. Strain PR3 produced DOD from PNO and PO only, with PNO being the more efficient substrate. An optimization study to achieve the maximum DOD yield from PNO revealed the optimal incubation time and medium pH to be 48 h and 8.0, respectively. Among the carbon sources tested, fructose was the most efficiently used, with a maximum DOD production of 130 mg/50 mL culture. Urea was the optimal nitrogen source, with a maximum product yield of 165 mg/50 mL culture. The results from this study demonstrated that PNO could be used as an efficient substrate for DOD production by microbial bioconversion.

The aims of this study were to compare the biomethanation of CO2 through specific methanogenic activity (SMA) test which was inoculated with four different types of mixed microbial culture obtained from full-scale anaerobic digestion (AD) plants. The experimental results showed that CH4 conversion was the highest in the samples inoculated by seed sludge taken from ADs of food waste and brewery; under this condition, the produced biomethane contains 89.3-91.9% of CH4. Meanwhile, the lowest level was obtained in the sample from sewage sludge. The measured ratio of CH4 production rate to CO2 consumption rate in all reactors was higher than the theoretical value (1) in the middle of the period and soon dropped to 0.7-0.8. It might be due to changed metabolic pathways in the reactor by the degradation of residual organic matter and the increased activity of homoacetogenic bacteria.

Bovine mastitis is an important microbial disease in the dairy industry. We investigated the frequencies of bacterial pathogens in 62 farms and pathogen antibiotic resistance from mastitis samples (n = 748). We tested the antimicrobial activity of chicken and duck egg white and lysozyme purified from chicken egg white. Moreover, we compared the microbiomes of normal and mastitic raw milk obtained by 16S rRNA gene pyrosequencing and culture methods. The results showed that the frequencies of Gram-positive pathogens (Enterococcus faecalis 37% and Staphylococcus aureus 36%) were higher than that of a Gram-negative pathogen (Escherichia coli 15%). Resistance frequencies to ampicillin and norfloxacin were lowest in Staphylococcus aureus (21%), Enterococcus faecalis (23%), and Escherichia coli (33%), and the antimicrobial activity of chicken egg white was higher than those of lysozyme and duck egg white. Pyrosequencing results revealed clear differences between the microbiomes of mastitic and normal raw milk samples and revealed a slightly similar, but clearly different, composition of pathogens compared to that from the culture method. Thus, pyrosequencing may be useful for elucidating changes in microbiomes during mastitis progression and treatment. A chicken egg white and antibiotic combination may help with mastitis treatment; however, further studies are needed.
This study was conducted to investigate the changes in the qualify of gamma-irradiated seasoned cuttle during storage. Seasoned cuttle packed in PVC film (0.06 win) was stored at $15{\pm}1^{\circ}C$ for 6 months after treatment with doses of 0 to 7 kGy. Microbial populations of seasoned cuttle were $1.6{\times}10^{5}CFU/g$ in total aerobic bacteria, $10^{4}{\sim}10^{5}CFU/g$ in yeasts &molds, and negative in coliforms, which were effectively reduced by 3 kCy or higher up to the undetectable level(<20 CFU/g). The pH and moisture content of the samples were not changed with irradiation, but moisture was some decreased during storage. The instrumental color (especially Hunter b value), pigments (lipophilic &hydrophilic) and TBA value of the samples increased with storage time as well as irradiation dose more than 3 kGy. The influence of storage condition, however, were more significant. Irradiation did not induce any changes in volatile basic nitrogen (VBN) and trimethylamine (TMA) contents, thereby maintaining their contents lower than those of the non-irradiated samples during storage by reducing the microbial load.
The effects of adding hull powders of black soybean on quality characteristics of tobu were studied. Proximate composition of the black soybean hull powders was crude ash, crude protein, crude fat, and carbohydrate at 2.60, 11.35, 2.18, and 73.97%, respectively. The addition of 5% black soybean hull powder decreased the production of whey and consequently increased the yield of tobu. Texture properties of evaluation hardness, adhesiveness, chewiness were the best in tobu which was made with 5% black soybean hull powders. Sensory evaluation showed texture, firmness were not significantly different among tobu with yellow soybean, black soybean, and added to 0.5% black soybean hull powders. At the beginning of the period of storage, there were not differences of microbial cell count as increasing black soybean hull powders, but observed after passing 5 days. The results of total microbial cell count showed that adding 5% black soybean hull powders in processing tobu extend storage time.

To evaluate microbial data and salivary measurements from clinically compatible, culture-based screening procedures employed with children younger than 36 months old. Plaque and stimulated saliva specimens were collected from 87 children. The pH of each saliva sample was measured before and after 0.94% lactic acid was added. Specimens were diluted and plated on selective media and non-selective media. Data collected were counts of mutans streptococci (MS) and lactobacilli (LB). In addition, total viable counts (TVC) of specimens, salivary pH and buffering capacity were also assessed. Each variable was compared to caries status of subjects. According to this study, the results were as followed: 1. Highly significant correlation with caries rates were found for counts of MS and LB. 2. The specific counts/ml saliva or plaque above which caries is predicted, or below which caries is not predicted were as follows: 1) Saliva MS; $10^5$ 2) Plaque MS; $2{\times}10^5$ 3) Saliva LB; $10^3$ 4) Plaque LB; $10^3$. 3. Salivary pH and buffering capacity versus caries status were not significant. 4. Microbial screening methods based on mutans streptococci had higher predictive values and odds ratios than methods for lactobacilli. 5. MS counts were clearly the best indicators of caries status in young children. This measurement can easily be obtained in a dental clinical setting both by conventional culture techniques, or commercial kits for MS recovery.
To develop multifunctional microbial inoculant, microorganisms with antagonistic activity and biofertilizing activity were screened. Pantoea agglomerans and Bacillus megaterium from our laboratory culture collection, and strain MF12 from soil near poultry farm in Miryang were selected. On the basis of morphological, physiological studies and 16S rDNA sequence analysis, isolate MF12 was identified as the Bacillus pumilis. Three strains were studied for insoluble phosphate solubilization, indole-3-acetic acid (IAA) and siderophore production, ammonification ability, hydrolytic enzyme production and antifungal activity against phytopathogenic fungi. P. agglomerans did not produce any visible clear zone on agar plate containing 0.5% $Ca_3(PO_4){_2}$ as a sole phosphorus source. However, this strain could solubilize insoluble phosphate in liquid medium. All strains produced IAA ranged from $3{\sim}639{\mu}g/ml$ depending on culture time and had ammonification ability. Among three strains, only P. agglomerans produced siderophore. P. agglomerans produced pectinase and lipase, B. megaterium produced amylase, protease and lipase while B. pumilis produced protease and lipase. P. agglomerans showed antifungal activities against phytopathogenic fungi, Fusarium oxysporum and Colletotrichum gloeosporioides. B. pumilis showed antifungal activities against Botrytis cinerea, Sclerotinia sclerotiorum and Phythium ultimum.

The purpose of this study was to determine the level of toothbrush bacterial growth, whether the dryness of the toothbrush head differs depending on the number of holes in the head, and to use these results as a reference for future toothbrush design. Two-millimeter holes were created on the head of the toothbrushes in groups of three, one, or zero holes. We made the solution with Streptococcus mutans, and the toothbrushes were placed in the solution and agitated. The toothbrushes were shaken to remove moisture and allowed to air-dry. The toothbrush heads were swabbed with saline and then placed in two inoculation groups. The first group was inoculated with a 102 dilution of the S. mutans culture and the second was inoculated with the original culture. After incubation, bacterial colony numbers were measured. The number of holes on the toothbrush head correlated with a decrease in number of cultured bacterial colonies. Our model of a toothbrush head with holes indicated that these holes in the toothbrush head were effective in reducing the level of microbial contamination and that a greater number of holes creates an improved toothbrush sanitation effect. The average number of colonies on the head of toothbrush by number of holes was high, followed by the number of holes 0, 1 and 3, and the average number of colony among toothbrush heads was same. The use of a toothbrush with holes between the toothbrush head indicates that it is effective in reducing the level of microbial contamination between the toothbrush head and toothbrush and the higher the number of holes, the better the effect.
Plant disease caused by root-knot nematode is a major problem in crop production. Using of chemical pesticides, one of the most efficient methods to control nematodes, have raised issues in toxicity to humans and animals and environmental pollution. In this study, to select actinomycete strains that have potential to serve as a microbial agent for control of nematodes, we investigated nematicidal activity of culture broth from 670 Streptomyces isolates. A culture filtrate of KRA15-528 isolate that was identified as S. flavogriseus on the basis of 16S rRNA sequence analysis, showed strong nematicidal activity against second stage of juveniles of Meloidogyne incognita and inhibited egg hatching; exposure to 10% of culture filtrate resulted in 71% juvenile mortality at 48 hours afters treatment and suppressed egg hatching by 54% at 9 days after treatment. When the KRA15-528 culture filtrate was partitioned with ethyl acetate and butanol, ethyl acetate layer exclusively showed strong activity; 91%, 53%, 30% of mortality at 1,000, 500, $250{\mu}g/ml$, respectively. Additionally, the culture filtrate suppressed gall formation on cucumber plant by M. incognita with no phytotoxicity. These results suggest that S. flavogriseus KRA15-528 has potential to serve as a microbial nematicide for the control of root-knot nematode disease.
