마그네슘함량이 높은 니켈 laterite광으로부터 습식공정에 의한 니켈과 코발트 회수 (Recovery of Nickel and Cobalt by a Hydrometallurgical Process from Nickel Laterite Ore with High Magnesium Content)

  • Leaching and solvent extraction experiments have been performed to develop a hydrometallurgical process for the recovery of nickel and cobalt from nickel laterite ore with high magnesium content. Most of the nickel and magnesium in the laterite ore dissolved at leaching conditions of $80^{\circ}C$ and 100 g/L sulfuric acid concentration. while half of the cobalt and iron were leached at the same conditions. Solvent extraction experiments were carried out with D2EHPA and saponifed D2EHPA from a synthetic solution containing Ni, Co, and Mg. The extraction percentage of Co, Mg. and Ni by D2EHPA was very low in a pH range of 4.4 to 7.3. while the extraction percentage sharply increased by using saponified D2EHPA. The stripping percentage of the metals from the saponified D2EHPA increased with sulfuric acid concentration and reached 99.9% at 1 M $H_2SO_4$ solution.

왕까마중(Solanum nigrum L.) 열매 분말의 추출 용매에 따른 항산화 및 항균 활성 (The Antioxidant and Antimicrobial Activity of Solanum nigrum L. Fruit Powder by Extraction Solvent)

  • This study was conducted in order to investigate the antioxidant and antimicrobial activity of Solanum nigrum L. fruit powder after undergoing different extraction solvent processes. The total phenolic content of Solanum nigrum L. fruit powder measured a 14.66 GAE mg/g after undergoing ethanol extraction, and the total flavonoid content measured at 201.23 mg CE/g when undergoing ethanol extraction. The ABTS radical scavenging activity was 160.38~209.53 TEAC umol/g, and the DPPH radical scavenging activity was 53.99~90.76 TEAC umol/g, which indicated a higher level of antioxidant power in the ethanol extract as opposed to in the water extract. The FRAP (ferric reducing antioxidant power) of Solanum nigrum L. fruit powder was 115.58~194.58 TEAC umol/g, and B. subtilis KCTC 2189 showed greater antimicrobial activity in the ethanol extract (concentration 200 ug/uL) as opposed to the water extract. Solanum nigrum L. fruit powder revealed differences in antioxidant and antimicrobial activity between the different extraction solvents. In particular, ethanol extract had higher antioxidant and antibacterial activity, meaning it is more favorable for usage as a functional food material.

부산 신항만 준설퇴적물로부터 중금속의 연속추출에 관한 연구 (A Study on Sequential Extraction of Heavy Metals from Marine Dredged Sediment at Busan New Port)

  • 본 연구에서는 부산신항만 준설퇴적물에 포함된 중금속의 총농도, 이동성, 생체이용률, 독성을 알아보기 위하여 총함량분석과 연속추출실험을 실시하였다. 준설퇴적물의 중금속 총농도는 다음과 같이 비교적 낮았다: Al (2.36~2.96 wt.%), As (1.6~3.3 mg/kg), Ba (30.0~33.8 mg/kg), Cd (0.12~0.18 mg/kg), Cr (27.5~35.0 mg/kg), Cu (11.3~15.0 mg/kg), Fe (2.91~3.51 wt.%), Mn (324~408 mg/kg), Ni (18.8~23.8 mg/kg), Pb (23.8~31.3 mg/kg), Zn (70.0~86.3 mg/kg). 또한 대부분의 중금속이 residual fraction으로 존재하였는데, 이는 오염원이 자연발생적일 가능성이 높음을 의미한다: Al (87.5~95.9%), As (74.1~93.8%), Ba (71.8~77.6%), Cr (69.5~94.3%), Cu (50.0~78.7%), Fe (70.8~87.6%), Ni (64.5~75.3%), Pb (53.4~64.3%), Zn (62.5~81.7%). 반면에 Cd과 Mn은 주로 non-residual fraction으로 존재했다. 준설퇴적물의 유해중금속 농도가 낮고 residual fraction 비율이 높기 때문에, 이를 벽돌, 도로포장재료 등으로 재활용하는 것이 가능하다고 사료된다.

Optimization of Ultrasound-assisted Extraction of Phenolic Compounds from Salicornia herbacea Powder

  • Salicornia herbacea is rich in natural minerals, dietary fibers, and potentially health-promoting phenolic compounds. In this paper, an experimental design was applied for the optimization of the ultrasound-assisted extraction of phenolic compounds from lyophilized Salicornia herbacea powder. The experiments were conducted in accordance with a five-level, three-variable central composite rotatable design (CCRD), and the effects of solvent concentration, extraction time, and extraction temperature were evaluated via response surface methodology (RSM). The optimal extraction conditions were as follows: ethanol concentration, 76.80%; extraction time, 20 min; and extraction temperature, $33.21^{\circ}C$; and the solvent concentration was the most significant parameter in this process, under which the predicted total phenolic content was 49.91 mg GAE/g sample.

Nannochloropsis oceanica로부터 용매추출법을 이용한 미세조류 오일 회수 (Microalgal Oil Recovery by Solvent Extraction from Nannochloropsis oceanica)

  • 본 연구에서는 Nannochloropsis oceanica로부터 바이오디젤 원료유로 활용하기 위한 미세조류 오일을 추출하였다. 용매추출법을 이용하여 오일을 추출하였으며, 미세조류의 건식 및 습식 조건에서 오일 추출률(yield) 및 오일 추출 효율(efficiency)을 비교하였다. N. oceanica는 지방산 함량이 317.8 mg/g cell으로 건조중량 대비 30% 이상의 높은 오일함량을 나타내었으며, 미세조류의 건식 조건에서 습식 조건보다 높은 오일 추출률을 나타내었다. 사용된 용매에 대해서 헥산 < 헥산-메탄올 < 클로로포름-메탄올 순으로 오일 추출률이 증가하였다. 그러나 추출된 오일의 지방산 함량을 분석한 결과, 오일 추출률이 증가할수록 지방산 함량은 감소하여 엽록소와 같은 불순물을 포함하고 있는 것으로 나타났다. 따라서 오일 추출률과 지방산 함량을 고려한 오일 추출 효율은 건식 조건에서 헥산-메탄올 이용 추출에서 82.6%의 가장 높은 효율을 나타내었고, 습식 조건에서는 클로로포름-메탄올 이용 추출에서 88.0%로 가장 높은 효율을 나타내었다. 따라서 경제적으로 미세조류의 건조가 가능한 경우에는 헥산-메탄올을 사용하고, 건조 비용이 높은 경우에는 습식 조건에서 클로로포름-메탄올을 사용한 용매추출법이 바람직하다.

추출조건에 따른 차전초 추출물이 3T3-L1 세포의 지방 축적에 미치는 영향 (The effect of Plantaginis asiaticae Folium on lipid accumulation in 3T3-L1 adipocytes by extraction conditions)

  • Objectives : Plantaginis asiaticae Folium (PA) has been widely used in Korean medicine for treatment of liver disease, stomach troubles and inflammation. We investigated the effect of PA on lipid accumulation in 3T3-L1 according to extraction conditions. Methods : The MTT assay was employed to evaluate the cytotoxicity of PA extracted by different solvents (water, 50% ethanol, and 95% ethanol) on 3T3-L1 preadipocytes. Oil red O staining was used to identify intracellular lipid accumulation in 3T3-L1. 3T3-L1 adipocytes were treated with PA at concentration ranging in 0.1, 0.2 and $0.4mg/m{\ell}$. PA was extracted by different extraction conditions such as extraction solvents, extraction time, and extraction temperature. In addition, UPLC analysis was used for determination of candidates of active ingredients in PA. Results : 3T3-L1 preadipocytes were treated with PA extracted by different solvents (water, 50% ethanol, and 95% ethanol) and there was no cytotoxicity. Oil red O staining was employed to identify the effect of PA on lipid accumulation in 3T3-L1. In the present study, PA water extraction at $70^{\circ}C$ for 6 hours decreased greatly in lipid accumulation. The range of concentrations was 0.1, 0.2 and $0.4mg/m{\ell}$. Concentration at $0.2mg/m{\ell}$ was the most effective one among them. Candidates of active ingredients in PA were shown plantamajoside and acteoside through UPLC. Conclusions : These results suggest that the effect of PA water extraction at $70^{\circ}C$ on lipid accumulation in 3T3-L1 is superior to other extraction conditions. We suppose that plantamajoside and acteoside may be candidates of active ingredients in PA.

Vibrio parahemolyticus에 대한 한방처방 "방약합편(方藥合編)${\rfloor}$ 및 그 단미제의 항균활성에 관한 연구 (Antimicrobial Activity of Korea Traditional Prescription(Bangyak-Happyon) and Herb Simplexes Extracts to Vibrio parahemolyticus)

  • Vibrio are become prevailing if superficial temperature of ocean is raised and their activities of area are expanded and most of ocean creatures(fishes, oysters etc) are polluted with vibrio. The one who has taken these poluted fishes and aysters uncooked caused foodpoisoning and diarrhea from Vibrio. Frequencies of these diseases breakout is disposed in westsea shore of Korea. According to ancient and traditional Korean medical book -${\ulcorner}$Bangyak Happyeon${\lrcorner}$(Collection of Local Medicines, 1884) - and their single prescribes, we carried out experiment check the activities of natural medicinal effects on Vibrio parahemolyticus. The prescriptions of trial materials are processed from extraction boiling water and 80% methanol and followed freeze dried and adsorbed to every discs in dosage of 10mg. Gentamycin of 10mg were used for control. The result of compound prescription displayed special diseases in antimicrobial activities of boiling water and MeOH extraction compared with control. In compound prescription, extraction MeOH of Sashinhwan(clear zone : 17mm) presented extraordinaire antimicrobial activity. In single prescription, extraction of boiling water(clear zone : 16mm) and MeOH(clear zone : 18mm) of Fructus Chebulae presented extraordinaire antimicrobial activity. The MBC of Fructus Chebulae extracts was expressed in boiling water(1.28mg/ml) and MeOH(0.64mg/ml).

Optimal Hot Water Extraction Conditions of Mixed Herbs Extract Mixture Using Response Surface Methodology

  • Human needs energy to maintain metabolisms, and these energy sources were uptake foods or nutritions. The most effective source was known for glucose among the nutrients, furthermore the glucose is an important source of energy for blood cells and control brain maintenences cells. But as food is plentiful and eating habits become more westernized, fast food and irregular meal times by works. Nowadays, diabetes were rapidly increased by malnutriton and obesity. Diabetes was the sixth highest on the list of causes of death in Korea, released by the Statistics Korea in 2015, which is considered a serious social problem for adult diseases. Therefore, this study aims to establish the optimal hot water extraction conditions of mixed herbs extract mixture compounds that are effective in diabetes. The independent factors were extraction temperature (X1: $40-120^{\circ}C$), extraction time (X2: 2-10 hrs.), and the ratio of water to sample (X3: 40-200 mg/mL). Their effects were assessed on dependent variables of the extract properties, which included soluble solid contents, Brix of sample extract, total polyphenols content, total flavonoids content and DPPH Radical scavenging activity. As a result, the content of total polyphenol content was the highest in No.12(6 hrs, $120^{\circ}C$, 67 mg/mL) and the highest total flavonoid contents was found in No.16(6 hrs, $80^{\circ}C$, 40 mg/mL). DPPH Radical scavenging activity showed the highest activity in No.7(8 hrs, $100^{\circ}C$, 100 mg/mL).

중금속에 의해 오염된 토양에 대한 토양세척기법의 적용성 연구 (Application of Soil Washing Technology to the Soil Contaminated by Heavy Metals)

  • 중금속에 의해 오염된 토양에 대해 토양세척기법을 적용하기 위한 최적의 조건을 도출하고자 회분식 및 실험실 규모의 파이롯트 실험을 수행하였다. 대상 중금속으로는 구리, 납, 아연을 선정하였고 각 중금속의 농도는 구리 500mg/kg dry soil, 납 1,000mg/kg dry soil, 아연 500mg/kg dry soil로 결정하였다. 세정제로는 유기산 중 citric acid와 oxalic acid를 선정하였다. 회분식 실험 결과, 구리, 납, 아연의 세정효율은 각각 79%, 72%, 72%로 나타났으며, 구리와 납에 대한 세정제의 농도와 진탕비는 citric acid 50mM과 1:5(중량/체적비)가 최적조건으로 판명되었고, 계면활성제 OA-5 및 SDS l∼2%를 첨가함으로써 세정효율을 향상시킬 수 있었다. 파이롯트 규모실험에서는 납으로 오염된 토양에 대하여 1차 토양세척시 59%의 세정효율을 얻었으며, 2차 토양세척으로 78%까지 세척할 수 있었다.

추출조건에 따른 노랑느타리버섯 추출물의 항산화 활성 (Antioxidant Activities of Pleurotus cornucopiae Extracts by Extraction Conditions)

  • 노랑느타리버섯을 물, 30%, 60% 및 90% 에탄올로 추출용매를 달리하여 추출한 추출물을 각각 농도를 달리하여 항산화 및 생리활성을 측정하였다. 그 결과 DPPH radical 소거능은 각 추출물 농도가 1 mg/mL일 때 모든 추출조건에서 24.34~31.15%의 다소 낮은 활성을 보였다. 총 폴리페놀 함량은 에탄올 추출물에 비해 물 추출물이 더 많은 페놀을 함유하고 있었고 이때 물 추출물이 26.34 mg/g으로 가장 많았다(P<0.05). SOD 유사활성 측정 결과 추출조건에 따른 경향은 보이지 않았으나, 표준물질인 ascorbic acid와 비교하였을 때 노랑느타리버섯의 활성이 2배 이상 높은 것으로 나타났다. ACE 저해 활성의 경우 에탄올 추출물의 활성이 높은 경향을 보였으나 모든 추출물이 35.99~57.42%로 비교적 낮은 활성을 보였다. 노랑느타리버섯의 아질산염 소거능 측정에서는 pH 3.0에서 높게 나타났고, 추출조건에 따라 60% 에탄올 추출물의 소거능이 가장 우수하였다. 이와 같이 노랑느타리의 다양한 항산화 및 생리활성을 기초자료로 한 천연물 유래의 기능성 소재 발굴 및 연구 활용이 가능할 것으로 사료된다.
