The anaerobic digester with sludge from sewage treatment plant was operated in the laboratory for two year to investigate the enumeration and activity of methanogenic microorganisms. In this experimental study, the effects of HRT on the degradation characteristics of organic materials and on the number of methanogenic bacteria produced were investigated. By making the media with the repeated wxperiment, the number and activity of methanogenic bacteria were measured. The increase of the removal rate of organic acid in the digester was oberved at HRT of 2 days. The total number of methane forming bacteria estimated by the MPN method showed 2.3 $\times$ $ at HRT of 3 days, 7$\times$$ of 5 days and 7.9$\times$$ $/ml of 10 days. The optimum incubation time for measuring the number of methanogenic bacteria was found as more than four weeks. The PMA revealed 161ml CH$/l day at HRT of 10 days and the PUA 290mg COD/l day. At the incubation time 4.3 days, the maximum value of CH$ *59.1%) was found. At this time, $ was found as 15.3% and $ 25.6%.
Effects of carbon dioxide partial pressure(PCO2) on bacterial population, methane production rate and matter degradation in anaerobic digestion were investigated by using anaerobic chemostat type reactors at 35$\pm$1$^{\circ}C$, at the HRT of 7 days. At PCO2 of 0.5 atm, the specific methane production rate and specific substrate removal rate reached the maximum rates. The methane production rates in the reactors fed by mixed substrate were 26% higher than those obtained under the controlled condition. The number of acetate consuming methanogenic bacteria enumerated by the MPN(most probable number) method, decreased when PCO2 exceeded 0.7 atm. Hydrogen consuming methanogenic bacteria and homoacetogenic bacteria increased as PCO2 increased from 0.1 to 0.6 atm, however, decreased slightly at PCO2 above 0.7 atm. The number of hydrolytic bacteria, sulfate-reducing bacteria and H2-producing acetogenic bacterial were not much influenced by the change of PCO2. The potential methanogenic activity reached the maximum at PCO2 0.5 atm, however, decreased significantly when PCO2 exceeded 0.7 atm, would depend on free PCO2 concentration in solution.

In order to investigate the effect of the methanogenic bacteria in bacteria in leachate on the degradability of landfill waste, this study has created 4 cylinder-shape PVC lysimeters (Diameter: 30cm, Height: 200cm, Volume: 140L) and for the biological treatment and recirculation of the leachate, two anaerobic batch reactors (Diameter: 20cm, Height: 30cm) were created. To simulate a conventional landfill, no recycling was done in L1. In L2, 1,068ml of leachate (twice of rainfall amount) was recycled. In L3 and L4, the leachate was anaerobically digested in a dark room (with $35{\pm}1^{\circ}C$) for a week and them recycled by 1,064ml and 2,128ml, respectively, with recycled water only. In terms of cumulative $CH_4$ production, however, L3 and L4 were much higher (three times) than L1 and L2. Between L3 and L4, the latter was 1.23 times higher than the former in terms of cumulative CH4 production. In other words, the more the methanogenic bacteria-activated leachate is recycled, the more active the degradation due to active methane fermentation by the recyled methanogenic bacteria. And methane recovery is different according to the amount of recycled the methanogenic bacteria in leachate.

In this paper, the microscopic results of granular sludge from the two-phase UASB reactors are presented, particularly on the acidogenic phase where the distillery waste water is directly delivered. Distillery wastewater as a feed contained SS of 3.6~10.6 g/l and COD of 17.3~30.4 g/l. A 12.5 l and a 4.7 l UASB reactors were used for acidogenic and methanogenic phases, repectively. Morphological studies on the granular sludge consortia were made with both scanning and transmission electron microscopies. It was interesting enough to find the different colored and shaped granules at the acidogenic on the 90th day and at the methanogenic phase on the 120th day. The acidogenic granular sludge mainly consisted of acidogenic bacteria, such as long chains of large rods, short plump rods, and various size of cocci. Whereas the filamentous bacteria of Methanothrix spp. prevail in the methanogenic granule, with dirverse bacteria entrapped therein.
Microstructural examinations were performed on the anaerobic biofilm from reactor filled with PE support media. Optical microscope, SEM and fluorescent microscope were used for qualitative and morphological studies on the attached microorganism under anaerobic condition. Microorganisms were attached in crevices where protection from shear forces of surfaces where easy to contact with support media surface. A hypothesis for biofilm accumulation occurs on a surface such as polymer support media is presented schematically : 1st step ; cell-support media attachment, 2nd step ; cell-support media attachment and cell-cell attachment, 3rd step ; attached biofilm from neighboring crevices joins together and growing, 4th step ; mature and irregualar biofilm was formed. In SEM photographs, shape and structures of biofilm were observed, but microorganism species and methanogens were not identified. During the operation, maintenance and decommissioning of nuclear power plant radioactive contaminated waste is produced. This waste is stored in an underground repository 60-100 meters below the surface. The metallic portion of this waste comprises mostly carbon and stainless steel. A long-term field exposure showed high corrosion rates, general corrosion up to 29 ㎛ a-1 and localized corrosion even higher. High corrosion rate is possible if microbes produce corrosive products, or alter the local microenvironment to favor corrosion. The bacterial and archaeal composition of biofilm formed on the surface of carbon steel was studied using 16S rRNA gene targeting sequencing, followed by phylogenetic analyses of the microbial community. The functional potential of the microbial communities in biofilm was studied by functional gene targeting quantitative PCR. The corrosion rate was calculated from weight loss measurements and the deposits on the surfaces were analyzed with SEM/EDS and XRD. Our results demonstrate that microbial diversity on the surface of carbon steel and their functionality is vast. Our results suggest that in these nutrient poor conditions the role of methanogenic archaea in corrosive biofilm, in addition to sulphate reducing bacteria, could be greater than previously suspected.

The substrate metabolism and bacterial population in an anaerobic digestion with the submerged separation membrane were investigated by using a laboratory-scale reactor at the hydraulic retention time(HRT) 1.0 and 0.5 day. The removal efficiencies of carbohydrate at the HRT 1.0 and 0.5 day were 99.8~99.9% and 98.0~99.6%, respectively. After the 58 days, the mixed liquor volatile suspended solids(MLVSS) concentration at the HRT 1.0 and 0.5 day were approximately 6,050 and 7,750 mg/L, respectively. According to the measurement by the most probable number(MPN) method, the numbers of acidogenic bacteria, $H_2$-utilizing and acetate-utilizing methc.nogenic bacteria were found to be $10^9$, $10^7{\sim}10^8$ and $10^6{\sim}10^8MPN/mL$, respectively. The composition of $CH_4$ in the produced gas was 46~50%. It is suggested that sulfate-reducing bacteria $10^7{\sim}10^8MPN/mL$ play an important role in producing $H_2$ and acetate in sulfate-depleted environment.

Two UASB reactors were operated to investigate the effect of high concentration of sulfate on anaerobic digestion of propionate using an upflow anaerobic sludge blanket (UASB) reactor. An organic loading rate of $1.2kg\;COD/m^3{\cdot}d$ and a hydraulic retention time of 1.6 d were maintained during this study. In the absence of sulfate, the UASB reactor achieved about 95% removal of chemical oxygen demand whereas in the presence of $2,000\;SO_4^{2-}mg/L$, the COD removal rate decreased to 83% due probably to the inhibition of dissolved sulfide inhibition. Interactions between the methane producing bacteria (MPB) and sulfate reducing bacteria (SRB) were measured to investigate the competition between MPB and SRB. The MPB consumed average 58% of the available electron donors at $COD/SO_4^{2-}$ ratio of 1. Propionate was consumed mainly by SRB, converting sulfate into sulfide and suppressing the methane production. The specific methanogenic activity (SMA) using acetate and propionate increased as microorganism acclimated to the substrate.
