Prediction of Energy Requirements for Maintenance and Growth of Female Korean Black Goats (번식용 교잡 흑염소의 유지와 성장을 위한 대사에너지 요구량 추정)

  • This study was conducted to predict the energy requirements for maintenance and growth of female Korean black goats during their growth and pregnancy phases. Fifty female goats ($18.7{\pm}0.27kg$) in their growth phase with an average age of 5 months were stratified by weight and randomly assigned into 5 groups. They were fed 5 diets varying in metabolic energy (ME) [2.32 (G1), 2.49 (G2), 2.74 (G3), 2.99 (G4), and 3.24 (G5) Mcal/kg] until they were 9-month-old. After natural breeding, 50 female goats ($30.7{\pm}0.59kg$) were stratified by weight and randomly assigned into 5 groups. They were fed 5 diets varying in ME [2.32 (P1), 2.43 (P2), 2.55 (P3), 2.66 (P4), and 2.78 (P5) Mcal/kg]. The average feed intake ranged between 1.5 and 2.0% of the body weight (BW), and there was no significant difference between the treatment groups with goats in growth or pregnancy phases. Average daily gain (ADG) in diet demand during the growth phase increased with an increasing ME density and ranged from 46 to 69 g/d (p<0.01). Feed conversion ratio (FCR) improved with the ME density during the growth phase (p<0.01). The intercept of the regression equation between ME intake and ADG indicated that energy requirement for maintenance of goats during growth and pregnancy phases was $103.53kcal/BW^{0.75}$ and $102.7kcal/BW^{0.75}$, respectively. These results may serve as a basis for the establishment of goat feeding standards in Korea. Further studies are required to assess the nutrient requirement of goats using various methods for improving accuracy.

Inhibitory Effects of Locusta migratoria Ethanol Extracts on RANKL-induced Osteoclast Differentiation (RANKL 유도된 파골세포 분화에 대한 풀무치 에탄올 추출물의 분화 억제 효과)

  • Recently, there has been an increase in the elderly population of the world. Consequently, bone metabolic diseases such as osteoporosis are emerging as a social problem. Osteoclasts play a role in bone resorption, and osteoporosis is induced when bone resorption occurs excessively. Because currently used bone resorption inhibitors may cause side effects when used for a long period of time, it is necessary to develop a new material that effectively inhibits osteoclast differentiation. This study aimed to confirm the inhibitory effect of ethanol extract of Locusta migratoria on RANKL-induced osteoclast differentiation and its mechanism. The toxicity and proliferation effects of LME on RAW264.7 osteoclasts were measured by an MTS assay. There was no cytotoxicity or proliferation when the osteoclasts were treated with up to $2,000{\mu}g/ml$ of LME. In order to confirm the effect of LME on the differentiation of osteoclasts, osteoclasts were treated with RANKL alone or with LME for 3 days. As a result of a TRAP (tartrate-resistant acid phosphatase) assay, the increasing osteoclast differentiation by RANKL decreased in a concentration-dependent manner with the treatment of LME. In addition, LME suppressed the expression of differentiation-related marker genes (TRAP, RANK, NFATc1, and CK) and proteins (NFATc1 and c-Src) that had been increased by RANKL. Also, LME influenced the $NF-{\kappa}B$, ERK and JNK signaling pathways, resulting in the inhibition of osteoclast differentiation. These results suggest that LME may be used as a novel functional material for the prevention and treatment of osteoporosis by playing a role in inhibiting bone absorption.

Tenebrio molitor (Mealworm) Extract Improves Insulin Sensitivity and Alleviates Hyperglycemia in C57BL/Ksj-db/db Mice (C57BL/Ksj-db/db 제 2형 당뇨모델을 이용한 갈색거저리 유충(밀웜) 추출물의 인슐린 감수성 및 혈당개선효과)

  • Diabetes is one of the serious chronic metabolic diseases caused by Westernized eating habits, and the goal of diabetes treatment is to keep blood glucose at a normal level and prevent diabetic complications. This study was designed to investigate the anti-diabetic effects of a mealworm (Tenebrio molitor larva) extract (MWE) on hyperglycemia in an animal model with type 2 diabetes. Diabetic C57BL/Ksj-db/db mice were divided into three groups: diabetic control, rosiglitazone, and MWE. The mice supplemented with MWE showed significantly lower blood levels of glucose and glycosylated hemoglobin when compared with the diabetic control mice. The homeostatic index of insulin resistance was significantly lower in mice supplemented with MWE than in diabetic control mice. MWE supplementation significantly stimulated the phosphorylation of insulin receptor substrate-1 and Akt, and activation of phosphatidylinositol 3-kinase in insulin signaling pathway of skeletal muscles. Eventually, MWE increased the expression of the plasma membrane glucose transporter 4 (GLUT4) via PI3K/Akt activation. These findings demonstrate that the increase in plasma membrane GLUT4 expression by MWE promoted the uptake of blood glucose into cells and relieved hyperglycemia in skeletal muscles of diabetic C57BL/Ksj-db/db mice. Therefore, mealworms are expected to prove useful for the prevention and treatment of diabetes, and further studies are needed to improve type 2 diabetes in the future.

Hepatoprotective Effects of Semisulcospira libertine Hydrolysate on Alcohol-induced Fatty Liver in Mice (알코올성 지방간 유발 마우스에서 다슬기 유래 가수분해물의 간 보호 효과)

  • Alcoholic steatosis is a fundamental metabolic disorder and may precede the onset of more severe forms of alcoholic liver disease. In this study, we isolated enzymatichydrolysate from Semisulcospira libertine by alcalase hydrolysis and investigated the protective effect of Semisulcospira libertine hydrolysate on liver injury induced by alcohol in the mouse model of chronic and binge ethanol feeding (NIAAA). In an in vitro study, the hydrolysate protects HepG2 cells from ethanol toxicity. Liver damage was assessed by histopathological examination, as well as by quantitating activities of serum aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP). After the administration of S. libertina hydrolysate, fat accumulation and infiltration of inflammatory cells in liver tissues were significantly decreased in the NIAAA mouse model. The elevated levels of serum AST, ALT, and ALP activities, along with the lipid contents of a damaged liver, were recovered in experimental mice administrated with S. libertina hydrolysate, suggesting its role in blood enzyme activation and lipid content restoration within damaged liver tissues. Moreover, treatment with S. libertine hydrolysate reduced the expression rate of cyclooxygenase (COX-2), interleukin $(IL)-1{\beta}$, and IL-6, which accelerate inflammation and induces tissue damage. All data showed that S. libertine hydrolysate has a preventive role against alcohol-induced liver damages by improving the activities of blood enzymes and modulating the expression of inflammation factor, suggesting S. libertine hydrolysate could be a commercially potential material for the restoration of hepatotoxicity.

Comparison of Housewives' Agricultural Food Consumption Characteristics by Age (주부의 연령대별 농식품 소비 특성 비교)

  • Lifestyle is changing rapidly, and food consumption patterns vary widely among households as dietary and food processing technologies evolve. This paper reclassified the food group of consumer panel data established by the Rural Development Administration, which contains information on purchasing agricultural products by household unit, and compared the consumption characteristics of agricultural products by age group. The criteria for age classification were divided into groups in their 60s and older with a prevalence of 20% or more metabolic diseases and groups in their 30s and 40s with less than 10%. Using the LightGBM algorithm, we classified the differences in food consumption patterns in their 30s and 50s and 60s and found that the precision was 0.85, the reproducibility was 0.71, and F1_score was 0.77. The results of variable importance were confectionery, folio, seasoned vegetables, fruit vegetables, and marine products, followed by the top five values of the SHAP indicator: confectionery, marine products, seasoned vegetables, fruit vegetables, and folio vegetables. As a result of binary classification of consumption patterns as a median instead of the average sensitive to outliers, confectionery showed that those in their 30s and 40s were more than twice as high as those in their 60s. Other variables also showed significant differences between those in their 30s and 40s and those in their 60s and older. According to the study, people in their 30s and 40s consumed more than twice as much confectionery as those in their 60s, while those in their 60s consumed more than twice as much marine products, seasoned vegetables, fruit vegetables, and folioce or logistics as much as those in their 30s and 40s. In addition to the top five items, consumption of 30s and 40s in wheat-processed snacks, breads and noodles was high, which differed from food consumption patterns in their 60s.

  • The purpose of this perspective research is to discuss the potential role of exercise-interventions in COVID-19, terms of prevention and prognosis in the periods of the COVID-19 vaccine. SARCO-CoV-2. COVID-19 was detected as a new virus causing severe cardiovascular and respiratory complications. It emerged as a global public health emergency and national pandemic. It caused more than 1 million deaths in the first 6 months of the pandemic and resulted in huge social and economic fluctuations internationally. Unprecedented stressful situations, such as COVID-19 blue and COVID-19 red impact on many health problems. In healthy individuals, COVID-19 infection may induced no symptoms (i.e., asymptomatic), whereas others may experience flu-like symptoms, such as ARDS, pneumonia, and death. Poor health status, such as obesity and cardiovascular and respiratory complications, are high risk factors for COVID-19 prevention, occurrence, and prognosis. Several COVID-19 vaccines are currently in human trials. However, the efficacy and safety of COVID-19 vaccines, including potential side effects, such as anaphylaxis (a life-threatening allergic reaction) and rare blood clots, still need to be investigated. On the basis of direct and indirect evidence, it seems that regular and moderate physical exercise can be recommended as a nonpharmacological, efficient, and safe way to cope with COVID-19. Physical inactivity and metabolic abnormalities are directly associated with reduced immune responses, including reduced innate, CMI, and AMI responses. Due to prolonged viral shedding, quarantine in inactive, obese and disease people should likely be longer than physical active people. Multicomponent and systemic exercise should be considered for the obese, disease, and elderly people. More mechanism research is needed in this area.

Transcriptomic Analysis of Triticum aestivum under Salt Stress Reveals Change of Gene Expression (RNA sequencing을 이용한 염 스트레스 처리 밀(Triticum aestivum)의 유전자 발현 차이 확인 및 후보 유전자 선발)

  • As a cultivar of Korean wheat, 'Keumgang' wheat variety has a fast growth period and can be grown stably. Hexaploid wheat (Triticum aestivum) has moderately high salt tolerance compared to tetraploid wheat (Triticum turgidum L.). However, the molecular mechanisms related to salt tolerance of hexaploid wheat have not been elucidated yet. In this study, the candidate genes related to salt tolerance were identified by investigating the genes that are differently expressed in Keumgang variety and examining salt tolerant mutation '2020-s1340.'. A total of 85,771,537 reads were obtained after quality filtering using NextSeq 500 Illumina sequencing technology. A total of 23,634,438 reads were aligned with the NCBI Campala Lr22a pseudomolecule v5 reference genome (Triticum aestivum). A total of 282 differentially expressed genes (DEGs) were identified in the two Triticum aestivum materials. These DEGs have functions, including salt tolerance related traits such as 'wall-associated receptor kinase-like 8', 'cytochrome P450', '6-phosphofructokinase 2'. In addition, the identified DEGs were classified into three categories, including biological process, molecular function, cellular component using gene ontology analysis. These DEGs were enriched significantly for terms such as the 'copper ion transport', 'oxidation-reduction process', 'alternative oxidase activity'. These results, which were obtained using RNA-seq analysis, will improve our understanding of salt tolerance of wheat. Moreover, this study will be a useful resource for breeding wheat varieties with improved salt tolerance using molecular breeding technology.

Studies on the Evaluation of the Spent Composts of Selenium-Enriched Mushrooms as a Feed Selenium Source (셀레늄강화 버섯폐배지에 대한 사료 셀레늄공급원으로의 평가 연구)

  • This study was conducted to evaluate the spent composts of selenium-enriched mushrooms as a feed selenium Source. Total selenium (Se) contents and Se profiles in the spent mushroom composts (SMC) were determined. In addtion, we also investigated the metabolism in relation to Se accumulation in the mushroom. Mushrooms used in this study were Flammulina velutipes and Se enriched mushrooms were grown for 60 days by adding 2 mg of inorganic Se (Na2SeO3) per kg of mushroom composts (MC) on as-fed basis and it was compared with mushrooms not to add Se to the MC. Total Se contents for Se-treated mushrooms were significantly increased (P<0.0001) by 20-fold (4.51 ㎍/g of dry) compared to Se-untreated (0.23 ㎍/g of dry). On the contrary, organic Se proportion was significantly lower (P<0.0001) in the Se-treated mushroom (72.3%) than Se-untreated (100%, not analytically detected of inorganic Se). Se distribution upon a length in the Se-treated mushrooms was the highest in the bottom part (6.86 ㎍/g of dry) near to MC, and top and middle parts were significantly lower (3.71 and 3.01 ㎍/g of dry, respectively) than the bottom (P<0.001). In the SMC from Se-treated mushrooms, a high concentration of Se (5.04 ㎍/g of dry) was still remained, but that from Se-untreated mushrooms was significantly low (P<0.0001) as 0.08 ㎍/g of dry. Se-treated SMC showed a high rate of organic Se (65.67%), suggesting that most of inorganic Se in the SMC was converted to organic Se by mushroom mycelia, and Se-untreated SMC showed 100% of organic Se, not being detected of inorganic Se. Prior to mycelia inoculation in the mushroom culture, the sterilization of MC brought approximately 18% of Se loss in the MC. This result is in accordance with facts generally known that Se is weak in the high temperature and it is consequently volatilized under that condition. Apparent and net accumulation rates (%) for Se into mushrooms were 14.81 and 10.14%, respectively and their difference (4.67%) is considered that it is due to the volatilization into the air via metabolic process of mushroom itself. From the result of this study, inorganic Se addition to MC for mushroom improved the Se content in the mushroom and SMC from Se-enriched mushrooms contained a high concentration of Se. Mycelium and fruiting body from mushrooms converted inorganic Se in MC to organic Se, indicating a high proportion of organic Se in the mushroom and SMC. Therefore, Se in Se-enriched mushroom and SMC was recognized as Se sources of food for human as well as feed for livestock.

Effects of Crude Protein and Phytase in the Diet on Growth Performance and Excretion Contents of Nitrogen and Phosphorus in Broiler Chicks (사료 내 단백질 및 Phytase가 육계 초생추의 생산성 및 분변 내 질소, 인 함량에 미치는 영향)

  • This study was conducted to investigate the appropriate levels of crude protein (CP) and phytase in the diet of broiler chicks in order to reduce nitrogen and phosphorus contents in feces while maintaining performance of broilers. Six hundred forty-eight 1-day-old male broilers (41.9±0.91 g) had a total of 3 × 3 complex factor of 3 levels of CP (22%, 21%, 20%) and 3 levels of phytase (1,000, 800, 500 FTU/kg) in the diet. Divided into 9 treatments, 4 replications per treatment, 18 birds per replication, were completely randomly assigned and reared in a metabolic cage for 7 days. Seven-day-old body weight (BW) and body weight gain (BWG) of broilers were significantly lower at CP 20% treatment (P<0.05), and feed conversion ratio (FCR) was significantly lower at CP 21% and phytase 800 FTU/kg treatment (P<0.05). Nitrogen and phosphorus contents in chicken excreta were significantly lower in CP 20% and phytase 500 FTU/kg treatment, respectively (P<0.05). Interactions between CP and phytase in the feed were shown for nitrogen and phosphorus in feces (P<0.05). In conclusion, considering the broiler performance and excretion contents of nitrogen and phosphorus, it is thought that CP and phytase levels of broiler chicks diet can be reduced by 21% and 800 FTU/kg, respectively.

The Influence of Long-term Aerobic Dancing Exercise on the Body Composition, Fitness and Mental Function of the Elderly Women (장기간에 걸친 에어로빅 댄스운동이 고령 여성의 신체조성, 체력 및 심리적인 요인에 미치는 영향)

  • The purpose of this study was to investigate the effects of long-term aerobic dancing exercise on body composition, physical fitness and mental function in older women over 70 years old. All subjects were limited to older women aged 70 to under 85. The experimental group performed aerobic exercise of 2-3 times a week for 9.2 ± 4.6 yrs, and the control group did not perform any regular exercise and spent 3-4 times a week mainly in the relaxing facility for older. As a results in this study, the weight, muscle mass and body fat percentage of the aerobic group were not significantly different from those of the control group, but there were significantly positive effects of aerobic group in visceral fat mass, abdominal obesity rate(%), body water content and systolic blood pressure(p < .05). In terms of physical fitness, Sit & reach, one-legged stand (right) and TUG of the aerobic group were significantly higher than those of the control group(p < .05), but there were no significant differences between two groups in the shoulder flexibilities and grip strength. MMSE as a cognitive function was not significantly different between the two groups, but GDS was significantly lower in the aerobic group than in the control group(p < .05). According to the correlation analysis between all variables and MMSE and GDS, MMSE was significantly inversely correlated with height (r = -0.425)(p < .05), and no correlation was detected with other variables. However, GDS have signitively negative correlations with resting heart rate(r = -0.464), sit & reach(r = -0.525) left(r = -0.491) and right grip strengths(r = -0.635) as well as positive correlation systolic blood pressure(r = 0.482) and TUG (r = 0.479), respectively(p < .05). In conclusion, long-term aerobic exercise of elderly women over 70 older had positive effects on variables related to metabolic disease (visceral fat and resting heart rate), silver fitness variables related to falls, and depression level in the elderly.
